Combined Effects of Groundwater Abstraction and Irrigation Reservoir on Streamflow (지하수 이용과 농업용 저수지가 하천유량에 미치는 복합 영향)

  • In this study, a watershed-based surface-water and groundwater integrated model, SWAT-MODFLOW was used to evaluate streamflow depletion induced by groundwater withdrawals and irrigation reservoirs for the Juksan-cheon watershed in South Korea. The streamflow responses to groundwater pumping and irrigation reservoirs were simulated under several different scenarios. The scenarios were (1) current pumping well withdrawals with reservoirs; (2) current pumping well withdrawals without reservoirs; (3) no pumping well withdrawals with reservoirs; (4) no pumping well withdrawals without reservoirs (natural condition). The simulated results indicated that the effects of groundwater pumping on streamflow depletion are a little more significant than those of irrigation reservoirs. Particularly, the groundwater withdrawals with irrigation reservoirs at current status (scenario 1) has induced the decrease of more than 20% in drought flow against the natural condition (scenario 4) at the outlet of the watershed. The specific drought flows through the main stream of Juksan-cheon watershed were simulated in order to assess the irrigation effects on downstream flows. It was found out that the specific drought flows are increasing as the distance from the reservoir increases due to the accumulation of the return flows to stream.

  • In this paper, the reasons of damages and the case study are review in which bridge pier with debris accumulation, and safety Influence factors by debris around the bridge piers are review. Also experiment Is conducted for the characteristic of flow around piers by different area and angle of debris and the basic characteristics was review for safe design of bridge and embankments. As result of review of several standards of design, hydraulic structure's freeboard is simply decided by discharge, so it needs more detail standards. And as result of experiment, in the case of that water depth is deep and velocity is slow, variation of water depth Is more increase as increasing of debris. Therefore the variation regime of flow characteristics like velocity and water depth by debris is more large in the stream of small or medium size, which streams have large water depth and slow velocity so Froude Number Is expressed as small in the flood. Also when Froude Number is about 0.5, the water elevation is over freeboard in the standard if the debris over 20%. Therefore when hydraulic structure is constructed in the stream of small or medium size, it need to conduct more detail experiments about influence of debris, distribution of velocity and variation of elevation, and than the more safe freeboard will be presented using the experimental results.

  • Sasa quelpaertensis Nakai (Korean name, Jeju-Joritdae) is one of the most abundant plants on Mt. Halla, Jeju Island, and it has long been used in traditional medicines. Recent studies have reported it as possessing various beneficial functions, including anti-inflammatory, anti-diabetic, anti-hypertension, anti-gastritis, anti-oxidant, and anti-cancer effects. However, the molecular mechanisms of its anti-cancer activity have not been clearly elucidated. In this study, we investigated the anti-cancer effects and mechanism of S. quelpaertensis on human colon cancer HT-29 cells. Cell growth inhibition by S. quelpaertensis was determined by MTT assay. Apoptosis was performed by DNA fragmentation, flow cytometry with propidium iodide staining (PI), and reverse transcription-polymerase chain reaction (RT-PCR) to confirm the anti-apoptotic factors, such as inhibitor of apoptosis (IAP) family members. $NO^{\bullet}$ production was determined by Griess assay. S. quelpaertensis treatment resulted in the time- and dose-dependent inhibition of the cell viability of HT-29 cells by inducing apoptosis, as evidenced by the accumulation of the sub-G1 cell population stained by PI, as well as the ladder-like DNA fragmentation in a dose-dependent manner. S. quelpaertensis-inducing apoptosis was accompanied by the induction of S cell cycle arrests, increasing $NO^{\bullet}$ concentrations, and the down-regulation of IAPs, including X-chromosome-linked IAP (XIAP), cellular IAP-1 (cIAP-1), cIAP-2, and survivin. Taken together, these findings have important implications for future clinical developments of S. quelpaertensis in colon cancer treatment.

  • To determine the cause of the population decline in Gobiobotia naktongensis, substrate preference and burying behaviour were investigated in this study. In general, the species was shown to prefer a substrate size of 1 mm or less, depending on the flow. In addition, the burying depth varied according to the size of the fish and increased with a decrease in water temperature. Our findings showed that the main cause of the population reduction was the physical changes in the substrate structure due to the dams or barrages construction. Notably, the accumulation of silt and mud in the substrate upon the formation of an upstream lentic water region for structural construction and bed armouring caused by scouring and reduced downstream inflow of fine sediment were deterministic in the fish habitat changes, causing problems in burying. As sand substrate structure is critical for the survival and inhabitation of psammophilous species, efficient strategies should be developed with proper habitat management to reduce the anthropogenic damage

A Study of the Changes of Breast Uptake in Menstrual Cycle on 18F-FDG PET/CT (월경 주기에 따른 18F-FDG PET/CT에서 유방 섭취 변화에 관한 고찰)

