Characterization of Nutritional Value for Twenty-one Pork Muscles

  • A study was conducted to evaluate nutritional value for twenty-one pork muscles. Ten market-weight crossbred pigs (five gilts and five barrows) were used for evaluating proximate chemical composition, cholesterol, total iron, calorie and fatty acid contents. As preliminary analysis revealed no noticeable sex effect, pooled data from both sexes were used for the final analysis. M. rectus femoris had the highest moisture content, while m. latissimus dorsi was lowest in moisture content (p<0.05). Protein content was highest for m. longissimus dorsi and lowest for m. supraspinatus (p<0.05). The tensor fasciae and latissimus dorsi muscles contained the highest intramuscular fat (p<0.05), while rectus femoris, adductor and vastus lateralis were lowest in intramuscular fat content. When simple correlations between chemical values were computed for the pooled dataset from all muscles, intramuscular fat had significant (p<0.05) negative linear relationships with moisture (r = -0.85) and protein (r = -0.51) contents. Calorie levels were not significantly affected by fat content, while rectus femoris and latissimus dorsi muscles showed lowest and highest calorie contents, respectively (p<0.05). Polyunsaturated fatty acid content was highest (p<0.05) for both m. adductor and m. rectus femoris, while it was lowest for m. longissimus dorsi. Collectively, the current study identified a large amount of variation in nutritional characteristics between pork muscles, and the data can be used for the development of muscle-specific strategies to improve eating quality of meats and meat products.

Characteristics of Wet Distillers Grains on In vitro Ruminal Fermentation and Its Effects on Performance and Carcass Characteristics of Finishing Hanwoo Steers

  • Two experiments were conducted to determine the nutrient composition, in vitro ruminal ammonia concentrations and pH of wet distillers grains (WDG, produced from tapioca 70% and rice 30%) and to evaluate dietary effects of fermented total mixed ration (TMR) using WDG on the performance, blood metabolites and carcass characteristics of Hanwoo steers from mid fattening to slaughter. In Exp. I, average dry matter (DM), crude protein, ether extract, crude fiber, ash, neutral detergent fiber, acid detergent fiber, and nitrogen free extract of seven WDG samples from an ethanol plant with different sampling dates were 19.9%, 24.8%, 3.8%, 21.8%, 8.87%, 60.3%, 34.5%, and 40.7% (DM basis), respectively. For in vitro ammonia concentrations and pH, each sample was assigned to 7 incubation times (0, 4, 8, 12, 24, 48, and 72 h). Linear increase was observed between 12 and 48 h for ammonia concentrations, but final ammonia concentrations (72 h) were not significantly different among WDG samples and fermentation patterns of WDG samples showed similar tendency. In vitro pH varied among treatments from 0 to 24 h, but were not different statistically after 48 h. In Exp. II, 45 Hanwoo steers of 23 months ($641{\pm}123kg$) from mid fattening period to slaughter (248 days) were randomly divided into three groups of 15 pens each (five repetitions/each treatment) and assigned to one of three dietary treatments; i) Control (TMR), ii) WDG 15 (TMR containing 15% of WDG, as fed basis) and iii) WDG 28 (TMR containing 28% of WDG, as fed basis). The body weight (BW), ADG, and feed conversion ratio (FCR) of control and WDG 15 and 28 during 248 days were 760.8, 740.1, and 765.5 kg, and 0.50, 0.50, and 0.52 kg/d, and 18.6, 17.6, and 17.1, respectively. The dry matter intake (DMI) (kg/d) of control (9.11) was higher (p<0.05) than WDG treatments (WDG 15%, 8.57; 28%, 8.70). Nevertheless, DMI did not affect BW, ADG, and FCR of Hanwoo finishing steers. Blood metabolites were in normal ranges and were not different among treatments except the albumin concentration. In carcass characteristics, WDG 15 (30%) showed higher frequency of A-carcass yield grade than WDG 28 (15%) and control (7%), and WDG 28 (61%) showed higher frequency of $1^{{+}{+}}$ and $1^+$-carcass quality grade than WDG 15 (40%) and control (60%). In conclusion, using WDG up to 28% in TMR did not show any negative effect on the performance and blood metabolites, and improved carcass quality of Hanwoo steers. Therefore, WDG can be a useful feed ingredient for Hanwoo steers in mid-fattening period to slaughter.

