  • 최근 중합효소연쇄반응을 이용한 분자생물학적 유전자분석기술의 발달로 성염색체상의 유전좌위 증폭을 통한 성별감정이 활발히 이루어지고 있다. 그중 사람 Y염색체상에 존재하는 남성 고환의 형성을 유도하는 sex-determining region Y(SRY) gene이 규명되어 유전질환의 조기 발견이나 예방 및 태아의 성별판정 등에 응용되고 있다. 그러나, 치아는 외부 환경에 대한 저항성이 가장 높은 장기로 성별감정 등 법의치과학적 개인식별에 널리 이용되고 있음에도 불구하고, SRY 유전자를 이용하여 치아에서의 성별감정에 대한 연구는 시도된 바 없다. 따라서, 본 연구에서는 사람 치아에서 중합효소연쇄반응법을 이용한 SRY 유전자를 검출하여 성별판정에 용용하고자 하였다. 남녀 각각 20개 치아의 치수와 상아질에서 DNA를 추출하여 중합효소연쇄반응 을 시행하고 SRY 유전자를 검색한 결과, 남성에서는 치수 13개중 8개, 상아질 7개중 4개에서 SRY 유전자가 검출되었고, 여생에서는 검출되지 않았다. 이러한 결과는 중합효소연쇄반응법을 이용하여 사람 치아에서 SRY 유전자를 검색할 때, 남성판별에 유용하고 치아를 이용한 성별감정시 기존의 성별감정에 이용되고 있는 다른 유전자와 함께 SRY 유전자를 검색함으로써 성별감정의 신뢰도를 높힐 수 있을 것으로 사료된다.

  • SOX9과 SRY 유전자는 척추동물에서 남성고환의 형성을 유도하는 요소로 알려졌다. SOX9 유전자는 SRY related HMG box gene중 하나로 유전질환의 XY성전환 및 성을 결정하는 데에 관여하며 성결정시기에 그 양에 따른 성전환 발생등 연구가 진행되고 있다. 그러나 이 유전자가 성별판정에 유용할 지는 확실치 않다. 반면 SRY 유전자는 포유동물에서의 배형성시기 고환형성을 결정하는 Y염색체 유전자로 남성에만 존재하고 여성에는 존재 않는다. 현재까지 이을 이용하여 법의학적 검체에서 남성판별에 유용하게 사용되고 있다. 본 실험에서는 X, Y와 같은 성염색체가 아닌 상동염색체상에 있으면서 SRY 유전자와 더불어 남성고환을 결정하는 또다른 요소로서의 기능을 가진 SOX9 유전자를 치아에서 검출하여 법의학적 성별판정에 유용할 수 있는지 알아보고자 본 연구를 수행하였다. 남녀각각 5개의 치아에서 치수와 상아질을 분리한 후 DNA를 추출하여 SOX9과 SRY 유전자의 특이적인 시발체를 제작하고 중합효소연쇄반응을 시행하여 증폭하고 전기영동을 시행하였다. 그 결과 SOX9 유전자는 남녀모두에서 유전자가 검출되었고, SOX9 유전자산물과 SRY 유전자를 혼합하여 사용시 남자에서만 유전자가 검출되었다. 이는 법의치과학적 성별판정에 있어 SOX9 유전자는 사람의 치아에서는 남녀 모두 존재하며 남녀 구별을 위한 성별판정에는 이용할 수 없으며 SRY 유전자와 함께 적용시 남성 특이적 SRY 유전자 검사중 발생할 수 있는 가성 음성 반응여부를 확인하는 데 유용할 것으로 사료된다.

  • Forensic odontology in individual identification has been progressing rapidly and its significance has been increasing. But not all individual identification can be done single method solely. Therefore, understanding the advantage and disadvantage of each method is necessary to achieve forensic individual identification properly in each case for it to be applied promptly, economically and precisely. Now 4 cases of forensic odontology are studied. 2 cases are age identification and 2 cases is forensic confirmation.

  • Forensic odontology in individual identification has been progressing rapidly and its significance has been increasing. But not all individual identification can be done single method solely. Therefore, understanding the advantage and disadvantage of each method is necessary to achieve forensic individual identification properly in each case for it to be applied promptly, economically and precisely. Now We have identified almost 20 case in Busan and Kyeong Nam area in 2008, 2009 and we would like to report some of cases. Finally We will consider role of forensic odontology.

  • Forensic odontology can be defined in many ways. One of definitions is simply that forensic odontology represents the overlap between the dental and the legal professions. The most important function of forensic odontology to determine of human identity from dental character. We had found 7 unidentified human bodies in Busan metro politan city, 2007 and we have identified 5 bodies among them.

