Background: The FimA of Porphyromonas gingivalis is a crucial pathogenic component of the bacteria and has been implicated as a target for vaccine development against the periodontal diseases. Methods: In this study, the purified fimbriae (FimA subunit polymers) protein was used for immunization in their native form and B hybridoma clones producing antibodies specific to FimA were established. Results: The monoclonal antibodies prepared from selected two clones, designated #123 (IgG2b/ kappa) and #265 (IgG1/kappa), displayed different patterns of binding activity against the cognate antigen. Both antibodies reacted with conformational epitopes expressed by partially dissociated oligomers, but not with monomer as elucidated by Western blot analysis. Ascites fluid containing the monoclonal antibodies showed the inhibitory activity against P. gingivalis to saliva-coated hydroxyapatite beads, an in vitro model for the pellicle-coated tooth surface. Conclusion: These results suggest that the monoclonal antibodies could be used as vaccine material against the periodontal diseases through passive immunization.

Staining profiles and bacterial morphology were compared in Xanthomonas citri pv. citri by a transmission electron microscopy. Four types of negative staining regimes were employed depending on culture media and heavy metal stains. The bacterial cells grown on LB agar media often appeared clustered on the supporting film. Meanwhile, individual bacterial cells could be readily found on the preparations from LB broth media. Typical rod-shaped cells (ca. $1\;{\mu}m$ in length) and their flagella were observed in either 2% uranyl acetate (UA) or 2% neutralized potassium phosphotungstate (PTA) staining. The UA-stained bacteria often showed relatively intact cell morphology and rather positively stained cells with a thin electron-dense stain depth around bacteria. The PTA-stained bacteria were characterized by the wrinkled cell surface where the stain was entrapped in grooves. In addition, distinct electron-dense stain depth was evident around the PTA-stained preparations. Numerous fimbriae could be mostly observed from the PTA-stained preparations of the two culture media, but not from the UA-stained preparations.
This paper deals with the isolation of Salmonella on 3 herds during the period from May 1984 to May 1985. Isolated Salmonella were examined for serotypes, adhesive fimbriae, antibiotic susceptibility and detection of R plasmid. The results were as follows; Of total 1505 cattle, 24 Salmonella were isolated from 18 cattle (1.2%) and their serotypes were S. enteritidis (9 strains), S. derby(4 strains), S. infantis (1 strain), $C_1$ group(8 strains), $C_2$ group(1 strain) and untypable(1 strain). Twelve strains of Salmonella were isolated from 227 cattle with diarrhea and their serotypes were S. enteritidis(4 strains), S. derby(3 strains), S. infantis(1 strain), $C_1$ group(3 strains) and untypable(1 strain). The isolation rate of Salmonella in cattle varied from 1.6 to 0.9% in 3 herds, it was higher in summer and autumn and lower with the age. Of total 24 strains, 23 were adhesive type 1 fimbriae. Antibiotic susceptibility test of Salmonella isolated was performed by the agar dilution method, using 9 antibiotics as follows: ampicillin(Am), chloramphenicol(Cm), gentamicin(Gm), kanamycin(Km), nalidixic acid(Na), rifampicin(Rf), streptomycin(Sm), sulfadimethoxine(Su) and tetracycline(Tc). All the strains tested were susceptible to Am, Cm, Gm, Km, Na, Rf and Tc. Of total 24 strains, 23(95.8%) were resistant to Su and 14(58.3%) to Sm. Of the 23 resistant Salmonella strains, all the strains were found to carry R plasmid. Among them, two strains which had the R plasmid conferring SmSu resistance was transferable at $25^{\circ}C$ only.
Fimbriae (fimA) of Porphyromonas gingivalis are filamentous components on the cell surface and are thought to play an important role in the colonization and invasion of periodontal tissue. P. gnigivalis fimA gene encoding fimbrillin, a subunit of fimbriae, has been classified into 5 genotypes (types I to V) based on the nucleotide sequences. In the present study, we examined the prevalence of these fimA genotypes in patients with dental implant and the relationship between prevalence of these genotypes and peri-implantitis. Dental plaque specimens obtained from 80 peri-implant sulci of 50 patients with dental implants were analyzed by 16S rRNA fimA gene-directed PCR assay. P. gingivalis were detected in 74.4% of the samples of the control group (healthy peri- implant sulci; probing depth<5mm) and in 92.0% of the samples of the test group (peri-implant sulci with peri-iimplantitis; probing $depth{\geqq}5mm$). Among the P. gingivalis-positive samples of the control group, the most prevalent fimA type was type I (29.3%), followed by type II (26.8%). In contrast, a majority among the P. gingivalis-positive samples of the test group was type II (56.S%), followed by type I (43.5%). TypeII fimA genotype organisms were detected more frequently in the test group and a significant difference in the occurrence of type II was observed between test and the control groups. A correlation between specific fimA types and peri-implant health status was found in type II (OR 3.545) and only a weak relationship was revealed in typeIV(OR 3.807). These findings indicate that P. gingivalis strains that possess type II fimA are predominant in peri-implant sulci with peri-implantitis and are closely associated with peri-implant health status. P. gingivalis with type II fimA may be involved in peri-implantitis.
