Effects of Feeding Herb Resources Powder on Meat Quality and Sensory Properties in Korean Native Black Goat (한방제재 급여가 재래 흑염소육의 육질 및 관능적 특성에 미치는 영향)

  • This study was carried out to investigate the effect of different herb resource powders on meat quality and sensory properties in Korean native black goat for 500 d. The experimental treatment was arranged with 24 heads (3 treatment/8 heads) by control group, T1 and T2 group respectively (Control, not added herb powders; T1, 1% added herb powders; T2, 2% added herb powders). Total weight gain for the Korean native black goat was the highest in T2 group feeding for 500 days, and daily gain tended to be similar to the total weight gain. The total feed intake were the highest as 519.20 kg in T2 group, although feed conversion showed 18.35 in the T2 group, which means it had the best feed efficiency compared to the other treatment groups. The carcass rate was higher in the T1 group (51.10%) than in the other groups (p<0.05). The cooking loss and drip loss of Korean native black goat was the highest as 34.72% and 3.83% in the control group (p<0.05). However, total cholesterol amounts in the treatment group were not significantly different from, although tended to be higher than, the control group (p>0.05). Also, the overall sensory evaluation of the treatment group revealed low scores, meaning more meat flavor than those of the control in tenderness, flavor, texture, and black goat off-flavor and overall evaluation (p<0.05). Total synthesis evaluation was higher for the treatment group (3.71, 3.90 point) than that of the control group (4.82 point) (p<0.05). Processing of Functional Porridge with Optimal Mixture Ratio of Mulberry Leaf Powder and Mulberry Fruit Powder (뽕잎분말과 오디분말의 최적 혼합비율을 이용한 기능성 죽 제조)

  • The purpose of this study was to develop a functional porridge prepared with mulberry leaf and mulberry fruit powder, which can ameliorate hypertension. The experiment was designed according to the central composite design. For optimization of the mixture ratio of mulberry leaf powder (MLP) and mulberry fruit powder (MFP), the independent variables were defined as MLP (X1) and MFP (X2) and the dependent variables were defined as K (Y1), Na (Y2), ${\gamma}$-aminobutyric acid (GABA) (Y3), cyanidin-3-glycoside (C3G) (Y4), rutin (Y5), and flavonoid (Y6). The optimal MLP to MFP mixture ratio according to the response surface method were 5.41 g of MLP and 2.65 g of MFP. The amounts of K, Na, GABA, C3G, rutin, and flavonoid in the optimal MLP and MFP mixture were 1,844.22 mg/100 g, 52.74 mg/100 g, 139.98 mg/100 g, 1,134.89 mg/100 g, 101.56 mg/100 g, and 201.28 mg/100 g, respectively. The amounts of Ca, K, Mg, and Na in the functional porridge at this optimal point were 27.66 mg/100 g, 131.32 mg/100 g, 19.57 mg/100 g, and 3.59 mg/100 g, respectively. Overall, this functional porridge can help reduce hypertension.

Enhancement of Skin Immune Activation Effect of Collagen Peptides Isolated from Asterias amurensis (불가사리 유래 콜라겐 펩타이드의 피부 면역 증진 효과)

  • Low molecular peptides were isolated from Asterias amurensis via SDS-PAGE. The peptides were separated via consecutive gel filtration as five fractions (F1-F5) according to molecular weights, based on the results of MALDI-TOF MS analysis. The molecular weight of the most active peptide was estimated as 15,000 daltons. The peptide showed cytotoxicity on normal human fibroblast cells at levels as low as 20% when 1.0 mg/mL of the samples was added. The peptide also exhibited higher levels of nitric oxide production from macrophages than the lipopolysaccaharides. It was determined that prostaglendin $E_2$ production was significantly inhibited, up to 127.8% as compared to the control. The low molecular peptide inhibited hyaluronidase activity as 535.7 ${\mu}g/mL$ of $IC_{50}$. It can be concluded that the relatively low molecular weight peptide, fucoidan, from A. amurensis has excellent cosmetic and immunomodulatory activities, which can be considered as a possible resource of new cosmetic agents for skin immunomodulation.

