During the screening or antiviral substances having inhibitory effect on tobacco mosaic virus(TMV) infection to tobacco plants, we found that a bacterial isolate, KTB61, which was identified as a Pseudomonas sp., strongly inhibited the formation of TMV local lesions. When the culture filtrate from KTB61 was applied on the upper surface of leaves of N. tabaccum Xanthi-nc tobacco at the same time of or 24 hours before TMV inoculation, almost complete inhibition was achieved. Incidence of systemic TMV infection to the susceptible tobacco cultivar, NC82, was reduced by 95% when TMV was inoculated onto the upper surface of leaves 24 hours after spraying the culture filtrate. Also 75∼80% of inhibitory effect was obtained by the inoculation of TMV onto the under surface of the leaves treated with culture filtrate 24 hours beforehand. In field trials, the TMV infection was reduced by 96.5% when the tobacco seedlings, N. tabaccum cv. NC82, were soaked with culture filtrate before transplanting.
To recover protopectinase (PPase) secreted from Bacillus subtilis IFO 12113, culture filtrate of the microorganism was treated with acetone, methanol, and ethanol, respectively. In the case of treatment with acetone at a ratio of 1: 1 (culture filtrate: acetone, v/v), PPase was purified 1.7-fold with 59.2% recovery The recovery of PPase was increased by increasing the acetone concentration. PPase was purified 4-fold with 100% recovery when the culture filtrate was precipitated with methanol at a ratio of 1 : 2 (culture filtrate: methanol, v/v). However, recovery of PPase was decreased by increasing the methanol concentration. PPase was purified 13.5-fold resulting in 68% recovery by the addition of ethanol with the final ratio 1 : 1(culture filtrate: ethanol, v/v) to the supernatant, which was obtained after precipitation of the culture filtrate with ethanol at a ratio of 1 : 0.5. These results show that methanol treatment is better than other organic solvent treatments for the simple recovery of PPase, whereas fractionated treatment of ethanol can recover PPase with higher purification fold.
Using the filtrate of loess suspension, we cultivated soybean sprouts and investigated its effect on growth and quality in soybean sprouts. In comparison with soybean sprouts cultivated by tap water, the soybean sprouts cultivated by the filtrate of loess suspension at $20^{\circ}C$ showed increases in its weight by 11.4% and length by 14.9%. When cultivated at $25^{\circ}C$, the soybean sprouts by filtrate of loess suspension also showed increases in its weight by 9.9% and length by 11.0%. We compared inorganic element contents and pH level between the filtrate of loess suspension and tap water. Contents of Ca, Mg, K, Na, Zn, Mn, and Cu did not show any difference, while only P was higher in the tap water. pH value did not show much difference either. Consequently, it seemed that inorganic element contents and pH in the filtrate of loess suspension did not give any effect on the growth of the soybean sprouts. And also there was no any significant difference in inorganic element and amino acid contents in two kinds of soybean sprouts. However in a sensory test, the color and overall acceptability of the soybean sprouts cultivated by the filtrate of loess suspension showed better than the soybean sprouts cultivated by the tap water.

The purpose of this study is to evaluate a de-watering filtrate treatment and the possibility of securing biological treatment capacity by changing the structure of the secondary clarifier. Accordingly, the column test was conducted to determine the effect of polymer in the de-watering filtrate on sludge sedimentation. Also, the characteristics of de-watering filtrate processing was evaluated through batch test and continuous processing operation. The results showed that sludge settling velocity increased with higher polymer concentration, and that effluent SS concentration was found to decrease. Regarding processing characteristics of de-watering filtrate, the removal efficiency of TSS and TBOD5 increased as the length of secondary clarifier was longer. Also, comparing injections into anoxic tank and secondary clarifier, de-watering filtrate by continuous infusion treatment process showed stability in both conditions. Therefore, by modifying the structure of secondary clarifier, efficient processing of de-watering filtrate is expected to be possible and processing capacity of small sewage treatment plants is considered to be improved.
