A series of experiments were conducted to determine the suitable freezing and thawing temperatures for the freezing of boar semen in 5 ml maxi-straws. The ultrastructure, in vitro fertilization (IVF) and artificial insemination (AI) of frozen-thawed semen were also be evaluated. The 5 cm freezing height gave the best results not only in post-thaw motility rate (54.00%), but also in normal acrosome morphology rate (NAR) (80.23%). There was no significant difference in the post-thaw motility between different thawing temperatures and corresponding thawing times (p>0.05); the group of $52^{\circ}C$ and 25 s gave the highest motility rate (45.00%). As a whole, not only from the motility but also the NAR, thawing at $42^{\circ}C$ was better than the other two treatments. In the freezing packages, 5 ml maxi-straw gave a little lower mobility (40%), viability rate (49.58%), plasma membrane integrity rate (53.91%) and NAR (52.65%) than the 0.25 ml straw, but there was no significant difference between the two straw volumes (p>0.05). The IVF capacity of frozen-thawed semen in this experiment was similar to fresh semen. From ultrastructure observation, the main damage to boar spermatozoa after freezing was seen in the acrosome, such as swelling and formation of vesicles. After AI in recipient Shanghai White sows, frozen-thawed semen from 5 ml maxi-straws and pellets produced 72.2% and 80% conception rate and 7.8 and 8 litter sizes, respectively, and there was no significant difference between the 5 ml maxi-straw and the pellet (p>0.05).
Chaenogobius laevis inhabit brackish waters. Adult male protects egg mass laid under the stones. The egg with small oil globule varied from 3.40 to 4.04 mm in diameter. When water temperature is $22.0\~23.0^{\circ}C$, larvae hatched about 113 hours after fertilization. The newly hatched larvae reared for 33 days grow up to 12.10 mm in total length and developed to the jevenile. When larvae length attained about 8.0 mm, jaw bones were more rapidly ossified than vertebrae and cranium.

The objective of this study was comparison of seed yields according to different seeding rates, seeding time and application of chemical fertilizations in Yanji region of China. The experiment was conducted on commercial farmland at Yanji region in 2013. All the experimental fields were designed following randomized block design with 3 replicates. The plant spacing was applied as $65{\times}1cm$ distance. To check the effect of seeding rate, two different seeding rate, 0.2 kg/0.1ha and 0.5 kg/0.1ha were applied. The higher seed yield was observed in low seeding rate (0.2 kg/0.1ha). The application of fertilizer (mixture of N and P) showed different results following different experimental fields. However, there were little positive effects following fertilizer application into commercial farmland. When we compare seed yields between two different seeding dates, $4^{th}$ May and $29^{th}$ May, the earlier seeding date ($4^{th}$ May) showed higher seed yields. In considering these results, low seeding rate and early seeding time is important for getting high camelina seed yields.

This study was initiated to investigate soil chemical properties under different soil systems. Data such as soil acidity(pH), electrical conductivity(EC), organic matter content(OMC), and cation exchange capacity(CEC) were analyzed with samples from multi-layer, USGA, and mono-layer systems. N, P, K and micronutrients were also measured. Multi-layer system was built up to 60-cm depth with rootzone layer, intermediate layer and two drainage layers. USGA system 45 centimeters deep was constructed with rootzone layer, intermediate layer and drainage layer. Mono-layer system, however, was made only with a 30-cm rootzone layer. Differences were observed in soil pH, EC, OMC, CEC and micronutrients. Soil pH was acceptable for turfgrass growth a year after establishment, being 5.5 to 6.5 in the study. Differences were greatly observed for EC among soil systems. Values of EC for multi-layer, USGA, and mono-layer systems were 39.79, 31.26 and 103.54 uS/em, respectively. The increase rate was approximately 4 to 8 times greater with mono-layer system than those with other two systems. Therefore, it was necessary to avoid micronutrient deficiency such as Fe, Mn etc. through an effective management program in mono-layer system because of its faster potential feasibility of salt accumulation. The greatest OMC was associated with USGA system, being 0.97% which was 11% over that of the other systems. Slight differences were observed for CEC among them. Mono-layer system produced 1.45 me/100g, 10.3% and 8.9% lower in CEC than those of multi-layer and USGA system, respectively. Micronutrients such as Fe, Zn, and Mn etc. were below the level required for turf growth, regardless of soil systems. It was considered that one year after turf establishment was not enough to build up micronutrients in sand-based soil systems to the normal level for a turf growth. These results demonstrate that intensive management program including grow-in concept fertilization should be integrated into sand-based soil systems, even after a year in establishment. Regular nutrient monitoring by soil analyses is a strong necessity to decide the kinds and amount of fertilizer. Also, strategic management program must be selectively employed according to sports turf soil systems.