Effect of increased dietary crude protein levels on production performance, nitrogen utilisation, blood metabolites and ruminal fermentation of Holstein bulls

  • Objective: This study investigated the effect of dietary crude protein (CP) supplementation on nutrient intake, nitrogen (N) utilisation, blood metabolites, ruminal fermentation and growth performance of young Holstein bulls. Methods: Twenty-one young bulls weighing $277{\pm}11.2kg$ were equally divided into three groups and were offered diets formulated with low CP (LCP; 10.21% CP and 4.22% rumen degradable protein [RDP]), medium CP (MCP; 12.35% CP and 5.17% RDP) and high CP (HCP; 14.24% CP and 6.03% RDP). Yellow corn silage was used as a unique forage source and was mixed with concentrate. This mixed feed was given ad libitum to the young bulls included in the study. Results: Results showed that CP intake, blood urea nitrogen, N intake, total N excretion and N balance increased linearly with an increase in dietary CP level (p<0.05). However, no significant difference was observed in nutrient digestibility among the bulls receiving the different diets. Ruminal pH (p<0.05) and ammonia nitrogen ($NH_3-N$) concentration (p<0.01) were significantly higher in the bulls receiving the MCP and HCP diets than in those receiving the LCP diet. The bulls receiving the HCP diet showed significantly higher ruminal bacterial protein level, propionate, acetate and total volatile fatty acid (TVFA) concentrations than bulls receiving the LCP diet (p<0.05). Moreover, dietary CP level exerted a significant positive effect on the final body weight, average daily gain and gain-to-feed ratio of the bulls (p<0.05). Conclusion: High dietary CP level is optimal for achieving maximum growth and high profitability without exerting a negative effect on the physiology of growing Holstein bulls.

폴리아닐린을 함유한 도전성 복합필름의 제조 및 특성 연구(1) (Characterization of Biodegradable Conductive Composite Films with Polyaniline(1))

  • 생분해성 고분자인 PLA를 매트릭스로 용액 중합된 DBSA가 15 mole% 정도 도핑된 PAni를 첨가하여 도전성 PAni/PLA 복합 필름을 제조하여 기계적, 전기적 특성 및 표면 morphology를 고찰하였다. FE SEM 측정 결과 PAni/PLA 복합 필름은 PAni 입자들이 매트릭스 고분자인 PLA에 비교적 잘 분산된 상태를 보였으며, 인장강도는 PAni 함유량 15 wt% 인 경우 $565.3kg_f/cm^2$에서 $309.7kg_f/cm^2$로 급격히 감소하였으며, 신율은 모든 필름에서 3-6%로 크게 변화하지는 않았다. 전기전도도는 PAni의 함량이 증가함에 따라 상승하였으며, PAni 함유량이 15 wt% 인 복합필름의 경우 $2.9{\times}10^{-3}$ S/cm의 전기전도도를 나타내었다. PAni 함유량에 따른 복합 필름의 열적 안정성은 $300^{\circ}C$ 이하에서는 약간 낮아졌으며, 고온 처리 후의 최종 잔존 타르의 함량은 PAni의 함량에 비례하였다. 결과적으로 PAni를 15 wt% 이상 함유한 복합필름은 전자파차폐 및 정전기방지 소재로 응용될 수 있다.

