Assessment of the Potential Allergenicity of Genetically Modified Soybeans and Soy-based Products

  • A comprehensive safety evaluation was conducted to assess the potential allergenicity of newly introduced proteins in genetically modified (GM) crops. We assessed the allergenicity of CP4 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) in GM soybeans. This assessment was performed by IgE immunoblotting with soy-allergic children's sera, amino acid sequence homology with known allergens, and the digestibility of CP4 EPSPS. No differences in IgE-antigen binding by immunoblotting were found between GM soy samples and the corresponding non-GM samples. Based on the comparison of EPSPS amino acid sequence homology with current allergen databases, no known allergen was found. In addition, CP4 EPSPS protein was rapidly digested by simulated gastric fluid (SGF). Taken together, these results indicate that GM soybeans have no allergenicity in children and are as safe as conventional soybeans.

  • Various insects have emerged as novel feed resources due to their economical, eco-friendly, and nutritive characteristics. Fish, poultry, and pigs are livestock that can feed on insects. The digestibility of insect-containing meals were presented by the species, life stage, nutritional component, and processing methods. Several studies have shown a reduced apparent digestibility coefficient (ADC) when insects were supplied as a replacement for commercial meals related to chitin. Although the expression of chitinase mRNA was present in several livestock, indigestible components in insects, such as chitin or fiber, could be a reason for the reduced ADC. However, various components can positively affect livestock health. Although the bio-functional properties of these components have been verified in vitro, they show positive health-promoting effects owing to their functional expression when directly applied to animal diets. Changes in the intestinal microbiota of animals, enhancement of immunity, and enhancement of antibacterial activity were confirmed as positive effects that can be obtained through insect diets. However, there are some issues with the safety of insects as feed. To increase the utility of insects as feed, microbial hazards, chemical hazards, and allergens should be regulated. The European Union, North America, East Asia, Australia, and Nigeria have established regulations regarding insect feed, which could enhance the utility of insects as novel feed resources for the future.

생선의 Allergen성 판정과 Allergen성을 감소시키는 가공학적 방법 (Evaluation of Allergenicity for Fish and Method for Reduction of Allergenicity by Food Technological Treatment)

  • In this research the results showed that Evans blue stain causes vascular permeation at antibody injection site by the passive cutaneous anaphylaxis(PCA) screening of octpus minor so we concluded. Octopus minor causes allergy. Psedosciaena Polyactis, Raja Kenojei, Metapenaeus joyneri also showed allergenicity. Microwave and autoclaving appeared to reduced allergenicity(up to 99%) during the technological treatment processing. On the other hand, UV light didn't seem to change the protein structure of allergens affect the allergenicity. Therefore, the technological treatment processing of fish such as canning and microwave would possibly reduce the allergenicity. Among the ultrafiltration fraction of Octopus minor, Pseudosciaena Polyactis, Raja Kenojei and Metapenaeus joyneri, those fraction over 100,000 contained allergen and those under 100,000 and when screening allergenic fish went through 10,000∼100,000 ultrafiltration, only the fraction of over 100,000 showed the anaphylactis activity for PCA. However whether screening fish would cause anaphylaxis in human or not is questionable. The future clinical experiment will verify this result with clinical experiment patients. 본 연구 결과로 낙지의 PCA검색으로써 항체주사 부위에 Evan's Blue 착색으로 인하여 vascular permation이 일어나 allergynicity 반응이 인정되므로 낙지는 allergy가 있다고 할 수 있다. 조기, 홍어, 새우도 역시 allergenicity성이 나타난 것을 볼 수 있었다. 또한 공정 중 microwave와 autoclaving은 4가지 수산식품 낙지, 조기, 홍어, 새우의 allergenicity를 현저히 감소시키는 것으로 나타났다. 반면에 자외선은 단백질로 구성된 allergen의 구조는 크게 변화시키지 못하는 것으로 보여진 것을 알 수 있다. 그러므로 생선의 가공방법은 통조림 가공이나 microwave 처리가 allergenicity성을 감소시킬 수 있는 것으로 보여진다. 낙지, 조기, 홍어, 새우의 한외여과 fraction중 고분자인 100,000이상에서만 allergenicity가 나타났다. 따라서 이 allergen들의 분자량은 100,000 이상으로 추정되며 검색된 allergynicity 생선을 10,000∼100,000으로 한외여과 하였을 때 100,000 이상의 fraction에서만 PCA에 의해 allergenicity가 있는 것으로 인정되었다. 그러나 이 검색된 생선이 anaphylaxis가 인간에게서까지도 반응할지는 의문이다. 이 연구의 결과는 차후 임상실험환자에 의하여 최종 검증되어야 한다.