  • $^{18}F-FDG$ PET/CT has been known a useful modality to diagnose high-glucose-using cells such as cancer cells by glucose metabolism of FDG. Mainly, FDG takes on cancer and inflammatory cells; however, there have been FDG uptakes on normal tissues by individual physiological characteristics, occasionally. Especially, in fertile females, unusual FDG uptake of breast changes as the menstrual cycle, and disturb diagnosis. Therefore, the study aimed to evaluate the change of breast FDG uptake in menstrual cycle on $^{18}F-FDG$ PET/CT. 160 females ($34{\pm}3.5$ years old) who do not undergo a gynecologic anamnesis and have regular menstrual cycle over the previous 6 months were examined. They were divided 4 groups (each 40 patients) as flow phase, proliferative phase, ovulatory phase and secretory phase using Pregnancy Calculator 0.14. and history taking. Discovery Ste (GE Healthcare, Milwaukee, Mi, USA) was used a s PET/CT. We analyzed SUVs on accumulated region on breast, and 3 nuclear medicine specialists did the Blind test. SUVs on the Breast were flow phase ($1.64{\pm}0.25$), proliferative phase ($0.93{\pm}0.28$), ovulatory phase ($1.66{\pm}0.26$) and secretory phase ($1.77{\pm}0.28$). It showed high uptake value in secretory, flow phase and ovulatory phase (p<0.05). In gross analysis, the accumulation of breast was divided into 3 grades as comparing with lung and liver. The breast's uptake was equal to lung (Grade I); between lung and liver (Grade II); equal to or greater than liver (Grade III). The results showed high uptake value in secretory, flow phase and ovulatory phase (p<0.05). In fertile females, FDG uptake of breast changed as menstrual cycle, and it available to diagnose breast disease. Therefore, we consider reducing false-negative finding of breast disease, by doing examination on appropriate period through history taking about individual menstrual cycle.

A Case Study for the Determination of Time Distribution of Frequency Based Rainfall (확률강우의 적정시간분포 결정에 관한 연구)

  • In recent, the heavy rainfall is frequently occurred and the damage tends to be increased. So, more careful hydrologic analysis is required for the designs of the hydraulic or disaster prevention structures. The time distribution of a rainfall is one of the important factors for the estimation of peak flow in hydrologic and hydraulic designs. This study is to suggest a methodology for the estimation of a rainfall time distribution which can reflect the meteorologic and topographical characteristics of Daejeon area. We collect the 34 years' rainfall data recorded in the range of 1969 to 2002 for Daejeon area and we performed the rainfall analysis with the data in between May and October of each year. According to the Huff method, the collected data corresponds to the first quartile which the rainfall is concentrated in the primary stage but the suggested method shows the different rainfall distribution with the Huff method in time. The reason is that the Huff method determines the quartile in each storm event while the suggested one determines it by estimating the dimensionless distribution of rainfall in duration after the accumulation of rainfall in time. The rainfall distributions estimated by two methodologies were applied to the Gabcheon basin in Daejeon area for the estimation of flood flow. Here we use the SCS method for the effective rainfall and unit hydrograph for the flood discharge. As the results, the peak flow for 24-hour of 100-year frequency was estimated as a $3421.20m^3/sec$ by the Huff method and $3493.38m^3/sec$ by the suggested one. We can see the difference of $72.18m^3/sec$ in between two methods and thus we may carefully determine the rainfall time distribution and compute the effective rainfall for the estimation of the peak flow.

  • Mammary epithelial cells contain a subpopulation of cells with a large proliferativ potential which are responsible for the maintenance of glandular cellularity and are the progenitor cells of mammary cancer. These clonogens give rise to multicellular clonal alveolar or ductal units(AU or DU) on transplantation and hormonal stimulation. To isolate putative mammary clonogens, enzymatically monodispersed rat mammary epithelial cells from organoid cultures and from intact glands are sorted by flow cytometry according to their affinity for FITC labeled peanut lectin(PNA) and PE labeled anti-Thy-1.1 antibody(Thy-1.1) into four subpopulations : cells negative to both PNA and Thy-1.1(B-), PNA+cells, Thy-1.1+cells, and cells positive to both reagents(B+). The in vivo transplantation assays indicate that the clonogenic fractions of PNA+cells from out-growths of organoids in primary cultures for three days in complete hormone medium(CHM) are significantly higher than those of cells from other subpopulations derived from cultrues or from intact glands. Extracellular matrix(ECM) is a complex of several proteins that regulated cell function ; its role in cell growth and differentiation and tissue-specific gene expression. It can act as a positive as well as a negative regulator of cellular differentiation depending on the cell type and the genes studied. Regulation by ECM is closely interrelated with the action of other regulators of cellular function, such as growth factors and hormones. Matrigel supports the growth and development of several different multicellular colonies from mammary organoids and from monodispersed epithelial cells in culture. Several types of colonies are observed including stellate colonies, duct-like structures, two- and three-dimensional web structures, squamous organoids, and lobulo-duct colonies. Organoids have the greatest proliferative potential and formation of multi-cellular structures. Phase contrast micrographs demonstrate extensive intracellular lipid accumulation within the web structures and some of duct-like colonies. At the immunocytochemical and electron micrograph level, casein proteins are predominantly localized near the apical surface of the cells or in the lumen of duct-like or lobulo-duct colonies. Squamous colonies are comprised of several layers of squamous epithelium surrounding keratin pearls as is typical fo squamous metaplasia(SM). All-trans retinoic acid(RA) inhibits the growth of SM. The frequency of lobulo-ductal colony formation increased with the augmentation of RA concentration in these culture conditions. The current study models could provide powerful tools not only for understanding cell growth and differentiation of epithelial cells, but also for the isolation and characterization of mammary clonogenic stem cells.

