어류의 조직중에 분포하는 알카리성 단백질분해효소의 활성조건 (Alkaline Pretense Activity of the Tissue Extracts from Some Different Kinds of Fish)

  • To check the differences of the digestive enzymes by the bait habits and the proteolytic activities of the fissile extracts from the fish, omnivorous filefish (Navodon modestus), carnivorous cat shark (Scilliorhinus tarazame) and bloodsucking hag fish (Eptatretus burgeri) were sampled for this experiment. The activity of crude alkaline protease extracted from the muscle and the internal organs of the samples was determined with casein as substrate. The activity of the proteolytic enzymes showed remarkable differences by the organs of the fish. The optimum condition of the pretenses from the muscle revealed in range of pH 7.8-8.3, at $60-65^{\circ}C$, while those of the enzymes from the internal organs were at about pH 8.2, $45-55^{\circ}C$, but those of hag fish were at about pH 6.7, $45-55^{\circ}C$. The proteolytic activity of the enzyme of alimentary canal in filefish and in hag fish was 57 and 11 times stronger than that of muscle, respectively. The crude enzyme from the alimentary canal of file fish showed the strongest proteolytic activity in samples submitted and that of cat shark was the lowest. The activity of pancreatic alkaline protease in cat shark was 50 fold higher than that of muscle alkaline protease in the fish.

2012년 제주의 양식 넙치 및 자연산 어류의 Kudoa septempunctata 감염 조사 (Monitoring of Kudoa septempunctata in Cultured Olive Flounder and Wild Fish in Jeju Island during 2012)

  • Kudoa septempunctata가 한국의 양식 넙치의 근육에 기생하는 것이 보고되어, 본 연구에서는 2012년 동안 제주 지역의 양식 넙치 및 자연산 어류에서 K. septempunctata의 감염 현황을 조사하였다. 제주지역 26개소의 넙치 양식장으로부터 143마리를 조사한 결과, 4개소 7마리(4.9%)에서 쿠도아 감염이 확인되었으며, 넙치 치어 67마리에서는 쿠도아가 검출되지 않았다. 또한, 제주 지역에서 어획 된 자연산 어류 8종에서도 쿠도아가 검출되지 않았다. 쿠도아가 검출 된 7마리 어류에 대하여 등근육 부위별 쿠도아 검출을 비교하기 위하여, 넙치의 두부쪽에서 채취한 등근육, 미부쪽에서 채취한 등근육, 그리고 등근육전체를 긁어 모은 샘플을 사용하여 검출률을 비교한 결과, 전체를 긁어 모든 샘플을 사용할 경우 검출률이 100%로 가장 높아, 어느 한 쪽에서 근육을 채취하는 것 보다 등근육 전체 부위를 골고루 채취하는 것이 쿠도아 검출에 더 효율적임을 확인하였다.

HPLC를 이용한 어류 중의 Spiramycin 분석 (Analysis of Spiramycin in Fish Using High Performance Liquid Chromatography)

  • A high performance liquid chromatography assay method for spiramycin in fish muscle was developed. The developed method was evaluated and validated by monitoring spiramycin in olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli) and in live conger eel (Anguilla japonica) in fish farms and distribution centers. Using the developed method, the recovery rate was up to 82.4-88.8%, which was higher than that of conventional methods (77.6-87.1%). In particular, the proposed sample treatment protocol was suitable for use with fish samples to remove low molecular weight materials and pigments that could interfere an accurate analysis. The prepared stock solution was very stable, and it remained chemically stable for 5 weeks at $4^{\circ}C$. The performance limit of the developed method for spiramycin acid in fish muscle was 0.05 ppm. One hundred thirty-four fish samples including olive flounder, black rock fish and live conger eel were analyzed to evaluate the overall efficiency of the modified method and to monitor the actual condition of spiramycin usage in fish farms. Olive flounder and black rock fish were collected from fish farms in coastal areas of Korea, and live conger eels were purchased from a fish market in the Busan area from September 2001 to March 2002. According to the overall performance of the developed method, it was considered suitable for the monitoring of spiramycin in fish muscle. The suggested method of analysis for spiramycin in fish muscle is registered in the Korean Official Methods of Food Analysis and is currently tieing utilized for fishery products inspection by the Korea Food and Drug Administration and the National Fisheries Products Quality Inspection Service.

Dual effect of Low- frequency Electromagnetic Field on Muscle Histopathology of Caspian Sea Cyprinus carpio

  • The effect of electromagnetic field on aquatic organisms has received little attention. In the current study, the effect of 50Hz electromagnetic field on muscle histopathology of Caspian Sea Cyprinus carpio, a species of economic importance, was investigated. A total of 120 healthy fish were used in this study. They were classified randomly in one of two groups as follows: Control or unexposed EMF group and experimental group with 5 different magnetic field intensities (0.1, 1, 3, 5 and 7mT) at 2 different exposure times including 30 and 60 minutes. Fish in the experimental group were exposed only once. Two weeks after exposure, dorsal muscles sectioned transversely, stained and were examined using a light microscope. Histopathologic assessments showed significant difference between control and EMF exposed groups at both 30 min. (p<0.01) and 60 min. (p<0.001) exposure times. We report for the first time that electromagnetic field in interaction with muscular tissue of Cyprinus carpio exhibits a dual effect which depends on the field intensity, and exposure time. At short exposure time (30 min.), EMF stimulates muscle growth process. At longer exposure time (60 min.), EMF can damage muscle tissue and result in muscle necrosis. More research is required to elucidate precise mechanisms involved in muscle hypertrophy and pathologic changes.

