Toxicity of the Puffer Fish Takifugu porphyreus and Takifugu rubripes from Coastal Areas of Korea (한국 연안산 검복(Takifugu porphyreus)과 자주복(Takifugu rubripes)의 독성)

  • Toxicity of two species of puffer fish, Takifugu porphyreus and Takifugu rubripes, collected from coastal regions of Korea, was determined using a mouse bioassay, In T. porphyreus, the proportion of toxic specimens containing ${\ge}$ 10 MU/g was 58.3% for the ovary, 32.6% for the skin, 12.0% for the gallbladder, 11.6% for the liver and intestine, and 9.3% for the fin; no toxicity was detected in the muscle and testis using the mouse bioassay. The highest toxin levels were 531 MU/g in the liver, 253 MU/g in the intestine, 136 MU/g in the gallbladder, 118 MU/g in the skin, 116 MU/g in the ovary, and 108 MU/g in the fin. The skin, which is used for human consumption, showed significantly high toxicity with an average of $11{\pm}3\;(mean{\pm}SE) MU/g$. Takifugu porphyreus toxicity also exhibited remarkable regional variation. In T. rubripes, the proportion of toxic specimens was 25.0% for the ovary, 15.8% for the liver, 11.1% for the gallbladder, and 5.3% for the fin and intestine; no toxicity was detected in the muscle, skin, or testis. Among the organs, the highest toxin levels were 228 MU/g in the ovary, followed by 112 MU/g in the liver, 28 MU/g in the gallbladder, 18 MU/g in the intestine, 11 MU/g in the fin, and 8 MU/g in the skin. Thus, we found acceptable toxin levels in the edible muscle and skin of T. rubripes and in the muscle of T. porphyreus. However, the skin of T. porphyreus, which showed significantly high toxicity, requires special attention when used for human consumption.

Analysis for cancer-related studies using Puffer fish poison (복어독의 종양 관련 기존연구 논문에 대한 분석)

  • Objective: The aim of this study is to analyze the previously published research articles related to puffer fish toxin focusing on tumor. Method: Literatures were searched in PubMed database, published since 2000, using the keyword; Puffer fish, Fugu and tetrodotoxin (TTX) with cancer or tumor. Research papers were classified by year, country, study model, used material, kind of tumor and study subject. Finally, a total of 41 studies were analyzed in this study. Results: From 2000 to 2018, the most abundant papers were published in 2009 (6 studies) and almost half of the papers were studied in United Kingdom (20 studies). The 39 studies used TTX purified from puffer fish while 2 studies used crude extract of skin and gonad of puffer fish. The most used target cell line was prostate cancer (15 studies), and the next was breast cancer (14 studies). The study methods were classified into 4 clinical studies, 2 animal studies and 35 cell-based studies. Conclusions: Our results show that the overview of cancer-related studies using puffer fish poison. This information would be helpful for the puffer fish-derived drug researches in the future.

Henneguya sp. infection of cultured snakehead, Channa argus fingerings (양식 가물치, Channa argus 치어의 Henneguya sp.에 의한 감염)

  • The present work describes myxozoan parasite found in the skin of cultured snakehead, Channa argus fingerings (total length, 5.5~7.2 cm) from Busan, Korea. Nature spores and plasmodia were found in the skin mucus of infected fishes. In fresh state, the total length of the spore was $27.96{\pm}2.50{\mu}m$. The size of spore body was $14.16{\pm}1.78{\mu}m{\times}4.88{\pm}0.61{\mu}m$. The polar capsules were pyriform and the size was $5.57{\pm}0.66{\mu}m{\times}1.36{\pm}0.33{\mu}m$. This is the first report of Henneguya sp. from cultured fish species in Korea, and further studies are necessary for definitive identification.

Occurrence and treatment of mixed infections with Vbrio anguillarum and Ichthyobodo sp. from cultured oliver flounder (Paralichthys olivaceus) (양식 넙치에서 Vbrio anguillarum 및 Ichthyobodo sp.에 의한 혼합 감염증의 발생 및 치료)

  • The high mortality (65.5%) of young olive flounder (Paralichthys olivaceus) showing skin ulcers occurred in fish obtained from a commercial cultured farm at Donghae on the eastern coast of Korea at March 2006. The dorsal skin showed ulcer lesions and large amount of mucus. We isolated Ichthyobodo sp. from the gill, and Vibrio anguillarum from the liver and kidney of dead and diseased fish. All of the fish infected with the V anguillarum and Ichthyobodo sp. were treated with 100 ppm formalin for 1 h, and treated with oxolinic acid for 4 h at $22^{\circ}C$, respectively. The formalin treated group was observed the low mortality (2.9%) when compared with high mortality of non-treated (82.9%), and of antibiotic treated group (85.9%) against mixed infections by Vibrio anguillarum and Ichthyobodo sp. organisms. We confirmed complete elimination against mixed infections of Vibrio anguillarum and Ichthyobodo sp. with 100 ppm formalin treatment.

  • Histopathological observations on a epidermal papilloma of wild red halibut (Hippogossoides dubius) caught at the area around Echung Island and Gogunsan Islands were done. The papilloma formed on the dorsal fin was the same color with the body on the ocular side but black on the non-ocular side. Under light microscopy, the tumor on the skin was very similar to epithelial papilloma supported by connective tissue. X-cell-like cells in the tumors were large and eosinophilic with a centrally-located, large nucleus. But the characteristic large nucleolus in the nucleus of the x-like cells was indistinct clear in this specimen because about 6-9 hours have passed after catching the fish and autolysis have occurred.