Effect of processed foods on serum levels of eosinophil cationic protein among children with atopic dermatitis

  • The prevalence of atopic dermatitis (AD) in school-age children has increased in industrialized countries. As diet is one of the main factors provoking AD, some studies have suggested that food additives in processed foods could function as pseudoallergens, which comprise the non-immunoglobulin E-mediated reaction. Eosinophil cationic protein (ECP) is an eosinophil granule protein released during allergic reactions to food allergens in patients with AD. Thus, serum ECP levels may be a useful indicator of ongoing inflammatory processes in patients with AD. The purpose of this study was to investigate the effect of consuming MSG in processed foods on serum ECP levels among children with AD. This study was performed with 13 patients with AD (age, 7-11 years) who had a normal range of total IgE levels (< 300 IU/ml). All participants ate normal diets during the first week. Then, six patients were allocated to a processed food-restricted group (PRDG) and seven patients were in a general diet group (GDG). During the second week, children in the PRDG and their parents were asked to avoid eating all processed foods. On the third week, children in the PRDG were allowed all foods, as were the children in the GDG throughout the 3-week period. The subjects were asked to complete a dietary record during the trial period. Children with AD who received the dietary restriction showed decreased consumption of MSG and decreased serum ECP levels and an improved SCORing score on the atopic dermatitis index (P < 0.05). No differences in serum ECP levels or MSG consumption were observed in the GDG. Serum total IgE levels were not changed in either group. In conclusion, a reduction in MSG intake by restricting processed food consumption may lead to a decrease in serum ECP levels in children with AD and improve AD symptoms.

Parthenolide Inhibits Ovalbumin-Induced cyclooxygenase-2 and Inducible Nitric Oxide Synthase Expression

  • In recent years, egg allergy has been raised as the most prevalent food hypersensitivity in the pediatric population. One of the major egg allergens is ovalbumin (OVA) which is the major protein in egg white, comprising 54% of its total protein content. Parthenolide isolated from Tanacetum partheniumb has been used to treat many chronic diseases for many years. In the present report, we present biochemical evidence that parthenolide inhibits the nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activation induced by OVA. Parthenolide also inhibits OVA-induced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression. These data suggest new approaches for the efficient alleviation of the allergic symptoms.

Isoliquiritigenin (ILG) is an active component present in licorice and has been used to treat many chronic diseases for many years. In this study, we present biochemical evidence that ILG inhibits OVA-induced nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activation. ILG also inhibits OVA-induced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression. These data suggest that ILG has anti-allergy effects.

Nitric Oxide Signal Transduction and Its Role in Skin Sensitization

  • Nitric oxide (NO) is a signaling molecule that plays a crucial role in numerous cellular physiological processes. In the skin, NO is produced by keratinocytes, fibroblasts, endothelial cells, and immune cells and is involved in skin functions such as vasodilation, pigmentation, hair growth, wound healing, and immune responses. NO modulates both innate and adaptive immune responses. As a signaling molecule and cytotoxic effector, NO influences the function of immune cells and production of cytokines. NO is a key mediator that protects against or contributes to skin inflammation. Moreover, NO has been implicated in skin sensitization, a process underlying contact dermatitis. It modulates the function of dendritic cells and T cells, thereby affecting the immune response to allergens. NO also plays a role in contact dermatitis by inducing inflammation and tissue damage. NO-related chemicals, such as nitrofatty acids and nitric oxide synthase (NOS) inhibitors, have potential therapeutic applications in skin conditions, including allergic contact dermatitis (ACD) and irritant contact dermatitis (ICD). Further research is required to fully elucidate the therapeutic potential of NO-related chemicals and develop personalized treatment strategies for skin conditions.

  • This review summarizes the cause of allergy and control by lactic acid bacteria. Atopic diseases such as asthma, rhinitis, eczema and food allergy have increased in most industrialized countries of the world during the last 20 years. The reasons for this increase are not clear and different hypotheses have been assessed including increased exposure to sensitizing allergens or decreased stimulation of the immune system during critical periods of development. Probiotic bacteria, which beneficially affect the host by improving its microbial balance, may mediate anti-allergenic effects by immune stimulation. Although more clinical evidences are required, the possible role of specific LAB strains in the prevention of allergic diseases has become more evident. since the role of functional food is important for prevention, it is expected for the more anti-allergy fermented milk products to be on the market.

Detection of a Thermal Stable-Soluble Protein (TSSP) as a Marker of Peanut Adulteration Using a Highly Sensitive Indirect Enzyme-Linked Immunosorbent Assay based on Monoclonal Antibodies

  • Food allergy represents a severe problem for many societies, including sensitive populations, academies, health authorities, and the food industry. Peanut allergy occupies a special place in the food allergy spectrum. To prevent consumption by consumers suffering from a peanut allergy, a rapid and sensitive detection method is essential to identify unintended peanut adulteration in processed foods. In this study, we produced four monoclonal antibodies (MAbs; RO 3A1-12, PB 4C12-10, PB 5F9-23, and PB 6G4-30) specific to thermo-stable and soluble proteins (TSSPs) of peanut and developed an enzyme-linked immunosorbent assay (ELISA) based on the MAbs. Among them, PB 5F9-23 MAb was firmly bound to Ara h 1, and other MAbs strongly reacted to Ara h 3 in the Western blot analysis. An antibody cocktail solution of the MAbs was used to enhance the sensitivity of an indirect ELISA, and the limit of detection of the indirect ELISA based on the antibody cocktail solution was 1 ng/ml and improved compared to the indirect ELISA based on the single MAb (11 ng/ml). The cross-reaction analysis revealed the high specificity of developed MAbs to peanut TSSPs without cross-reaction to other food allergens, including nuts. Subsequently, analyzing processed foods by indirect ELISA, all foods labeled as containing peanuts in the product description were confirmed to be positive. The results indicate that the developed antibodies exhibit high specificity and sensitivity to peanuts and can be used as bio-receptors in immunoassays or biosensors to detect intentional or unintentional adulteration of peanuts in processed foods, particularly heat-processed foods.