배합사료와 생사료로 사육한 넙치의 체조성 및 육질 비교 평가 (Comparative Evaluation of Proximate Composition and Muscle Quality according to the Olive Flounder (Paralichthys olivaceus) fed Extruded Pellets and Raw Fish-Based Moist Pellet)

  • This study was conducted to evaluate the quality characteristics (proximate composition, fatty acids, amino acids and sensory properties) of whole body and dorsal muscle in olive flounder, which is switch size from juvenile to growing fed extruded pellets (EP) and raw fish-based moist pellet (MP) for 9 months. Sample was collected at 5 and 9 months to compare with those of EP and MP. Whole body crude protein of fish fed EP at 9 months was significantly higher than that of fish fed MP (P<0.05). Linolenic acid(18:3) and n-6/n-3 ratio in dorsal muscle of fish fed EP were significantly higher than those of fish fed MP (P<0.05). No significant differences were observed in the results of the flavor, color, taste and texture of the muscle of the fish fed EP compared to those fed MP (P>0.05). Based on the experimental results, we concluded that olive flounder cultured with EP is not inferior to those fed MP for quality characteristics of whole body and muscle.

Effects of Dietary Protein Sources on Growth and Body Composition in Olive Flounder (Paralichthys olivaceus)

  • A 6-week feeding trial was conducted to investigate the effects of six different dietary animal protein sources on growth and body composition of olive flounder, Paralichthys olivaceus in recirculating system. White fish meal (WFM), flounder muscle (FLM), carp muscle (CM), blood meal (BM), squid liver powder (SLP) and casein (CA) were used as the main animal protein sources in the six experimental diets. Fish averaging $2.9\pm0.03g$ $(mean\pm SD)$ were distributed to each aquarium as a group of 15 fish and were fed one of the six experimental diets to each treatment of triplicate groups. After 6-week of the feeding trial, fish fed white fish meal (WFM) and flounder muscle (FLM) diets showed a significant higher weight gain $(WG\%)$ (P<0.05) than those of fish fed the CM, BM, SLP and CA diets. Fish fed BM diet showed the lowest WG among all the dietary treatments. Feed conversion ratio (FCR) and protein efficiency ratio (PER) showed the similar trend as WG. Hematocrit and hemoglobin were not affected by the dietary treatments. Fish fed the FLM and CM diets showed significant higher survival rate than those of fish fed BM diets, and there was no significant difference in survival of fish fed WFM, FLM, CM, SLP and CA diets. These results indicated that WFM and FLM are the best dietary protein sources tested in olive flounder.

72종 어류의 일반성분조성과 콜레스테롤 및 $\alpha$-토코페롤함량 (Proximate Composition, Cholesterol and $\alpha$-Tocopherol Content in 72 Species of Korean Fish)

  • 통영 연안해역 및 경남 내륙지방에서 어획된 72종 (해산어 59종, 담수어 13종)의 어류에 대한 식품영양학적 기초자료를 얻기 위하여, 그들의 일반성분조성과 CHOL 및 $\alpha$-Toc 함량의 분석결과를 요약하면 다음과 같다. 지질 함량은 표 중층 회유어 (평균 $6.09\%$) > 담수어(평균 $4.40\%$) > 연안 암초어 (평균 $3.41\%$) > 저서어(평균 $2.21\%$) 순으로 많았으며, 수분 함량과는 역상관관계 (p<0.001)를 나타냈다. 단백질은 전 어류에서 평균 약 $17\~18\%$를 함유하였으나, 수분 함량과의 상관성은 저서어를 제외한 다른 어류에서는 인정되지 않았고, 회분 함량은 전 어류에서 $1\~2\%$로 거의 일정하였다. CHOL 함량은 6.7(꼼치)$\~$249mg(뱀장어)/100g muscle의 범위로 어종에 따라 큰 차이를 보였으며, 지질함량과 정의 상관관계를 보였다. $\alpha-Toc$ 함량은 뱀장어 (4.1 mg/100 g muscle)를 제외한 나머지 어종에서는 흔적 또는 3mg/100g 이하였으며, 지질 함량과 대체로 비례하는 경향을 나타냈다.

