The Anti-depressive Effect of Rehmanniae Radix Preparata via Anti-inflammatory Activity (숙지황 추출물의 항염증 작용을 통한 항우울 효과)

  • Objectives: Rehmanniae Radix Preparata (RRP) has been used as a traditional remedy to treat gynecology and endocrine diseases. Recently, studies on antioxidant and anti-inflammatory effects of RRP have been reported, so it was judged that RRP extracts would have an anti-depressive effect. Methods: We investigated the anti-neuroinflammatory and anti-depressive effect of RRP on lipopolysaccharide (LPS)-induced depression and LPS-stimulated BV2 microglia. RRP inhibited the LPS-stimulated excessive release of nitrite in the BV2 cells. RRP also significantly inhibited the inflammatory cytokines such as tumor necrosis factor (TNF)-alpha, interleukin (IL)-1beta and IL-6 in LPS-stimulated BV2 microglial cells. Results: RRP significantly suppressed the LPS-induced mitogen-activated protein kinase (MAPKs) and nuclear factor (NF)-𝜅B activation. In addition, administration of RRP not only inhibited the immobility time in the forced swimming test (FST) but also increased the total travel distance in the open field test (OFT). Also, RRP inhibited the elevation of TNF-alpha, IL-1beta, and IL-6 in brain of LPS-injected mice. Conclusions: Considering the overall results, our study showed that RRP exhibited the anti-neuroinflammatory and anti-depressive activities via deactivation of MAPKs and NF-𝜅B.

Prediction of radiation dose to adult human from radiopharmaceutical manufactured by third generation bisphosphonate labeled with Rhenium

  • Introduction: The crucial step in preclinical process of radiopharmaceutical production is internal dosimetry evaluation by different ways to realize radiobiological dose-response relationships and to extract the results for clinical use. Till now several bone-seeking radiopharmaceuticals have been developed for bone metastasis. Interesting features of bisphosphonates attracted attentions to them in the field of radiopharmaceutical therapy and studies on new generation of them have been doing too. Materials and methods: In this study, we used ZNA as representative of the third generation. The radiopharmaceutical 188Re-ZNA was produced and its radiochemical purity was investigated. Then, the biological distribution of the produced radiopharmaceutical at 1, 2, 4 and 24 h after injection on different organs of mice were investigated. Finally, the absorbed dose of organs in the human body was assessed using the RADAR method. Results: The results show 96% radiochemical purity of the 188Re-ZNA radiopharmaceutical. The amount of %ID/g in bone is 1.131% after 1 h and in 24 h it has a significant amount compared to other organs, that is 0.516%. Also dosimetric results show that the highest absorption dose is related to bone and the amount of this dose is 0.050 mGy/MBq. Conclusion: Considering the possibility of producing the 188Re-ZNA radiopharmaceutical, as well as the proper distribution of this radiopharmaceutical in target and non-target organs and increasing the absorbed dose in bone, it can be concluded that this radiopharmaceutical can be useful in the "radiopharmaceutical therapy" in metastases.

  • Recently, there has been an increasing trend in the role of social media in tourism marketing. We analyze changes in tourism marketing trends using tourism marketing keywords through social media networks. The aim is to understand marketing trends based on the analyzed data and effectively create, maintain, and manage customers, as well as efficiently supply tourism products. Data was collected using web data from platforms such as Naver, Google, and Daum through TexTom. The data collection period was set for one year, from December 1, 2022, to December 1, 2023. The collected data, after undergoing refinement, was analyzed as keyword networks based on frequency analysis results. Network visualization and CONCOR analysis were conducted using the Ucinet program. The top words in frequency were 'tourists,' 'promotion,' 'travel,' and 'research.' Clusters were categorized into four: tourism field, tourism products, marketing, and motivation for visits. Through this, it was confirmed that tourism marketing is being conducted in various tourism sectors such as MICE, medical tourism, and conventions. Utilizing digital marketing via online platforms, tourism products are promoted to tourists, and unique tourism products are developed to increase city branding and tourism demand through integrated tourism content. We identify trends in tourism marketing, providing tourists with a positive image and contributing to the activation of local tourism.

