식물 잎을 이용한 중금속 이온의 제거에 관한 연구 (Removal of Heavy Metal Ions from Aqueous Solution by Leaves)

  • In this research, I carried out the adsorption and removal test of Pb, Ni, Co and Cu ions using organic substances spread out any where in the nature which can be obtained easily from our neighbor-such as Paulownia coreana, Pinus densiflora, Juniperus chinesis, Quercus dentata, Magnolia kobus, Platanus occidentalis, Gingko biloba, Diospyros kaki leaves. As the result of the research to find the best optional condition for the adsorption and removal, shows that the adsorption and removal ratio of Pb ion by a Paulownia coreana raw leaves is 99% at $70^{\circ}C$, those of Ni ion and Co ion by Magnolia kobus formalin treatment leaves are 79% at $70^{\circ}C$, 97% at $40^{\circ}C$ respectively. And that of Cu ion by Platanus occidentalis treatment leaves is 97% at $50^{\circ}C$ in mixed solution. As the result of comparing the removal ratio by raw leaves and formalin treatment leaves, the removal ratio of treatment is 30~90% more effective than raw leaves in most cases. And I concluded Pb > Cu > Co > Ni ion in multiple solution and Co > Ni > Cu >Pb ion in single solution after testing adsorption and removal ratio of mixed solution separately as time goes by. In general, the reactions were completed within first 5 minutes. The test result of measuring the hydrolysable tannin content of each leaf shows that an overcup Quercus dentata is 11.36%, a Diospyros kaki is 10.81% and the rest of them are 2.49~4.12% in raw leaves cases. In treatment leaves cases, an overcup Quercus dentata is 3.23% and the others are less than 1%.

내,외인성으로 유도된 Nitric Oxide가 흰쥐의 통각전달에 미치는 효과

  • 흰쥐의 뒷발바닥에 carragennan(cg)을 피하투여하여 염증성 통증을 유발한 뒤, 그 통증이 최고에 달하는 3시간 뒤 Ach이나 SNP를 동일한 항법으로 투여해서 30분 후 cg유발 통증에 대하여 그 효과를 Randall-Selitto paw pressure test로 검색하였고, 체내 Nitric Oxide Synthase(NOS) 억제제인 L-NAME과 L-NOARG를 용량별로 적용하여 Ach의 진통효과를 억제하는 정도와 Methylene Blue 및 Hemoglobin을 투여해서 SUP효과 억제를 검사하였다. 그리고 척수 지주막하강내로 catheter를 삽입하여 위의 NO donor를 주입하고, NO의 중추신경계에서의 통각전도에 미치는 효과를 Tail-Flick test로 살펴보았다. 끝으로 NO가 가진 급,만성 통각관련효과를 희석한 formalin을 써서 검색하였다. 결 과: Ach과 SNP는 흰쥐에게 말초경로투여시 유의성있는 진통효과를 보였으며, NOS inhibitor와 NO scavenger로 그 효과가 역전되었다. 또한 NOS inhibitor간에는 억제효과가 용량의존성이 유사하게 나타났고, 중추신경계로의 직접투여때는 여러 생리적 조건이 직, 간접으로 관여함이 확인되었다.

Investigations of Pet Ether Extract of Caesalpinia Pulcherrima (L.) Swartz Leaves Extract on Analgesic, Anti-inflammatory, and Antipyretic Properties

  • The pet ether extract of Caesalpinia pulcherrima, leaves was studied for its antinociceptive, anti-inflammatory and antipyretic property. The extract at doses of 50 and 200 mg/kg, p.o., significantly (p<0.05) reduced the number of writhing induced by acetic acid and inhibited the late phase (20-30 min) in formalin test in mice. The extract failed to increase the pain threshold level in tail immersion test in mice. In carrageenan induced paw edema in rats and in acetic acid induced increase in vascular permeability test in mice, the extract (50-600 mg/kg, p.o.) failed to produce any significant activity. While in cotton pellet granuloma test, the extract at doses of 200 and 600 mg/kg (p.o.) significantly (p<0.05) reduced the granuloma formation and was comparable to reference drug, dexamethasone. In ethylpheylpropiolate ear edema test 0.5 mg and 1 mg/ear application of extract significantly (p<0.05) inhibited ear edema. In yeast induced hyperthermia in rats, the extract did not produce any reduction in temperature. The results suggest that the extract acts peripherally to produce analgesic action and anti-inflammatory activity through steroidal mechanism.