본 연구는 세 종류의 Porphyromonas gingivalis(Pg) antigen의 IgG subclass associated recognition을 평가하기 위해 수행했었다. 총 35명의 조기발병형치주질환자중, Pg381에 대한 IgG2항체의 증가를 보이는 5명이 급속진 행형 치주질환자, IgG4의 증가를 보이는 6명의 환자(국소유년형 치주질환자 2명과 급속진행 형 치주질환자 4명), IgG2+4의 증가를 보이는 2명의 급속진행형 치주질환자 그리고 IgG1+2+4의 증가를 보이는 8명의 환자(국소 유년형 치주질환자 2명과 급속진행형 치주질환자 6명)으로 구성된 21명의 환자를 dot immunoblot analysis를 위해 선택했다. 실험에 사용된 정제된 항원은 Pg381에서 추출한 43-kd fimbrilin protein과 lipoplysaccharide(LPS), Pg A7A1-28(ATCC 53977)에서 추출한 capsularpolysaccharide(CPS)였다. Immunoblotting pattern은 IgG4 antibody가 fimbrial antigen에 강력히 반응함을 보여주었다. Fimbriae에 잘 반응하는 몇몇의 IgG4 antibody역시 antigen에 대해 양성반응을 보였다. 대조적으로 IgG2는 CPS antigen을 일차적으로 인식했다. 전부는 아니지만 대부분의 경우, single이나 group화된 IgG subclass는 모두 LPS antigen을 인식하지 못했다. 같은 group에서 염색강도의 개인적인 차이는 증명되었다. 이런 결과는 조기발 병형 치주질환에서 Pg의 fimbriae와 CPS가 immunodominant antigen이 될 수 있음을 제시한다. 더욱이 IgG subclass antibody가 이런 Pg의 immunoglobulin antigen을 선택적으로 인식함을 알았고, 이는 조기발병형치주질환의 병리에 immunodominant antigen과 함께 IgG의 기능적인 역할을 고려해야 함을 제시한다.
Image contrast of whole bacteria was compared in Staphylococcus aureus and Escherichia coli depending on pre-stain suspension liquids by energy-filtering transmission electron microscopy. The two bacterial strains were suspended in three most commonly used liquids for negative staining (triple distilled water [DW], phosphate-buffered saline [PBS], and nutrient broth [NB]) and directly observed without staining or stained with neutralized potassium phosphotungstate (PTA), respectively. Even though in low contrast, unstained bacteria were observed owing to their inherent electron density and cell shape in zero-loss (elastic scattering) images. After being suspended in PBS, unstained bacteria appeared to have higher contrast and more refined periphery than DW-suspended ones, and extracellular appendage structures such as fimbriae and flagella could be discerned. The unstained bacteria appeared to be invariably surrounded with electron-lucent precipitates, possibly from PBS. As far as delineation of the structures, the combination of DW or PBS suspension with subsequent staining provided the most satisfactory results, as evidenced by the high contrast of bacterial morphology and appendage structures. However, after being suspended in NB and stained with PTA, bacteria often had too high contrast or poor staining, with electron-dense aggregates around the bacteria. These results suggest that suspension with concentrated organic aliquots including broth media before PTA staining could deteriorate image contrast, and should be used only in dilute form for visualizing bacterial morphology and appendage structures. Moreover the contrast enhancement of unstained bacteria by salt granules would be advantageous in demonstrating bacterial sorption of environmental particles like heavy metals, maintaining minimal contrast for cell imaging.
Porphyromonas gingivalis is a gram negative. black-pigmented anaerobe, associated with periodontitis & peri-implantitis. P. gnigivalis fimA gene encoding fimbrillin(structural subunit of fimbriae) has been classified into 5 genotypes(types I to V) based on the nucleotide sequences. In the present study, we examined the prevalence of these fimA genotypes in patients with dental implant and the relationship between prevalence of these genotypes and a condition of peri-implant tissue. Dental plaque specimens obtained from 189 peri-implant sulci of 97 patients with dental implants were analyzed by 16S rRNA fimA gene-directed PCR assay. P. gingivalis were detected in 86.2% of the alll samples. Among the P. gingivalis-positive samples, a significant difference in the occurrence of typeII was observed between test and the two control groups. In two control groups, typeII fimA were detected in 6.3%(PD<5mm/BOP-). 18.7%(PD<5mm/BOP+). In the test $group(PD{\geqq}5mm/BOP+)$, type II fimA genotype were detected most frequently in 50.0% . And a correlation between specific fimA types and peri-implantitis was found in $typeII(R^2=l.105)$. These results suggest that P. gingivalis strains that possess typeII fimA are gradually increased, as a condition of peri-implant tissue is getting complicated and are closely associated with peri-implant health status. Cocconeis coreana and C. sijunghoensis are described as new based on micromorphological and molecular data. C. coreana is represented by five unialgal cultures from four different freshwater bodies, two from North Korea and three from South Korea. C. sijunghoensis is represented by two unialgal cultures from a brackish water body in North Korea. Except for one, all of the strains auxosporulated and showed an almost quadrupling of size in length and width. Morphologically, these species with their two different elliptical valves belong to the Cocconeis placentula group. The raphe valve has striae with uniseriate areolae continuing across a pronounced submarginal hyaline rim to the edge of the valve. The sternum valve has uniseriate dash-like areolae continuously from the valve face until the valve edge. Micromorphologically, these species possess two different open valvocopulae: only the raphe valvocopula has fimbriae; the sternum valvocopula has none. Based on p-distances of currently available DNA sequence data, i.e., rbcL and 18SV4, both species are pronouncedly different from the epitype strain of C. placentula, with C. coreana closest to the published molecular data of the strain UTEX FD23 named C. placentula from Iowa, USA, while C. sijunghoensis is closest but not the same as the published molecular data of strain D36_012, the epitype strain of C. placentula from Berlin, Germany. Based on scanning electron microscope observations, differentiating features are discussed concerning valvocopula fimbriae, central area, areolation of the sternum valve and on the raphe valve especially between the submarginal hyaline rim and edge.
