The beneficial effects of lactic acid bacteria (LAB) have been intensively investigated in recent decades with special focus on modulation of the host intestinal microbiota. Numerous discoveries of effective probiotics are driven by a significantly increasing demand for dietary supplements. Consequently, technological advances in the large-scale production and lyophilization are needed by probiotic-related industries for producing probiotic LAB for commercial use. Our study had a dual objective, to determine the optimum growth medium composition and to investigate appropriate cryoprotective additives (CPAs) for Lactobacillus salivarius, and compare its responses with other Lactobacillus species. The one-factor-at-a-time method and central composite design were applied to determine the optimal medium composition for L. salivarius cultivation. The following composition of the medium was established (per liter): 21.64 g maltose, 85 g yeast extract, 1.21 ml Tween 80, 6 g sodium acetate, $0.2g\;MgSO_4{\cdot}7H_2O$, $0.02g\;MnSO_4{\cdot}H_2O$, $1g\;K_2HPO_4$, $1.5g\;KH_2PO_4$, $0.01g\;FeSO_4{\cdot}7H_2O$, and 1 g sodium citrate. A cryoprotective additive combination comprising 10% (w/v) skim milk and 10% (w/v) sucrose supplemented with 2.5% (w/v) sodium glutamate was selected for L. salivarius, and its effectiveness was confirmed using culture-independent methods in the freeze-dried cells of the Lactobacillus strains. In conclusion, the optimized medium enhanced the species-specific cultivation of L. salivarius. On the other hand, the cryoprotective effects of the selected CPA mixture may also be dependent on the bacterial strain. This study highlights the necessity for precise and advanced processing techniques for large-scale production of probiotics in the food and feed industries.
The effects of sucrose fatty acid ester (SE, 1% w/w) and glycerin (GL, 1% v/w) additions, storage temperature$(0,\;20\;and\;70^{\circ}C)$, and time $(0{\sim}6\;day)$ on texture properties, hardness(H), cohesiveness(O), chewiness(C) and rheological property(R) of Baikseolgi were studied. The H of Baikseolgi increased sharply in the early stage of storage at 0 and $20^{\circ}C$, while increased gently at $70^{\circ}C$ with increasing storage time. After 6 days of storage, the H of Baikseolgi at $20^{\circ}C$ had a little lower than that at $0^{\circ}C$. However, the H of Baikseolgi at $70^{\circ}C$ was 10.7% of that at $0^{\circ}C$. The addition of GL had greater effect on the reduction of H than that of SE. The H of control, SE and GL additions were 336, 216 and $$174\;g_f, respectively, after 6 days at $70^{\circ}C$. The O of Baikseolgi at $70^{\circ}C$ were higher than those at $0^{\circ}C$. The O of GL added Baikseolgi had the highest value and the second and the third were SE added and control, respectively. The O of Baikseolgi decreased with increasing storage time. The C of Baikseolgi of increased with increasing storage time, which had similar curve patterns to the H of Baikseolgi. Instantaneous stress and equilibrium stress of Baikseolgi decreased with increasing storage temperature. The affection of viscous element increased and that of elastic element decreased with increasing storage temperature.
As mulberry fruit's functionalities are known to the media, the interest in promoting of consumption and processed products is increasing. But there is no C3G(Cyanidin-3-glucoside) data based on the stability of the pigment during processing. To solve this problems, and to expand the use of mulberry fruit, processing methods was developed for mulberry fruit juice improved cyanidin-3-glucoside(C3G) stability. The results obtained are summarized as follows. The food additive citric acid with 0.3 % improved C3G content and antioxidant ability in the treatment of mulberry fruit and sucrose the ratio of 50 % : 50 %(w/w). In the case of the addition of oligosaccharides, citric acid decreased antioxidant ability. Xylitol treatment showed up the lowest of C3 content, but by the addition of citric acid improved the pigment content and antioxidant activity of the mulberry fruit juice. Addition of citric acid was more affected C3G stability than containers (clear glass bottles, brown glass bottles, aluminum foil, green glass bottle, translucent glass bottles). In the processing of mulberry juice, 3 minutes blanching treatment using microwave dropped C3G content somewhat. Therefore, using this method is not recommended. However in the antioxidant ability, microwave blanching showed a stabilizing effect compared to the other treatments.
