The antibiotic meroparamycin was produced in the free culture system of Streptomyces sp. strain MAR01. Five solid substrates (rice, wheat bran, Quaker, bread, and ground corn) were screened for their ability to support meroparamycin production in solid-state fermentation. In batch culture, wheat bran recorded the highest antibacterial activity with the lowest residual substrate values. The highest residual substrate values were recorded for both ground corn and Quaker. On the other hand, no antibacterial activity was detected for rice as a solid substrate. The use of the original strength of starch-nitrate medium in the solid-state fermentation gave a lower antibacterial activity compared with the free culture system. Doubling the strength of this medium resulted in the increase in the activity to be equivalent to the free culture. The initial pH (7.0) of the culture medium and 2 ml of spore suspension (1 ml contains $5{\times}10^{9}spores/ml$) were the optima for antibiotic production. The water was the best eluent for the extraction of the antibiotic from the solid-state culture. Ten min was enough time to extract the antibiotic using a mixer, whereas, 60 min was required when shaking was applied. Semicontinuous production of meroparamycin using a percolation method demonstrated a more or less constant antibacterial activity over 4 runs ($450-480{\mu}g/ml$). The semicontinuous production of the antibiotic was monitored in a fixed-bed bioreactor and the maximum activity was attained after the fourth run ($510{\mu}g/ml$) and the overall process continued for 85 days.
Attempts were made in the laboratory to produce bypass fat using acid oil by precipitation and fusion methods. The degree of saponification by both of these methods was above 80 percent. Where heating facilities are not available, precipitation method could be used, otherwise, fusion method of bypass fat production is found to be more convenient, especially for commercial scale operations as handling of large volume of solutions is eliminated. Bypass fat thus produced was tested in vitro for rumen fermentation. Incorporation of acid oil in the incubation medium reduced TVFA conc. from 127.06 to 124.09 mM/l SRL and increased ammonia-N levels from 210.50 to 223 mg/l SRL indicating that the microbial activity was affected on incorporation of acid oil in the incubation medium. However, incorporation of bypass fat in the incubation medium did not significantly affect TVFA conc. as well as ammonia-N levels. In another experiment, nine rumen fistulated sheep in three groups of three each were fed bypass fat at two different levels. Dry matter disappearance in 24 h from the nylon bags suspended in the rumen of animals under different groups was found to be $47.74{\pm}1.10$, $47.55{\pm}0.21$ and $50.74{\pm}1.11$ in group I (control), group II (fed bypass fat 50 g/day) and group III (fed bypass fat 100 g/day), respectively. These studies indicated that it is possible to produce bypass fat from acid oils, a by-product of oil refining process, and its feeding did not affect rumen fermentation.
Prodigiosin as a high-valued compound, which is a microbial secondary metabolite, has the potential for antioxidant and anticancer effects. However, the large-scale production of functionally active Hahella chejuensis-derived prodigiosin by fermentation in a cost-effective manner has yet to be achieved. In the present study, we established carbon source-optimized medium conditions, as well as a procedure for producing prodigiosin by fermentation by culturing H. chejuensis using 10 L and 200 L bioreactors. Our results showed that prodigiosin productivity using 250 ml flasks was higher in the presence of glucose than other carbon sources, including mannose, sucrose, galactose, and fructose, and could be scaled up to 10 L and 200 L batches. Productivity in the glucose (2.5 g/l) culture while maintaining the medium at pH 6.89 during 10 days of cultivation in the 200 L bioreactor was measured and increased more than productivity in the basal culture medium in the absence of glucose. Prodigiosin production from 10 L and 200 L fermentation cultures of H. chejuensis was confirmed by high-performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LC-MS) analyses for more accurate identification. Finally, the anticancer activity of crude extracted prodigiosin against human cancerous leukemia THP-1 cells was evaluated and confirmed at various concentrations. Conclusively, we demonstrate that culture conditions for H. chejuensis using a bioreactor with various parameters and ethanol-based extraction procedures were optimized to mass-produce the marine bacterium-derived high purity prodigiosin associated with anti-cancer activity.