Streptomyces sp. Y9343이 生産하는 齒面細菌膜 分解酵素의 精製와 特性 (Purification and Characterization of Degradative Enzyme of Dental Plaque from Streptomyces sp. Y9343)

  • Streptococcus mutans has been implicated as primary causative agents of dental caries by insoluble glucan (IG) in human and experimental animals. An attempt was made to search for the ${\alpha}$-1,3 glucanase that degrades IG produced by S. mutans. ${\alpha}$-1,3 glucanase was detected in the culture supernatant of microorganisms, which are isolated from soils on agar medium containing IG as a sole carbon source. This Streptomyces sp. hydrolysed IG produced by immobilized S. mutans and was named as Y9373. This enzyme required ${\alpha}$-1,3 glucan (IG) as an inducer. The optimum conditions for enzyme production were studied. The enzyme was purified by 30~70% $(NH_4)_2SO_4$ precipitation, anion exchange chroma tography on DEAE-cellulose and gel filtration on Sepadex G-75. The purified enzyme has a specific activity of 7840.0 U/mg protein giving 32.1-fold purification and final yield of 0.53%. The molecular weight was estimated to be about 22.5 kDa by SDS-PAGE. The optimum pH and temperature for enzyme reaction were 6.5 and 37$^{\circ}C$, respectively and the enzyme was relatively stable at the temperature below 60$^{\circ}C$. The activity of purified enzyme was enhanced by adding $Co^{2+},\;Mn^{2+}\;and\;Mg^{2+}$ into the medium, whereas inhibited by adding $Hg^{2+},\;Zn^{2+}$ and SDS. The $K_m\;and\;V_{max}$ value of ${\alpha}$-1,3 glucanase for IG were estimated to be 2.50 mM and 0.0431 mM/min, respectively. The thin layer chromatographic analysis of hydrolysates from IG with ${\alpha}$-1,3 glucanase showed that glucose was the main product of reaction. This enzyme activity was about 14 times higher than marketing dextranase as preventive agent against artificial dental caries by S. mutans in TH medium including 5% sucrose after 30 minutes.

Chemical and Fatty Acid Composition of Longissimus Muscle of Crossbred Bulls Finished in Feedlot

  • This work was carried out to study the chemical and fatty acid composition of Longissimus muscle (LM) of crossbred young bulls finished in a feedlot. After weaning (at 8 months old), the bulls were kept in a feedlot for 180 days. The bulls were kept in individual pens and fed (twice daily) with corn silage, soybean hulls, cracked corn, limestone, urea and mineral salt. The bulls were slaughtered with a final weight of 464 kg. Forty bulls were used: 10 Caracu (CAR), 10 Canchim (CAN), 10 Caracu vs. Charolais (CCH) and 10 Canchim vs. Aberdeen Angus (CAA). The percentages of moisture, ash, crude protein, total lipids, as well as the fatty acid composition, were measured in the LM. The moisture percentage was lower (p<0.05) for bulls from CAA genetic group (71.2%) in comparison to bulls from CAR (74.2%), CAN (74.9%) and CCH (74.7%) genetic groups. On the other hand, there was no difference (p>0.05) among bulls from CAR, CAN and CCH genetic groups. Ash percentage was lower (p<0.05) for CAR bulls (0.96%) in comparison with the other genetic groups. There was no difference (p>0.05) among CAN, CCH and CAA genetic groups. Similarly, there was no difference (p>0.05) in crude protein among the different genetic groups. Total lipids percentage was higher (p<0.05) for CAA bulls (5.35%) and lower (p<0.05) for CAN (1.85%) and CCH (1.41%) genetic groups. Genetic group has little effect on the fatty acid composition of Longissimus muscle of bulls. However, CLA (C 18:2 c-9 t-11) percentage was higher (p<0.05) for CAR (0.33%) and CCH (0.37%) in comparison to CAN (0.27%) and CAA (0.29%) genetic groups. Saturated, monounsaturated and polyunsaturated fatty acids, n-6 and n-3 percentages did not differ (p>0.05) among genetic groups. PUFA/SFA ratio ranged from 0.10 to 0.15, with no difference (p>0.05) among genetic groups. Similarly, n-6/n-3 ratio ranged from 12.6 to 16.3, without difference (p>0.05) among genetic groups.