Anti-nociceptive Properties of Ribes fasciculatum

  • Ribes fasciculatum (Saxifragaceae) has been widely used as a traditional medicine for the treatment of cough, antidote, cold, lacquer poison, and sore throat. In the present study, we evaluated the anti-nociceptive effects of ethyl acetate fraction of Ribes fasciculatum (ERF) in mice. Test results of tail-immersion test and hot plate test revealed that the ERF had strong anti-nociceptive activities on thermal nociception in a dose dependent manner, indicating ERF's anti-nociception on the central pain. Moreover, the acetic acid-induced chemical nociception was also significantly reduced by ERF treatment. This result shows that ERF may also work on the peripheral pain. We further performed formalin test to confirm ERF's anti-nociceptive properties and found that pain responses were significantly decreased by ERF treatment. Interestingly, in the combination test with naloxone, the analgesic activity of ERF was not changed, indicating that the opioid receptor was not involved in the ERF-mediated anti-nociception. These results indicate that ERF might be possibly used as a painkiller for the treatment of nociceptive pains.

진행성 치주염이 지수 조직에 미치는 영향 (The influence of Advanced Adult Periodontitis on the pulp)

  • The purpose of this study was to observe histopathologically the influence of advanced periodontitis on pulp tissue, and to conclude the correlation between the results with clinical manifestations. The samples were teeth with over 7mm pocket depth and over 50% radiographic bone loss. These were diagnosed to have very poor prognosis and thus planned to be extracted. Those with any of following conditions were excluded from the samples, loss of vitality, periapical pathology, restoration or prosthesis, dental caries, and attrition or abrasion. It was because these conditions could affect pulp without any correlation with periodontal disease. For the experiment, 17 teeth from 11 patients were selected. Average age of patient was 47. Each tooth was examined for following categoris; pocket depth, gingival recession, electric pulp test, mobility, percussion test, sensitivity test. The extracted teeth were fixed buffered neutral formalin solution. It was decalcified using 4% nitric acid. Sliced histological samples observed using light microscope, for pulp status, and severeity of inflammation. 4 samples were excluded due to histologic sample discrepency. Thus 13 samples were subject to observation. 4 showed normal conditions. Focal reversable pulpitis was shown in 5 samples. Chronic pulpitis was observed 1 sample. Pulpal abscess was observed in 3 samples.

팥배나무의 진통 효과 (Anti-nociceptive Effects of Sorbus alnifolia)

  • In this study, we evaluated the anti-nociceptive activities of Sorbus alnifolia. To investigate the anti-nociceptive properties of the methanolic extract of Sorbus alnifolia (MSA), we conducted several tests using various experimental mouse pain models. Herein, MSA significantly delayed the latency time and writhing motion in the hotplate test and acetic acid test, respectively. These result indicated that MSA has an ability to manage both peripheral and central nociception. We could further confirm the analgesic effects of MSA by performing formalin test. In combination test using naloxone, a non-selective opioid receptor antagonist, analgesic activity of MSA was partly antagonized by naloxone, but not completely, indicating that the MSA acts as a partial opioid receptor agonist. Out results suggest that the S. alnifolia may be possibly used as valuable anti-nociceptive agent.

  • 앞의 연구에서 우리는 진세노사이드 Rc, Rd, Re 및 Rr를 복강내 전 처리할 경우 포르말린으로 유도된 통증을 억제하다는 것을 보고하였다 그러나 이러한 진세노사이드가 어느 위치에서 항통증작용을 발휘하는가에 대하여서는 아직 알려지지 않고 있다. 본 연구에서는 이들 진세노사이드를 뇌실내, 척수강내 혹은 피하내 전처리한 다음 포르밀린에 의하여 유도되는 통증이 어느 위치에서 억제되는가를 연구하였다. 연구 결과 이들 진세노사이드는 척수강내 전처리할 경우 포르말린에 의하여 유도되는 통증을 억제하는 것으로 나타났다. 급성 통증 phase에서 ED$_{50}$는 Rc가 1.0 (0.SS~l.75mg/kg)이었고, Rd가 1.15 (0.6~2.25 mg/kg)이었고, Re가 8.9(3.9~20.5 mg/kg)이었다. 지속성 통증 phase에서 ED$_{50}$는 Rc가0.3 (0.1~0.85 mg/kg)이었고, Rd가 0.6 (0.35~l.1 mg/kg)이었고, Re가 2.45 (1.2s~4.65 mg/kg)이었고, Rf가 1.9(1.5~4.25 mg/kg)인 것으로 나타났다. 또한 뇌실내 전처리할 경우에도 이들 진세노사이드들은 포르말린에 의하여 유도되는 통증을 억제하였다. 급성통증 phase에서 ED$_{50}$는 Rc가 0.9 (0.55~l.4 mg/kg)이었고, Rd가0.9 (0.45~l.7 mg/kg)이었고, Re가 0.93 (0.Sol.75 mg/kg), Rf가1.85 (0.95~3.5 mg/kg)인 것으로 나타났다. 지속성 통증 phase에서는 ED$_{50}$는 Rc가 0.7 (0.45~l.05 mg/kg)이었고, Rd가 1.25(0.7~2.2 mg/kg)이었고, Re가 0.85 (0.45~l.6 mg/kg)이었고, H의 경우에는 0.8 (0.4~l.45 mg/kg) 이었다. 항통증 효능 potency는 두 가지 투여 경로에 있어서 Rc$\geq$Rd>Re>Rf인 것으로 나타났다. 그러나, 피하내 주사는 포르말린에 의하여 유도되는 통증을 억제하지 않은 것으로 나타났다. 이러한 연구 결과는 진세노사이드에 의한 항통증 작용은 척수 수준 및 척수위 수준에서 이루어진다는 것을 보여주고 있다.보여주고 있다.