Evaluation of Removal Efficiencies of Heavy Metals Using Brown Seaweed Biosorbent Under Different Biosorption Systems (폐미역을 이용한 생물흡착 시스템별 중금속 제거 효율 평가)

  • BACKGROUND: Heavy-metal pollution represents an important environmental problem due to the toxic effects of metals, and their accumulation throughout the food chain leads to serious ecological and health problems. METHODS AND RESULTS: Optimum conditions in continuous-flow stirred tank reactor (CSTR) and packedbed column contactor (PBCC) using brown seaweed biosorbent were investigated. Under optimum conditions from both lab-scale biosorbent systems, removal efficiency of copper (Cu) in a large-scale PBCC system was investigated. Removal capacity of Cu using brown seaweed biosorbent in a lab-scale CSTR system was higher than that in a lab-scale PBCC system. On the other hand, over 48 L/day of flow rate in Cu solution, removal efficiency of Cu in a lab-scale PBCC system was higher than that in a lab-scale CSTR system. Optimum flow rate of Cu was 24 L/day, optimum Cu solution concentration was 100 mg/L. Removal capacity of Cu at different stages was higher in the order of double column biosorption system > single column biosorption system. Under different heavy metals, removal capacities of heavy metal were higher in the order of Pb > Cr > Ni > Mn ${\geq}$ Cu ${\geq}$ Cd ${\fallingdotseq}$ Zn ${\geq}$ Co. Removal capacity of Cu was 138 L in a large-scale PBCC system. Removal capacity of Cu a large-scale PBCC system was similar with in a lab-scale PBCC system. CONCLUSION(s): Therefore, PBCC system using brown seaweed biosorbent was suitable for treating heavy metal wastewater.

Anti-obesity effect of EGCG and glucosamine-6-phosphate through decreased expression of genes related to adipogenesis and cell cycle arrest in 3T3-L1 adipocytes (Adipogenesis관련 유전자발현감소와 Cell Cycle Arrest를 통한 EGCG와 Glucosamine-6-Phosphate의 Anti-Obesity 효과)

  • Purpose: Several studies have proven that EGCG, the primary green tea catechin, and glucosamine-6-phosphate (PGlc) reduce triglyceride contents in 3T3-L1 adipocytes. The objective of this study is to evaluate the combination effect of EGCG and PGlc on decline of accumulated fat in differentiated 3T3-L1 adipocytes. Methods: EGCG and PGlc were administered for 6 day for differentiation of 3T3-L1 adipocytes. Cell viability was measured using the CCK assay kit. In addition, TG accumulation in culture 3T3-L1 adipocytes was investigated by Oil Red O staining. We examined the expres-sion level of several genes and proteins associated with adipogenesis and lipolysis using real-time RT-PCR and Western blot analysis. A flow cytometer Calibar was used to assess the effect of EGCG and PGluco on cell-cycle progression of differentiating 3T3-L1 cells. Results: Intracelluar lipid accumulation was significantly decreased by combination treatment with EGCG $60{\mu}M$ and PGlc $200{\mu}g/m$ compared with control and EGCG treatment alone. In addition, use of combination treatment resulted in directly decreased expression of $PPAR{\gamma}$, $C/EBP{\alpha}$, and SREBP1. In addition, it inhibited adipocyte differentiation and adipogenesis through downstream regulation of adipogenic target genes such as FAS, ACSL1, and LPL, and the inhibitory action of EGCG and PGlc was found to inhibit the mitotic clonal expansion (MCE) process as evidenced by impaired cell cycle entry into S phase and the S to G2/M phase transition of confluent cells and levels of cell cycle regulating proteins such as cyclin A and CDK2. Conclusion: Combination treatment of EGCG and PGlc inhibited adipocyte differentiation through decreased expression of genes related to adipogenesis and adipogenic and cell cycle arrest in early stage of adipocyte differentiation.
