This study was designed to observe the effect of green tea on colon tumor incidence and biomarkers of colon carcinogenesis in 1, 2-dimethlhydrazine-treated rats. Male Sprague Dawley rats at 7 weeks of age were divided into two groups: control and green tea(GT) groups. Control rats had distilled water as drinking water but GT group received green tea extracts(2.5%, w/v water) as drinking water throughout the experiment periods. All rats were fed the experimental diet containing 15% fat by weight for 20 weeks. and were i.m. injected with DMH for 6 weeks to give total dose of 180mg/kg body weight. Tumor incidence was reduced in GT group (39%) compared with control group (56%) Green tea significantly reduced cell proliferation (total cells per crypt, crypt length and proliferative zone) in colonic mucosa and also significantly reduced the levels of preformed prostalandin E2(PGE2) and thromboxance B2(TXB2) in colonic mucosa but the fatty acid profile of total lipid in colonic mucosa was not significantly influenced by green tea. However the relative percent of C20:4 and the levels f preformed PGE2 and TXB2. were significantly higher in tumor tissue compared with normal surrounding mucosa.Green tea increased the fecal excretion of total bile acid but not scondary bile acid which is known as one of promoters for colon cancer,. These results suggest that green tea could have preventive effect against colon cancer when consumed daily by influencing on antioxidant effect and the metabolism of arachidonic acid.
The effects of diet induced hyperlipidemia on kidney function were studied in Sprague-Dawley rats fed high fat diet containing 20% beef tallow and high cholesterol diet containing 5% cholesterol for 8, 12, 16 weeks, respectively. The concentrations of serum total lipid, total cholesterol and LDL-cholesterol were significantly high cholesterol diet groups during all experimental periods (p<0.05). HDL-cholesterol concentration was the lowest value in high cholesterol diet group of 16 weeks(p<0.05). Triglyceride concentration was not affected by experimental diets. Serum total protein, albumin and creatinine concentration tended to higher in high cholesterol diet groups than those in high fat diet groups. And serum urea-N concentration was higher in high fat diet group of 16 weeks than that in other diet groups. Urinary total protein and urea-N were higher in high cholesterol diet groups than those in other diet groups regardless of experimental period period. There was no significant difference in urinary creatinine concentratin among diet groups(p<0.05). GFR was lower in high cholesterol diet groups than that in high fat diet groups at 8, 16 weeks, respectively. Wet weight per body weight, total lipid, triglyceride, total cholesterol concentations of liver tissue were apparently high in high cholesterol diet groups(p<0.05). Kidney wet weight per body weight wer not affected by experimently diets, total lipid concentration of kidney tissue was significantly high in high fat diet groups of 12 weeks(p<0.05), kidney tissue triglyceride concentrations of high cholesterol diet groups of 12, 16 weeks apparently low, and total cholesterol concentration of kidney tissue was higher in experimental diet groups than that of control groups at 12, 16 weeks(p<0.05). Fecal excretion, total lipid, triglyceride and cholesterol concentrations of feces were markedly high in all high cholesterol diet groups except high fat diet group of 16 weeks. The results of light microscopic examination indicated that glomerulosclerosis was not obsrved in rats fed experimental diets.
This study was conducted to fine out the preventive effects of chitosan and chitosan oligomer on the disorders of hepatic functions and lipid metabolism induced by 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) using adult male rats (SD) for four weeks. Rats were fed chitosan ($4\%$) or chitosan oligomer ($4\%$) diets respectively before 3weeks of TCDD treatment (50 ug/kg BW) by intraperitoneal injection and then continually supplied these diets for one week until being sacrificed. The elevation of serum total and LDL cholesterol levels induced by TCDD treatment was significantly reduced in the rats fed chitosan diets. The increment of liver triglyceride levels caused by TCDD treatment was tended to suppress in all rats fed chitosan and chitosan oligomer diets. Fecal total lipid and cholesterol excretion were high levels in the rats fed chitosan diets. The hepatic cytosolic catalase activities significantly decreased by TCDD treatment appeared recovering trend by chitosan diets. In hepatic microsomal cytochrome p-450, NADPH cytochrome p-450 reductase, ethoxycoumarin-o-deethylase (ECOD) and benzphetamin N-demethylase (BPND) chitosan than chitosan oligomer diets apparently decreased the increasing levels by TCDD treatment. In histochemical observation the fat droplets and apoptosis of hepatocytes by TCDD treatment were markedly alleviated by chitosan and chitosan oligomer diets. These results indicate that chitosan, more than chitosan oligomer can exert preventive effects on some disorders of hepatic functions and lipids accumulation by TCDD.