This study was designed to determine the mutagenic potential of hydrolyzed glycomacropeptide (GMP) powder (hereafter referred to as 23%-GNANA; product name: HELICOBACTROL-23) in a micronucleus test using bone marrow in ICR mice. Three experimental groups were used: a 3-step concentration group, with a maximum concentration of 2,000 mg/kg, and other sequentially two-fold lower concentrations, a negative control group, and a positive control group. The test material was administered for 2 d to observe the frequency of micronucleus formation up to 48 h after the test material was absorbed by the body. When the polychromatic erythrocyte (PCE) content of erythrocytes was compared, no significant differences were noted between the negative control group and the test group (p<0.05). Similarly, when the average numbers of micronucleated PCE (MNPCE) in 2,000 PCE per animal were compared, no significant difference was observed between the negative control group and the test group (p<0.05). No dose-response relationship with regard to the concentration of the test material administered was noted. These results allow us to conclude that hydrolyzed whey protein powder does not cause formation of micronuclei in mouse bone marrow cells under the applied conditions. In this study, the average frequency of micronucleus formation in PCE was significantly higher in the positive control group compared with the negative control group; thus, the test conditions were appropriate for detecting the frequency of micronucleus formation induced by the test material. In conclusion, the safety of 23%-GNANA test substance was verified in an in vivo micronucleus test in mice, conducted before the registration of HELICOBACTROL-23 as a food additive.
Dietary flavonoids have antioxidant and antitumor promoting effects. Rhus verniciflua Stokes (RVS) is a flavonoid-rich herbal medicine and has long been used as a food additive and an antitumor agent in Korea. Previous study demonstrated that a purified flavonoid fraction prepared from RVS, herein named RCMF (the RVS chloroform-methanol fraction), exhibited growth inhibition and induced apoptosis in human osteosarcoma(HOS) cells. This study evaluated if p53-mediated pathway is associated with the RCMF-induced apoptosis in HOS cells. RCMF was shown to be capable of inducing apoptosis of the cells, as expected, and transparently increased p53 expression in the cells. However, the RCMF-induced cytotoxicity was suppressed by transfecting the cells with antisense p53 oligonucleotide, which also inhibited the decrease of Bcl-2 and the increase of Bax in mitochondria, and the release of cytochrome c into cytosol. This finding suggests that p53-mediated mitochondrial stress is required for RCMF-induced apoptosis in HOS cells.
This study was to improve the quality of Kochuzang and utilize red pepper seed. Kochuzang were prepared with the addition of red pepper seed and garlic (2%), compared the changes in the various chemical components and enzyme activity during the aging period of Kochuzang, and also organoleptic values of the products. Enzyme activities of liquefying and saccharogenic amylase, protease and lipase were increased by addition of garlic pulp and the suvival activities of enzyme except liquefying amylase were lasted high the late period of aging. Also the addition of red pepper seed was effective in maintaining the enzyme activities Change of titration acidity and pH of kochuzang were little when red pepper seed was added, but in case of a garlic additive it showed no difference at the late period of aging. Total nitrogen and amino nitrogen were increased by the addition of red pepper seed or garlic until the late period of aging, and ammonia nitrogen also increased during the middle period of againg, but showed no difference at the late period. Alcohol content was decreased by the addition of garlic or red pepper seed. but crude lipid was increased by the audition of red pepper seed. but crude lipid was increased by the addition of red pepper seed. Generally, taste, flavor and color of garlic added group were superior to the non-garlic added group for the products which aged for 10 weeks. Therefore, The quality of Barley Kochuazng may be improved by adding 2% garlic to the whole red pepper.
Decay during soybean sprout culture detracts their quality as well as increases their production costs. This study was done to determine the effects of acetic and propionic acids on growth and morphological characters of the sprouts. The soybean seeds of 3 cultivars (cv. Eunhakong, Pungsannamulkong and Orialtae) imbibed for 2 minutes at their different concentrations (0, 0.1 and 0.2%) were soaked for 6 hours in 4 ppm BA solution after the first 5.5 hour water imbibition and 0.5 hour aeration, and cultured at $20^{\circ}C$. On the 6th day, harvested soybean sprouts were classified into 4 categories on the base of hypocotyl length;>7cm, 4 to 7cm,<4cm and not germinated, and their morphological characters, fresh weights, lesion spots on cotyledons were measured or analyzed. The stronger concentration the higher rate of longer than 4cm although there was no significant difference between the two acids. Pungsannamulkong showed the longest hypocotyls but Eunhakong did the thickest ones, and Orialtae did the greatest total fresh weight, in which all the characters were not influenced by the two acids and their concentrations. Lesion spots on the cotyledons were equal to the two acids although less in their treatments than in no treatment. Utilization of propionic acid was more desirable Dan acetic acid through their treatment cost analysis.