Catalpol and Mannitol, Two Components of Rehmannia glutinosa, Exhibit Anticonvulsant Effects Probably via GABAA Receptor Regulation

  • Epilepsy is a brain disorder that affects millions of people worldwide and is usually managed using currently available antiepileptic drugs, which result in adverse effects and are ineffective in approximately 20-25% of patients. Thus, there is growing interest in the development of new antiepileptic drugs with fewer side effects. In a previous study, we showed that a Rehmannia glutinosa (RG) water extract has protective effects against electroshock- and pentylenetetrazol (PTZ)-induced seizures, with fewer side effects. In this study, the objective was to identify the RG components that are responsible for its anticonvulsant effects. Initially, a number of RG components (aucubin, acteoside, catalpol, and mannitol) were screened, and the anticonvulsant effects of different doses of catalpol, mannitol, and their combination on electroshock- and chemically (PTZ or strychnine)-induced seizures in mice, were further assessed. Gamma-aminobutyric acid (GABA) receptor binding assay and electroencephalography (EEG) analysis were conducted to identify the potential underlying drug mechanism. Additionally, treated mice were tested using open-field and rotarod tests. Catalpol, mannitol, and their combination increased threshold against electroshock-induced seizures, and decreased the percentage of seizure responses induced by PTZ, a GABA antagonist. GABA receptor binding assay results revealed that catalpol and mannitol are associated with GABA receptor activity, and EEG analysis provided evidence that catalpol and mannitol have anticonvulsant effects against PTZ-induced seizures. In summary, our results indicate that catalpol and mannitol have anticonvulsant properties, and may mediate the protective effects of RG against seizures.

Radiation Effect on Body Weight and Peripheral Blood Picture Induced by Whole-Abdominal X-ray Irradiation with Different Fractionation in Mice (백색마우스에 대한 전복부 조사에서 상이한 분할조사가 체중과 말초혈액 소견에 미치는 효과)

  • The object of this study was to determine the difference of radiation effect in different fractional doses and to establish optimal fractionation schedule on the whole-abdominal X-ray irradiation. Total 160 mice were irradiated with 150 KVP, 15 mA orthovoltage x-ray machine and two different fractionation (100 cGy/Fr. and 200 cGy/Fr.) were used. Body weight, hemoglobin and WBC count with differential count were analyzed according to the same amount of total dose, same field size and two different fractionation schedules. The result of this study were summarized as follows: There was no significant difference in body weight and hemoglobin concentration by sex or fractional dose. Leukopenic change was prominent in the 3,000 cGy irradiation group and the proportion of decrease was remarkable in the 200 cGy/Fr, group than 100 cGy/Fr, group. Progressive decrease of lymphocyte count and reciprocal increase of neutrophil count were noted as dose increment. The effect of the fractional does on WBC count and proportion of lymphocyte were significant. This suggests that judicious selection of fractional dose may be important in clinical radiotherapeutic practice.

  • Development of supervoltage treatment machine may minimize skin reaction by skin-sparing effect, but skin damage is still one of "the dose limiting factor" in radiation therapy. In spite of these importance, systemic histopathologic studies of skin in similar conditions which used in clinical treatment has not been performed so far. 60mice were irradiated with conventional fraction ($200{\times}5/wk$) and whole abdominal field ($2{\times}3cm$, from symphysis pubis to xyphoid process). Used machine was 250KV, 24mA, orthovoltage x-ray machine. Histopathological changes of acute skin reaction at the level of total irradiation dose were analyzed and the possible mechanism of later chronic changes were investigated. Obtained results are as follows: 1. In 1,000 rad irradiated group, only mild epidermal edema is noted. 2. In 2,000 rad irradiated group, slightly decreased number and size of hair follicles and appendages, dermal edema and scanty infiltration of inflammatory cells are visible. 3. In 3,000 rad irradiated group, marked increased capillary congestion and prominant infiltration of inflammatory cells are observed. 4. In 4,000 rad irradiated group, vascular wall thickening with proliferation of endothelial cells are prominant. Dermal thinning and hyalinization are newly developed. 5. In 5,000 rad irradiated group, complete desquamation of epidermis is not seen, despite of acceleration of all above mentioned changes.

  • Probiotics have many effects such as antihypertensive, prevention of cancer, antioxidation, reduction of dermatitis symptoms, improvement of mineral absorption, reduction of allergic symptoms, and decrease of cholesterol, However, the main role of probiotics is that they balance intestinal microbials proportion. L. acidophilus is one of probiotics and microflora in intestine. It has an acidification activity, aroma production, texture formation and probiotics properties. We studied on the roles of L. acidophilus in mice. In this study, body weights of mice were decreased when administration of L. acidophilus ($1{\times}10^{10}$ CFU) and swimming ability has been raised than a normal group after feeding on L. acidophilus ($1{\times}10^{10}$ CFU). After taking L. acidophilus ($1{\times}10^{10}$ CFU), total white cells were increased than a normal group; hemoglobin and thrombocytes were increased. The level of cholesterol and triglyceride were decreased in blood analysis. We knew L. acidophilus is related to innate immune system. We found out the secretion of cationic peptide was increased in the Lysoplate assays as a result of L. acidophilus ($1{\times}10^{10}$ CFU) administration. Appearance rate of lysozyme was also increased than the normal group on an immunohistochemistry stain. We confirmed L. acidophilus contributes to host health through innate immune system stimulation. L. acidophilus more than $1{\times}10^{10}$ CFU are thought to be beneficial for the host health and prevention of intestinal diseases in field condition.

