Bacillus 균주를 이용한 검정콩 청국장의 생리활성 및 Isoflavone 함량 (Some Biological Activities and Isoflavone Content of Chungkugjang prepared with Black Beans and Bacillus Strains)

  • Bacillus 균주를 이용하여 42$^{\circ}C$에서 72시간동안 발효된 검정콩 청국장의 몇 가지 생리활성 및 sioflavone(daidzein, genistein) 함량을 조사하였다. B. subtilis 보다는 분리균주 Bacillus megaterium SMY-212 균주를 첨가한 청국장이 항균활성이 매우 높았고, 대립 검정콩의 메탄올 추출물이 소립 검정콩의 것보다 항균 효과가 우수하였다. 청국장의 수소공여능은 대립 및 소립 검정콩 청국장에서 각각 76.4 및 75.5%를 나타내었으며, B. subtilis 및 SMY-212 를 첨가한 것은 무첨가보다 약간 높게 나타났다. 아질산염 소거능은 대립 검정콩이 소립 검정콩보다 그 효과가 크게 나타났으나, 균종간에는 차이가 나타나지 않았다. Linoleic acid 에 대한 과산화물가 및 흰쥐의 liver homogenate에 의한 TBARS 함량으로부터 볼 때 검정콩 청국장의 메탄올 추출물은 상당한 항산화효과가 있었다. 그 효과는 소립 검정콩의 청국장 추출물이 대립 검정콩의 청국장 추출물보다, 균주 첨가 청국장 추출물이 균주 무첨가 청국장 추출물보다 크게 나타났다. 청국장의 daidzein 및 genisteing함량은 발효 중 증가하였으며, 대립 검정콩 청국장이 소립 검정콩 청국장에 비하여 약간 높았다.

검정콩 청국장의 생리활성 및 발효중 Phytoestrogen 함량의 변화 (Biological Activities of Chungkugjang Prepared with Black Bean and Changes in Phytoestrogen Content during Fermentation)

  • 검정콩을 이용하여 제조된 청국장의 기능적 특성을 평가하기 위하여 검정콩 청국장의 메탄올 추출물에 대한 몇가지 기초적인 생리활성과 발효중 phytoestrogen화합물(daidzein, genistein)의 함량변화를 조사하였다. 대두 청국장의 추출물은 대부분의 시험균주에 대하여 거의 항균활성이 없거나 약하게 나타났으나, 검정콩 청국장의 추출물은 시험균주 모두에서 항균활성을 나타내었다. 특히, 소립 검정콩의 청국장 추출물의 항균활성이 대립 검정콩보다 강하였다. 대립 및 소립 검정콩 청국장의 수소공여능은 각각 76.4및 75.5%로서 대두 청국장의 67.3%에 비하여 더 높게 나타났다. 또한 아질산염 소거능은 각 청국장 추출물에서 대조구에 비하여 모두 90%이상을 나타내었다. Linoleic acid에 대한 과산화물 및 흰쥐의 간 지질에 대한 TBA의 수치는 청국장의 추출물을 첨가한 시험구가 첨가하지 않은 대조구에 비하여 낮았으며, 그 항산화 효과는 소립 검정콩, 대립 검정콩 및 대두 청국장의 추출물 순이었다. Daidzein 및 genistein의 함량은 청국장 발효중 점진적으로 증가하였으며, 대두 청국장에 비하여 검정콩 청국장의 함량이 훨씬 높았고, 대립 검정콩이 소립 검정콩으로 제조한 청국장의 함량보다 높게 나타났다. 또한 genistein 함량은 대체로 daidzein 함량의 2배정도였다.

Monascus Pilosus로 발효시킨 검정콩 첨가 식이가 고지방식이 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향 (Dietary effects of black bean fermented by Monascus pilosus on body weight, serum lipid profiles and activities of hepatic antioxidative enzymes in mice fed high fat diets)

