Analysis of erectile dysfunction drugs and their analogues in counterfeit drugs and herbal medicines by LC-ESI-MS/MS

  • Distribution of various illegal or counterfeit drugs of seven approved erectile dysfunction drugs and their analogues has been increased, causing health problems such as cardiovascular disorder, tachycardia, headache, or vision disturbance. We used liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) to determine the erectile dysfunction drugs and their analogues in various counterfeit drugs. Eleven erectile dysfunction drugs and their analogues were detected, with sildenafil and its analogues being the most counterfeited compounds (73.8 %), followed by tadalafil and its analogues (25.4 %). The limits of detection (LOD) and the limits of quantitation (LOQ) of liquid-type and solid-type negative samples ranged from 0.1 to 3.3 ng/mL or ng/g and from 0.3 to 10.0 ng/mL or ng/g, respectively. The recoveries ranged from 84.3 to 112.3 % and 83.2 to 110.2 %, respectively. The contents of sildenafil and tadalafil in the various counterfeit drugs ranged from 21.0 to 947.5 mg/g and from 0.2 to 170.2 mg/g, respectively.

한국, 미국, 일본의 식품성분표 비교 (International Comparison of Food Composition Table)

  • This study was conducted to compare the composition table of Korean food with that of foreign food. Analysis was made for Korean food composition table($5^{th}$ revision), Korean food composition table in Appendix of Recommended Dietary Allowances for Korean(6$^{th}$ th/ edition), Standard tables of food composition in Japan($5^{th}$ revised edition) and USDA Composition of Foods - Raw, Processed, Prepared. The method of content analysis was applied for this study and such differences were pointed out as the classification of food items, food items enlisted, the content unit of food and food components presented etc. To improve Korean food composition table, new food items and components should be added and old food items be eliminated based on the change of people's food consumption pattern. Also analysis for the domestic foods consumed by local people should be accomplished rather than borrowing foreign country's data.

LC-MS/MS를 이용한 한국에서 유통되는 농산물의 잔류 농약 평가 (Evaluation of Residual Pesticides in Commercial Agricultural Products Using LC-MS/MS in Korea)

  • 본 연구에서 한국 유통 농산물 17품목(쌀, 사과, 배, 오렌지, 복숭아, 딸기, 바나나, 파인애플, 포도, 키위, 수박, 토마토, 오이, 호박, 참외, 깻잎, 상추 등)을 대상으로 다성분 분석농약 60종을 분석하기 위하여 대상농산물의 대분류별로 곡류에서는 현미, 두류는 메주콩, 과실류에서는 감귤, 채소류는 고추, 서류에서는 감자, 버섯류에서는 느타리버섯을 선정하여 회수율 실험을 수행하였다. 60% 이상의 낮은 회수율을 나타내는 성분 중 fluquinconazole, methomyl은 4가지 이상의 농산물에서 공통적으로 낮은 회수율을 나타내었다. fluquinconazole은 식품에 적용하는 식품공전 4.1.3.22의 개별 시험법을 이용한 전처리 과정을 통해 과일류, 과채류, 콩류에서 개선된 회수율 결과를 얻을 수 있을 것으로 예상되며 methomyl 은 고추, 대두, 버섯 등 식품에 적용하는 식품공전 4.1.3.15의 개별 시험법을 통해 개선된 회수율 결과를 얻을 수 있을 것으로 예상된다. 버섯, 고추에서 공통적으로 60% 미만의 낮은 회수율을 나타내었던 butocarboxim은 현재 식품공전상 개별 시험법이 존재하지 않는 항목이므로 이에 대한 개별시험법을 개발하여 다종농약다성분 시험법을 보완할 필요가 있다고 사료된다. 한국 유통 농산물 17품목(쌀, 사과, 배, 오렌지, 복숭아, 딸기, 바나나, 파인애플, 포도, 키위, 수박, 토마토, 오이, 호박, 깻잎, 상추)을 대상으로 다성분 분석농약 60종을 분석하여 농약 잔류실태를 조사하였다. 조사대상 농산물 358건 중 대부분의 농산물인 291건(81.3%)의 농산물은 농약이 검출되지 않았으며, 67건(18.7%)의 농산물에서 농약이 검출되었으나, 잔류허용기준 미만으로 검출되었다. 쌀 등 17품목에의 다소비 농산물의 농약 잔류실태를 조사한 결과 농약의 잔류량은 잔류허용기준 미만으로 ADI 대비 EDI는 0.000009%에서 0.0861%로 위해도가 아주 낮은 것으로 조사되었으며 비교적 안전한 수준으로 농산물에 대한 농약관리가 이루어지는 것으로 판단되었다.