Antioxidant activity and physicochemical composition of fermented Vigna angularis using Bacillus subtilis KCCM 11965P (Bacillus subtilis KCCM 11965P를 이용한 붉은팥 발효물의 항산화 활성 및 이화학성분 분석)

  • Health of human intestine has deteriorated due to excessive stress and western diet. In this study, Vigna angularis was fermented by Bacillus subtilis KCCM11965P in order to develop prebiotic resource for improving bowel movement. The contents of ash and crude protein were $3.35{\pm}0.04%$ and $21.1{\pm}0.19%$ respectively. Vigna angularis extract (1, 3, 5%) were incubated with 3% (v/v) Bacillus subtilis KCCM 11965P for 0, 24, 48, and 72 h. Total bacterial numbers showed that the combination of 3% powdered seeds and 72 h incubation time was optimum condition for this experiment. Total polyphenol content increased from $0.18{\pm}0.010mg/mL$ in pre-incubation to $0.23{\pm}0.007mg/mL$ in post-incubation with the condition mentioned above. DPPH radical scavenging activity also increased from $36.1{\pm}6.0%$ to $63.6{\pm}5.2%$. Analysis of protease activity showed $2.69{\pm}0.003unit/mL$ in combination of 5% powdered seeds and 72 h incubation time. Amylase activity increased from $1.0{\pm}0.1unit/mL$ in pre-incubation to $26.0{\pm}0.2unit/mL$ in post-incubation. The analysis of free amino acids after incubation with Bacillus subtilis KCCM 11965P showed that leucine increased from 5.22 mg/L to 67.59 mg/L and tyrosine, one of non-essential amino acid also increased 10.08 mg/L to 259.35 mg/L by incubation with 5% powdered seeds. Most of organic acid were reduced by incubation for 72 h. These results suggest that Vigna angularis could be utilized most as a prebiotic resources.

Effect of Achyranthis Radix and Drynariae Rhizoma Extracts on Antioxidant Activity and Antioxidant Enzymes (우슬과 골쇄보의 추출물이 항산화 활성 및 항산화 효소 대사에 미치는 효과)

  • In vitro and in vivo experiments using Achyranthis radix and Drynariae rhizoma extracts were conducted. Antioxidant properties were analyzed and the effects on bone, glucose and lipid metabolism were investigated. Drynariae rhizoma (64.67%) obtained higher DPPH radical scavenging activity compared to Achyranthis radix (19.03%). Similar results were obtained in the reducing power. No differences were observed on the ABTS radical scavenging ability and SOD. In contrast, Achyranthis radix (77.60%) has higher chelating ability compared to Drynariae rhizoma (46.21%). In vivo experiments revealed higher plasma TBARS in OVX-DR than in OVX-AR. Opposite result was seen in erythrocyte TBARS. Hepatic, nephritic and erythrocyte enzymes were considered for the antioxidant enzyme activities. GSH-Px and PON of hepatic enzymes were higher in OVX-AR. While the CAT and GR were higher in OVX-DR. SOD, GSH-Px, GR and PON of nephritic enzymes of OVX-DR were higher compared to OVX-AR. Almost similar values were obtained in CAT using both extracts. The OVX treated rats obtained higher CAT and GR in the erythrocyte enzymes compared to SHAM. The SOD of erythrocyte enzymes in OVX-DR was higher compared to OVX-AR. On the other hand, the GSH-Px was higher in OVX-AR.

