Effect of Aesculetin and $O_2$Concentrations on In Vitro Development of Preimplantation Embryo in Hanwoo (Korean Native Cattle) (한우 수정란의 체외발육에 있어서 Aesculetin과 $O_2$농도의 영향)

  • The present study was examined effects of aesculetin and $O_2$concentrations on in vitro development of Hanwoo (Korean Native Cattle) embryos derived from in-vitro matured and fertilized (IVM-lVF) oocytes. The oocytes were cultured for the first 40~44 h after in vitro fertilization, then embryos of 2 to 8 cell stages were cultured under the different culture conditions for another 6 days. In experiment 1, the higher rates of morulae and blastocysts were produced in 5% $O_2$, than in 20% $O_2$(P<0.05). There was significantly (P<0.05) higher in embryos cultured with 1 $\mu\textrm{g}$/$m\ell$ than with 0, 5 and 10 $\mu\textrm{g}$/$m\ell$ of aesculetin. In experiment 2, the proportions of embryo developed with blastocysts and morulae plus blastocysts in 5% $O_2$, again was significantly (P<0.05) higher in 20% $O_2$, during the culture with aesculetin and/or taurine. In the 5 and 20% $O_2$atmosphere, the inclusion of 1 $\mu\textrm{g}$/$m\ell$ aesculetin or 2.5mM taurine increased significantly (P<0.05) the percentages of blastocysts and morulae plus blastocysts. In experiment 3, in medium with aesculetin plus PDGF and taurine plus EGF than other treatment groups, significantly (P<0.05) higher developmental rates were obtained. Number of blastomere in balstocyst stage were also higher in medium with that than without aesculetin. However, there were no significant differences in all culture conditions. In experiment 4, the proportions of embryo developed to the morulae and blastocyst stages were significantly (P<0.05) higher rates in medium with natural and commercial aesculetin than in control medium. No significant differences, however, were observed in between natural (71%) and commercial (70.0%) aesculetin. Number of blastomere in blastocyst stage were also higher in medium with natural and commercial aesculetin than in control medium. However, there was no effect on the number of blastomeres by these treatment. These data indicate that preimplatation embryos are very sensitive to condition that can cause oxygen concentration and show that efficiency role of aesculetin for improving bovine embryo development in vitro.

Effect of Cysteamine on In Vitro Maturation of Porcine Oocytes and Development of Porcine IVM/IVF Embryos (Cysteamine의 첨가배양이 돼지 난포란의 체외성숙과 배발달에 미치는 영향)

  • The present study was carried out to examine the effect of cysteamine in vitro maturation (IVM) of porcine oocytes and development of porcine IVM/IVF Embryos. The results were summarized as follows : 1. The rates of nuclear maturation, penetrated oocytes, pronuclear formation, polyspermic oocytes and mean numbers of the penetrated sperm were not different in NCSU23 maturation medium with 0, 25, 50 and 100 $\mu$M cysteamine (P〉0.05). 2. The rates of blastocyst formation at day 7 after in vitro fertilization in 0, 25, 50 and 100 $\mu$M cysteamine were 17.9$\pm$6.1, 17.4$\pm$6.3, 24.2$\pm$1.9 and 16.9$\pm$2.0%, respectively. And the total cells were 30.7$\pm$2.4, 34.9$\pm$2.8, 39.6$\pm$2.3 and 36.8$\pm$3.6, respectively. Fifty $\mu$M cystealnine group was significantly higher than those of any other treatment groups (P<0.05). 3. The ratios of ICM/total cells in 20~40% category were 20.5, 41.6, 19.5 and 31.5%, respectively. Twenty five $\mu$M cysteamine group was higher than those of other groups. 4. The rates of blastocyst formation at day 7 in the NCSU-23 culture medium of porcine IVF-produced embryos with 0, 25, 50, and 100 $\mu$M cysteamine were 16.0$\pm$0.2, 13.6$\pm$1.7, 25.0$\pm$0.8 and 15.7$\pm$4.5%, respectively. And the total cells were 27.0$\pm$3.7, 36.1$\pm$4.8, 34.0$\pm$3.8 and 25.2$\pm$4.4, respectively. Fifty $\mu$M cysteamine group was significantly higher than those of any other treatment groups (P<0.05). 5. The ratios of ICM/total cells in 20~40% category were 53.8, 30.0, 16.6 and 11.1%, respectively. The addition groups of cysteamine were lower than those of control group. In conclusion, these results suggested that the addition of 50 $\mu$M cysteamine in the IVM medium and 25~50 $\mu$M cysteamine in IVC medium were effective on the blastocyst formation and total cells of blastocysts.

