A Novel Approach for Assessing the Proteolytic Potential of Filamentous Fungi on the Example of Aspergillus spp.

  • Proteolytic enzymes produced by filamentous fungi can degrade various fibrous and globular proteins along with other metabolites that may also find application in biotechnology. In this study, the effect of proteolytic enzymes of 22 Aspergillus strains on various proteins was investigated using protein-containing diagnostic media. Subsequently, a new parameter estimating secreted proteinases specificity towards fibrous or globular proteins without its advanced biochemical research - index of severity of proteolytic action (ISPA) - was suggested. This index determines mycozymes specificity in following manner: its value increases with greater affinity to fibrous proteins, decreases if there is higher affinity to globular proteins. ISPA value was the lowest (0.52) for Aspergillus domesticus, indicating the highest specificity to globular proteins, the highest one (1.26) for A. glaucus, whose proteinases best hydrolyzed fibrous proteins. However, the highest overall proteolytic potential was observed for Aspergillus melleus. The ability to produce acid, alkali and extracellular pigments was evaluated for all isolated strains as well.

  • 본 연구는 크로마토그래피적 방법에 의한 단백질 분리를 규모화시키기 위한 기초적인 실험을 행한 것이다. 단백질 분리를 위해서 화학적 처리된 폴리에틸렌섬유를 미국 Millipore사의 여과 Cartridge에 장착시키고, 그 시스템에서 단백질인 5%의 Bovine Serum Albumin을 통과시켜 섬유 표면의 단백질 흡착능력과 결합 가역성 등을 관찰하였다. 또한 실험자료들을 Omega 프로그램이 내장된 컴퓨터와 맥킨토시의 Kaleidagraph로 자료처리 한 것을 보여주었다.

후두에 발생한 악성 섬유성 조직구종 (A Case of Malignant Fibrous Histiocytoma of the Larynx)

  • Malignant fibrous histiocytoma is one of the rare types of larynx tumor. The most common sites of the tumor are limbs, trunk, and retroperitoneal space, but tumor localization within head and neck are very rare. It is built of histiocytes, fibroblasts and multinuclear giant cells. A diagnosis of the tumor includes microscopic and immunohistologic examination with identification of specific tissue markers and intermediate filaments of proteins. This disease has been treated by several methods combining radical surgery, radiotherapy, and chemotherapy, but the prognosis is poor. We present 74-year-old Asian man with dysphonia for 2 years. The tumor of the larynx was examined on laryngoscopy. The radical surgery rendered the final pathological diagnosis, confirmed histologically and immunohistochemically as malignant fibrous histiocytoma. This tumor was treated with laser cordectomy followed by radiotherapy. 3.5 year's observation of the patient didn't either show any signs of recurrence or dysphonia.

반복단위 단백질 고분자의 유전공학적 합성 및 응용 (Genetic Synthesis and Applications of Repetitive Protein Polymers)

  • 본 연구는 특정 아미노산들로 구성된 단위체가 반복되는 형태를 가지는 반복단위 단백질을 유전공학적으로 합성하는 방법들과 응용사례들을 소개하고 있다. 유전공학적 합성법은 단위체의 반복횟수를 정확하게 제어하면서 인식부위의 제한을 없애서 원하는 단백질만을 발현할 수 있도록 발전해왔으며, 최근 소개된 RDL과 CCM 방법에 의하여 가능해졌다. 반복단위 단백질의 응용사례로는 대표적으로 ELP, SLP, Prolamin 등의 단백질을 합성하여 생체재료나 약물전달시스템을 개발하는데 응용하거나, ELFSE의 drag-tag 개발에 응용되는 연구들이 진행되고 있다. 화학적으로 합성된 고분자에 비해 유전공학적으로 합성된 반복단위 고분자의 경우, 고유의 물리적 성질과 함께 환경에 미치는 유해함이 상대적으로 적다는 점 때문에 미래의 신소재로 기대되고 있다.