Background: Diagnosis by direct microscopy and/or by culture of the Mycobacterium tuberculosis from body fluids or biopsy specimens is "Gold standard". However, the sensitivity of direct microscopy after Ziehl-Neelsen staining is relatively low and culture of mycobacteria is time consuming. Detection of mycobacterial DNA in clinical samples by the polymerase chain reaction is highly sensitive but laborious and expensive. Therefore, rapid, sensitive and readily applicable new tests need to be developed. So we had evaluated the clinical significance of serologic detection of antibody to 38 kDa antigen, which is known as the most specific to the M. tuberculosis complex, and culture filtrate antigen by ELISA in sputum AFB smear negative patients. Method: In this study, culture tests for acid fast bacilli with sputa or bronchial washing fluids of 183 consecutive patients who were negative of sputum AFB smear were performed. Simultaneously serum antibodies to 38 kDa antigen and unheated culture filtrate of M. tuberculosis were detected by an ELISA method. Results: The optical densities of ELISA test with 38 kDa and culture filtrate antigen were significantly higher in active pulmonary tuberculosis cases than in non tuberculous pulmonary diseases (p<0.05), but in patients with active pulmonary tuberculosis, those of the sputum culture positive patients for M. tuberculosis were not significantly different from those of the sputum culture negative cases(p>0.05). In the smear-negative active pulmonary tuberculosis patients, the sensitivity of the ELISA using 38 kDa antigen and culture filtrate was 20.0% and 31.4%. respectively. The specificity was 95.3% and 93.9%. respectively. Conclusion : In active pulmonary tuberculosis but smear negative, the serologic detection of antibody to 38 kDa antigen and culture filtrate by ELISA cannot substitute traditional diagnostic tests and does not have clinically significant role to differenciate the patient with active pulmonary tuberculosis from other with non-tuberculous pulmonary diseases.
This study Provides the treatment methods of liquid wastes from the dissolution/purification process of nuclear fuel raw material and the fabrication process of nuclear fuel powder. One of the treatment methods is to process liquid waste from uranium raw material dissolution/purification process. This waste, of the strong acid, can be reused to dissolve the fine ADU particles in filtrate which is ADU waste of pH 8.0 converted from AUC waste after recovery of uranium. To dissolve the fine ADU particles, ADU filtrate was pretreated to pH 4.0 with the dissolution/purification waste, and then mixed with the lime to pH 9.2 and aged for 30 minutes. From this processing, uranium content of the filtrate was decreased to below 3ppm. The waste from fuel powder fabrication is emulsified solution dispersed with fine oil droplets. This emulsion was destroyed effectively by adding and mixing the nitric acid with rapid heating at the same time. After this processing, $Na_2U_2O_7$ compound is produced by addition of NaOH. Optimum condition of this processing was shown at pH 11.5, and uranium content of the filtrate was analyzed to 5ppm. To remove the trace of uranium in the filtrate, lime should be added. Otherwise, 4N nitric acid was used to destroy the emulsion directly, and then lime was added to this waste. Uranium content of the treated filtrate was below 1 ppm. In addition to these wastes, the trace of uranium in filtrate after recovery of uranium from the AUC waste which is produced during PWR power preparation, is treated with NaOH to takeup fluorine(F) in the waste because fluorine is valuable and toxic material. In the finally treated waste, uranium was not detected.
Uranyl-peroxide compound was prepared by the reaction of excess hydrogen peroxide solution and trace uranium in filtrate from nuclear fuel conversion plant. The $CO_3{^{2-}}$ in filtrate was removed first by heating more than $98^{\circ}C$, because uranyl-peroxide compound could not be precipitated by $CO_3{^{2-}}$ remaining in filtrate. The optimum condition for uranyl-peroxide compound was ageing for 1 hr after controling the pH with $NH_3$ gas and adding the excess $H_2O_2$ of 10ml/lit.-filtrate. Uranium concentration in the filtrate was appeared to 3 ppm after the precipitation of uranyl-peroxide compound, and the chemical composition of this compound was analyzed to $UO_4{\cdot}2NH_4F$ with FT-IR, X-ray diffractometry, TG and chemical analysis. Also, this fine particle, about $1{\sim}2{\mu}m$, could be grown up to $4{\mu}m$ at pH 9.5 and $60^{\circ}C$. The separation efficiency of precipitate from mother liquor was increased with increase of pH and reaction temperature. Otherwise, the crystal form of this particle showed octahedral by SEM and XRD, and $U_3O_8$ powder was obtained by thermal decomposition at $650^{\circ}C$ in air atmosphere.
