Characterization of a peroxidase in excretory-secretory product of adult Parasonimus westermani (폐흡충 성충이 분비배설하는 anti-oxidant ensymes의 특성 관찰 및 peroxidase의 정제)

  • When activity of peroxidase in auld Pnrqfonimn westermqni was monitored using o-dianisidine and $H_2O_2$ as substrates, its specific activity was 1.5 times higher In excretory-secretory product (ESP) than in crude extract. The one was purified by two purification steps of Sephacryl S-300 Superfine gel permeation and DEAE-Trisacryl M anion exchange chromatographies. Its activity increased 16.9 fold with 32.3% recovery. The enzyme was inhibited totally by 1 millimoles of dithiothreitol (DTT), 2-mercaptoethanol and azide. Molecular mass was 16 kDa in reducing SDS-polyacrylamide gel electrophoresis (PAGE) or 19 kDa in TSK-Blue gel filtration high performance liquid chromatography (HPLC). respectively. Special staining for peroxidase by diaminobenzidine on SDS-PAGE confirmed the activity. The peroxidase was less reactive to a paragonimiasis serum when observed by SDS-PAGE/immunoblot. In addition, specific activities of superoxide dismutase (SOD) and catalase were also identified in the ESP. High activities of these antioxidant enzymes in ESP indicate that they are parts of defense mechanisms against reactive oxygen intermediates from host.

Strength properties of composite clay balls containing additives from industry wastes as new filter media in water treatment

  • Pebble matrix filtration (PMF) is a water treatment technology that can remove suspended solids in highly turbid surface water during heavy storms. PMF typically uses sand and natural pebbles as filter media. Hand-made clay pebbles (balls) can be used as alternatives to natural pebbles in PMF treatment plants, where natural pebbles are not readily available. Since the high turbidity is a seasonal problem that occurs during heavy rains, the use of newly developed composite clay balls instead of pure clay balls have the advantage of removing other pollutants such as natural organic matter (NOM) during other times. Only the strength properties of composite clay balls are described here as the pollutant removal is beyond the scope of this paper. These new composite clay balls must be able to withstand dead and live loads under dry and saturated conditions in a filter assembly. Absence of a standard ball preparation process and expected strength properties of composite clay balls were the main reasons behind the present study. Five different raw materials from industry wastes: Red Mud (RM), Water Treatment Alum Sludge (S), Shredded Paper (SP), Saw Dust (SD), and Sugar Mulch (SM) were added to common clay brick mix (BM) in different proportions. In an effort to minimize costs, in this study clay balls were fired to $1100^{\circ}C$ at a local brick factory together with their bricks. A comprehensive experimental program was performed to evaluate crushing strength of composite hand-made clay balls, using uniaxial compression test to establish the best material combination on the basis of strength properties for designing sustainable filter media for water treatment plants. Performance at both construction and operating stages were considered by analyzing both strength properties under fully dry conditions and strength degradation after saturation in a water bath. The BM-75% as the main component produced optimum combination in terms of workability and strength. With the material combination of BM-75% and additives-25%, the use of Red Mud and water treatment sludge as additives produced the highest and lowest strength of composite clay balls, with a failure load of 5.4 kN and 1.4 kN respectively. However, this lower value of 1.4 kN is much higher than the effective load on each clay ball of 0.04 kN in a typical filter assembly (safety factor of 35), therefore, can still be used as a suitable filter material for enhanced pollutant removal.

A Feasibility Study on Sewage Discharge Water Treatment for Water Reuse by Direct Contact Membrane Distillation (하수처리수 재이용을 위한 직접접촉식 막증발법 적용 가능성 연구)

  • The reuse of wastewater is being diffused to manage and develop the water resources. Generally, the treated wastewater is discharged to the river after being treated to meet the effluent quality standard or reused for diverse uses through the reprocessing. And recently, as the reuse of the treated wastewater is activated, the technologies to utilize for the high quality water resources such as industrial water by reusing the wastewater with Membrane Distillation (MD) are under development. In this study, the direct contact membrane distillation (DCMD) process has been applied to treat sewage discharge water for water reuse. The laboratory scale experiment was performed by using a hydrophobic PVDF membrane with the pore size of $0.22{\mu}m$. The influence of operating parameters, such as feed temperature, feed flow rate, feed concentration, on the permeate flux and rejection has been investigated. All filtration tests were conducted till the feed volume reached a concentration factor of 3.0. Thus, the operating period ranged between 19 hr and 49 hr depending on filtration performance. The results showed that above 92% of TN, TP, COD and TOC in the feed could be rejected regardless of an operating condition. The water flux was ranged from 13.8 to 20.3 LMH. The lowest flux was obtained at the operating condition with the feed temperature of $50^{\circ}C$ and feed flow velocity of 500 mL/min while the highest one was measured with $60^{\circ}C$ and 900 mL/min. When the concentration factor reached 3.0, water flux declined by approximately ranged from 14.5% to 33.3%. But the fouling in MD is almost fully reversible, with more than 90% recovery of permeate water flux following a DI water rinse without the addition of chemical cleaning reagents.