The Effects of yeast $\beta$-glucan in the Diet on Immune Response of Japanese eel, Anguilla japonica, by Oral Administration (Yeast $\beta$-glucan 첨가 사료가 뱀장어의 비특이적 면역 반응에 미치는 영향)

  • The effects of dietary yeast β-glucan administration on growth, nonspecific immune responses, serum lysozyme, skin mucous lysozyme, NBT (nitroblue tetrazolium) reduction by phagocytes, and disease resistance against Edwardsiella tarda in Japanese eel, Anguilla japonica were evaluated. Fish were fed the diets supplemented with 0%, 0.1% and 0.5% of yeast β-glucan to a commercial diet for 6 weeks. The body weight gain from the fish fed on the 0.5% supplemented diet for 6 weeks was significantly higher than the control. Both serum and skin mucous lysozyme were significantly higher in the all experimental groups except 2 weeks of 0.5% group. The bactericidal activity of serum was slightly increased at 6 weeks. Also, The intracellular superoxide anion production of kidney phagocytes was significantly higher in the all experimental groups. The diet supplemented with 0.1% were also found to raise the relative percent survival (RPS) of Japanese eel after an artificial challenge with 1×107 cells of Edwardsiella tarda per fish. The results suggested the potential of yeast β-glucan to activate some innate immune responses and to improve the growth in Japanese eel.

Skin Transcriptome Profiling of the Blass Bloched Rockfish (Sebastes pachycephalus) with Different Body Color Patterns (체색 패턴이 다른 개볼락(Sebastes pachycephalus) 피부 전사체 프로파일링)

  • The body color pattern in fish is a distinctive feature for species identification. The blass bloched rockfish Sebastes pachycephalus is a commercially important marine fish species, distributed in the central and southern parts of Korea and south Hokkaido of Japan. It has a morphological feature divided into four subspecies according to with or lacking distinct spots on the body surface, and to the location of markings on the body surface. However, the genetic basis of body color pattern of S. pachycephalus is still unknown. Thus we analyzed the transcriptome of S. pachycephalus skin samples using RNA-seq analysis to investigate functional genes related to body color patterns. The experimental skin samples were prepared by classified into 'Wild type' (lacking distinct spots and markings) and 'Color type' (with distinct spots and marking). Two skin sample transcriptomes were compared pairwise and the results revealed that were 164 differentially expressed unigenes in the skin samples of 'Wild type' and 'Color type'. Gene Ontology analysis of 164 differentially expressed unigenes showed that these genes were included in the functional group of molecular function (2 genes), biological process (46 genes), and cellular component (6 genes). There were several genes that body color type skin specific expression and the genes were CTL (Galactose-specific lectin nattectin), CUL1 (Cullin-1), CMAS (N-acylneuraminate cytidylyltransferase), NMRK2 (Nicotinamide riboside kinase 2), ALOXE3 (Hydroperoxide isomerase ALOXE3), SLC4A7 (sodium bicarbonate cotransporter 3). Our study is the first attempt to search for functional genes involved in the formation of body color patterns in S. pachycephalus. The differentially expressed unigenes obtained in this study can be used as candidate genes for functional gene study related to body coloration of fish.

Mass Mortality of Doctor Fish(Garra rufa obtusa) Caused by Citrobacter freundii Infection (Citrobacter freundii 감염에 의한 Doctor fish(Garra rufa obtusa)의 집단 폐사)

  • In this paper, we described a case of mass mortality of doctor fish from a private fish hatchery farm in Korea with a history of abnormal swimming behavior, diffuse bleeding on the skin and fins and sudden death caused by fish pathogenic bacteria, Citrobacter freundii. Twelve moribund fish fingerling samples were submitted to College of Veterinary Medicine, Seoul National University in October 2008 for diagnostic examination. Diagnostic results showed that the morphological and biochemical properties of the bacteria isolated from the moribund fish were C. freundii. The remaining diseased fish from the hatchery farm were given treatment based on our recommendation and successfully recovered.

Effect of Calcium Compounds from Oyster Shell Bouind Fish Skin Gelatin Peptide in Calcium Deficient Rats (어피 젤라틴 펩티드와 결합한 굴껍질 유래 칼슘 화합물이 칼슘 결핍 흰쥐에 미치는 영향)

  • To utilize the oyster shell containing a lot of calcium, we investigated the bioavailability of calcium compounds from oyster shell. First, calcium oxide was prepared by burning of oyster shell at $1200^{\circ}C$. Its purity was approximately $98.5\%$. Calcium compounds, $CaCl_2$, and $CaHPO_4$, from the calcium oxide were prepared by chemical reaction. Effect on calcium absorption by the calcium compounds from oyster shell was improved using fish skin gelatin peptides.(FSGP), which was prepared by enzymatic hydrolysis of fish skin gelatin for 4hr with tuna pyloric caeca crude enzyme (TPCCE). in vitro experiment, calcium absorption by addition of FSGP in a mixture solution of calcium and phosphate was higher approximately $70\%$ than that by control. in vivo using calcium deficient rats, a group taken the diets with $3\%$ FSGP and $CaHPO_4$ was significantly improved amount of calcium and ash in femur and strength of femur. These results suggest that calcium compounds from oyster shell and FSGP could be used as an effective dietary calcium source.
