식이성 윤충류질환의 관리전략 수립을 위한 감염원의 역학 및 병원체의 생물학적 특성에 관한 조사연구 - 한국산 민물어류에 기생하는 Metagonimus속 피낭유충의 숙주특이성과 감염실험을 통한 성충의 분류 (Classification and host specificity of Metagonimus spp. from Korean freshwater fish)

  • 한국인에 널리 유행하는 Metagonimus속 흡충류의 분류학적 문제점들을 해결하기 위해 다양한 지역에서 채집한 여러 종의 민물어류로부터 Metugonim속 피낭유충을 검출하여 햄스터에 감염 실험을 하였다. 감염실험 결과 은어는 기존에 알려져 있던 M. yokogawnia 피낭유충 외에 M. tokqhashii의 피낭유충에도 감염되어 있는 것이 밝혀졌으며 붕어에는 M. takohoshii의 피낭유충만 이 감염되어 있었다 피라미, 끄리, 갈겨니등에서 검출된 Metqgonim속 피낭유충의 감염실험 결과, 이들 어종은 모두가 Metosonim Miyata type의 피낭유충에만 감염되어 있는 것으로 나타났다. Metagonimus Miyata type의 성충은 여러 가지 특징 및 숙주특이성 등에 의해서 다른 두 종과 구별되었으며. 그 분류학적 위치에 대해서 몇 가지 가능성을 고찰하였다.

  • This study was studied in Tamjin liver that is situated jangheung-gun Jeollanm-do. Fishways that was studied with the emphasis on Ice harbour type fishway analyzed efficiencies. In result, Ice harbor type fishway discovered 12species of fish among 13species in Tamjin liver. But, most other fishways have discovered a few fishes. Because most fishways established ill station and managed badly.

국가습지유형분류체계의 습지 유형 (하천형과 호수형)에 따른 경남지역 습지의 어류군집 특성 분석 (The Analysis of the Fish Assemblage Characteristics by Wetland Type (River and lake) of National Wetland Classification System of Wetlands in Gyeongsangnam-do)

  • 습지 유형에 따른 어류군집 특성을 파악하고 이를 통해 관리 전략을 마련하기 위해 경상남도에 위치한 29개의 습지 (하천형 20개소, 호수형 9개소)를 대상으로 조사를 실시하였다. 조사결과 하천형 습지에서는 평균(${\pm}SD$) $10.3{\pm}4.8$종이, 호수형 습지에서는 평균 $9.1{\pm}4.1$종이 출현하였으며, 출현 종수의 차이는 확인되지 않았다(Mann-Whitney U test, P>0.05). 반면 두 습지 유형의 어류군집을 구성하는 종들은 통계적으로 유의한 수준의 차이를 보였으며(PERMANOVA, Pseudo-F=2.9555, P=0.007), 각 유형의 어류군집에 가장 크게 기여하는 종은 참갈겨니(하천형, 28.51%)와 블루길 (호수형, 23.21%)로 확인되었다 (SIMPER). 지점별 어류군집을 활용한 NMDS 분석결과 총 3개의 그룹(하천형, 호수형, 기타)으로 구분되어 기존의 유형 구분과 차이를 확인할 수 있었다. 현재 습지 관리는 멸종위기종을 중심으로 한 일원화된 방법이 제시되고 있으나, 본 연구에 의하면 습지 유형별 어류군집에 있어서 차이가 있기 때문에 고유종, 외래종, 주요 기여종에 대한 정보를 활용한 관리방법이 마련되어야 한다. 또한 현재 지형을 기반으로 한 습지의 유형 분류가 이루어지고 있으나 일부 습지의 유형에 대한 분류가 모호한 경우가 확인되었으며, 이에 대해 생물상 분석을 통한 보완이 이루어질 필요가 있다. 본 연구는 두 개의 습지 유형에 대한 분석결과로 한계가 있기 때문에 향후 모든 유형의 습지를 대상으로 연구를 실시하여 각 습지의 유형을 대변할 수 있는 세부적인 관리 방법 마련이 이루어져야 할 것이다.