In-situ TiO2 Formation and Performance on Ceramic Membranes in Photocatalytic Membrane Reactor (광촉매 반응기용 세라믹 막에의 TiO2 층 형성과 성능평가)

  • Fabricating photocatalytic composite membrane with a mesoporous and tailored morphological structure would have significant implication for environmental remediation. In this study, we reported hybrid $TiO_2$ immobilized photocatalytic membrane and its application for the treatment of dye solution. Photocatalytic film with high porosity and homogeneity was fabricated by graft copolymer as polymer template. Hybridization of membrane filtration with photocatalysis was successfully achieved by photocatalytic membrane reactor developed. Result showed that membrane permeability was significantly reduced after immobilizing the $TiO_2$ film on bare $Al_2O_3$ support. The membrane characterization indicated that well organized $TiO_2$ film was successfully formed on $Al_2O_3$ support. Benefiting from the controlled morphology of $TiO_2$ film, the composite membrane exhibited almost complete degradation of organic dye within 5 h of filtration under UV illumination. Langmuir-Hinshelwood model explained degradation of organic dye. First-order rate constant was approximately six times with $TiO_2$ immobilized composite ceramic membrane, higher than the one with the bare $Al_2O_3$ support (0.0081 vs. $0.0013min^{-1}$).

Purification and Characterization of Antioxidative Peptides from Enzymatic Hydrolysate of Cod Teiset Protein (대구고니 단백질의 효소적 가수분해물로부터 항산화성 펩타이드의 분리${\cdot}$정제 및 특성)

  • In order to utilize by-products which would normally be discarded in marine processing plants, cod teiset protein was hydrolyzed and antioxidative actiTity of the hydrolysate was investigated. AntioxidatiTe peptide was isolated using ultrafiltration membrane, ion-exchange chromatography on a SP-Sephadex C-25 column, gel filtration on a Sephadex G-15 column, high performance liquid chromatography on an ODS column, and capillary electrophoresis chromatography. Antioxidative activities of the cod teiset hydrolysate were compared with ${\alpha}-tocopherol$, one of the commercial antioxidant. The hydrolysate passed through a membrane with molecular weight cut-off (MWCO) 1 kDa was shown the strongest antioxidative activity, and the activity was higher $10{\%}$ as compared with ${\alpha}-tocopherol$. In addition, the peptide isolated by ion-exchange chromatography, gel filtration, and HPLC, respectively, was higher $53{\%}$ as compared with ${\alpha}-tocopherol$, and the amino acid sequence was Ser-Asn-Pro-Glu-Trp-Ser-Trp-Asn.

Effects of Membrane Filtration Concentrate of Sewage Reuse Facility on Performance of Bioreactor in Sewage Treatment Facility (하수재이용시설 농축수가 하수처리장 생물반응조 운영에 미치는 영향)

  • This study investigated the effect of concentrate of membrane filtration of sewage reuse facilities on bioreactor's microorganisms in sewage treatment facility, and concentrate was analyzed, oxygen uptake rate and continuous activated sludge experiment were performed. As a results, the concentration of organic matter and TP concentration in concentrate was closely related to the concentration of SS and the concentration of ionic substances in concentrate was higher in order of $Cl^-$ > $Na^+$ > $Ca^{2+}$ > $K^+$ > $Mg^{2+}$ > $F^-$. And the analysis value of analytical items was greatly fluctuated according to sampling time of concentrate. Result of OUR test according to the mixing ratio of concentrate to sewage, it was found that the concentrate acts as an organic matter in the activated sludge microorganism and it increases with the increase of the concentrate mixing ratio. As a result of continuous activated sludge experiment, MLSS concentration, organic removal efficiency and TN removal efficiency gradually decreased with increasing concentrate mixing ratio. Based on the experimental results, expected mixing ratio of concentrate to affect the treatment efficiency of activated sludge is 25%.

  • The application of electro-coagulation has been attempted to control the membrane fouling problem in a MBR (Membrane Bio-Reactor). This study examined the effects of the operating parameters (current density and contact time) of the electro-coagulation process on the change in the characteristics of activated sludge. The current density changed from 2.5 to 12, $24A/m^2$, and the contact time was varied from 0 to 2 and 6 hr, respectively. At a current density of $24A/m^2$ and 6 hr of operation, the MLSS changed from 6,800 to 7,000 mg/L (3% increase), but the MLVSS did not increase significantly. After 6 hr of operation, the soluble COD decreased from 71 to 37 mg/L under the $24A/m^2$ condition, from 113 to 67 mg/L under the $12A/m^2$ condition, and from 84 to 80 mg/L under the $2.5A/m^2$ condition. On the other hand, soluble-TN and -TP concentration showed slight changes. The soluble-EPS and Bound-EPS concentration decreased slightly with increasing current density. The membrane filtration performance of activated sludge before and after electro-coagulation was compared. The filtration resistances after electro-coagulation decreased from 6 to 61 %, particularly as the current density and contact time were increased. This indicates that electro-coagulation can be used to control membrane fouling in the MBR process.

