Development of Eggs, Larvae and Juvenile of the Korean Bullhead, Pseudobagrus fulvidraco (Richardson) reared in the laboratory (동자개의 형태발달과 성장특성)

  • The early developmental stages, growth and morphological changes of the Korean bullhead, Pseudobagrus fulvidraco, were studied from a series of reared specimens. Details of the early developmental stages are illustrated with special reference to morphological transformations. Egg and sperm of Korean bullhead were obtained from mature adults under hormonal treatment, fertilized artificially, and incubated in the aquarium. The incubation period of fertilized eggs was 55 to 66 hours at a temperature of 24.9${\pm}$0.34$^{\circ}$. Larvae were fed successively with Artemia salina and Daphnia magna for 2 to 15 days and artificial food after 20 days. Fertilized eggs were adhesive and spherical with a diameter of 2.04mm(n = 100). The mean total length of newly hatched larvae was about 4.92${\pm}$0.33 mm. Mouth opening occurred on one-day-old yolk-sac larvae, and initial feeding was observed on the third day after hatching. The morphological transitions from larvae to juvenile and juvenile to young stages occurred when the fish reached about 17 mm in total length (about 13days after hatching) and about 32 mm in total length (about 30 days after hatching), respectively. Many changes in proportion of body parts to total length were observed at about 7~8 mm and 30~32 mm, corresponding to the transformations from larvae to juvenile and from juvenile to young, respectively. In comparing relative growth of each body part against total length, those characteristics related to head parts showed positive growth in the pre-larval stages, while those concerning mobile abilities showed positive growth in the post-larval stage.

Embryonic Development and Metamorphosis of the Ascidian Halocynthia aurantium (붉은멍게(Halocynthia aurantium)의 배발생과 변태)

  • The ascidian Halocynthia aurantium (sea peach), which belongs to the phylum Chordata, is thought to be a valuable organism of aquaculture like H. roretzi (sea pineapple), but its biological characteristics such as development and ecology are not well known. In this study, in order to obtain basic data for H. aurantium farming, the development processes of H. aurantium inhabiting the east coast of Gangwon-do were investigated and compared with those of H. roretzi, a related species. As a result, the morphology and developmental stages of the fertilized eggs, embryos and larvae of H. aurantium were very similar to those of H. roretzi. Fertilized eggs of H. aurantium took about 42.1 hours to hatch at 11℃, almost similar to 40.9 hours of H. roretzi. The time required for larvae to metamorphose into juveniles after hatching was very similar between the two species. The hatched larvae of the two species became juveniles with oral and atrial siphons after 23 days at 11℃. Both types of embryos developed slowly in seawater at low temperatures and rapidly developed at high temperatures. Fertilized eggs of H. aurantium hatched in an average of 62.3 hours at 9℃, 42.1 hours at 11℃, and 36.3 hours at 13℃, whereas those of H. roretzi hatched in an average of 60.4 hours, 40.9 hours, and 35.2 hours. Most of H. aurantium embryos did not develop normally above 15℃, so it is thought that attention is needed in the seed production processes.

Prevention Strategies for Viral Nervous Necrosis (VNN) in Sevenband Grouper Epinephelus septemfasciatus Aquaculture Farms (능성어(Epinephelus septemfasciatus) 양식장에서의 바이러스성신경괴사증(VNN) 예방대책)

  • Viral nervous necrosis (VNN) is a serious disease of sevenband grouper Epinephelus septemfasciatus in Korean aquaculture farms. However, we suggest the following preventative methods for hatcheries: 1) disinfecting rearing water, 2) selecting spawners via ELISA and PCR, 3) selecting eggs via PCR, 4) disinfecting fertilized eggs, and 5) proper facilities management. When these methods are implemented, nervous necrosis virus (NNV)-free fish are produced because vertical and horizontal transmission is prevented. However, horizontal transmission of NNV through rearing seawater sourced from the environment during grow-out stages in sea cages can still occur. Live NNV vaccines with a low rearing temperature or Poly(I:C) immunization are very effective at preventing horizontal transmission of NNV in rearing farms. Furthermore, even after VNN is contracted, fish mortality can be reduced by administering Poly(I:C).

  • Objective: To verify the effect of Matrigel, a ECM complex from Engelbreth-Holm-Swarm (EHS) mouse sarcoma on the preimplantation development and apoptosis of mouse fertilized eggs. Method: Late pronucleus stage eggs were cultured through the blastocyst stage in the presence of Matrigel (0.5%, v/v). Characteristics of apoptosis and cell number assesed by Hoecst staining and TUNEL labeling at the blastocyst stage, respectively. Results: Morphological development, number of cells per embryo was significantly increased but rate and number of TUNEL positive nuclei of the embryo were decreased in the presence of Matrigel. Conclusion: This result suggested that at low concentration of Matrigel improves both viability and morphological development in the preimplantation mouse embryos.