Quantitative and qualitative evaluation on the accuracy of three intraoral scanners for human identification in forensic odontology

  • The purpose of this study was to analyze the accuracy of intra oral scanner (IOS) to confirm the applicability of IOS for the recording and analysis of tooth morphology in forensics. The less damaged mandible specimen with many teeth remaining was scanned three times using three types of intraoral scanners (CS3600, i500, and Trios3). For quantitative comparisons of the scanned images produced by these intraoral scanners, root mean square (RMS) values were computed using a three-dimensional analysis program and a one-way ANOVA was conducted with Tukey HSD (honestly significant difference) as a post-hoc analysis (α=0.05). The repeatability of the full scan data was highest with the i500 (0.14±0.03 mm), and the post-hoc analysis confirmed significant differences between the CS3600 and the i500 outcomes (P-value=0.003). The repeatability of the partial scan data for the teeth in the mandible was highest with the i500 (0.08±0.02 mm), and the post-hoc analysis confirmed significant differences between the CS3600 and the i500 (P-value=0.016). The precision of the full scan data was highest with the i500 (0.16±0.01 mm) but the differences were not statistically significant (P-value=0.091). Meanwhile, the precision of the partial scan data for the teeth in the mandible was highest with the Trios3 (0.22±0.02 mm), but the differences were not statistically significant (P-value=0.762). Considering that the scanning of other areas of the oral cavity in addition to the teeth is important in forensic odontology, the i500 scanner appears to be the most appropriate intraoral scanner for human identification. However, as the scope of oral scanning is generally limited to teeth in the practice of dentistry, additional discussions of how to apply the IOS in forensic odontology are needed. Ultimately, the results here can contribute to the overall discussion of the forensic applicability dental data produced by intraoral scanners.

Allele Frequency of the Short Tandem Repeat Locus Human Lipoprotein Lipase(LPL) Gene by Polymerase Chain Reaction in the Korean Population (한국인에서 중합효소연쇄 반응법에 의한 STR 유전좌위 LPL의 유전자빈도 검색)

  • 한국인 집단에서 개인식별의 기초자료로 활용하고자 한국인 201명을 대상으로 STR 유전좌위 중 하나인 LPL 유전좌위의 유전자 빈도 및 유전자형 분포를 구하였다. 혈액으로부터 추출한 핵 DNA를 중합효소연쇄반응으로 증폭시키고 폴리아크릴아마이드 겔 상에서 전기영동하여 은염색한 후 관찰하여 다음의 결과를 얻었다. 1. 한국인 집단 201명의 LPL 유전자에서 5개의 대립유전자, 7개의 유전자형을 검출하였으며, 이형접합도는 50.7%로 나타났고 대립 유전자다양성 (allelic diversity value)은 0.454, 개 인식 별력 (PD)은 0.674를 보였다. 2. 대립 유전자 및 유전자빈도는 9, 10, 11, 12, 13 대립 유전자에서 각각 0.020, 0.714, 0.100, 0.164, 0.002로 나타났으며, 대립유전자 7, 8, 14는 관찰되지 않았다. 이상의 결과를 볼 때 한국인 집단에서 STR LPL유전좌위의 유전자빈도는 친자감정 등 개인식별에 유용하게 사용할 수 있으나 감정실무에 응용시 다수의 STR유전좌위 및 VNTR유전좌위의 분석을 병행하여야 할 것으로 사료된다.

  • In order to be utilized as a database in forensic identification and parentage test, allelic frequency and genotype distribution of short tandem repeat(STR) F13B locus was analysed by polymerase chain reaction in 210 Korean adults who are not related. The results were as follows. 1. 3 alleles and 56 genotypes of F13B locus were detected and heterozygosity value was 48.6% and allelic diversity value was 0.639 and the power of discrimination was 0.804. 2. The observed each alleles and allelic frequency was 8(0.069), 9(0.193), 10(0.738). In conclusion, the allelic frequency of STR F13B locus in the Korean is considered as an useful DNA allelic profile for forensic identification, but it should be used with several other STR locus to get definitive conclusion of analysis for individual identification and parentage testing.

  • Allelic frequency and genotype distribution of short tandem repeat(STR) F13A01 locus was analysed by polymerase chain reaction, polyacrylamide gel electrophoresis and silver staining from human genomic deoxyribonucleic acid(DNA) was extracted from 205 unrelated Korean to be applied to forensic identification and parentage testing as a database. The results were as follows : 1. 5 alleles and 11 genotypes of F13A01 locus were detected and heterozygosity value was 62.0% and the observed each alleles and allelic frequency was 3.2(0.363), 4(0.105), 5(0.063), 6(0.466), 16(0.002). 2. The allelic diversity value was 0.639 and the power of discrimination was 0.804.3. Compared with observed number of alleles and allele frequency in ethnic difference, result was appeared to be similar to that of Japanese and Asians, while was appeared to be much different to that of Blacks and Caucasians in the observed number of alleles and frequency of allele 3.2, 5, 7. From the above result of this investigation, the allelic frequency of STR F13A01 locus in the Korean was considerd to be useful for individual identification and parentage testing as a database.