Effects of clomiphene citrate on ovarian function and embryo developmental capacity in the rat (랫드에 있어서 클로미펜 시트레이트가 난소기능 및 수정란 발육성에 미치는 영향)

  • The effects of CC the ovulatory response, oocyte normality, ovarian steroidogenesis and subsequent embryo developmental potential were examined in PMSG-treated rats. On Days of 25~27 of age, immature female Sprague Dawley rats were treated with three different doses(0.05, 0.1 or 1.0mg /day) of clomiphene citrate or vehicle. The females subsequently received 4IU PMSG on Day 28 and/or 10IU hCG on Day 30, and were killed on Day 31. Some females given 0.1mg CC or vehicle with 4IU PMSG were then mated and killed on Days 2, 3, 4 and 5 of pregnancy. Compared to vehicle(control) group, by increasing the doses of CC, there were a significant decrease in the ovulatory response as judged by both the proportion of rats ovulating and the mean number of oocytes per rat and a marked reduction of ovarian weight. The increasing doses of CC substantially promoted the degeneration(%) of oocytes ovulating in a dose-dependent manner. The CC-mediated inhibitions of the ovulatory response and ovarian weight were oompletely overcome by a subsequent treatment of hCG. Increasing doses of CC resulted in a siginificant elevation of serum estradiol with the decreased levels of progesterone and androgens. The additive treatment with hCG was effective to reduce the elevation of estradiol and to increase the reduction of progesterone produced by high dose(1.0mg) of CC. The preimplantation embryos recovered from 0.1mg CC-treated pregnant rats demonstrated a progressive early loss from Day 3 of pregnancy with a significant increase in the percentage of degeneration during all periods examined, compared to controls. The rate of progressive embryo cleavage in the CC-treated rats were slower than that in controls from Day 3 of pregnancy. Additionally, the percentage of the cleaved embryos recovered from the CC-treated rats remained significantly lower consistently from Day 2 of pregnancy, compared to control regimen. These results demonstrate a possible mechanism of CC-mediated inhibition of ovulatory response in the rats which may include the attenuation or blockade of the endogenous secretion of gonadotropins and also suggest that its detrimental effects observed on oocyte normality and embryonic development may be caused by abnormal follicular steroidogenesis( especially elevated estradiol) preceding fertilization.

Studies on Mixed-Seeding of Native Woody and Herb Species (녹화용(綠化用) 자생(自生) 목본식물(木本植物)과 초본식물종자(草本植物種子)의 혼파처리(混播處理)에 관(關)한 연구(硏究))