  • The anti-obesity effects of fermented black bean were tested with mice fed a high fat diet for seven weeks. Body weight gain and feed efficiency ratio (FER) in the high fat diet control (HC) group were markedly higher, compared with those of the normal control (NC) group, but were significantly lower in the 2% black bean powder supplemented high fat diet (BB) group and 2% black bean powder fermented by M. pilosus supplemented high fat diet (BBM) group, compared with those of the HC group. Food intake in the HC and BB groups was significantly lower than that of the NC and BBM groups. Water intake in the HC group was significantly lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. On the other hand, relative liver and kidney weight in the HC group was lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. In addition, whereas epididymal fat weight in the HC group was markedly higher than that of the NC group, it was significantly lower in the BB and BBM groups, compared with that of the HC group. Meanwhile, hepatic GSH in the HC group was significantly lower than that of the NC group, but was slightly higher in the BB and BBM groups, compared with that of the HC group. Although hepatic LPO in the HC group was dramatically higher than that of the NC group, it was significantly lower in the BB and BBM groups, compared with that of the HC group. In addition, serum TG, total cholesterol, and LDL-cholesterol in the HC group was significantly higher than that of the NC group, but was significantly lower in the BB and BBM groups, compared with that of the HC group. On the contrary, HDL-cholesterol in the HC group was significantly lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. In addition, activity of XOR D type in the HC group was lower than that of the NC group, but was slightly higher in the BB and BBM groups, compared with that of the NC group. Activities of ROS scavenging enzymes, such as SOD, GPX, and GST in the HC group were significantly lower than those of the NC group, but were significantly higher in the BB and BBM groups, compared with those of the HC group. In addition, serum ALT activity in the HC and BB groups was higher than that of the NC group, but was significantly lower in the BB and BBM groups, compared with that of the HC group. In histopathological findings, hepatic fat accumulation in the HC group was higher than that of the NC group, but was lower in the BBM group, compared with that of the HC and BB groups. In particular, antiobese, hypolipidemic, and antifatty liver effect of black bean powder fermented by M. pilosus was specifically higher than that of non-fermented steamed black bean. In conclusion, the constituents of black bean fermented by Monascus pilosus have been proven to not only inhibit obesity and hyperlipidemia but also decrease hepatic fat accumulation in high fat diet-induced obese mice.

Bacillus subtilis에 의해 발효된 다양한 콩의 품질 특성 (Quality Characteristics of Various Bean Varieties Fermented with Bacillus subtilis)

  • Purpose: For dissecting the possible benefits and usages of Cheonggukjang, five different beans, soybean, black soybean, red bean, chickpea, and lentil bean, underwent the fermentation process along with Bacillus subtilis. Methods: Resultant Cheonggukjang physicochemical and sensory properties such as pH, amino type nitrogen, slime contents, rheological properties, and sensory strength and consumer acceptance were analyzed. Results: Values of pH were found to be highest in Cheonggukjang made with soybean (SC), followed by Cheonggukjang made with black bean (BSC) and Cheonggukjang made with red bean and soybean mixture (RBSC), and lastly Cheonggukjang made with chickpea (CBC) and lentil bean (LBC). Amino type nitrogen values were shown to be highest in SC with a value of $394.74{\pm}28.80mg%$, subsequently were values taken from RBSC, followed by BSC, CBC, and lastly LBC. Slime contents were the highest in SC with a value of $3.44{\pm}0.36%$, with RBSC, BSC, CBC, and LBC without statistical difference. Total colony count was found to show higher value in BSC, SC, and RBSC, in contrast to the lower values found in CBC and LBC. Color was shown to be lightest in CBC and with darker shades shown that of SC, LBC, RBSC, and BSC in descending order. In rheological properties, hardness, adhesiveness, and viscosity were found to be highest in SC. Sensory strength analysis and sticky appearance were highest in SC, with fermented odor being lowest in CBC and BSC. The strength of sweetness, sourness, and umami taste did not show statistical difference, however, bitterness was shown to be highest in LBC and lowest in CBC, SC, and BSC. In the consumer acceptance test, overall acceptance was highest in SC and CBC. Conclusion: In conclusion, SC was found to be the best bean variety for making Cheonggukjang fermented with Bacillus subtilis, however, CBC may act as an alternative for manufacture of Cheonggukjang resulting in sound consumer acceptance.

