Effect of Food and Salinity on Larval Growth and Survival of the River Puffer, Takifugu obscurus (황복, Takifugu obscurus의 초기 발달 동안 성장 및 생존에 있어 먹이와 염분의 효과)

  • In the present study, we investigated the effects of food and salinity on growth and survival of river puffer, Takifugu obscurus offsprings in indoor land-based tank during the early development. In the food experiment, the river puffer larvae (TL 8.7$\pm$0.1 mm, BW 20.0$\pm$3.2 mg) were fed with tubificid, Limnodrilus gotoi, water flea, Daphnia carinata, mysid, Neomysis awatschensis, Manila clam, Ruditapes philippinarum and artificial food together Artemia nauplii for 30 days, and the growth and the survival of larvae were investigated. In the salinity experiment, the river puffer larvae and juvenile at three stages (Stage I: TL 5.8$\pm$0.4 mm; Stage II: TL 12.0$\pm$0.2 mm, Stage III: TL 44.5$\pm$0.7 mm) were supplied with Artemia nauplii, water flea and/or artificial food and were reared in 0, 10, 20 and 30 psu at respective stages. The results shows that the growth rates of larvae feeding with artificial food and tubificid together Artemia nauplii were higher than those of larvae in other groups, but the survival rates of larvae feeding with water flea and mysid together Artemia nauplii were higher than those of larvae in other groups. In salinity, the results shows that the growth and survival rates of offsprings were significantly higher in 10∼20 psu than those in 0 and 30 psu at Stage I and n. At Stage III, the growth and the survival rates were the highest in 20 psu, but the lowest in 0 psu. However, it should be noted that the rates in 0 and 30 psu were significantly enhanced at Stage III in comparison with those at Stage I and II. Therefore, it is concluded that river puffer, T. obscurus is euryhaline marine species, which can normally grow and live at salinities from 0 to 30 psu, and that a combinative supplement with Artemia nauplii and water flea, D. carinata may confer an advantage on growth and survival of the river puffer offsprings in indoor land-based tank.

Reproduction of Water Flea by the Culture Conditions (배양조건에 따른 물벼룩의 개체생산 특성)

  • Four species of water fleas (Daphnia magna, Daphnia pulex, Daphnia galeata, and Moina macrocopa) were examined for the clarification of their reproduction with culture conditions. The reproduction tests of water flea by the culture conditions were carried out. For the comparison of the reproduction rate, five media (manure-soil medium, DIN medium, M4 medium, EPA medium, fertilizer medium) were applied to determine the best medium. Daphnia magna, Daphnia pulex, and Moina macrocopa were appeared the best reproduction in the manure-soil medium at $20^{\circ}C.$ The lifespan and young reproduction were better in manure-soil medium than the others. But Daphnia galeata was lived for 34 days in the fertilizer medium at $20^{\circ}C.$ The culture of Daphnia galeata was difficult to rear than the other species. In the current study, the microcystin of Microcystis sp. did not particularly affect on the survival of water fleas. But the lifespan was short and the reproduction rate was low. Therefore water flea have a preference for Scenedesmus sp. than Microcystis sp. On the condition of the feeding Scenedesmus sp., all examined water fleas appeared to have the longest lifespan and the most young water fleas produced at any medium and temperature as compared with the feeding the Microcystis sp. For the culture temperature, the lifespan was longer on $20^{\circ}C$ than $15^{\circ}C$.

Time-dependent Effects of Bisphenol Analogs on Ecdysteroid Pathway Related Genes in the Brackish Water Flea Diaphanosoma celebensis (Bisphenol 구조 유사체가 기수산 물벼룩 Ecdysteroid 경로에 미치는 영향)

  • Bisphenol A is a representative endocrine disruptor and continuously detected in aquatic environment due to wide use, resulting in adverse effects on growth, development, and reproduction in diverse organisms as well as human. Structural analogs have been developed to substitute BPA are also suspected to have endocrine disrupting effects. In the present study, the time-dependent expression patterns of ecdysteroid synthesis (nvd, cyp314a1), receptors (EcRA, EcRB, USP, ERR), and downstream signaling pathway - related genes (HR3, E75, Vtg, VtgR) were investigated using quantitative real time reverse transcription polymerase chain reaction (qRT-PCR) in the brackish water flea Diaphanosoma celebensis exposed to Bisphenol analogs (BPs; BPA, BPF, and BPS) for 6, 12, and 24 h. As results, the expression of nvd, cyp314a1, EcRs, USP, ERR and E75 mRNA was upregulated at 6 h exposure to BPF, which is earlier than BPA and BPS (12 h). On the other hand, HR3, E75 and VtgR mRNA levels were elevated at 6 h earlier at BPS and BPF than at BPA (12 h), but Vtg mRNA level was slightly changed within 24 h. These findings suggest that like BPA, BPF and BPS can also modulate the transcription of ecdysteroid pathway - related genes with different mechanisms, and have a potential as endocrine disruptors. This study will provide a better understanding the molecular mode of action of bisphenols on ecdysteroid pathway in the brackish water flea.