  • This study was conducted to find out the appropriate rates of mixed-seeding of native woody and herb species. It was carried out in greenhouse from 1994 to 1995. The early days germination individuals were $60no./m^2$ in case of woody species for mixed seeding treatment and the treatments were fertilization, soil surface treatment, seeding amount. The investigation factors are the number of individuals of woody and herb species, biomass, height, number of tillers, etc. The mean individuals of Lespedeza bicolor and Amorpha frutzcosa were investigated $1.14no./m^2$, $0.496no./m^2$ in 1995, and Spiraea prunifolia var. simpliciflora and Alnus hirsuta were withering to death. In 1995, the Individuals of Oenothera odorata, Themeda triandra var. japonica, Cymbopogon tortilis var. goeringii, Miscanthus sinensis, Artemisia princeps var. orientalis, Lespedeza cuneata, and Arundinella hirta were investigated $5.06no./m^2$, $1,072no./m^2$, $0no./m^2$, $412.53no./m^2$, $88.6no./m^2$, $8.9no./m^2$, $57.46no./m^2$, respectively, in case of herb species. The height and biomass of Lespedeza bicolor and Amorpha fruticosa showed very fast growth, and those of Themeda triandra var. japonica, Miscanthus sinensis had a similar tendency. For the changes in woody species, according to the seeding amount, there was no significant relationship between species, but It was highly significant in 1995. In case of the herb species, the relationship between the number of individuals and seeding amount was significant for both 1994 and 1995. The most number of tiller was Miscanthus sinensis(21), and showed Themeda triandra var. japonica, Arundinella hirta in descending order. For the mixed seeding of wood and herb species, the number of individuals, height, biomass, tiller of herb species diminished as the seeding amount of herb species increased. It can be concluded that using Lespedeza bicolor and Amorpha fruticosa for woody species and like Arundinella hirta in herb species will be efficient for revegetation measures. It seems, therefore, that the plants of revegetation methods will be used to Lespedeza bicolor, Amorpha fruticosa and Arundinella hirta, and further study is needed on the seeding amount.

Effects of Application Method of Pig Compost and Liquid Pig Manure on Yield of Whole Crop Barley (Hordeum vulgare L.) and Chemical Properties of Soil in Gyehwa Reclaimed Land (계화간척지에서 돈분뇨 퇴.액비 시용이 청보리 (Hordeum vulgare L.) 수량 및 토양화학성에 미치는 영향)

  • In order to develop the application method of pig compost (PC) and liquid manure (LM) for whole crop barley cultivation, experiments were conducted at Munpo series (coarse loamy, mixed, nonacid, mesic family of Typic Fluvaquents) soil in Gyehwa-reclaimed land, six plots, a LM applied rate as N% ; non-application, chemical fertilizer (CF)100, 100, 50+50, 50+CF50 and (PC30+LM40)+LM50 as basal and additional fertilizer. $NO_3^-$-N content in soil was decreased as along with the growth of plant, highest in LM100% as basal fertilization at early growth stage and highest in (PC30%+LM40%)+LM40% and CF100% at last growth stage. Amount of $NO_3^-$-N and $NH_4^+$-N in soil was high in (PC30%+LM40%)+LM40% and CF100% of top soil but in subsoil significant difference was little in all treatment. Amount of OM, $A_V.P_2O_5$, T-N, exchangeable Ca and Na in soil was higher (PC30%+LM40%)+LM40% than non-application after harvest. Amount of nutrient uptake in plant was higher in CF100% and split application of LM than LM 100% application. Nitrogen utilization rate was in the order of CF100% >LM50%+LM50%=LM50%+CF50%>(PC30%+LM40%)+LM40% >LM100%. The yield of whole crop barley in (PC30%+LM40%)+LM40% and CF100% was 3.2 times more than in non-application ($309kg\;10a^{-1}$). Feed values such as crude protein and TDN was increased 1.0% ~ 1.4% in LM as split application than basal 100% treatment. Accordingly, in order to increase yield of a whole crop barley with application PC+LM in reclaimed land treat split application rather than to treat LM 100% into the land.

The Effect of Application Levels of Slurry Composting and Bio-filtration Liquid Fertilizer on Soil Chemical Properties and Growth of Radish and Corn (총각무와 옥수수 재배시 SCB액비 시용수준이 토양화학성과 생육에 미치는 영향)