FUNCTIONAL PROPERTIES OF SAENGSIK (SUBSTITUTE MEAL) FORMULATED WITH UNCOOKED CEREALS FERMENTED BY LACTIC ACID BACTERIA

  • The objective of this study was to improve the qualities of Saengsik (substitute meal formulated with uncooked cereal powder) such as functionality, safety and sensory preference by using Lactic Acid Bacteria (LAB). The optimum formulation of fermented Saengsik (substitute meal) was verified as $40\%$ of fermented brown rice, $13\%$ of fermented job tears, $5\%$ of yellow soybean, $5\%$ of waxy rice, $5\%$ of sorghum, $3\%$ of Seumok bean, $1\%$ of sesame, $3\%$ of barley, $2\%$ of red bean, $5\%$ of prosomillet, $2\%$ of foxtail millet, $9%$ of black rice, $3\%$ of buckwheat, $1\%$ of chestnut, $2\%$ of pumpkin, $3\%$ of peanut. Fermented Saengsik decreased significantly serum glucose in STZ-induced diabetic rats. However, serum lipid levels were not affected by fermented Saengsik. In rats fed with high-fat diet, serum triglyceride level was slightly decreased and the contents of liver triglyceride were significantly decreased by supplementation of fermented Saengsik.

Components in Commercial Douchi-a Chinese Fermented Black Bean Product by Supercritical Fluid Extraction

  • Douchi, a popular seasoning agent in Chinese dishes prepared by the Aspergillus oryzae fermentation of black beans, was subjected to supercritical fluid extraction (SFE) and gas chromatography-mass spectrometry analysis for its volatile components. A total of 73 components were identified in two commercial brands, which were positively confirmed and quantified. Among the common components in the two brands were 18 acids, 12 alcohols, 11 aldehydes, 9 esters, 1 furan, 11 other oxygen-containing compounds, 4 pyrazines, 2 pyridines and 5 miscellaneous compounds. The most abundant components found were acetic acid, benzoic acid, 2,6-dimethylpyrazine, 2-piperidinone, 3-methylbutanoic acid, maltol, 4-ethylphenol, 2-methylpropanoic acid, butanoic acid, 2-pyrrolidinone, all fatty acids and some esters.

김치 유래 Lactobacillus plantarum을 이용한 팽화 검은콩 발효물의 제조 및 품질특성 (Manufacturing and Quality Characteristics of Puffed Black Bean Fermented by Lactobacillus plantarum Strains Isolated from Kimchi)

  • 본 연구에서는 유산균에 의한 검은콩의 최적 발효조건을 확립하고 발효된 검은콩의 품질특성을 평가하였다. 가정에서 제조된 배추김치로부터 분리한 Lactobacillus plantarum SU22는 병원성 세균에 대해 강한 항균 활성을 보였고 생체 아민과 발암성 효소인 β-glucuronidase를 생성하지 않았기 때문에 검은콩 발효의 스타터로 선정되었다. 검은콩은 5 kgf/cm2, 150℃로 조절된 팽화기에서 10분 동안 팽화 처리한 것과 처리하지 않은 것으로 각각 10-30% 혼합액을 만들고 L. plantarum SU22를 1%(v/v) 접종하여 37℃로 48시간 동안 진탕 배양하면서 유산균의 생육특성을 비교하였다. 유산균의 생균수는 L. plantarum SU22로 발효된 팽화 검은콩(20%) 배양액에서 9 Log CFU/mL 이상이었다. 20% 팽화 검은콩 배양액을 alcalase로 가수분해한 후 L. plantarum SU22로 발효하는 것이 최적 조건인 것으로 밝혀졌으며 유산균의 생균수가 10.30 Log CFU/mL까지 증가하였다. 최적 조건에서 총 폴리페놀 함량(94.02 mg GAE/g)과 DPPH 라디칼 소거능(92.50%)이 비 발효 대조군(87.74 mg GAE/g, 83.14%)에 비해 현저하게 증가하였다(P<0.05). 결론적으로 검은콩을 팽화한 후 alcalase 처리와 L. plantarum SU22를 병행하여 발효할 때 바이오제닉 아민이 없는 검은콩 발효액을 효과적으로 제조할 수 있으며, 총 폴리페놀과 DPPH 라디칼 소거능을 유의적으로 증가시킬 수 있다는 것을 알 수 있었다.