This study was conducted to establish the optimal conditions for in vitro embryo production using oocytes derived from follicles of slaughter-house ovaries. The ovaries of Hanwoo were obtained from a local slaughter-house. The oocytes were aspirated from visible follicles of 2~7mm in diameter. The recovered oocytes which were completely surrounded by at least 2 layers of cumulus cells and a homogeneous cytoplasmic pigmentation were used. The selected oocytes were washed 3 or 4 times with D-PBS containing 10% bovine calf serum (BCS) and matured in vitor (IVM) in Ham's F-10 supplemented with 10% BCS or 0.01 $\mu\textrm{g}$/ml epidermal growth factor(EGF) at 39$^{\circ}C$ under 5% CO2 in air for 24 hours. They were fertilizqed in vitro (IVF) with fresh sperm separated by Percoll density gradient or swim-up in TALP media. The zygotes were cultrued with or without bovine oviductal epitherial cells(BOEC) in media(HECM-6 supplemented with 11 amino acid and / or TCM-199 supplemented with 10% BCS) for 7 to 10 days. The results obtained were as follow: The cleavage rate and developmental rate to blastocyst after maturation and IVF were not significantly different between Ham's F-10 with EGF(76.0% vs. 44.0%) and BCS(75.9% vs. 43.6%)(P<0.05). The cleavage rate and development rate to blastocyst after fertilizing by swim-up or Percoll method were not signifciantly(P<0.05) different between swim-up (80.2% vs. 29.2%) and Percoll(81.9% vs. 26.5%) (P<0.05). The cleavage rate in TCM 199(80.5) was signficiantly higher than that in HECM-6 (72.0%) (P<0.05). However, developmental rate to blastocyst using TCM 199 following HECM-6 for 3 or 4 days (42.2%) was significantly higher than that in TCM-199 alone(26.7%)(P<0.05). The cleavage rate and development rate of embryos produced in vitro by exchange timing for HECM-6 media were not significantly different between in day 3(78.6% vs. 45.5%) and day 4(75.0% vs. 43.2%)(P<0.05). The cleavage rate and developmental rate to blastocysts according to co-culture system were not significantly different between with (74.2% vs. 41.4%) and without BOEC(73.95 vs. 43.5%) (P<0.05). The number of blastomere in blastocyst stage after co-culture with or without BOEC was not significantly different (106.7$\pm$5.1 and 96.6$\pm$4.0). In conclusion, the most transferable IVP embryos could be produced from Ham's F-10 medium for IVM, Percoll density gradient method for IVF sperm separation and in vitro culture in HECM-6 until day 3 or day 4, and then transferred into TCM-199 until day 9 within adequate embryo density in culture droplets after insemination.
Spawning induction and early growth of the sea urchin, Strongylocentrotus intermedius were studied with the purpose of artificial seedling production. Gonadosomatic index(GSI) of the sea urchin showed the highest value in October, and rapidly decreased in December. It means that October and November is the peak of spawning season of the sea urchin in the latitude. Spawning induction by injection of potassium chloride solution in October has showed 44.0~100.0% reaction rate, and were produced 6,300$\times$10$^4$ eggs. Spawned eggs have shown the fertilization rate of 92.3~98.2% and the hatching rate of 78.2~87.0%. The metamorphosis of larvae after hatching in the seawater temperature of 13.7~17.1$^{\circ}C$ resulted in early eight-armed larvae in 13 days and late eight-armed larvae in 20 days. The collection of progenies was possible in 24~25 days after hatching and collection rate was 18.5~26.1% (mean 22.3%). Test diameter immediately after collection had a mean 350 ${\mu}{\textrm}{m}$. Survival rate and test diameter of juvenile sea urchin after collection were 58.5%, 1.32 mm in 30 days, 27.7%, 3.82 mm in 92 days and 15.6%, 11.70 mm in 181 days, respectively.