  • A liquid fertilizer treated with slurry composting and biofiltration (SCB) process has been applied increasingly on agricultural field but the effects on the soil properties and crop production has not been throughly evaluated. This study was conducted to investigate the effect of the SCB application on soil chemical properties and the growth of radish and corn. SCB liquid fertilizer as a basal fertilization was treated with five levels based on $6kg\;10a^{-1}$ for radish and $10kg\;10a^{-1}$ for corn. The experimental design was the completely randomized block design with five levels and three replicates. Electrical conductivity (EC), $NO_3$-N, Exch. K and Exch. Na increased depending on the treatment levels of SCB. There were no changes in soil organic matter, Avail. $P_2O_5$, Exch. Ca and Exch. Mg. EC, $NO_3$-N and Exch. Na content decreased as precipitation increased. Especially, they decreased up to the initial condition before the treatment after the heavy rainy season in 2008. Although Exch. K decreased at the rainy season, they remained relatively higher content after the experiment on August, 2008. Fresh weight and the amount of N uptake of radish increased due to the levels of SCB, but corn did not present any significant increase. It is recommended that we need to decide the proper amount of SCB as well as the application method on the field to increase the productivity and decrease environmental stress. Additional experiments also need to clarify the effect of the trace element and heavy metal accumulations due to long term application of SCB.

Evaluation of N2O Emissions with Different Growing Periods (Spring and Autumn Seasons), Tillage and No Tillage Conditions in a Chinese Cabbage Field (배추의 재배시기와 경운 유.무에 따른 아산화질소 배출 평가)

  • Importance of climate change and its impact on agriculture and environment has increased with a rise of greenhouse gases (GHGs) concentration in Earth's atmosphere. Nitrous oxide ($N_2O$) emission in upland fields were assessed in terms of emissions and their control at the experimental plots of National Academy of Agricultural Science (NAAS), Rural Development Administration (RDA) located in Suwon city. It was evaluated $N_2O$ emissions with different growing periods (spring and autumn seasons), tillage and no tillage conditions in a chinese cabbage field. The results were as follows: 1) An amount of $N_2O$ emissions were high in the order of Swine manure compost>NPK>Hairy vetch+N fertilizer. By tillage and no tillage conditions, $N_2O$ emissions were reduced to 33.7~51.8% (spring season) and 31.4~76.7% (autumn season) in no-tillage than tillage conditions. 2) In autumn season than those spring season, $N_2O$ emissions at NPK, hairy vetch+N fertilizer and swine manure compost were reduced to 49.6%, 39.0% and 60.0%, respectively, in tillage treatment and 59.5%, 70.6% and 58.7%, respectively, in no-tillage treatment. 3) $N_2O$ emission measured in this study was 15.2~86.4% lower with tillage and no tillage treatments than that of the IPCC default value (0.0125 kg $N_2O$-N/kg N).

Growth of 'Nokkwang' Hot Pepper Plug Seedlings as Influenced by Various Ratios of Pre-planting NH4+:NO3- in Root Substrate (상토에 기비로 혼합된 NH4+:NO3- 비율에 따른 '녹광' 고추 플러그 묘의 생장)

  • This study was conducted to determine the effects of a pre-planting fertilizers with various $NH_4{^+}:NO_3{^-}$ ratios in a coir dust:peatmoss:perlite (3.5:3.5:3.0, v/v/v) medium on the growth of hot pepper (Capsicum annuum L. cv. Nokkwang) plug seedling. Nitrogen levels were fixed to $300mg{\cdot}L^{-1}$ and the $NH_4{^+}:NO_3{^-}$ ratios were varied to 0:100, 27:73, 50:50, 73:27, and 100:0. The 50-cell trays were filled with treatment media containing pre-plant fertilizers, then seeds were sown. After seeds were germinated, the trays were moved to greenhouse and seedlings were feed with 13-2-13 and 20-9-20 fertilizers, alternatively. The changes in pH and EC were measured every week and soil solution for nutrient concentrations were analyzed in week 0, 3, and 7. The measurements of seedling growths as well as analysis of tissue nutrient contents were also conducted in week 7. The varied $NH_4{^+}:NO_3{^-}$ ratios did not influence on the pHs of root media after incorporation of pre-planting fertilizers, but the ECs were heightened as proportion of $NH_4{^+}$ to $NO_3{^-}$ were elevated. During the raising of seedlings, the pHs rose over time in the treatments of 0:100 and 27:73 ($NH_4{^+}:NO_3{^-}$). The concentrations of all macro-elements in root media decreased gradually as seedlings grew in all treatments. The seedling growths 7 weeks after seed sowing were the highest in the treatments of 27:73 and 50:50 ($NH_4{^+}:NO_3{^-}$) and those became worse in the treatments of higher $NH_4{^+}$ ratios than 73%. In terms of inorganic element contents based on the dry weight of above ground tissue, the treatment of 0:100 showed the lowest content of Ca, Mg, Na, Cu, Mn, and Zn. Based on the results, it is desired that $NH_4{^+}$ ratio in pre-planting fertilization is maintained to be 50% or less for the raising of hot pepper plug seedlings.