Molecular cloning of peroxidase cDNAs from dehydration-treated fibrous roots of sweetpotato and their differential expression in response to stress

  • Three peroxidase (POD) cDNAs were isolated from dehydration-treated fibrous roots of sweetpotato (Ipomoea batatas) plant via the screening of a cDNA library, and their expressions were assessed to characterize functions of each POD in relation to environmental stress. Three PODs were divided into two groups, designated the basic PODs (swpb4, swpb5) and the anionic PODs (swpa7), on the basis of the pI values of mature proteins. Fluorescence microscope analysis indicated that three PODs are secreted into the extracellular space. RT-PCR analysis revealed that POD genes have diverse expression patterns in a variety of plant tissues. Swpb4 was abundantly expressed in stem tissues, whereas the expression levels of swpb5 and swpa7 transcripts were high in fibrous and thick pigmented roots. Swpb4 and swpa7 showed abundant expression levels in suspension cultured cells. Three POD genes responded differently in the leaf and fibrous roots in response to a variety of stresses including dehydration, temperature stress, stress-associated chemicals, and pathogenic bacteria.

  • Extracellular matrix(ECM) is composed of the ground materials(proteoglycan) and nano size diameter fibrous proteins(ex. collagens) that together form a composite-like structure. In this study, fibrous scaffold with biomimetic architecture based on collagen nanofibers interpenetrated in PLGA/chitosan microfibrous matrix. Chitosan was selected for its structure similarity to glycosaminoglycan and neutralizing capacity for PLGA acidic metabolite. Collagen nanofiber were prepared by electrospinning. (omitted)

High-Level Production of Spider Silk Protein by Fed-Batch Cultivation of Recombinant Escherichia coli and Its Purification

  • Silk proteins from Nephila clavipes are fibrous proteins containing repetitive sequences with both crystalline and amorphous domains. In order to obtain high-level production of silk protein, the synthetic genes had 16 contiguous units of the consensus repeat sequence of the silk protein were expressed in Escherichia coli BL21(DE3) under the strong inducible T7 promoter. For production of recombinant silk protein in large amounts, pH-stat fed-batch cultures were carried out. The recombinant silk protein was produced as soluble forms in E. coli, and the recombinant silk protein content was as high as 11% of the total protein. When cells were induced at $OD_{600}$ of 60, the amount of silk protein produced was 6.49 g/L. After simple purification steps, 9.2 mg of silk protein that was more than 80% pure was obtained from a 50 mL culture, and the recovery yield was 26.3%.

Characterization of Silk Fibroin/S-carboxymethyl Kerateine Surfaces: Evaluation of Biocompatibility by Contact Angle Measurements

  • Surface characterization of materials has been considered critical in the development of biomaterials, as many unfavorable responses from the body occur at the interface between a material and the body component. The contact angle measurement is one means to characterize the surface properties and to correlate them to the biocompatibility of materials. In this paper, surface characteristics of silk fibroin/S-carboxymethyl kerateine, representative fibrous proteins, were investigated by contact angle measurements of ESCA. The biocompatibility of the blends was evaluated based on minimal interfacial free energy concept, and compared with other potential biomaterials. It was also hypothesized that the enhanced surface polarity of the blends was generated from the conformational transition of proteins. This approach to evaluate the biocompatibility of materials based on surface characteristics may find wide utility in many biomedical applications.

The Localization of Cytokeratin 19 and Vimentin in Sprague Dawley Albino Rat Skin Tissue

  • Cytokeratin 19 (CK19) expressed in epidermis of skin, bulge region of hair follicle, outermost layer of outer root sheath and proximal and distal to bulge. Vimentin is a fibrous protein that localized in cytoplasm of fibroblast and forms cytoskeleton to maintain shape of cell and nucleus. In this study, CK19 and vimentin in skin were confirmed with light, fluorescence and transmission electron microscope. As a result, CK19 was localized epidermis, hair follicles, outer root sheath and nucleus of Merkel's cell. However, vimentin was localized some epidermis, dermis, hypodermis and nucleus of Merkel's cell. The role of CK19 is self-renewal and homeostasis in skin. Also, hair follicle regeneration and hair growth is known to be related. It is supposed that required of structural proteins that make up cytoskeleton is increased. Thereby, expression of CK19 is increased. It is considered that vimentin localized in order to stabilize structure of cell and cytoskeleton of fibroblasts. Also, CK19 and vimentin present in nuclei of Merkel's cell, and to act as a fibrous protein that make up end of a nerve fiber present in Merkel's cell and paracrine function of Merkel's cell.