Effects of different space allowances on growth performance, blood profile and pork quality in a grow-to-finish production system

  • Objective: This experiment was conducted to evaluate the optimal space allowance on growth performance, blood profile and pork quality of growing-finishing pigs. Methods: A total of ninety crossbred pigs [$(Yorkshire{\times}Landrace){\times}Duroc$, $30.25{\pm}1.13kg$] were allocated into three treatments (0.96: four pigs/pen, $0.96m^2/pig$; 0.80: five pigs/pen, $0.80m^2/pig$; 0.69: six pigs/pen, $0.69m^2/pig$) in a randomized complete block design. Pigs were housed in balanced sex and had free access to feed in all phases for 14 weeks (growing phase I, growing phase II, finishing phase I, and finishing phase II). Results: There was no statistical difference in growing phase, but a linear decrease was observed on average daily gain (ADG, p<0.01), average daily feed intake (ADFI, p<0.01), and body weight (BW, p<0.01) with decreasing space allowance in late finishing phase. On the other hand, a quadratic effect was observed on gain to feed ratio in early finishing phase (p<0.03). Consequently, overall ADG, ADFI, and final BW linearly declined in response to decreased space allowance (p<0.01). The pH of pork had no significant difference in 1 hour after slaughter, whereas there was a linear decrease in 24 h after slaughter with decreasing space allowance. Floor area allowance did not affect pork colors, but shear force linearly increased as floor space decreased (p<0.01). There was a linear increase in serum cortisol concentration on 14 week (p<0.05) with decreased space allocation. Serum IgG was linearly ameliorated as space allowance increased on 10 week (p<0.05) and 14 week (p<0.01). Conclusion: Data from current study indicated that stress derived from reduced space allowance deteriorates the immune system as well as growth performance of pigs, resulting in poor pork quality. Recommended adequate space allowance in a grow-to-finish production system is more than $0.80m^2/pig$ for maximizing growth performance and production efficiency.

Effects of feeding corn naturally contaminated with aflatoxin on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of Na+, K+-ATPase in ducklings

  • Objective: A 14-d trial was conducted to determine the effects of feeding corn naturally contaminated with aflatoxin $B_1$ ($AFB_1$) on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of $Na^+$, $K^+-ATPase$ in ducklings. Methods: A total of 704 ducklings were blocked on the basis of sex and body weight (BW), and then allocated randomly to one of the following two treatments: i) CON, basal diet and ii) $AFB_1$, diets with 100% of normal corn replaced with $AFB_1$ contaminated corn. There were 22 pens per treatment and 16 birds per pen. The concentration of $AFB_1$ was 195.4 and $124.35{\mu}g/kg$ in the contaminated corn and $AFB_1$ diet, respectively. Results: The $AFB_1$ decreased average daily gain, average daily feed intake, d 7 BW, final BW in the whole trial, and feed conversion ratio (FCR) during d 8 to 14 and d 1 to 14 by 10% to 47% (p<0.05), while FCR during d 1 to 7 was increased (p<0.05). $AFB_1$ did not affect mortality to 7 d of age, and then increased to 5.8% from 8 to 14 d of age (p<0.01). Apparent ileal gross energy digestibility was reduced by $AFB_1$, whereas apparent ileal digestibility of dry matter, nitrogen, and amino acid was improved (p<0.01). Feeding $AFB_1$ diets increased serum concentration of leptin and insulin-like growth factors-1 (IGF-1) (p<0.05), but had no effect on neuropeptide Y, ghrelin, cholecystokinin-8 or insulin (p>0.05). Dietary treatments did not influence relative expression of jejunal $Na^+$, $K^+-ATPase$ gene (p>0.05). Conclusion: Taken together, feeding corn naturally contaminated with $AFB_1$ reduced growth performance, improved apparent ileal digestibility, and affected serum leptin and IGF-1 in ducklings from d 1 to 14.