식품의 선택 동기가 유기농 식품에 대한 태도 및 구매의도에 미치는 영향에 관한 연구 (The Effect of Food Choice Motive on Attitude and Intention of Purchasing Organic Food)

  • Due to an overall increase of income, the general standard of living has improved and people have begun to be interested in being more healthy in their lives. This tendency has affected the food market, especially in relation to organic and eco-friendly food. Thus, the overall market size for those products has grown to give more choices to consumers. To examine the effect of the motive for choosing certain food products on the actual attitude and intent to purchase the products, a survey was given to 330 people living in Seoul, which resulted in 235 usable responses. The content of the questionnaire consisted of 18 questions on food choice motives, 3 questions on the attitude toward organic foods and 3 questions on the intention of purchasing for organic foods. The SPSS 12.0 statistics program was used to analyze of following: frequency analysis, factor analysis, reliability analysis, t-test, one way ANOVA and regression analysis. Five factors of food choice motives were obtained from the analysis: health, convenience, price, familiarity and environmental protection. The regression analysis showed that food choice motive, health and environmental protection factors have a positive relationship with organic food attitudes and organic food attitudes have a positive relationship with the intent to purchase organic food.

  • Metabolomics aims at the comprehensive, qualitative and quantitative analysis of wide arrays of endogenous metabolites in biological samples. It has shown particular promise in the area of toxicology and drug development, functional genomics, system biology and clinical diagnosis. In this study, analytical technique of MS instrument with high resolution mass measurement, such as time-of-flight (TOF) was validated for the purpose of investigation of amino acids, sugars and fatty acids. Rat urine and serum samples were extracted by selected each solvent (50% acetonitrile, 100% acetonitrile, acetone, methanol, water, ether) extraction method. We determined the optimized liquid chromatography/time-of-flight mass spectrometry (LC/TOFMS) system and selected appropriated columns, mobile phases, fragment energy and collision energy, which could search 17 metabolites. The spectral data collected from LC/TOFMS were tested by ANOVA. Obtained with the use of LC/TOFMS technique, our results indicated that (1) MS and MS/MS parameters were optimized and most abundant product ion of each metabolite were selected to be monitorized; (2) with design of experiment analysis, methanol yielded the optimal extraction efficiency. Therefore, the results of this study are expected to be useful in the endogenous metabolite fields according to validated SOP for endogenous amino acids, sugars and fatty acids.

Quantitative Analysis and Enantiomeric Separation of Ephedra Alkaloids in Ma Huang Related Products by HPLC-DAD and UPLC-MS/MS

  • Ephedra is a genus of the Ephedraceae family and is found in temperate regions, such as Central Asia and Europe. Among the various ephedra species, Ma Huang (Ephedra herb) is derived from the aerial parts of Ephedra sinica S tapf, Ephedra equisetina Bunge, and Ephedra intermedia Schrenk & C.A. Mey. Ma Huang contains various ephedra alkaloids, including (-)-ephedrine, (+)-pseudoephedrine, (-)-norephedrine, (+)-norpseudoephedrine, (-)-methylephedrine, and (+)-methylpseudoephedrine, which are found naturally as single enantiomers, although they can be prepared as racemates. Although the use of Ma Huang in foods is prohibited in Korea, products containing Ma Huang can be imported, and so it is necessary to develop a suitable analytical technique for the detection of Ma Huang in foods. Herein, we report the development of analytical methods for the detection of ephedra alkaloids in products containing Ma Huang. Following sample purification by solid phase extraction, quantitative analysis was performed using ultra-performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-MS/MS). Additionally, the enantiomers were successfully separated using HPLC-DAD. We successfully analyzed various food samples, where the ephedra alkaloids were qualitatively and quantitatively determined, and the enantiomers were separated. It is expected that these methods may contribute toward preventing the distribution of illegal products containing Ma Huang.