Implantation rates remain low following human in vitro fertilization (IVF). Suboptimal culture conditions may limit the ability of embryos to hatch as blastocysts, and artificial opening of the zona pellucida has been proposed as a means to promote subsequent hatching (assisted hatching). In this study, assisted hatching (AH) by zona drilling using acidic Tyrode's solution was performed in 320 patients, due to their age of more than 38 years (group A), the thick zona pellucida (group Z; $ZP\geq0.18{\mu}m$), and failures in implantation more than 3 times in previous IVF-ET trial (group P). This study was designed firstly, to study the effects of AH on the outcomes of IVF-ET according to the indications and secondly, to verify the appropriate application of AH. The results were as follows; 1. There was no difference in pregnancy rate between AH group (26.6%) and non-AH group (26.5%). 2. Assisted hatching (AH) showed significantly higher pregnancy rate of the patients with thick zona pellucid a than those of the patients with age factor and with the history of repeated implantation failure. But in the patients with age factor only, AH resulted in higher pregnancy rate. 3. Interestingly, the patients with complex factors including zona factor (Z: 33.9%; ZA: 30.4%; ZP: 31.6%; ZAP: 21.4%) showed higher pregnancy rates than other complex factors excluding zona factor (A: 24.4%, P: 0%; AP: 10.8%). From these results, AH is more helpful to the patients with thick zona pellucida rather than patients with older age and/or previous repeated implantation failure.

Forty schools having natural turf playgrounds were investigated by on-spot investigation and oral interviews with relevant school officials to find out basic information on turfgrass management practices of school playgrounds in Gyeonggi province. Average area of playground was $3,890m^2$ per school and $12m^2$ per student. Ninety five percent of turf playgrounds were managed by school staff and ninety percent of schools spent less than 5 million won per year for turf management. The difficulties in turfgrass management were considered as a major challenge for the schools, followed by turfgrass management cost. Among the management practices, school officials pointed out weed management as the most difficult work, followed by irrigation. The average number of fertilization and mowing was 2 and 6 times per year, respectively. About the half of playgrounds were irrigated only when there was wilting symptom. Zoysiagrass was the most popular choice for the school playgrounds and only three school playgrounds were established with pop-up irrigation system. Fourteen school playgrounds had good turf quality but the rest of school playgrounds had inadequate turf quality requiring minor or full renovation.

We had already reported the successful germination for green pods of purple lady's slipper orchid (Cypripedium macranthos Sw.). The green pod methods is to take immature seeds in green capsules, sterilize the capsule, and take out the sterile seeds. This method, however, needs very critical time of harvest. The critical time of seed harvest changes depending upon the species, condition of the specimen, and climatic influence, and the right time lies between 5 and 12 weeks after fertilization. In this study, the mature seeds were collected after 120-130 days with hand-polination of lady's slipper orchids. Mature seeds are usually dormant and it has to be overcome, either with hormone or storing the seeds near freezing for two or three months to break dormancy. The seeds were first surface sterilized with 70% ethanol and then transferred 1% NaOCl for 10-15 minutes, followed by rinses 3 times with sterilized distilled water. The cypripedium seeds consists of an embryo within a seed coat known as a testa. The testa is water repellent and the seed has a large air space between the embryo and testa so the seed tends to float on water. We had resolved the problems with vacuum pump to soak water into the testa before sterilization. The seeds were placed on liquid or agar solidified germination media. Cultures were incubated at $24{\pm}1^{\circ}C$ in dark. The seeds were germinated in 6-8 weeks in liquid suspension culture (germination percentage over 18%); however, the seeds on agar solidified media took more than 5 months to germinate and the germination percentage less than 5%. The most effective media for liquid culture was 1/4 strength Murashige and Skoog (MS) medium with 50 ml/l coconut water ($4brix^{\circ}$) at pH 5.8.
The spawning behavior, development of eggs and larvae of the Korea freshwater goby, Rhinogobius brunneus (Temminck et Schlegel) were studied. The eggs were spawned as a one-layer mass, hanging from the underside of a small pebble, and guarded by one male. The eggs were elliptic, about 1.48 mm in length and 0.65 mm in breadth, with a round top and a somewhat flat base with glutinous fibers. Hatching in the indoor tank with $17.0^{\circ}C$ in mean water temperature started from the 146 hours after fertilization. In the late developing stages, the embryo moved and freely changed its head to face the free tip of the chorion (normal egg) or to the basal end with a boundle of adhesive filaments (agrippa egg). Newly-hatched larvae were $3.10\~3.30$ mm in total length (mean: 3.22 mm), and mouth and anus were not yet open. Melanophores were present on the air- bladder, around the anus, and on the ventral part of the caudal region. The larva $3\~4$ days old transformed to postlarval stage, and they were $3.30\~3.85$ mm in total length (mean: 3.60 mm). As yolk sac and oil globules werw nearly absorbed, mouth and anus were open, and they fed rotifers actively. In $20\~22$ days after hatching the larvae grew to 5.85 mm in 71, and the caudal notochord flex at $45^{\circ}$.