Biotreatment Technologies for Air Pollution Control (생물학적 처리기술을 이용한 대기오염 제어)

  • Biological treatment is a relatively recent air pollution control technology in which off-gases containing biodegradable odors and volatile organic compounds(VOCs) are vented through microbes. It is a promising alternative to conventional air pollution control methods. Bioreactors for air pollution control have found most of their success in the treatment of dilute and high flow waste air streams containing VOCs and odor compounds. They offer several advantages over traditional technologies such as incineration or adsorption. These include lower treatment costs, absence of formation of secondary pollutants, no spent chemicals, low energy demand and low temperature treatment. The three most widely used technologies are described, namely biofiltration, biotrickling filtration, bioscrubbing. The most widely used bioreactor for air pollution control is biofilter, but it has several limitations. In the past years major progress has been accomplished in the development of vapor phase bioreaction systems, for solving problems of biofilter. Biotrickling filters are more complex than biofilters, but are usually more effective, especially for the treatment of compounds which are difficult to degrade or compounds that generate acidic by-products. This, paper reviews fundamental and theoretical/practical aspect of air pollution control in biofilter, biotrickling filter and bioscrubber, focusing more extensively on biotrickling filtration. Special emphasis is given to the operating parameters and the factors influencing performance for air pollution control, and cost estimation in biotreatment technologies.

Use of Hydrogen Peroxide with Ozone to Simultaneously Reduce MIB and Quench Ozone Residual in Existing Water Treatment Plants Sourcing Water from the Han River (한강을 원수로 하는 오존/과산화수소 고도정수처리공정에서의 MIB제거 및 잔류오존 농도에 관한 연구)

  • This paper provides a detailed account of pilot testing conducted at South Lake Tahoe (California), the Ddukdo (Seoul) water treatment plant (WTP) and the Bokjung (Seongnam) WTP between February, 2010, and February, 2012. The objectives were first, to characterize the reactions of ozone with hydrogen peroxide (Peroxone) for Han River water following sand filtration, second to determine empirical ozone and hydrogen peroxide doses to remove a taste-and-odor surrogate 2-methylisoborneol (MIB) using an advanced oxidation process (AOP) configuration and third, to determine the optimum dosing configuration to reduce residual ozone to a safe level at the exit of the process. The testing was performed in a real-time plant environment at both low- and high seasonal water temperatures. Experimental results including ozone decomposition rates were dependent on temperature and pH, consistent with data reported by other researchers. MIB in post-sand-filtration water was spiked to 40-50 ng/L, and in all cases, it was reduced to below the specified target level (7 ng/liter) and typically non-detect (ND). It was demonstrated that Peroxone could achieve both MIB removal and low effluent ozone residual at ozone+hydrogen peroxide doses less than those for ozone alone. An empirical predictive model, suitable for use by design engineers and operating personnel and for incorporation in plant control systems was developed. Due to a significant reduction in the ozone reaction/decomposition at low winter temperatures, results demonstrate the hydrogen peroxide can be "pre-conditioned" in order to increase initial reaction rates and achieve lower ozone residuals. Results also indicate the method, location and composition of hydrogen peroxide injection is critical to successful implementation of Peroxone without using excessive chemicals or degrading performance.

Characteristic of the Permeation Flux of Hollow Fiber Membranes by Process Pressures Change (공정압 변화에 따른 중공사막의 투과플럭스 특성)

  • This study was carried out to evaluate the performance of the separate membrane (HF; hollow fiber membrane with polysulfone) process applied with the external membrane types, internal pressure membrane types and external-internal types according to the variations of pressure and membrane pore size in the purification treatment process of the lake water. The maximum permeate flux was average values of 282 LMH and 234 LMH with the pore size of 0.3 and 0.05 ${\mu}m$ respectively in the external pressure membrane process, and 443 LMH and 522 LMH with the pore size of 0.3 and $0.05{\mu}m$ respectively in the internal pressure membrane process. In addition, the maximum permeate flux of the process that was applied with external and internal membrane pressure simultaneously showed the average values of 674 LMH with the pore size of $0.3{\mu}m$, and 648 LMH with the pore size of $0.05{\mu}m$. Therefore, maximum yield per unit area is supposed when the separate membrane that was applied with external and internal pressure simultaneously are used to treat the lake water.
