Purification and Characterization of Lipoxygenase from Melania Snail (다슬기에서 추출한 Lipoxygenase의 정제와 특성)

  • Melania snail(Semisulcopira bensoni) is used as ingredient in Korean traditional soup and nutritional foods. Generally, lipoxygenase in several food products may produce off-flavors during their processing and storage. Therefore, the inactivation of lipoxygenase is required to make the better extracts from Melania sanil. Also, the quality on freshness of Melania snail may be evaluated by lipoxygenase activity. The lipoxygenae activity was the highest at 40~60% saturation among several concentrations in salting-ouot saturated solution of ammonium sulfate. The partial purification of lipoxygenase was successfully obtained by Sephacryl S-200 gel chromatography. The first peak among three peaks for protein determination showed the highest activity of lipoxygenase in 13~16 fractions among 100 fractions. The highest peak of lipoxygenase activity by ion exchange chromatography was shown at 0.1M NaCl. In the purification step, the specific activity was 20.8U/mg and activity yield was 19.8%. The optimum pH and temperature were pH6.0~8.0 and 3$0^{\circ}C$, respectively. Molecular weight of the lipoxygenase was estimated about 35kDa by SDS-PAGE.

Changes of Physicochemical and Flavor Components of Ume According to Varieties and Picking Date (매실 품종과 수확 시기에 따른 이화학적 특성과 향기성분의 변화)

  • This study aims to determine the optimal picking time and the favorate variety for Ume processing. The changes of physicochemical components and flavors of 6 varieties of the Ume were investigated during maturing. Average weight of the fruit increased to the range of 151-292% from 70 days to 90 days after blooming. The hardness of fruit decreased during maturing and Koume variety was not suitable for processing among the last harvesting samples. Moisture and ash were reached to 89-91%, 0.57-0.69%, respectively, and the ash content increased during maturing. Among the Ume varieties, relatively high content of total acid was observed in Oshuku and Koume. The major organic acid were malic, citric, succinic, and tartaric acid. The content of malic acid decreased significantly, whereas citric acid increased during maturing. Thirty five kinds of flavor components were identified from the Ume fruit and main components were ethyl acetate, butyl acetate, and organic acid derivatives. Koume contained a large amount of flavor components among the Ume and the content increased during maturing.

Schisandra Fructus Butanol Fraction Reduces Serum Total Cholesterol and Triglyceride Levels in Hyperlipidemic Mice (오미자 부탄올 분획물이 고지혈증이 유도된 생쥐의 지질대사 및 간조직 유전자 발현에 미치는 영향)

  • The berries of Schisandra chinensis (Schisandra fructus) are given the name Omiza in Korean (五味子), which translates as "five flavor fruit" because they possess all five basic flavors in Korean traditional herbal medicine: salty, sweet, sour, pungent (spicy), and bitter. It is used as a remedy for many ailments: to resist infections, increase skin health, and cure insomnia, coughing, and thirst. This study was designed to investigate the effects of Schisandra fructus butanol fraction (SFB) on serum lipid levels in hyperlipidemic mice. In this experiment, effects on total cholesterol, HDL-cholesterol, and triglyceride in serum were measured. In our results, SFB did not affect weight gain in hyperlipidemic mice. Oral administration of SFB lowered levels of total cholesterol and triglyceride, which were elevated by induction of hyperlipidemia. Finally, administration of SFB regulated changes in gene expression which were related to cell growth and differentiation.

Salt reduction in foods using protein hydrolysates (단백질 가수분해물을 이용한 식품 내 소금 저감화)

  • As excessive intake of salt is regarded as a reason for health problems, the tendency of people to attempt to reduce intake of salt in their everyday lives is on the rise. In Korea, where many people have a higher intake of salt compared to those in other countries, there have been diverse efforts to improve on this eating habit. Protein hydrolysates are chemically, physically hydrolyzed protein that have been widely utilized as a material for not only regular food but health functional food due to have diverse biological effects such as anti-oxidation, anti-inflammation, prevention of diabetes, and regulation of blood pressure. Various amino acids such as glutamic acid, arginine and arginine dipeptides, which exist in the components of protein hydrolysates, have also been recently recognized as being helpful in decreasing the use of salt in foods as they can greatly enhance salty taste when used concurrently with salt due to having both salty and palatable flavors. In the case of protein hydrolysates that decompose soy protein or fish protein such as anchovy, they could reduce consumption of salt by as much as 50% without affecting people's food preferences when applied to food as they boost salty taste by approximately 10% to 70%. Although there are only a few studies on protein hydrolysates as a salty taste enhancer or salt substitute, the results of several studies are indicative of the potential of protein hydrolysates as a salty taste enhancing ingredient.

  • The olfactory bio technology is largely based on its corresponding recognition technology and smell stimulus that acquires, analyzes, and processes volatile organic compounds present in chemical molecules, which are present in the breath or air evoked by an electronic nose artificially imitating the human biological nose. The olfactory bio technology is also based on a scent display technology that automatically diverges various digital flavors based on aesthetics, concentration, duration, and intensity information required to enhance the sensibility using a computer. Recently, attempts have been made to apply noninvasive screening of dementia by sensing, analyzing, encoding, and transmitting bio information obtained through an olfactory interface, both domestically and externally; further, the olfactory medical content technology has been applied to delay or reduce the onset of dementia. In this study, we will focus on early screening of dementia using olfactory biology information and dementia cognitive enhancer content that delays or reduces the onset of dementia.