Toxicity assessment of food additive(E171) in aquatic environments (식품첨가물 E171이 수생물에 미치는 독성 평가)

  • E171, a mixture of titanium dioxide, has been widely used as a food additive due to its whitening effect and low toxicity. However, it has been proven that E171 is no longer safe for public health. So far, there are insufficient studies on the toxic effects of E171 on organisms especially using standardized test methods. In this study, toxicity assessments of E171 to two aquatic species, water flea (Daphnia magna) and zebrafish (Danio rerio), were performed using modified standardized test methods based on the physicochemical properties of E171. The hydrodynamic diameter, polydispersity index, and turbiscan stability index (TSI) were measured to ensure the dispersion stability of E171 in exposure media during the test period. The EC50 for immobilization of water flea was 141.7 mg L-1 while zebrafish was not affected until 100 mg L-1 of E171. Measurements of reactive oxygen species (ROS) and antioxidant enzyme activities confirmed that E171 induced oxidative stress, leading to the activation of superoxide dismutase and catalase in both water flea and zebrafish, although the expression of antioxidant enzyme genes differed between species. These results suggested the potential risk of E171 to aquatic organisms and provided toxicological insights into the impacts of E171 on the environment.

Acute Toxicity Test of Agricultural Chemicals to Water Fleas (물벼룩을 이용한 농약의 급성 독성에 관한 연구)

  • There are concerns that chemical residues could harm the consumer on the environment, although 50 to 80% of the crops would be destroyed by pests and others without agrochemicals. Environmental fate and ecotoxicity studies are usually carried out to assess the impact on the human and the environment. A comparision of the Daphnia magnia and Simocephalus mixtus toxicity was performed to study the relative sensitivities and discrimination abilities to agriculture chemicals. The species of Simocephalus mixtus was more sensitive to agriculture chemicals than Daphnia magnia. Simocephalus mixtus was approved to be a water flea in determining insecticide and pesticide toxicity by heart-beat rate in a consistency and repeatability. The order of acute toxicity to water flea Daphnia magnia for ecotoxicity test was carbaryl>benomyl>amtirole with both Daphnia magnia and Simocephalus mixtus. The heartbeat pattern after the exposure to agrochemicals was different from that of exposure to heavy metals. Agrochemical leathal concentration test with heartbeat rate measurement was found to be more appropriate than inhibition concentration test with respect to toxicological endpoint.

Toxicity Assessment of PFOA and PFOS Using Freshwater Flea Hyalella azteca (담수산 물벼룩인 Moina macrocopa를 이용한 과불화화합물 PFOA와 PFOS의 독성평가)

  • Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) are fully flurorinated organic compounds which are highly persistent in environment and accumulated in organism. These chemicals are released to the environment at their manufacture, during their use in industrial and consumer applications and from disposal after their use. The purpose of this study was to determine the effects of PFOA and PFOS on the freshwater flea (Moina macrocopa). Acute toxicity test and chronic toxicity test were performed for 2 days and 10 days, respectively. Acute toxicity was assessed on the basis of mortality, while chronic toxicity was assessed by fecundity as well as mortality. The acute toxicity studies on PFOA and PFOS showed that the values of $LC_{50}$ were $73.9\;mg/L\;and\;27.7\;mg/L$ respectively. In the chronic toxicity test, fecundity was reduced significantly at 24.1 mg/L of PFOA and 9.3 mg/L of PFOS, respectively. Conclusively, the results of this work suggest that Moina macrocopa could be a suitable model organism for screening and assessing of environmental pollutants in water.