Effects of Traditional Sauce Type and Storage Time on Quality Characteristics, Shelf-life and Flavor Compounds of Marinated Pork Cooked by Sous Vide Method

  • The present study aimed at evaluating effects of traditional sauce type and storage time on shelf-life and flavor compounds of marinated pork cooked by Sous-Vide method. Five different traditional sauces (Meju soy sauce, Brewed soy sauce, Fish-soy sauce, Ishiru fish sauce and Anchovy fish sauce) purchased from Asian countries were used. After marination with the sauces, polyvinylchloride film bags containing the marinated pork samples were cooked using Sous-Vide method for $55^{\circ}C$ for 5 h and $60^{\circ}C$ for 30 min, and were then stored for 8 wk at $10^{\circ}C$. Results showed that the pork samples marinated with the sauces retarded the growth of total plate counts (TPC) during storage. At $8^{th}$ wk storage, TPC counts were significantly lower in all samples marinated with the sauces compared to control (p<0.05). Lipid oxidation level was significantly lower in the T2 (Meju soy sauce) and T6 (Anchovy fish sauce) compared to those of T3 (Brewed soy sauce) and T5 (Ishiru fish sauce) or control after 8 wk storage (p>0.05). Forty volatile flavor compounds were detected from the control and marinated samples at $4^{th}$ wk of storage. The pork marinated with Anchovy fish sauce presented significantly higher amounts of importantly pleasant flavor compounds such as; pyrazines and sulfurcontaining compounds than those marinated with other remaining sauces and control. It is concluded that the marination with Anchovy fish sauce partly improved the shelf-life and increased amounts of pleasant flavor compounds of Sous-Vide cooked pork products during storage in comparison to the other remaining sauces.

Long Double-stranded RNA Induces Sequence-specific RNA Interference and Type I Interferon Responses in Rock Bream (Oplegnathus fasciatus)

  • To determine whether long double-stranded RNA (dsRNA) induces RNA interference and type I interferon (IFN) responses in fish, long dsRNAs encoding enhanced green fluorescent protein (EGFP), GFPuv, and polyinosinic-polycytidylic acid sequences were co-injected with an EGFP expressing plasmid, into rock bream (Oplegnathus fasciatus). We investigated the EGFP mRNA and protein levels, and the transcriptional responses of dsRNA-dependent protein kinase and Mx1 genes. Long dsRNAs were strong inducers of a type I IFN response in rock bream, resulting in nonspecific suppression of exogenous gene expression. Furthermore, sequence-specific knockdown of exogenous gene expression at the mRNA level was detected at an early phase (24 h). These results suggested that long dsRNA may inhibit exogenous gene expression through an early mRNA interference response and a later type I IFN response in fish.

Dimethylhydrazine으로 처리한 쥐에서 식이섬유소와 지방종류가 대장의 종양발생율과 세포증식에 미치는 영향 (Effect of Dietary Fiber and Fat on Tumor Incidence and Cell Proliferation of Colonic Mucosa in DMH-Treated Rats)

  • This study was designed to observe the effect of dietary fiber and fat on colon tumor incidence and cell proliferation. Male Sqraue Dawley rats(n=225) at 7 weeks of age, were divided into 3 groups depending on the type of fat b(beef tallow, corn oil and DHA-rich fish oil) and each group was again divided into 3 groups depending on type of fiber(fiber-free, perctin and cellulose) . The experimental diet containing dietary fat at 15%(w/w) and fiber at 6%(w/w) levels was fed for 25 weeks. At the same time, each rats was intramuscularly injected with DMH two times a week for 6 weeks to geive total dose of 180mg/kg body weight. Cell proliferation was measured by in vivo incroporation of bromodeoxyuridine (BrdU) into DNA. Fish oil decreased the tumor incidence (9.67%) compared with beef talow (33.39%) and corn oil (21.21%). Tumor incidence was decreased in all groups that fed cellulose (11.67%) compared with those of fiber-free(21.74%) and pectic(19.70%). Most of tumors was distributed at the site of the distal colon. The rats fed both fish oil and cellulose significantly decreased th enumber of tumors and tumor incidence compared to other groups. Fish oil was more effective in preventing cell prolofieration by decreasing crypt length and labeling index(LI) compared with beef tallow(p<0.05). Cell proliferation in distal colon was more developed to the upper part of the crypt compared to proximal colon. Overall tumor incidence and cell proliferation were more affected by dietary fat. But the effect of dietary fiber was different depending on type of fat in the experimental diet. These results suggest that a DHA -rich fish oil may has more decisive effect in inhibiting the cell proliferation in colon.