A Study on Xu Dachun's Understanding of Medicinals According to the Shennong Bencao Baizhonglu (서영태(徐靈胎)의 본초관(本草觀)에 대한 연구(硏究) - 『신농본초경백종록(神農本草經百種錄)』을 중심으로 -)

  • Objectives : Based on the entries on each medicinal's 'specification' and the phrase '所以然' in the Shennong Bencao Baizhonglu, this paper aims to examine Xu Dachun's understanding of herbal medicinals. Methods : One hundred medicinals from the text were sorted into eight categories: Qi, flavor, color, form, quality, nature and characteristic, time of growth, and producer, then examined. Results : 1. Medicinals related to Qi were aromatic and pungent and warm. 2. Medicinals related to flavor were explained according to the Five Flavors' nature in terms of the thing and its functions theory. 3. Medicinals related to color were explained in relation to the Five Colors, whose interpretations were rather erratic. 4. Medicinals related to form were explained as it being possible to figure out their effects based on each medicinal's shape and form. 5. Medicinals related to quality were those that were used to manage dryness and dampness using their dry or wet qualities. 6. Medicinals related to nature and characteristics needed to be understood in their originality to grasp their unique abilities. 7. Both producer and time of growth were closely related to each medicinal's efficacy. Conclusions : Understanding of the application of medicinals in the Shennong Bencaojing through continuous examination into each medicinal's 'specification' and 'its reasoning', together with their clinical application, will allow for learners to build an understanding of herbal medicinals.

History of kimchi industry (김치산업의 발달사)

  • Kimchi has been one of the main menu on our dining table at all seasons as the best matching side dish with boiled rice. Kimchi was privately made in every household, and it has been commercially made for military feeding since Korean Civil War in 1950. Commercialized kimchi in can was also made for soldiers sent to Vietnam in late 1960s and for workers sent to Middle East in 1970s. As industry was growing, the number of people dining out increased, and with export to Japan in 1980s. Kimchi industry had greatly expanded until 1990s. However. the growth of industry has slowed down as individual consumption decreased and import from China radically increased. Since 2000, kimchi industry has been growing again focusing on quality improvement, which is possible due to up-to-date system and high standard sanitation control in manufacturing. Kimchi becomes the most important cultural product at the core of our food culture as all Korean people realize that it contains not just a wide variety of ingredients but unique flavors of each local areas and sincere heart of local people. Commercialization of kimchi will increase even though its consumption decreases as people's dietary life changes.

Comparison of quality and bioactive components of Korean green, white, and black teas and their associated GABA teas

  • Various types of tea have been cultivated to obtain different flavors and enhance their functional properties. The objective of this study was to investigate the physicochemical properties of γ-aminobutyric acid (GABA) teas produced from commercial Korean green, white, and black teas. The concentration of total minerals was reduced in GABA green tea and GABA white tea but was improved in GABA black tea. The essential, non-essential, and total free amino acid contents were remarkably increased in the GABA teas. The amino acid GABA content was increased by 561.00 and 294.20 times in GABA white tea and GABA black tea, respectively. The antioxidant potential was not reduced, although the total polyphenol and total flavonoid contents decreased in GABA green tea and GABA black tea. The results indicated that the overall nutritional value of commercial green, white, and black teas could be improved by processing them into GABA teas.

Antecedents of Customer Loyalty and Perceived Service Quality: A SEM Analysis of Thai Restaurant Brands

  • With over 15,000 Thai restaurants worldwide and 5,342 in the USA alone, Thai cuisine has been repeatedly internationally acclaimed for its taste, flavors, smell, and nutritional values. The Thai Ministry of Commerce has also established a global objective to find and award the best Thai food restaurants using Thai Select Premium (TSP), Thai Select Unique (TSU), and Thai Select brand labels. Therefore, using systematic random sampling, 620 diners were selected from certified Thai Select restaurants (TSR) in either Los Angeles, New York, Miami, or Chicago to ask for their opinions concerning factors important to each restaurant's service loyalty (SL). The four constructs, 18 observed variables, and six hypotheses were analyzed using LISREL 9.1. Results revealed a significant positive effect (88% R2) on the causal factor interrelationships on TSRs' SL. Additionally, the three factors affecting SL the most were service tangibility, taste perceptions, and brand image, with total effect values of 0.94, 0.43, and 0.31, respectively. Finally, when each US diner was asked why they chose to eat Thai food, respondents overwhelmingly indicated that taste (49.50%) and healthiness (32.90%) were most important. The study also adds to the literature by highlighting the importance of Thai cuisine contributing to healthier lifestyles.

A Method for Making Kimchi Containing Snow Crab in a COVID-19 Environment

  • Purpose: The purpose of this study is to develop a kimchi recipe for immunity enhancement using snow crab, which has high food value in the era of COVID-19. Research design, data and methodology: The snow crab kimchi manufacturing method of this study includes the steps of preparing a kimchi seasoning containing snow crab seasoning and chitosan powder. Kimchi seasoning is made by adding 5 parts by weight of crab seasoning and 1 part by weight of chitosan powder to 100 parts by weight of the basic kimchi seasoning prepared by mixing radish, minced garlic, minced ginger, onion juice, anchovy sauce, red pepper powder and glutinous rice paste. Results: It was possible to develop new flavors, possibilities and characteristics of snow crab kimchi by extending the health and functional effects, taste, and preservation period without significantly changing the unique manufacturing method, taste and function of kimchi, including snow crab. Conclusions: Snow crab kimchi was excellent in taste and aroma while enhancing the health functions of the body, such as improving people's immunity. The developed snow crab kimchi manufacturing method can not only improve people's health, but also expand the choice of preference for kimchi taste and shelf life.