  • Kudoa septempunctata에 대한 PCR 검사에서 양성으로 확인된 개체를 조직 내 포자가 확인된 개체(3마리)와 포자가 확인되지 않은 개체(4마리)를 선별하여 순치 후 혈액과 조직검사를 실시하였다. 혈액에 대하여 PCR 검사를 실시하여 포자가 명확하게 확인되었던 2개체에서는 음성이 확인되었으며, 다른 5개체는 감도는 각각 차이는 있었지만 양성으로 확인되었다. 포자가 확인된 개체의 각 장기(간, 비장, 신장, 심장, 위, 소화관, 근육)에 대하여 PCR 검사를 실시한 결과, 심장과 근육에서 명확한 양성이 나타났다. 근육조직의 H&E 염색과 ISH를 실시하여 K. septempunctata의 감염으로 근섬유에 괴사가 발생하고, 이 부위에 포자의 슈도시스트가 형성되는 것을 확인하였으나 심한 염증반응을 유도하지는 않는 것이 확인되었다. 이후 혈액에 대한 더욱 면밀한 조사가 필요할 것으로 보이며, 연구내용은 넙치양식에서의 쿠도아감염증의 예방과 구제를 위한 기반자료가 될 것으로 기대된다.

  • 우리나라의 하천에 분포(分布)하고 있는 담수어(談水魚)의 식품학적(食品學的)인 기초자료(基礎資料)를 얻고져 비교적(比較的) 희귀종(稀貴種)에 속(屬)는 은어, 누치, 쏘가리 및 밀어 배육중(背肉中)의 핵산관련물질(核驗關聯物質), 유리(遊離)아미노산(酸) 및 구성(構成)아미노산(酸)을 분석(分析)하였다. 핵산관련물질(核酸關聯物質)은 IMP가 월등히 많아 은어, 누치, 쏘가리 및 밀어 배육중(背肉中)에 $10.7{\sim}19.5{\mu}mole/g$의 범위였다. UMP 및 hypoxanthine은 $2.0{\mu}mole/g$ 이하였고. GMP와 inosine은 흔적량에 불과하였다. 그리고 AMP는 은어 배육(背肉) 및 밀어 배육(背肉)에서 각각 2.0, $2.7{\mu}mole/g$으로 검출(檢出)되었으나 누치와 쏘가리 배육(背肉)에서는 흔적량으로 검출(檢出)되었다. 유리(遊離)아미노산(酸) 조성(組成)을 보면 은어 배육(背肉)의 엑스분중(分中)에는 taurine 및 histidine이 각각 전체(全體)의 36.6%, 15.4%, 누치 배육(背肉)에는 taurine, glycine 및 histidine이 각각(各各) 21.2${\sim}$29.0%, 쏘가리 배육(背肉)에는 taurine, histidine 및 alanine이 각각(各各) 14.5${\sim}$31.8%, 밀어 배육(背肉)에는 taurine, proline 및 threonine이 9.1${\sim}$31.2%로서 비교적(比較的) 많은 함량(含量)이 검출(檢出)되었다. 구성(構成)아미노산(酸) 은어 배육중(背肉中)에는 glutamic acid, glycine, aspartic acid 및 lysine이 총(總) 구성(構成)아미노산(酸)의 44.0%, 누치 배육중(背肉中)에는 43.5%, 그리고 쏘가리 및 밀어배육중(背肉中)에는 glutamic acid, aspartic acid, lysine 및 leucine이 각각(各各) 43.3%와 43.1%를 점유(占有)하고 있었다. glutamic acid는 어종(魚種)에 관계(關係)없이 그 함량(含量)이 가장 많았다.

망상어 척출자궁(剔出子宮)에 대(對)한 자율신경계(自律神經系)의 작용(作用) (Actions of Autonomic Drugs on the Uterine Muscle of Ditrema Temmincki Bleeker)

  • The author studied the actions of autonomic drugs on the uterine muscle isolated from Ditrema temmincki Bleeker, and the results obtained were summerized as follows. 1) The motility of the fish uterus was stimulated by epinephrine, norepinephrine and phenylephrine, but inhibited by isoproterenol. 2) The inhibitory effects of isoproterenol on the fish uterus was not affected by phenoxybenzamine, but blocked by propranolol. 3) The excitatory effects of phenylephrine on the fish uterus were blocked by phenoxybenzamine, but stimulated by propranolol. 4) The excitatory effects of epinephrine and norepinephrine were reversed by phenoxybenzamine and stimulated by propranolol. 5) The motility of the fish uterus pretreated with phenoxybenzamine and propranolol was not affected by isoproterenol, phenylephrine, epinephrine and norepinephrine. 6) It seemed that the uterine muscle of the fish had both alpha excitatory and beta inhibitory receptors. 7) The motility of the uterus of the fish was stimulated by acetylcholine. The stimulating action of acetylcholine was antagonized by atropine. 8) The motility of fish uterus was not affected by nicotine and DMPP. The actions of these drugs were not affected by pretreatment with hexamethonium and atropine. 9) It is, therefore, concluded that there are not present ganglia cells furnished with cholinergic fiber in the uterine wall of the fish.