On the Rat-Fleas in Ducksom and Kwangnaru Areas of Seoul City (서울시 뚝섬, 광나루 일대의 쥐벼룩에 대하여)

  • A rat-fleas survey was carried out in Ducksonm and Kwangnaru areas of Seoul city from Janurary to December 1955 . In this survey, 1,204 redents were trapped alive and 740 of which being Rattus norvegicus, 457 Apodemus agrarius, 5 Eutamiias asiaticus and 2 shrewes ((crocidura rusila). On these rodents 270 fleas were collected and identified thus : Ceratopylidae : Monopsyllus anisus (Rothschild 1908) ···200 Leptosylla segnis(Schonherr, 1832)···1 Hystrichopsyllidae : Ctenophthalmus congener, Rothschild ···43 Neopsylla bidentiformis (Wagner, 1893)···11 Stenoponia sidimi Marikovsky, 1936···9 Pulcidae : Xenopsylla cheopis (Rothschild, 1903)···6 On the average 8.5% of the rodents were infestated with fleas. The infestation rate showed a marked varying degree in a year and three high incidence peaks in Spring (10.3%), Summer (11.4%-11.8%) and Winter (13.7%). The average number of fleas per rat (flea-index) was 0.22 for the whole year with a maximum of 0.41 in December and a minimum of 0.03 in February. Of all, the most prevalent species are Monopsyllus anisus (74.1%) and secondly Ctenophthalmus congener (15.9%). both species occupied 90% of all. Xenopsylla cheopis, a known vector of plague, was found very rare in our survey and only appeared in Winter. The striking lower incidence of flea-index compared with previous data (6.6 by Nagahana, 1931-1936) appears to be due to the nationwide application of DDT and BHC insecticides since 1945.

Relative Effectiveness of Replacement Diets for Mass Production of the Brackish Water Flea Diaphanosoma celebensis (기수산 물벼룩(Diaphanosoma celebensis)의 대량생산을 위한 대체 먹이원의 효과)

  • This study examined the effect of formulated diets (composed of condensed freshwater $Chlorella$ or $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil sources derived from baker's yeast) on the population growth and fatty acid composition of the brackish water flea ($Diaphanosoma$ $celebensis$) to determine diet effectiveness for mass production. Results indicated that the highest maximum density, fecundity, and population specific growth rate (SGR) of the flea were observed using a diet of $Tetraselmis$ $suecica$ (control). However, results using condensed freshwater $Chlorella$ and the mixed treatment of $Schizochytrium$ sp.+$Crypthecodinium$ sp.+EPA oil did not significantly differ from the control treatment ($P$>0.05). Thus, $T.$ $suecica$ could be replaced with a combination diet of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil in condensed freshwater $Chlorella$ without growth interruption of $D.$ $celebensis$. As the percentage of combination diet (mix of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil) in condensed freshwater $Chlorella$ increased, the level of ARA, EPA, DHA, and n-3 HUFA in $D.$ $celebensis$ also increased. However, the population density of $D.$ $celebensis$ sharply decreased with the use of treated supplements C5M5 (CHL [50%] and $Crypthecodinium$ sp.+ $Schizochytrium$ sp. + EPA oil [MIX 50%]). The optimum proportions of CHL and MIX for $D.$ $celebensis$ within the mixture were 70% and 30%, respectively. Based on the results, the combination of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil in condensed freshwater $Chlorella$ appears to be a stable and effective food source for mass production of $D.$ $celebensis$.

Economic Injury Level of the Striped Cabbage Flea Beetle, Phyllotreta striolata (Coleoptera: Chrysomelidae), on Chinese Cabbage (시설배추에서 벼룩잎벌레의 경제적 피해수준 설정)

  • This study was conducted to determine the economic injury levels and control thresholds for the striped cabbage flea beetle, Phyllotreta striolata (Coleoptera: Chrysomelidae), on Chinese cabbage at two different planting times. The number of inoculated adults per 10 cabbages was 0, 2, 4, 8, and 16 at the early developmental stage of the cabbage5 days after planting) and 0, 10, 20, 30, and 40 at the middle developmental stage (30 days after planting). Damages to the leaves at the first inoculation were 2.5-21.1% and at the second inoculation were 1.8-26.3% after harvesting. The linear relationships between population density and yield reduction were as follows: Y = 1.3475X + 2.135 ($R^2$ = 0.8699) at the early developmental stage and Y = 0.703X - 1.78 ($R^2$ = 0.966) at the middle developmental stage. On the basis of these results, the economic injury levels caused 5% loss of yield; there were 2.1 adults per 10 Chinese cabbage at the early developmental stage and 9.6 adults per 10 Chinese cabbage at the middle developmental stage.