This study was conducted to investigate the supplementary effects of Suwon 464 developed by Rural Development Administration, which has over two times of dietary fiber content compared with normal rice (Ilpum), on lipid metabolism in diabetic mice. We supplied 5 kinds of experimental diets (com starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight, organ weights, and lipids levels of serum, liver and feces were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, 1.4, and 2.0% respectively. Body weight and liver and epididymal fat pad weights were lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentrations of serum triglyceride was lower in SP and SB groups than CO and IP groups. The levels of hepatic total lipid and total cholesterol were significantly lower in SP and SB groups than CO group, and the level of hepatic triglyceride was lower in IB, SP and SB groups than CO group. The levels of total lipid and triglyceride excreted in feces were higher in IB, SP and SB, and the level of total cholesterol in feces was higher in SP and SB groups than CO group. These results suggested that the high dietary fiber rice (Suwon 464) decrease the triglyceride or total cholesterol concentrations of serum and liver by increasing of fecal lipid excretion in diabetic mice. This study was designed to investigate the effects of caffeine intake on Ca and P metabolism in ovariectomized rats. For this purpose, the ovariectomized female rats weighing 230.8$\pm$3.5g were divided into four groups, eight rats each ; control, low caffeine(LC), medium caffeine(MC)and high caffeine(HC) groups. They were supplied for six weeks with the caffeine of 0mg, 66.8mg, 167mg and 334mg per kilogram of diet, respectively, and the results are summarized as follows. 1) There were no significant differences in feed intake, feed efficiency ratio and body weight change among all of experimental groups. But liver weight(both total weight and weight/body weight) was significantly decreased by caffeine in MC group. 2) The weight of tibia was decreased by caffeine intake in MC and HC groups. But the length, Ca and P content of tibia and femur was not changed in all of caffeine groups. 3) Ca, PTH and calcitonin levels in serum were not affected by caffeine. While, serum P level in HC group was significantly increased as compared with the control. 4) The fecal excretion of Ca and P tended to be higher in the caffeine groups, and as the result, the absorption rate, retention and retention rate of Ca and P tended to be decreased. It was noteworthy that P retention was significantly lowered in HC group as compared with LC group. The results showed that, when caffeine was taken by ovarietomized rats, the weight of tibia was decreased and the retention rate of Ca and P tended to be lowered. Therefore, too much intake of caffeine for women whose bone mineral density of tibia is decreased after postmenopause seems to accelerate the decrease of bone mineral density due to the negative effect on metabolism of Ca and P.
To evaluate long-term metabolic response of free-living Korean subjects consuming local sources adequately or slightly below habitual intake. Subjects, six male medical students aged 23-25 years were free-living, but consumed diets in the metabolic unit. They were healthy as determined from medical laborarory tests. They belong to the Korean middle-class. Temperature of outdoor during study were ranged between -2 to $26^{\circ}C$, whereas the indoor were $18-22^{\circ}C$. Animal protein ranged between 25-30% of total calorie. The three meals and a vitamin supplement to meet recommendation were given daily. Three 50day periods following the design were used to test the protein-energy adequacy of the diet. Urinary nitrogen excretion was measured daily. Fecal samples were pooled for the last 5 days of each week. Nitrogen balance was computed using 5mg N/Kg skin and miscellaneous losses. Body weight was measured daily. The study was covered on 126 metabolic observations weekly. Body weight changes were relatively stable with the energy intake of 37-44Kca1/kg, and N-balances were also relatively stable, but it was influenced by physical and or mental stress somehow. It could be assumed that the mean protein and energy requirement were $190{\pm}10mg$ N/Kg with $41{\pm}2$ Kcal/kg, and the mean digestibility of protein was 81% on these subjects with the experimental conditions. The protein requirement, however, could be cut down somewhat with some limited increaing of energy intake under parameters of N-balance and maintaining body weight. It was also observed a tendency of the requirement were getting smaller with the increase of body size.