Effects of Dietary Supplementation with Rosemary and α-Tocopherol Acetate on Performance and Meat Quality of Chicken Meat during Refrigerated Storage (로즈마리와 α-Tocopherol Acetate의 급여가 육계의 생산성 및 냉장 저장 중 계육의 품질에 미치는 영향)

  • The effects of rosemary and $\alpha$-tocopherol, added individually or in combination, on broiler performance, thiobarbituric acid reactive substance (TBARS), total plate count (TPC) and meat color of chicken thigh meat were investigated. Three hundred broiler chicks divided into five groups were fed a basal diet (control) or basal diet supplemented with 5 g rosemary/kg (T1), 10 g rosemary/kg (T2), 200 mg $\alpha$-tocopherol/kg (T3), or 5 g rosemary/kg + 200 mg $\alpha$-tocopherol/kg (T4) for 5 weeks. Following slaughter, chicken meat was stored at $4^{\circ}C$ for 10 days. All treatments did not influence the performance. Rosemary supplementation delayed lipid oxidation in thigh meat during refrigerated storage. T2 was significantly (p<0.05) more effective in delayed lipid oxidation compared to T1, but was inferior to T3. Samples containing a combination of antioxidant had lower TBARS values than those containing the individual antioxidants, indicating a synergistic effect. TPC was significantly increased (p<0.05) in thigh meat of all groups throughout the refrigerated storage. The T3 and control groups showed TPC counts that did not differ from each other during the entire storage period. However, rosemary supplementation was associated with bacterial counts that were significantly lower (p<0.05) than the control and $\alpha$-tocopherol groups at day 3 of storage and thereafter. For this period, T1 presented TPC counts that were significantly higher than the T2 group (p<0.05). At all storage times, the thigh meat of rosemary-fed chickens was redder than control (higher $a^*$), while no differences in $L^*$ and $b^*$ values were found. A synergistic effect was obtained from the combination of rosemary with $\alpha$-tocopherol, whereas individual use of the antioxidants significantly improved color stability compared to the control.

Study on the Screening System of Organic Resources for Agricultural Utilization (유기성 자원의 농업적 활용을 위한 선별체계 연구)

  • This study was conducted to find suitable methods for screening organic resources useful for compost. Twenty-seven industrial and domestic sludges were collected from various cities and industrial areas. Contents of organic matters in the sludges were in the range of 79.3-98.0%, and the contents were much higher than the regulation level (60%) for raw materials of compost. Contents of total nitrogen were in the range of 0.8-2.6%. Contents of Fe and Al were very high. Content of HEM was highest in textile sludge ($257mg\;kg^{-1}$) and the contents in the others were in the range of $12.6-90.3mg\;kg^{-1}$. Content of PAHs was lowest in food sludge ($739.1{\mu}g\;kg^{-1}$ and pulp-mill sludge had the highest PAHs content ($3461.8{\mu}g\;kg^{-1}$). $Microtox^{(R)}$ $EC_{50}$ values were higher in the sludges which were classified as a possible material in composting after analysis and investigation. Lettuce root elongation and $EC_{50}$ values were relatively lower in pulp-mill sludge, sewage sludge 3 (Large city), food sludge and leather sludge. Therefore, mineral nutrients, heavy metals, organic compounds (HEM, PAHs, PCBs), and bioassay ($Microtox^{(R)}$ $EC_{50}$, Relative root elongation test) are recommended to be included in the screening system of raw material of compost in addition to the current regulation with organic matter and 8 heavy metals.

Assessment of Nutritional Components, Antioxidant Contents and Physiological Activity of Purple Corn Husk and Cob Extracts (자색옥수수 포엽과 속대 혼합 추출물의 영양성분, 항산화 활성 물질 함량분석 및 생리활성 평가)

  • The objective of this study was to investigate the worth of extract husk and cobs of the Seakso 1 (EHCS) for the functional foods. We aimed to investigate the proximate composition, fatty acids, amino acids, antioxidant active substance contents, antioxidant activity, inhibitory activity of the ${\alpha}-amylase$ and ${\alpha}-glucosidase$. The proximate composition of the EHCS have represented 6.90% moisture, 7.31% crude ash, 0.52% crude fat and 7.07% crude protein. Among the 17 kinds of amino acids that were analyzed in thd EHCS, the glutamic acid was the highest, with 736.08 mg / 100 g. The fatty acids detected in the EHCS were palmitic acid oleic acid and linoleic acid. The proportion of the unsaturated fatty acids was 83.33%. We determined the contents of the antioxidant active substance by the total polyphenol and flavonoid. The total polyphenol and flavonoid contents were 99.87 mg/g and 25.02 mg/g, respectively. The antioxidative activity of the EHCS were determined using a DPPH and ABTS assay. In the antioxidative activity determination, the DPPH and ABTS radical scavenging activities were 95.62% ($1,000{\mu}g/mL$) and 92.00% ($10,000{\mu}g/mL$), respectively. The inhibitory activity of ${\alpha}-amylase$ and ${\alpha}-glucosidase$ (10 mg/mL) were 95.86% and 76.92%, respectively. These results suggest that the EHCS could be potentially used as a resource for the bioactive materials for health functional foods.