The solution polymerization was conducted to synthesize pressure sensitive adhesive for polarizer film using acrylic monomers. 2-Ethylhexylacrylate, butylacrylate, acrylic acid were used as acrylic monomers. The ratio was 2-ethylliexylacrylate:butylacrylate:acrylic acid=25:50:3.6 by reflecting $-40^{\circ}C$ of glass transition temperature in the pressure sensitive adhesive. When 1 wt% of coupling agent was added to the polymerized pressure sensitive adhesive, the light transmissivity was significantly increased. This result is due to the enhancement of adhesive power against liquid crystal cell by Si-O bond of coupling agents. Cross-linking agent was added by 0.5, 1.0, and 1.5 wt% with respect to the synthesized polymer. Initial tackiness decreased, while cohesion increased with increasing crosslinking agent. In the analysis of contact angle, the increase of crosslinking agents yielded the enhancement of surface energy, resulting in the decrease of contact angle. From the measurement of heat resistance, the acrylic pressure sensitive adhesive showed excellent heat resistance regardless of change in temperature and contents in crosslinking agent. In the observation of a cutting plane, the increased crosslinking agent represented a smoother and cleaner section. Comprehensively, the optimum additive amount of crosslinking agent was determined to be 1.0 wt% to monomer.
As a part of a study on the effective use of cultured oyster, oyster powder-added soybean pastes were prepared and its characteristics were examined. As the concentration of oyster powder added to commercial soybean paste was increased, the moisture level decreased, the crude and amino nitrogen content increased with no change in the salinity and titratable acidities. Also, the color of the soybean paste changed to a darker color as the concentration of oyster powder added to commercial soybean paste increased. Judging from the results of the above experiment and sensory evaluation, the optimal additive concentration of oyster powder to commercial soybean paste was revealed as 9%. The total amino acid content of oyster powder-added soybean paste was 2% higher than that of commercial soybean paste. The ratio of calcium and phosphorus of oyster powder-added soybean paste was about 1:2-2:1, an improvement in calcium absorption as compared to commercial soybean paste. Functional fatty acids, such as 20:5n-3 and 22:6n-3, were not detected in commercial soybean paste but detected in oyster powder-added soybean paste. The major free amino acids in oyster powder-added soybean paste were glutamic acid and aspartic acids. According to the above results, the nutritional, functional and taste properties of commercial soybean paste were improved while reducing preparation time of the housewife.
Anethole, eugenol, isoeugenol, safrole and isosafrole are ingredients of refined oils which are obtained from some plants and their chemical structures are very similar. They are mainly used as a flavoring agent, food additive, dental analgesics and for many drugs. But, there is no report about their effect on the intestinal motility. The result of examining the effect, potency and mode of action of anethole, eugenol, isoeugenol, safrole and isosafrole on motility of isolated rabbit jejunum segment, are as follows : 1. Single administration of anethole, eugenol, isoeugenol, safrole and isosafrole showed the inhibition of motility of isolated rabbit jejunum segment, degree of which was various. The $pD_2$ values of isoeugenol, isosafrole, eugenol, safrole and anethole in isolated rabbit jejunum segment were 4.22, 4.18, 4.17, 4.15 and 3.82 (in the descending order of potency). 2. The contracted rabbit jejunum segment : by carbachol, pilocarpine, barium chloride and histarmine were relaxed by five essential oil. 3. The relaxed rabbit jejunum segment by anethole was not recovered by carbachol, pilocarpine, barium chloride and histamine. The relaxed rabbit jejunum segment by eugenol, isoeugenol, safrole and isosafrole were recovered by carbachol, pilocarpine and barium chloride but partially recovered by histamine. 4. Judging from the facts above, it is thought that five essential oil are inhibit the motility of isolated rabbit jejunum segment by neurotropic and musculotropic action.