Indigenous strains of Trichoderma species isolated from rhizosphere soils of Tea gardens of Assam, north eastern state of India were assessed for in vitro antagonism against two important tea fungal pathogens namely Pestalotia theae and Fusarium solani. A potent antagonist against both tea pathogenic fungi, designated as SDRLIN1, was selected and identified as Trichoderma viride. The strain also showed substantial antifungal activity against five standard phytopathogenic fungi. Culture filtrate collected from stationary growth phase of the antagonist demonstrated a significantly higher degree of inhibitory activity against all the test fungi, demonstrating the presence of an optimal blend of extracellular antifungal metabolites. Moreover, quantitative enzyme assay of exponential and stationary culture filtrates revealed that the activity of cellulase, ${\beta}$-1,3-glucanase, pectinase, and amylase was highest in the exponential phase, whereas the activity of proteases and chitinase was noted highest in the stationary phase. Morphological changes such as hyphal swelling and distortion were also observed in the fungal pathogen grown on potato dextrose agar containing stationary phase culture filtrate. Moreover, the antifungal activity of the filtrate was significantly reduced but not entirely after heat or proteinase K treatment, demonstrating substantial role of certain unknown thermostable antifungal compound(s) in the inhibitory activity.

During the screening of antiviral substances having inhibitory effects on tobacco mosaic virus (TMV) infection to tobacco plants, we found a bacterial isolate KTGBP10, which was identified as a Stenotrophomonas sp., strongly inhibited the infection of TMV. When the culture filtrate from KTGBP10 was applied on the upper surface of leaves of Xanthi-nc tobacco plants at the same time or 24 hours before TMV inoculation, almost complete inhibition of TMV infection was achieved. And $40\%$ inhibition was shown with application of the culture filtrate to the under surface of leaves. In field trials, transmission of TMV from diseased seedlings to the healthy ones during transplanting work was reduced by $87.1\~92.6\%$ when the culture filtrate or cell suspension was sprayed onto the tobacco seedlings, cv. NC82, 24 hours before transplanting. No toxic effect was observed on the tobacco plants. When the broth filtrate of KTGBP10 was supplied by soaking through the cut-leaves before and/or after virus inoculation, the TMV infection was also inhibited by $50.4\~65.3\%$.

In order to obtain safe drinking water, free from surface contamination, a study to determine the potential for developing a bank-filtrate system in the Iryong and Yongsan, Nakdong River Basin, Korea was conducted. The main type of aquifer In the study area is alluvial, consisting mostly of sand and gravel. The hydraulic conductivity(k) of the Iryong and Yongsan test areas were 8.63${\times}$10$^-2$cm/s and 9.90${\times}$10$^-2$cm/s, respectively, indicating that these areas are satisfactory for bank filtrate production. Pilot plants(IRPL and YSPL) were set up In Iryong and Yongsan to monitor the change in the quality of bank-filtered water and to determine the effect pumping had on the surrounding hydrogeologic system. The pilot plants operated continuously for about two months and the data obtained were used to validate the groundwater flow model. Computer simulations were conducted to predict the effects of producing bank filtrate using MODFLOW. MODPATH was also linked with the flow model to analyze particle tracking. According to the results of the model simulations and the hydrogeologlc study, long-term pumping, the minimization of drawdown and the availability of uncontaminated sell and groundwater conditions for the catchment area were all Important factors for successful bank-titrate system development.