Occurrence and Molecular Identification of Anisakis Dujardin, 1845 from Marine Fish in Southern Makassar Strait, Indonesia

  • Anisakis spp. (Nematoda: Anisakidae) parasitize a wide range of marine animals, mammals serving as the definitive host and different fish species as intermediate or paratenic hosts. In this study, 18 fish species were investigated for Anisakis infection. Katsuwonus pelamis, Euthynnus affinis, Caranx sp., and Auxis thazard were infected with high prevalence of Anisakis type I, while Cephalopholis cyanostigma and Rastrelliger kanagurta revealed low prevalence. The mean intensity of Anisakis larvae in K. pelamis and A. thazard was 49.7 and 5.6, respectively. A total of 73 Anisakis type I larvae collected from K. pelamis and A. thazard were all identified as Anisakis typica by PCR-RFLP analysis. Five specimens of Anisakis from K. pelamis and 15 specimens from A. thazard were sequenced using ITS1-5.8S-ITS2 region and 6 specimens from A. thazard and 4 specimens from K. pelamis were sequenced in mtDNA cox2 region. Alignments of the samples in the ITS region showed 2 patterns of nucleotides. The first pattern (genotype) of Anisakis from A. thazard had 100% similarity with adult A. typica from dolphins from USA, whereas the second genotype from A. thazard and K. pelamis had 4 base pairs different in ITS1 region with adult A. typica from USA. In the mtDNA cox2 regions, Anisakis type I specimens from A. thazard and K. pelamis showed similarity range from 94% to 99% with A. typica AB517571/DQ116427. The difference of 4 bp nucleotides in ITS1 regions and divergence into 2 subgroups in mtDNA cox2 indicating the existence of A. typica sibling species in the Makassar Strait.

Association between fish and shellfish, and omega-3 PUFAs intake and CVD risk factors in middle-aged female patients with type 2 diabetes

  • BACKGROUND/OBJECTIVES: This study was performed to investigate the association between the dietary intake of fish and shellfish, and omega-3 polyunsaturated fatty acids (PUFAs) and cardiovascular disease (CVD) risk factors in the middle-aged Korean female patients with Type 2 diabetes (T2D). SUBJECTS/METHODS: A cross-sectional analysis was performed with 356 female patients (means age: 55.5 years), who were recruited from the Huh's Diabetes Clinic in Seoul, Korea between 2005 and 2011. The dietary intake was assessed by a validated semi-quantitative food frequency questionnaire and analyzed using the Computer Aided Nutritional Analysis program (CAN-Pro) version 4.0 software. RESULTS: In a multiple regression analysis after the adjustment for confounding factors such as age, BMI, duration of diagnosed T2D, alcohol consumption, fiber intake, sodium intake, and total energy intake, fish and shellfish intake of the subjects was negatively associated with triglyceride and pulse wave velocity (PWV). Omega-3 PUFAs intake was negatively associated with triglyceride, systolic blood pressures, diastolic blood pressures, and PWV. The multiple logistic regression analysis with the covariates showed a significant inverse relationship between the omega-3 PUFAs consumption and prevalence of hypertriglyceridemia [OR (95% CI) for greater than the median compared to less than the median: 0.395 (0.207-0.753)]. CONCLUSIONS: These results suggest that the consumption of fish and shellfish, good sources of omega-3 PUFAs, may reduce the risk factors for CVD in the middle-aged female patients with T2D.