Effects of Co-digestate application on the Soil Properties, Leachate and Growth Responses of Paddy Rice (통합혐기소화액의 시용이 벼 생육 및 논토양 환경에 미치는 영향)

  • Livestock manures have a potential to be a valuable resource with an efficient treatment. In Korea, 42 million tons of livestock manure were generated in 2008, and 84 % of them were used for compost and liquid fertilizer production. Recently recycling of livestock manure for biogas production through anaerobic digestion is increasing, but its utilization in agriculture is still uncertified. In this study, there was applied co-digestate to the paddy for rice cultivation based on N supplement. Co-digestate was fertilizer fermented with pig slurry and food waste combined with the ratio of 70:30(v:v) in its volumetric basis. For assessing the safety of co-digestate, it was monitored the contents of co-digestate for seasonal variation, resulted in no potential harm to the soil and plant by heavy metals. The results showed that soil applied with co-digestate was increased in exchangeable potassium, copper and zinc mainly due to the high rate of pig slurry in co-digestate applied. Considering high salt content due to the combination with food waste, strict quality assurances are needed for safe application to arable land though it has valuable fertilizer nutrient. Leachate after treatment showed that the concentration of nitrate nitrogen washed out within two weeks. Considering the salt accumulation results in soil, it is highly recommended that the application rate of co-digestate should not exceed the crop fertilization rate based on N supplement. With these results, it was concluded that co-digestate could be used as an alternative fertilizer for chemical fertilizer. More study is needed for the long-term effects of co-digestate application on the soil and water environment.

Studies on the Productivity of Individual Leaf Blade of Paddy Rice (수도의엽신별 생육효과에 관한 연구)