디프테리아 toxin 정제와 무독화 toxoid 백신 생산 (Purification of Diphtheia Toxin and the Production of Detoxificated Toxoid Vaccine)

  • 본 연구에서는 디프테리아 toxin을 정제한 후 무독화하여 toxid를 생산하기 위하여 crude toxin을 2중 U.F를 통해 분자량 100.000이상의 단백질과 30.000이하의 배지 유래 단백질 및 색소를 제거한 결과 순도 1,300 Lf/mg PN의 toxin을 정제하였다. 이를 다시 DEAE-ion exehange chromatography를 통해 toxin을 정제한 후 무독화하여 순도 2,560 Lf/mg PN의 toxoid를 얻을 수 있었다. 이와 같이 생산된 디프테리아 toxoid는 동물 실험 결과 toxin으로 reversion이 발견되지 않았으며, 역가에 있어서도 crude toxin을 무독화한 후 정제한 toxoid와 비교하여 더 우수하였고 대한민국 생물학적 제제 기준에 규정된 성인용 디프테리아 백신 순도 기준 2,500 Lf/mg PN 이상에 적합하였다. 따라서 본 연구를 통해 성인용 디프테리아 백신의 생산 가능성을 확인하였다.

Comorbid Gastric Adenocarcinoma and Gastric and Duodenal Strongyloides stercoralis Infection: A Case Report

  • Strongyloides stercoralis can cause systemic infection, termed strongyloidiasis, and gastrointestinal ulcer disease in immunocompromised patients. However, to our knowledge, there are no reported cases of comorbid gastric adenocarcinoma and S. stercoralis infection. Here, we report a case of an 81-year-old Korean man who presented with S. stercoralis infection coexisting with early gastric adenocarcinoma (T1aN0M0). S. stercoralis eggs, rhabditiform larvae, and adult females were observed in normal gastric and duodenal crypts. They were also observed in atypical glands representative of adenocarcinoma and adenoma. Preliminary laboratory tests revealed mild neutrophilic and eosinophilic leukocytosis. A routine stool test failed to detect rhabditiform larvae in the patient's fecal sample; however, S. stercoralis was identified by PCR amplification and 18S rRNA sequencing using genomic DNA extracted from formalin-fixed paraffin-embedded tissues. Postoperatively, the patient had a persistent fever and was treated with albendazole for 7 days, which alleviated the fever. The patient was followed-up by monitoring and laboratory testing for 4 months postoperatively, and no abnormalities were observed thus far. The fact that S. stercoralis infection may be fatal in immunocompromised patients should be kept in mind when assessing high-risk patients.

Performance of HPV E6/E7 mRNA Genotyping Test on Paired Cervical Cancer Exfoliated Cells and Formalin Fixed Paraffin Embedded Tissues

  • Investigation of human papillomavirus (HPV) in archival formalin-fixed paraffin-embedded (FFPE) material is important for understanding cervical carcinogenesis. The objective of the present study was to identify the high risk HPVs (HR-HPVs) using HPV E6/E7 mRNA testing from archival tissues in cervical cancer and the relation to HR-HPVs genotypes in paired cervical exfoliated cells. HPV E6/E7 mRNA testing and DNA chip testing were performed in 79 paired cervical FFPE tissues and exfoliated cells from women with histologically confirmed squamous cell carcinoma and adenocarcinoma. Overall agreement in HR-HPVs detection from FFPE samples and cytology samples were 98.5% in HPV 16, 100% in HPV 18, HPV 31, HPV 33, HPV 58, HPV 66, and HPV 68. Type-specific agreement between FFPE samples and cytology samples was 89.1% in HPV positive, 93.5% in HPV 16 and more than 70% in the other HR-HPVs. In conclusion, HR-HPVs were reliably detected in paired FFPE and cytology samples with some variation in type-specific detection.