Calcium Alginate로 제조한 어유 캡슐의 성질 및 산화안정성 (Properties and Oxidation Stability of Fish Oil Capsules Manufactured with Calcium Alginate Gels)

  • Alginates are used to encapsulate various materials, including food, cosmetics, and pharmaceuticals. This study examined the properties and oxidation stability of fish oil capsules manufactured with calcium alginate gels. The fish oil capsules were manufactured by dropping sodium alginate solution and fish oil into a calcium chloride solution through nozzles. The membrane thickness, sphericity, rupture strength and deformation depth of the fish oil capsules were determined. The peroxide value of the fish oil was assayed to determine the oxidation stability of the capsules. The capsules measured approximately 3 mm with a membrane thickness of 90 μm independent of the amount of fish oil added. As the amount of fish oil encapsulated increased, the sphericity, rupture strength and deformation depth of the capsules decreased. The encapsulation efficiency increased until the amount of fish oil was 30%. The oxidation stability of fish oil in capsules was dependent on the type of nozzle, e.g., the oxidation stability of fish oil in capsules made using a double nozzle was greater than with a single nozzle. These results should lead to industrial application of fish oils including eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, as nutraceuticals.

퍼스널 컴퓨터를 이용한 칼라 어군탐지기의 개발에 관한 연구 (Development of the PC Based Color Fish Finder)

  • 판되고 있는 어군탐지기는 칼라 모니터를 사용하든 기록지를 사용하든 현장에서 관측한 에코 데이터의 저장과 재생하는 기능이 없는 경우가 대부분이고, 어군탐지기의 기능은 제조회사에 의해 정하여져 있어 확장성이 약하다. 본 연구에서는 이 문제를 해결하는 하나의 방안으로 퍼스널 컴퓨터(386DX, 20 MHz), 디지털 I/O 보드, 동시 샘플링형 A/D 보드(12bit, 15 $\mu$sec/ch) 및 초음파 송수신 보드를 사용하여 어군탐지 정보의 저장과 재생이 자유롭고 확장성이 뛰어난 마스터형 칼라 어군탐지기를 개발하였고, 그 성능을 확인하는 실험을 행하였다. 또한, 퍼스널 컴퓨터와 A/D 보드를 이용하여 타 어군탐지기로부터의 신호를 입력하여 칼라 에코그램으로 재생할 수 있는 슬레이브형 칼라 어군탐지기를 개발하여 그 성능을 확인하는 실험도 행하였다. 에코신호의 샘플링 간격은 0.1m, A/D 변환속도는 30$\mu$sec로 하였다. 마스터형에서는 초음파 송수신 보드만 갖추면 사용자의 용도에 알맞은 칼라 어군탐지기를 구성할 수 있음을 공기중 실험 및 수조 실험에서 확인하였으며, 슬레이브형에서는 타 어군탐지기로 부터의 신호를 퍼스널 컴퓨터에 입력하여 재생할 수 있음을 확인할 수 있었다. 이 실험 결과로부터 본 연구에서 개발한 마스터형 칼라 어군탐지기는 사용자의 용도에 적합하게 개량할 수 있어 다양한 분야에 응용할 수 있을 것으로 기대된다. 슬레이브형에서는 에코 시뮬레터를 연결한 기록식 어군탐지기의 기록과 이 어군탐지기의 수신부에서 출력되는 에코 신호 및 트리거 신호를 시험 제작한 어군탐지기에 입력시켜 신호 처리한 칼라 에코그램을 비교한 결과, 해저 수심, 어군 등의 에코 기록이 완전히 일치하면서 해상력이 뛰어났으며, 키보드의 기능키에 할당된 에코그램의 shift 기능, 화면상의 최대 표시 심도 조정 기능, gate 기능 등이 정상적으로 작동함을 획인할 수 있었다. 자원조사용 어군탐지기의 에코 신호를 데이터 로거에 기록한 후 이 데이터를 본 연구에서 개발한 어군탐지기로 재생하는 실험을 행한 결과, 양호하게 재생할 수 있었으며, 무선 송수신기를 이용하여 에코 시뮬레이터의 신호를 원격으로 수신 처리한 결과도 양호하였다. 따라서, 슬레이브형 어군 탐지기는 흑백 기록식 어군탐지기를 이용하여 칼라 에코그램을 나타내는 상업용으로 혹은 어류 자원을 조하고 관리하기 위한 학술 연구용으로 활용될 수 있을 것으로 기대된다.