612 strains isolated from Korean traditional fermented food in Sunchang and their investigated biochemical characterization and ability of biogenic amines non-producing. We selected the SCJ4 having various activity by measurement of extracellular enzyme, antioxidant and antimicrobial activities. Selected strain SCJ4 by 16S rRNA sequencing and biochemical characterization was named Bacillus subtilis SCJ4. And then, we investigated cell growth of SCJ4, and optimized of culture medium constituents using response surface methodology as statistically method. Response surface methodology used Plackett-Burman experimental design for screening of medium constituent. Tryptone, peptone and $MgSO_4$ as medium constituent improving cell growth selected. In order to find out optimal concentration on each constituent, we carried out central composite design. Consequently, optimized concentrations of tryptone, peptone and $MgSO_4$ were predicted to be 15.35 g/L, 12.235 g/L, and 3.5 g/L respectively. Through the model verification, we confirmed about 1.28-fold improvement of the dried cell weight from 0.8767 g/L to 1.1222 g/L when compared to basal medium.
In order to find out the inhibitors of acetic acid fermentation in Korean ginseng (Panax Sin son C. A. Meyer), total aglycone, panaxadiol, panaxadiol, oleanolic acid and ${\beta}$ -sitosterol were added to the basal medium, respectively, and a surface culture was carried out at 30$^{\circ}C$. The results were as follows: 1 . Saponins lost their activity to inhibit the acetic acid fermentation by hydrolysis. 2 Panaxadiol inhibited slightly, and the degree of inhibition was about 1/300 of that of free saponins. 3. Panaxadiol and oleanolic acid inhibited silighly similar to total aglycone. 4. Acetic acid fermentation was stimulated at the early stage when ${\beta}$-sitosterol was added to the media below the level of 0.000815%. But the fermentation was inhibited when media contained it more than that media 5. An over-oxidation of acetic acid was observed when the media contained total aglycone. panaxadiol, panaxatriol, oleanolic acid and ${\beta}$-sitosterol, respectively, while the media which contained sucrose, ginseng extracts ginseng saponins was shown not to be over-oxidized.
In order to study the effect of Korean ginseng (Panax ginseng C. A. Meyer) components on acetic acid fermentation, ginseng extracts, sucrose, total can de saponins were added to the basal niedium respectively and surface culture was carried out at 30$^{\circ}C$. Lag ime, total acidity of the fermentation broth inhibitors and the degrees of inhibition were determined in tile course of fermentation . 1. Acetic acid fermentation was not inhibited by the addition of less than 1.93% of sucrose but the degree of inhibition was increased slightly by the addition of sucrose more than that. 2. Ginseng extract inhibited acetic acid fermentation slightly, and the degree of inhibition was similar to that of sucrose. Lag time was about 72 hours when a 20% of ginseng extract was added to the basal medium while that of the control was 22hours. 3. The free saponins inhibited acetic acid fermentation considerably, and the degree of inhibition of the saponins was about 400 folds of that of ginseng extracts. An increase of total acidity of the broth which contained 2.905% of the saponins was not observed even after one month. 4. It was presumed that some other components except saponins and sucrose in ginseng extracts counter the inbition effect of saponins on acetic acid fermentation

Effects of protease-resistant antimicrobial substances (PRA) produced by Lactobacillus plantarum and Leuconostoc citreum on rumen methanogenesis were examined using the in vitro continuous methane quantification system. Four different strains of lactic acid bacteria, i) Lactococcus lactis ATCC19435 (Control, non-antibacterial substances), ii) Lactococcus lactis NCIMB702054 (Nisin-Z), iii) Lactobacillus plantarum TUA1490L (PRA-1), and iv) Leuconostoc citreum JCM9698 (PRA-2) were individually cultured in GYEKP medium. An 80 ml aliquot of each supernatant was inoculated into phosphate-buffered rumen fluid. PRA-1 remarkably decreased cumulative methane production, though propionate, butyrate and ammonia N decreased. For PRA-2, there were no effects on $CH_4$ and $CO_2$ production and fermentation characteristics in mixed rumen cultures. The results suggested that PRA-1 reduced the number of methanogens or inhibited utilization of hydrogen in rumen fermentation.

