Germicidal Effect of Electrolyzed Seawater on Live Fish and Shellfish (전기분해 해수의 활어패류 살균 효과)

  • To secure the biological safety of live fish and shellfish for raw consumption, the germicidal effects of electrolyzed seawater were evaluated. Upon direct exposure to electrolyzed seawater, coliform group bacteria were killed and decreased to undetectable levels after 1 day. The physicochemical characteristics of the seawater were stable during the test period. A byproduct of chlorine disinfection, trihalomethane, was not generated by the electrolysis of seawater. Vibrio parahaemolyticus infection in a live fish was effectively resolved by electrolyzed seawater and became undetectable after 12-36 h of treatment. Bioaccumulation of coliform group and fecal coliform bacteria in live oysters Crassostrea gigas was removed within 18 h of treatment. This study demonstrated that electrolyzed seawater is an effective and safe germicidal agent for the traditional retail market and can help to prevent outbreaks of foodborne disease associated with the consumption of raw fish and shellfish.

Accumulation of Butyltin Compounds in Shellfish and Fish from Korean Coastal Areas

  • Butyltins (BTs) were measured in 2 species of shellfish and 16 species of fish collected along the Korean coast. Tributyltin (TBT) was detected in the mussels and oysters of sentinel organisms (20 to 940 ng Sn/g dry wt), indicating widespread contamination of TBT in the Korean coast. The elevated concentrations of TBT in the shellfish found in the sites near harbors or shipyards suggested that antifouling paints are probable major sources of butyltins in these areas. The TBT compound was detected in 12 out of 16 fish samples. The concentrations of TBT in fish muscles wee between 7 and 151 ng Sn/g dry wt, while the level in whole body of anchovy was very high (793 ng Sn/g dry wt). Exposure doses to Korean people via consumption of these marine products wee evaluated. The results suggested that the environmental levels of TBT were below the level of concern.

Comparison of Antimicrobial Resistance and Multi-Drug Resistance Patterns of Escherichia coli Isolated from Aquatic Organisms Off the West Coast of South Korea (서해안 수산생물에서 분리한 대장균(Escherichia coli)의 항생제 내성 및 다제 내성 양상 비교)

  • Antimicrobial resistance patterns of Escherichia coli were investigated. Strains were isolated from 310 shellfish, 36 crustaceans, and 12 fish collected off the West Coast of Korea from April 2019 to October 2020. Two hundred and ninety-five E. coli strains were isolated from shellfish, 100 from crustaceans, and 54 from fish. Strains isolated from shellfish showed the highest resistance to ampicillin (27.5%), whereas those from crustaceans were resistant to sulfisoxazole (30.0%) and those from fish were resistant to ampicillin (59.3%) and sulfisoxazole (59.3%). Ceftazidime resistance was observed in strains isolated from short neck and hard clams, whereas gentamicin resistance was observed in strains from fish. Multi-drug resistance was observed in 56 strains (48.7%) isolated from shellfish, 11 (28.2%) from crustaceans and 27 (73.0%) from fish. Depending on the source of isolation, the strains showed specific antimicrobial resistance tendency. Strains isolated from shellfish showed 12 different multi-drug resistance patterns, whereas those from crustaceans showed high resistance (59%) to a single antimicrobial agent and those from fish showed a broad trend of multi-drug resistance to more than eight antimicrobials.

A Study on Heavy Metal Contents of the Fresh Water Fish, and the Shellfish in Koran (국내 유통 민물어류와 연안산 패류의 중금속 함량에 관한 조사)

  • This study were conducted to estimate the values of the heavy metal in the fresh water fish and shellfish collected from fish market. The levels of the heavy metal were determined in a total of 319 samples of 11 species of the fresh water fish and 14 species of the shellfish by mercury analyzer and atomic absorption spectrophotometer. The results of this study were summerized as follows; The average contents of the heavy metal in the fresh water fish were Pb 0.075, Hg 0.05:3, Cd 0.002, Cr 0.135, Fe 8.695, Mn 1.078, Zn 9A91, Cu 0.548 mg/ kg. The average contents of the heavy metal in the shellfish were Pb 0.059. Hg 0.007, Cd 0.146, Cr 0.147, Fe 40.808, Mn 7.738, Zn 13.943, Cu 2.7:31 mg/kg. Being compared the average contents of the heavy metal of the fresh water fish with those of shellfish, the average contents of Cd, Fe, Mn, Cu in the shellfish were significantly higher than in the fresh water fish(P<0.001). Depending on the sampling areas, the average contents of the heavy metal were different by districts. The contents of the heavy metal were changed with the seasons(P<0.05).