  • Two human lactoferrin expression vectors(pCChcLf and pCChcLf-1) were constructed using rat $\beta$-casein gene and human lactoferrin cDNA. The recombinant DNAs containing human lactoferrin cDNA were microinjected into the fertilized eggs of hybrid mice (BDF1 : C57BL$\times$DBA) and the DNA-injected eggs were treansferred into the oviducts of foster mothers. Genomic DNAs were isolated from the tails of mice born from the microinjected eggs and analyzed by Southern blot analysis. As a result, 5 and 9 transgenic mice with CChcLf and CChcLf-1 gene were produced, respectively. To determine tissue-specificity of transgene expression, Northern blot analysis was performed. Female transgenic mice were killed at day 10 of lactation and total RNAs from various tissues were isolated. Based on Northern blot analysis, it was shown that transgene was mainly expressed in the mammary glands of transgenic mice. In addition, the human lactoferrin in milk was detected by enzyme-linked immunosorbent assay. For this study, milk was obtained from the mammary glands of the transgenic mice at day 10 of lactation. In line #2 of CChcLf and line #7 of CChcLf-1 transgenic mice, human lactoferrin was secreted into the milk at concentration levels of 340ng/ml and 60ng/ml, respectively.

  • To investigate the characteristics of hybrid eggs and larva produced by olive flounder Paralichthys olivaceus females and starry flounder Platichthys stellatus males, we examined the developmental speed of hybrid eggs at different water temperatures. The developmental speed of hybrid eggs tended to increase with increasing water temperature. Specifically, the hatching times were 91 hrs, 62 hrs and 43 hrs at $10^{\circ}C$, $15^{\circ}C$ and $20^{\circ}C$, respectively. The mean biological minimum temperature of the hybrid was $1.3^{\circ}C$, which is in between that of the olive flounder and the starry flounder. In high water temperatureseasons, slower growth was observed in hybrids of the starry flounder which is a coldwater fish.

  • The egg development and early life history of Korean endemic bitterling Rhodeus pseudosericeus were observed under the controlled water temperature, $17.0{\pm}0.5^{\circ}C$. Fertilized eggs are opaque yellow in color, measuring 2.9~3.1 mm in length and 1.6~1.9 mm in breadth. The eggs of this species began to hatch about seventy hours after insemination and the range of total length of larvae were 3.2~3.4 mm. The larvae reached at the heterotrophic stage about twenty-five days after hatching. R. pseudosericeus had a pair of wing-shaped yolk projection during the prelarval stage that is a common character of the genus Rhodeus.

Observed Pattern of Diel Variation in Specific Gravity of Pacific Mackerel Eggs and Larvae

  • Although Pacific mackerel (Scomber japonicus) is an important commercial species in Korea, its recruitment mechanism remains largely unknown. Diel vertical positioning of larvae in the water column, which is affected by their specific gravity and the surrounding water density, may help to provide an understanding on recruitment success through predator avoidance and prey availability. The specific gravity measurement on Pacific mackerel eggs and larvae would seem to be essential information necessary to learn about the transport process from spawning to nursery grounds, and consequently the recruitment success. Eggs were artificially fertilized, and larvae were fed with rotifer when their mouths opened 3-4 days after hatching. We conducted the experiment using a density gradient water column to measure the ontogenetic changes in specific gravity from fertilization to 10 days after hatching. Egg specific gravity was stable during most of the embryonic period, but a sudden increase to $1.0249g\;cm^{-3}$ happened just before hatching. However, the specific gravity of newly hatched larvae was much lighter ($1.0195g\;cm^{-3}$), and specific gravity tended to increase continuously after hatching. Comparison of specific gravity with seawater density reveals that eggs and newly hatched larvae can float in the surface layer of the ocean. For the later period of the experiment, the specific gravity showed a cyclic diel pattern: the highest in the evening while the lowest at dawn. The fullness of larval stomach may be responsible for the observed differences in specific gravity, because stomach fullness was lower (40-60%) at midnight, and higher (80-85%) in evening. The diel pattern of specific gravity might provide clues regarding how larvae match the diel vertical migration of prey organisms.

Eggs Development and Early Life History of Spine Loach, Cobitis hankugensis (Pisces: Cobitidae), Endemic to Korea (한국 고유종 기름종개 Cobitis hankugensis의 난발생 및 초기생활사)

  • The spine loach, Cobitis hankugensis endemic to Korea were collected at the Nokdong River, Unbong-eup, Namwon-si, Jeollabuk-do during June 2007 for study its eggs development and early life history. The eggs were obtained by injecting gonadotropin (LHRH-1) to female and were fertilized by dry method in the laboratory condition. The matured eggs were separative, demersal and light yellowish with $1.29{\pm}0.07mm$ in diameter average. Under $23{\sim}25^{\circ}C$, the eggs were hatched in 45~52 hours after fertilization, and just hatching larvae were $4.5{\pm}0.24mm$ in total length. On 5 days after hatching, they were $6.6{\pm}0.13mm$ in total length and their yolk sacs were completely absorbed. From 25 days after hatching, they became the juvenile stage with $9.0{\pm}0.49mm$ in total length. On the 100 days after hatching, their external forms and band patterns were similar to those of adults with average $27.3{\pm}2.82mm$ in total length.