Study on the Cultural Marketing Factors of Tourism Destination in the Era of Convergence : Focusing on the Brand Attitude and Brand Equity Differences by segmented groups (융복합시대의 관광지 문화마케팅에 관한 연구 : 세분집단별 브랜드태도 및 자산 인식 차이를 중심으로)

  • This study began with the focusing the growth of importance of cultural marketing and its successes by applying to branding in a phase in which the cultural production era has arrived after the industrial production era. This study aimed to reveal the effect of cultural marketing, which are used as marketing strategy in the tourism field, on brand attitude and awareness. Therefore, this study tried to investigate the brand equity and attitude in accordance with the cultural marketing factors by survey Chinese tourists visiting Korea. The result and contents of the study are as follows. First, it was confirmed that the research question on the possibility of market segmentation in accordance with cultural marketing factors; the market segments were classified as Cluster 1 general involvement perceived group, Cluster 2 high involvement perceived group, Cluster3 middle involvement perceived group. Second, it revealed significant differences on brand attitude among the segments; Cluster 2 showed high brand attitude and Cluster 3 showed average level on brand attitude. Third, it found a statistically significant differences in the perception of brand equity including perceived quality, brand awareness and brand image of tourism destination. In conclusion, this study was able to identify market segments in accordance with cultural marketing factors of tourism destination and revealed significant differences in the segments by brand attitude and brand equity. This could provide practical implication that various cultural marketing activities to generate positive results on brand equity and brand attitude and cultural marketing strategies is also needed to increase foreign tourists' awareness of these cultural marketing activities.

Antidepressant Effects of JG02 on Chronic Restraint Stress Animal Model (만성구속스트레스 동물모델에 대한 JG02의 항우울 효과)

  • Objectives: As a general emotion, everyone can temporarily experience depression, but depressive disorder is a disease that excessively affects daily life. Among the various causes of depression, the deficiency of monoamine-based neurotransmitters such as serotonin and epinephrine are considered significant. Thus, antidepressants that target monoamines are used frequently. However, side effects such as nausea, vomiting, insomnia, anxiety, and sexual dysfunction are observed. Thus, it is necessary to develop a new therapeutic agent with fewer side effects. In this study, we investigated the antidepressant effect of JG02, used to treat depression by normalizing the flow of qi (氣) in Korean medicine. Methods: C57BL/6 mice were selected and randomly divided into six groups: normal, control, amitriptyline, and JG02 (50, 125, 250 mg/kg), respectively. Except for normal, depression was induced by applying restraint stress at the same time for six hours daily for 14 consecutive days. Saline, amitriptyline or JG02 samples were orally administered two hours before applying the stress. After that, a forced swimming test and an open field test were performed. Additionally, serum corticosterone, serotonin mRNA, BDNF mRNA, and protein in the hippocampal region were measured and compared. Results: JG02 decreased immobility time rate in the FST and increased the zone transition number and travel distance in the OFT. Also, JG02 inhibited the release of serum corticosterone, and increased serotonin, BDNF gene expression, and BDNF protein in the hippocampus. Conclusions: In this study, JG02 showed significant antidepressant effects on the chronic restraint stress mice model. When further research is performed based on JG02, the development of a new antidepressant is considered highly possible.

Imputation of Missing SST Observation Data Using Multivariate Bidirectional RNN (다변수 Bidirectional RNN을 이용한 표층수온 결측 데이터 보간)

  • The data of the missing section among the vertex surface sea temperature observation data was imputed using the Bidirectional Recurrent Neural Network(BiRNN). Among artificial intelligence techniques, Recurrent Neural Networks (RNNs), which are commonly used for time series data, only estimate in the direction of time flow or in the reverse direction to the missing estimation position, so the estimation performance is poor in the long-term missing section. On the other hand, in this study, estimation performance can be improved even for long-term missing data by estimating in both directions before and after the missing section. Also, by using all available data around the observation point (sea surface temperature, temperature, wind field, atmospheric pressure, humidity), the imputation performance was further improved by estimating the imputation data from these correlations together. For performance verification, a statistical model, Multivariate Imputation by Chained Equations (MICE), a machine learning-based Random Forest model, and an RNN model using Long Short-Term Memory (LSTM) were compared. For imputation of long-term missing for 7 days, the average accuracy of the BiRNN/statistical models is 70.8%/61.2%, respectively, and the average error is 0.28 degrees/0.44 degrees, respectively, so the BiRNN model performs better than other models. By applying a temporal decay factor representing the missing pattern, it is judged that the BiRNN technique has better imputation performance than the existing method as the missing section becomes longer.