Effects of processing, particle size and moisturizing of sorghum-based feeds on pellet quality and broiler production

  • Objective: This study aimed to assess the effect of pelleted and expanded sorghum-based feeds prepared with different moisture levels and particle size of ingredients on metabolizable energy, ileal digestibility of amino acids and broiler performance. Methods: The experiment was performed with 720 male broiler chicks of the Cobb strain, with treatments of six replications, with 15 birds each; they were arranged in a completely randomized design and $2{\times}2{\times}2$ factorial scheme (pelleted or expanded feed processing, 0.8% or 1.6% moisture addition in the mixer, and particle size of 650 or 850 microns). Results: Higher pellet quality (pellets, % and pellet durability index [PDI]) was obtained in expanded diets and inclusion of 1.6% moisture. The particle size of 850 microns increased the PDI of final diet. All studied treatments had no significant effect on weight gain and broiler carcass and cut yields. Lower feed conversion occurred for birds fed pelleted feed at 42 d. The highest apparent metabolizable energy (AME) and apparent metabolizable energy corrected to zero nitrogen balance (AMEn) values of feed in the initial rearing phase (10 to 13 days) were observed in birds fed pelleted feed or for feed prepared with 1.6% moisture. The highest ileal digestibility coefficients of amino acids were obtained with the consumption of pelleted feed prepared with a particle size of 650 microns and 1.6% moisture. Conclusion: Pelleted feed prepared with a milling particle size of 650 microns and 1.6% moisture provided increased ileal digestibility of amino acids and AMEn in the starter period. However, the expanded feed improved pellet quality and feed conversion of broilers at 42 days of age. We conclude that factors such as moisture, particle size and processing affect the pellet quality, and therefore should be considered when attempting to optimize broiler performance.

Osmotic Priming 및 Solid Matrix Priming 처리에 의한 당근과 양파종자의 발아증진과 초기생육에 미치는 영향 (Effect of Osmotic and Solid Matrix Priming to Improve Germination and Early Growth of Carrot and Onion Seed)

  • 본 연구는 당근과 양파에서 발아력 증진에 미치는 osmotic priming과 SMP효과를 비교하기 위해 수행되었다. Osmotic priming과 SMP 처리과정중 작물별 수분흡수율은 처리 1시간 후 급속한 수분흡수가 이루어졌고, 두 작물 모두 osmotic priming이 SMP보다 수분흡수 속도가 빨랐다. 이러한 경향은 처리 4시간까지 유지되었다. 처리최종일의 수분흡수율은 당근과 양파에서 osmotic priming이 SMP보다 2%높았다. Osmotic priming과 SMP 처리는 두 작물 모두 발아율을 향상시키지 못했지만 $T_{50}$ 및 MDG는 단축되어 조기발아 하였으며, 이러한 경향은 저온인 $15^{\circ}C$에서 더욱 현저하였다. Osmotic priming과 SMP 처리 후 초기함수율로 재건조하면 당근에서는 발아속도에 큰 차이가 없었으나, 양파에서는 표면건조에 비해 발아속도가 지연되었다. Osmotic priming과 SMP 상호처리간 발아력을 비교한 결과 두 작물 모두 SMP 처리가 osmotic priming보다 발아촉진에 효과적이었다. 당근과 양파종자를 osmotic priming과 SMP 처리하면 묘출현율도 향상되었고, 묘출현에 소요되는 일수를 단축시켜 신속한 묘출현을 유도하였다. 그러나 묘출현에 이은 유묘의 초기생육에는 통계적인 유의성은 인정되지는 않았으나 osmotic priming과 SMP 처리된 종자는 무처리에 비해 향상되었다.