  • Experiment I: A field experiment was conducted in an attempt to find the effect of top-dressing at heading time in different levels of nitrogen application and of different positioned leaf blades formed by the treatment of leaf defoliation at heading time on the ripening and the yield of rice. The results obtained are as follows: 1. Average number of ears per hill and average number of grains per ear in different levels of nitrogen application were increased as the amount of nitrogen applied was increased. while the rate of ripened grains the yield of rough rice and the weight of 1, 000 kernels of brown rice were decreased respectively as the amount of nitrogen applied was increased. 2. The rate of ripened grains and the weight of 1.000 kernels of brown rice in different levels of nitrogen, top-dressing at heading time were larger than those in control and increased. The yield of rough rice although statistically significant differences were not recognized, were numerically increased. 3. The rate of ripened grains, the yield of rough rice, the weight of 1, 000 kernels of brown rice and the rate of hulling in different treatments of leaf defoliation were remarkably decreased as the degree of leaf-defoliation became larger. 4. The rate of ripened grains, the yield of rough rice, the weight of 1, 000 kernels of brown rice and the rate of hulling in different combinations of number of remained leaves positioned differently, formed the order of $L_1(flag leaf)>L_2>L_3>L_4$ when only one leaf blade was remained, and were increased as the positions of leaves were higher when two leaf blades. were, remained. 5. In case of decrease in the number of leaf blades positioned differently, by the treatment of leaf. defoliation, rate of ripened grains, the yield of rough rice, the weight of 1, 000 kernels of brown rice and the rate of hulling were increased as the area of remained leaves became larger and the nitrogen content of a leaf blade was increased. 6. There was a tendency that the increase in the amount of fertilizer application made the rate of ripened grains and the weight of 1, 000 kernels of brown rice reduced in any number of remained leaf blades, but the application of top-dressing at heading. time resulted in the reverse tendency. The yield of rough rice showed a tendency to be increased as the amount of basal dressing and top-dressing increased and for the application of top-dressing at heading time, the yield of rough rice was less at the smaller number of those. 7. The productivity effect of the rate of ripened grains and the yield of brown rice covered by leaf blades was more than 50 per cent and that of the. weight of 1, 000 kernels of brown rice was not more than 1.0 percent. As the amount of nitrogen application increased the. effect of leaf blades on the rate of ripened. grains and the weight of 1, 000 kernels of brown rice was increased. The effect of leaf blades on the weight of brown rice was increased as the amount of basal dressing-application, but the effect was decreased as the amount of top-dressing at heading time increased, 8. The productivity effects of different positioned leaf blades on the rate of ripened grains, the yield of rough rice and the weight of 1, 000 kernels of brown rice were in order of $L_1(flag leaf)>L_2>L_3>L_4$ the productivity effects of $L_1$ and $L_2$ had a tendency to be increased as the amount of nitrogen applied was increased. Experiment II: A field experiment was done in order to disclose the effect of the time of nitrogen application on yield component and the effect of different positioned leaves formed by leaf defoliation at heading time on the rate of ripened grains and the yield of rice. The results obtained are as follows: 1. Average number of ears per hill was increased in the treatment of nitrogen application from basal dressing to 22 days before heading and in the treatment of application distributed weekly. Number of grains was increased in the treatment of nitrogen application from 36 days to 15 days before heading. The rate of ripened grains was, lower in the treatment of nitrogen application from top-dressing to 15 days before heading than in that of non-application, was higher in the treatment of nitrogen application within 8 days before heading, and was the lowest in that of application 29 days before heading. The yield of rough rice was the highest in the treatment of nitrogen application from 29 days to 22 days before heading. The weight of 1, 000 kernels of brown rice was a little high in the treatment of application from 29 days to 8 days before heading. 2. The rate of ripened grains the yield of rough rice, the weight of 1, 000 kernels of brown rice and the rate of hulling in different treatments of leaf defoliation were remarkably decreased as the degree of leaf defoliation got larger and there were highly significant differences among treatments. There was also a recognized interaction between the time of nitrogen application and leaf defoliation. 3. In relation to the rate of ripened grains, the weight of 1. 000 kernels of brown rice and the rate of hulling in different numbers of remained leaves positioned differently and their combinations, the yield components were in order of $L_1(flag leaf)>L_2>L_3>L_4$ when only one leaf was remained, which indicated that the components were increased as the leaf position got higher. When two laves were remained, the rate of ripened grains, the yield of rough rice and rate of hulling were high in case of the combinations of upper positioned leaves, and the increase in the weight of 1, 000 kernels of brown rice appeared to be affected most]y by flag leaf. Discussing the productivity effects of different organs in different times of nitrogen application, the productivity effect of a leaf blade on the rate of ripened grains was higher as the time of nitrogen application got later, and in the treatment of non-fertilization the productivity effect of a leaf blade and that of culm were the same. In the productivity effect on the yield of brown rice, the effect of culm covered more than 50 percent independently on the time of nitrogen application, and the tendency was larger in the treatment of non-fertilizer. The productivity effect of culm on the weight of 1. 000 kernels of brown rice was more than 90 percent, and the productivity effect of a leaf blade was increased as the time of application got later. 7. The productivity effect of a leaf blade in different positions on the rate of ripened grains, the yield of rough rice and the weight of 1, 000 kernels of brown rice had a tendency to be increased as the time of application got later and as the position of leaf blades got higher. In the treatment of weekly application through the entire growing period, the rate of ripened grains and the yield of rough rice were affected by flag leaf and the second leaf at the same level, the but the weight of 1, 000 kernels of brown rice was affected by flag leaf with more than 60 percent of the yield of total leaves.