Garlic has been used for condiments and also for medicines to cure various diseases since ancient times. Many studies on the processing of garlic have been published, however, few of them were related with fermentation because of the antimicrobial action of the garlic. In this study, to conduct garlic fermentation, 4 lactic acid bacteria (LAB) strains with growth abilities in garlic medium were selected. Addition of various nitrogen, carbon, and mineral sources generally did not improve the growth of experimental strains during garlic fermentation except for Lactobacillus casei KFRI 704 by yeast extract and Lactococcus lactis subsp. cremoris ATCC 19257 by mineral sources. High performance liquid chromatography (HPLC) analysis of 32 phenolic compounds during fermentation showed that formononetin was decreased time dependently. The concentrations of volatile compounds and alliin did not change during fermentation. The results of this study would provide the basic understanding of garlic fermentation by selected strains of LAB.

In this study, we optimized the composition of the indole-3-acetic acid (IAA) production medium using response surface methodology on Pantoea agglomerans SRCM 119864 isolated from soil. IAA-producing P. aglomerans SRCM 119864 was identified by 16S rRNA gene sequencing. There are 11 intermediate components known to affect IAA production, hence the effect of each component on IAA production was investigated using a Plackett-Burman design (PBD). Based on the PBD, sucrose, tryptone, and sodium chloride were selected as the main factors that enhanced the IAA production at optimal L-tryptophan concentration. The predicted maximum IAA production (64.34 mg/l) was obtained for a concentration of sucrose of 13.38 g/l, of tryptone of 18.34 g/l, of sodium chloride of 9.71 g/l, and of L-tryptophan of 6.25 g/l using a the hybrid design experimental model. In the experiment, the nutrient broth medium supplemented with 0.1% L-tryptophan as the basal medium produced 45.24 mg/l of IAA, whereas the optimized medium produced 65.40 mg/l of IAA, resulting in a 44.56% increase in efficiency. It was confirmed that the IAA production of the designed optimal composition medium was very similar to the predicted IAA production. The statistical significance and suitability of the experimental model were verified through analysis of variance (ANOVA). Therefore, in this study, we determined the optimal growth medium concentration for the maximum production of IAA, which can contribute to sustainable agriculture and increase crop yield.

The aim of this study was to establish the optimal medium composition for enhancing L(+)-lactic acid (LLA) production using response surface methodology (RSM). Lactobacillus paracasei SRCM201474 was selected as the LLA producer by productivity analysis from nine candidates isolated from kimchi and identified by 16S rRNA gene sequencing. Plackett-Burman design was used to assess the effect of eleven media components on LLA production, including carbon (glucose, sucrose, molasses), nitrogen (yeast extract, peptone, tryptone, beef extract), and mineral (NaCl, K2HPO4, MgSO4, MnSO4) materials. Glucose, sucrose, molasses, and peptone were subsequently chosen as promising media for further optimization studies, and a hybrid design experiment was used to establish their optimal concentrations as glucose 15.48 g/l, sucrose 16.73 g/l, molasses 39.09 g/l, and peptone 34.91 g/l. The coefficient of determination of the equation derived from RSM regression for LLA production was mathematically reliable at 0.9969. At optimum parameters, 33.38 g/l of maximum LLA increased by 193% when compared with MRS broth as unoptimized medium (17.66 g/l). Our statistical model was confirmed by subsequent validation experiments. Increasing the performance of LLA-producing microorganisms and establishing an effective LLA fermentation process can be of particular benefit for bioplastic technologies and industrial applications.