콩의 종류에 따른 증편의 품질특성 (The quality characteristics of Jeung-pyun made with different kinds of beans)

  • Jeung-pyun, a very popular fermented rice cake consumed in Korea, consists mainly of rice, rice wine (Tak-Ju), and sugar. The effects of addition of different beans on the quality characteristics of the batter and Jeung-pyun were investigated. Six different beans were mixed with the rice flour at levels of 5% and 10% of the rice flour weight, respectively. The addition of Back-tae, Huk-tae, Sori-tae, which are types of soybeans, and black gram significantly increased the batter volume and viscosity. However, the fermented Back-tae (Cheongguk-jang) was not effective at increasing the batter volume and viscosity. The buffering effect of the beans was very significant on the fermented batter, and the decrease in pH of the fermented batter made with beans was less than that of the control batter without beans. Additions of the soybeans and Cheongguk-jang were most effective for the buffering effect in the fermented batter. Although the Back-tae, Huk tae, and Sori-tae were different colors and shapes, they were all soybeans and exhibited similar effects on the Jeung-pyun batter. However, the effects of the beans were not significant on the Jeung-pyun. The volume and moisture content of the Jeung-pyun made with beans were not significantly different from the volume and moisture of the Jeung-pyun made without beans. The above results suggest that the addition of different soybeans, mung beans, and black gram significantly effects on the properties of Jeung-pyun batter, but not Jeung-pyun itself.

서리태 발효추출물이 탈색을 통해 손상된 모발에 미치는 영향 (Effect of Extract from Fermented Black Soybean (Glycine max var. Seoritae) on the Hair Damaged by Decolorization)

  • 본 연구에서는 서리태 발효추출물의 모발보호효과를 확인하기 위한 연구를 수행하였다. 화학적 산화를 통해 손상된 모발을 준비한 뒤 서리태 발효추출물을 처리하였을 때 모발의 형태학적 특성, 인장강도, 구성성분의 변화를 분석하여 비교하였다. 그 결과 모발에 산화제를 처리하였을 때, 표피의 큐티클 층이 손상되고 모발의 인장강도가 $14.32{\pm}0.83g/cm^2$에서 $12.32{\pm}0.79g/cm^2$로 감소되었음을 확인하였다. FT-IR 분석결과 산화제를 처리한 모발은 버진 헤어에 비하여 1,077, 1,041, $801cm^{-1}$에 해당하는 피크가 증가하였으며, 이를 통해 케라틴 단백질 간의 이황화 결합에 필수적인 시스테인이 산화되는 것을 확인하였다. 반면, 손상된 모발에 서리태 발효추출물을 처리한 경우에는 표피의 큐티클 층의 틈이 메워지고, 인장강도가 $14.27{\pm}0.96g/cm^2$로 회복되었으며, 모발의 성분 중 시스테인의 산화물 비율이 감소하는 것으로 분석되었다. 이러한 결과들을 통해 발효 서리태 추출물은 산화제에 의해 손상된 모발의 보호 소재로 연구될 가치가 있는 것으로 기대된다.

키위와 무를 첨가한 소립 검정콩 청국장의 정미성분 (Taste Compounds of Small Black Bean Chungkugjang Added with Kiwi and Radish)

  • 소립검정콩 청국장의 품질개선관 불쾌취 억압을 목적으로 키위와 무를 첨가하여 42$^{\circ}C$에서 3일간 발효시킨 소립 검정콩 청국장의 몇 가지 맛성분을 조사하였다. 소립 검정콩 청국장의 황색대두 청국장보다 유리아미노산 함량이 적었지만, 키위와 무를 첨가하여 발효시키면 유리아미노산의 함량이 증가되어 콩단백질 분해에 효과적이었다. 유기산은 모든 청국장에서 citric acid가 가장 많았으며, 다음올 acetic acid, lactic acid 순으로 많았다. 지방산은 linoleic acid (50.82~54.51%)> oleic acid(17.76~22.10%)> plamitic acid(12.13~13.79%) 순으로 많았다. 청국장의 종류별로 유기산과지방산 비율은 유의적인 차이가 없었다. 청국장의 주요 휘발성 성분으로는 indole, 2,5-dimethyl pyrazine 및 tri-methyl pyrazine 였으며, 청국장의 독특한 향미성분의 pyra-zine 류는 대두 청국장보다 키위와 무를 넣은 소립 검정콩 청국장에서 약 3배 정도 감소하였다. 주요 핵산관련 물질은 uracil과 UMP였으며, 다른것은 그 함량에 유사한 경향이었다.