Effects of Co-digestate application on the Soil Properties, Leachate and Growth Responses of Paddy Rice (통합혐기소화액의 시용이 벼 생육 및 논토양 환경에 미치는 영향)

  • Livestock manures have a potential to be a valuable resource with an efficient treatment. In Korea, 42 million tons of livestock manure were generated in 2008, and 84 % of them were used for compost and liquid fertilizer production. Recently recycling of livestock manure for biogas production through anaerobic digestion is increasing, but its utilization in agriculture is still uncertified. In this study, there was applied co-digestate to the paddy for rice cultivation based on N supplement. Co-digestate was fertilizer fermented with pig slurry and food waste combined with the ratio of 70:30(v:v) in its volumetric basis. For assessing the safety of co-digestate, it was monitored the contents of co-digestate for seasonal variation, resulted in no potential harm to the soil and plant by heavy metals. The results showed that soil applied with co-digestate was increased in exchangeable potassium, copper and zinc mainly due to the high rate of pig slurry in co-digestate applied. Considering high salt content due to the combination with food waste, strict quality assurances are needed for safe application to arable land though it has valuable fertilizer nutrient. Leachate after treatment showed that the concentration of nitrate nitrogen washed out within two weeks. Considering the salt accumulation results in soil, it is highly recommended that the application rate of co-digestate should not exceed the crop fertilization rate based on N supplement. With these results, it was concluded that co-digestate could be used as an alternative fertilizer for chemical fertilizer. More study is needed for the long-term effects of co-digestate application on the soil and water environment.

Effects of the Artificial Culture Medium of Wild Ginsengs on Rumen Fermentation Characteristics In Vitro (산삼배양액 이용에 관한 반추위 미생물 대사 연구)

  • This study was conducted to determine the effects of the artificial culture medium of wild-ginsengs on in vitro fermentation characteristics. NH$_3$-N concentration was showed the highest in 3% WGM treatment among all treatments and control. In addition, microbial protein synthesis was significantly different in all treatments throughout the incubation time, and WGM 3% treatment was the highest at the 9 h incubation(P〈0.05). Protozoa numbers within rumen were decreased in all WGM treatments at 9 h incubation time, whereas WGM 3% treatment was always decreased throughout the incubation(P〈0.05). NDF and ADF digestibility were proportionally increased as the incubation time in both control and treatments. NDF digestibility showed no significantly difference between control and the 3% treatment, and ADF digestibility was similar in all. Total volatile fatty acid(VFA) concentrations of WGM treatments without 5% were significantly higher than control (P〈0.05). No differences were observed in total VFA, acetate, propionate and butyrate concentration among the WGM treatments. Acetate/Propionate ratio of WGM treatments was higher than control after 12 h incubation(P〈0.05). As a result of the artificial culture medium of wild-ginseng on rumen fermentation characteristics in vitro, microbial protein synthesis of WGM treatment was higher than control, and WGM 3% was the highest in all treatments(P〈0.05). The effect of saponin in artificial culture medium of wild-ginseng tended to decrease NH$_3$-N concentration, while it increases the microbial synthesis in early incubation. Therefore, artificial cultures medium of wild-ginseng can increase utilization of feed by microbial and anti-protozoal effects of saponin, which may enhance microbial synthesis capacity in early fermentation period in rumen.