Consumer Perception of GM Food: Factors that Influence Purchasing of GM Food in South Korea

  • This study examined how the consumers' perception of GM food affects their purchasing behavior. In addition, this study investigated how the amount of knowledge a consumer has regarding GM food affects their perception of this type of food. The quantitative research method was used to collect data. The data from a self-administered survey, which was conducted in South Korea, was analyzed using descriptive analysis, ANOVA, factor analysis, and multiple regression analysis methods. The results of the survey indicate that most respondents may have unknowingly purchased GM food. Further, the respondents reported that they likely had known about or had heard of GM food. In addition, the survey indicated that the amount of knowledge possessed by the respondents regarding GM food greatly affected their perceptions of this type of food. These findings will contribute to the current GM food market by providing the food market with additional information relating to the consumers' perceptions of GM food.

커피 및 과실류 가공품의 오크라톡신 A 모니터링 (Monitoring Ochratoxin A in Coffee and Fruit Products in Korea)

  • 국내 커피 및 과실류 가공품 중 오크라톡신 함량 조사를 위해 전처리법과 분석법을 확립하여 커피와 과실류 가공품 총 388건을 모니터링한 결과 7건에서 오크라톡신이 검출되었다. 커피 177건 중 7건에서 검출되었고, 평균 함량은 $0.139\;{\mu}g/kg$으로 EU 기준 $5.0\;{\mu}g/kg$ 보다 낮아 안전한 수준으로 판단되었다. 과실류 가공품에서는 211건 모두 오크라톡신이 검출되지 않았다. 이상에서 살펴본 바와 같이 커피 및 과실류 가공품의 오크라톡신의 오염수준은 전체적으로 높지 않지만, 최근 기호 식품으로서 커피 및 과실류 가공품의 섭취가 점차 늘어나고 있고, 오크라톡신 오염량이 비교적 높은 커피를 선호하는 계층에 있어서는 오크라톡신에 지속적으로 노출될 가능성이 있으므로 주의해야 할 것으로 생각되며, 앞으로도 이들에 대한 체계적인 관리와 안전성 확보를 위한 지속적인 노력이 필요할 것으로 생각된다.

죽순식초의 화학적 특성 및 향기분석 (Chemical Characteristics and Flavors of Bamboo-shoot Vinegar)

  • 담양군 죽순으로 제조한 죽순식초와 시판 과실식초의 이화학적 특성인 산도, 무기질, 아미노산, 플라보놀을 비교 분석하고 SBSE-GC/MS방법을 사용하여 향기성분을 분석하였다. 죽순식초의 산도는 4.49%로 제일 낮았으며 무기질 중 Na의 경우 8.36 mg/100 g으로 다른 과실식초에 비해 그 함량이 낮았다. 아미노산은 다른 시판 식초에 비해 tyrosine과 lysine의 함량이 높았으며 flavonol aglycone 중 quercetin이 3.29 mg/L 들어있었다. SBSE방법으로 향기성분을 분석한 결과 aldehyde류의 함량이 시판식초에 비해 높았으며 특이적으로 수면 유도물질로 알려진 oleamide가 처음으로 검출되었다.

녹차음료에서의 카테킨 및 카페인 함량 조사 (Determination of Amounts of Catechin and Caffeine in Green Tea Beverages)

  • The physiologically advantageous aspects of green tea have been identified recently and green tea has been a favorite drink of many people. Due to the increased awareness of green tea's positive effects on human health, the demand for foods containing green tea has increased. This has led to the development of diverse green tea-related beverages; thereby many companies in Korea have put a wide variety of manufactured green tea beverages on the market. However, the components within green tea beverages have not been examined in Korea yet. In this study, we investigated the contents of the physiologically functional materials found in green tea, such as catechin, catechin gallate, epicatechin, epicatechin gallate, epigallocatecin gallate, gallocatechin gallate and caffeine. Fifty-six green tea products purchased from the local grocery stores and cafes were analyzed using high performance liquid chromatography (HPLC) analysis. As a result, all tested products contained catechin and caffeine, although the amount of each component was largely different. The total amount of catechin derivatives in the manufactured green tea beverages purchased from cafes was 263.17 mg/L, while they were 61.99 mg/L in the beverages purchased from the local grocery stores. And, to the almost samples the amount of caffeine was proportional to the amount of catechin.