Investigation of Domoic acid in Shellfish Collected from Korean Fish Retail Outlets

  • The purpose of this study was to determine the extent of domoic acid (DA) a potent neurotoxin, responsible for the syndrome amnesic shellfish poisoning (ASP) contamination of various species of bivalve shellfish purchased from fish market in Korea and the implications for food safety. Liquid chromatography (LC) methods were applied to quantify DA in shellfish after sample clean-up using solid-phase extraction (SPE) with strong anion exchange (SAX) cartridges. Toxin detection was achieved using photodiode array ultraviolet (LC-UV) and electrospray ionization-mass (LC-ESI-MS). DA was identified in 4 bivalve shellfishes of 872 shellfishes collected from March, 2006 to October, 2007 in Korea. DA amount of 3 surf clams (Mactra veneriformis) collected at Seoul, Daejeon, and Daegu were 4.13, 1.99, and 1.94 mg/kg, respectively. DA amount of 1 pink butterfly shell (Peronidia venulosa) collected at Seoul was 3.02 mg DA/kg. The amounts of DA that were present in 4 bivalve shellfishes were within EU guideline limits for sale of shellfish (20 mg DA/kg).

Pathogenicity of new reassortant betanodaviruses to various juvenile fishes (새로운 betanodavirus 재편성체(reassortant)의 어류 치어에 대한 병원성 분석)

  • With the recent isolation of a new betanodavirus in shellfish, Korean Shellfish Nervous Necrosis Virus (KSNNV), it has also been identified the reassortant KSNNV of two RNA segments, in which one segment is KSNNV genotype but the other one is known genotype. In this study, we confirmed that the ressortant KSNNVs obtained in previous screening study of our laboratory for betanodaviruses in shellfish were KS/RGNNV and RG/KSNNV type by performing two consecutive multiplex RT-PCR on each RNA1 and RNA2 segment (R1- and R2-discriminative multiplex two-step RT-PCR, respectively) to determine the genotype of each segment based on the size of amplicon. In the pathogenicity analysis, none of the reassortants induced specific external symptoms or mortality of VNN, but viruses of 2 × 104~105 copies/mg or more were detected at 14 days after injection (107 copies/fish) in brain tissues of 4 species except for crucian carp and common carp among the 6 species of juvenile fish used. In addition, the histopathological features of weak but distinct vacuole formation were also found in the brain of these infected fish, but no difference was found between the two reassortants KS/RGNNV-KG and RG/KSNNV-CM.

On the Some Toxic Dinoflagellates (유독플랑크톤(과편모조류를 중심으로)에 관한 고찰)

  • Some species of dinoflagellates were considered as one of the causative organisms of PSP(Paralytic Shellfish Poison) or DSP(Diarrhetic Shellfish Poison). Fish and shellfish are intoxicated by feeding of toxic plankton, sometimes human is intoxicated by feeding on these intoxicated fish and shellfish. In past ten years, the physiological and ecological studies of the toxic plankton has been investigated for development of monitoring system and preventation and control measures of PSP. However, in our country still little is known on a research for the toxic dinoflagellates. This paper reviews the general biology, taxonomic problem, physioecology and culture method of the toxic planktons such as Protogony-aulax and Dinophysis.

Review on the Biological Effects of Suspended Solids on Shellfish, Fish, and Seaweed (부유물질이 패류, 어류 및 해조류에 미치는 생물학적 영향에 관한 고찰)

  • In order to obtain basic informations about impact assessment of marine life by suspended solids in coastal waters, the survey about recent study related with suspended solids effects on exploitable marine life (shellfish, fish, seaweed.) was tried. 1) The paper concerning impacts of suspended solids on marine life has been mainly focused on shellfish. 2) Effects of suspended solids on marine life mainly investigated for acute toxicity. 3) Experiments with suspended solids are intended on adult and juvenile in shellfish, egg and juvenile in fish, and spores in seaweed. Biological impacts of suspended solids can vary depending on concentrations, duration, and life stage, and these informations would improve a knowledge of the effects of suspended solids on marine life.

The Growth of Vibrio vulnificus in Meat Homogenates of Fish and Shellfish (어패육에서의 Vibrio vulnificus의 증식에 관한 연구)

  • The change of cell counts of Vibrio vulnificus in meat homogenates of fish and shellfish by the storage time and temperature was examined to get basic information for precautionary steps against septicemia from slices of raw fish (sashimi). Therefore, we inoculated raw and cooked meat homogenates of fish and shellfish with Vibrio vulificus M-8 (isolated from shellfish ) and stored them at $-20^{\circ}C,\;4^{\circ}C\;and\;30^{\circ}C$ for 72 hours. Vibrio vulnificus M-8 was not detected in 32 hours when it was frozen and stored at $-20^{\circ}C$ after inoculating them into phosphate buffer solution at concentration of $10^5\;cell/ml$, while the existance of Vibrio vulnificus was identified after 72 hours of storage at the same temperature in case of inoculation into the meat homogenate of yellow tail. The cell count of Vibrio vulnificus was decreased as about $20\%$ of initial count after 2 hours storage at $4^{\circ}C$ in phosphate buffer solution with fish and shellfish homgenates. From the experimental results it was recognized that Vibrio vulnificus was labile to the cold stress. In comparison to the growth of growth of Vibrio M-8 at $30^{\circ}C$ in the raw and cooked meat of the yellow tail(Seriola guingueradita), snapper(Chrysophrys major), ark shell(Anadra brouhgtonii), and oyster(Crassostrea gigas), the raw meat homogenates were more excellent than the cooked ones though all fish and shellfish meat homogenates were proves to be good for the growth of the microbe.