The study was conducted to investigate the effects of dietary calcium and soy isoflavone on body fat and lipid metabolism in high fat-induced obesity. Four week old female C57/BL6J mice, known as a good model of diet-induced obesity, were fed low Ca and high fat diet for 6 weeks. After induced obesity, mice were divided into six groups according to diets varying calcium contents (0.1 or 1.5%) and genistein contents (0 or 500 or 1,000 ppm). Body weight, fat pad (perirenal fat and parameterial fat), adipocyte size, serum total lipid and total cholesterol were significantly decreased by both high Ca intake and genistein supplementation. However, the effect of genistein supplementation showed in low Ca-fed groups. Serum LDL-cholesterol and TG were significantly decreased by high Ca intake and genistein supplementation, respectively. In liver, lipogenic enzymes (fatty acid synthase and malic enzyme) activity and TG were significantly decreased by both high Ca intake and genistein supplementation. This inhibitory effect of genistein on lipogenic enzymes showed in low Ca-fed groups. But liver total cholesterol and total lipid were significantly decreased by high Ca intake and genistein supplementation, respectively. Fecal excretion of total lipid, total cholesterol and TG were significantly increased by high Ca intake, not by genistein supplementation. In conclusion, high calcium intake and genistein supplement may be beneficial for suppression of obesity through direct anti-adipogenesis by decreasing fat weight and size and indirect anti-lipo-genesis by inhibiting lipogenic enzymes activity and improving lipid profile.
Dongchunghacho rice, produced by cultivating Dongchunghacho fungus on rice, could be an effective functional food because it offers added value to rice and thus increases rice consumption. However, the physiological effect of Dongchunghacho rice has not been reported yet although there is increasing consumers demand. Therefore, we investigated the effect of Dongchunghacho rice (unpolished rice cultivated with Cordyceps militaris) on lipid metabolism in hyperlipidemic rats. Forty of 8 wk-old male Sprague-Dawley rats were divided into four groups after a 1 week of adaptation period and fed either a normal diet (66% polished rice diet, NC), high fat (12 g/100 g)-high cholesterol (1 g/100 g) diet with 53% polished rice (HC), or high fat-high cholesterol diets supplemented with 30% of the total rice as either unpolished rice (DR) or Dongchunghacho rice (DR). After 4 weeks, rats fed the Dongchunghacho rice diet with high fat and cholesterol had dramatically lower plasma LDL cholesterol concentrations and atherogenic indexes and higher plasma HDL cholesterol levels compared with the rats consuming polished rice or unpolished rice with high fat and cholesterol diet. Dongchunghacho rice led to less total lipid and total cholesterol accumulation in liver. However, these significant reductions in plasma or hepatic lipid profiles were not closely correlated with fecal total lipid or total cholesterol excretion. The plasma concentration of total cholesterol and triglycerides were not affected by Dongchunghacho rice. This hypolipidemic effect of Dongchunghacho rice seemed to be unrelated to unpolished rice itself, because the plasma and hepatic lipid profiles of DR group were not different from that of the HC group. These results suggest that unpolished rice containing cultivated Cordyceps militaris can improve plasma and hepatic lipid profiles in rats fed with high fat-high cholesterol diet.
This study was carried out to investigate the supplementary effects of soybean oil and rice gem oil compared with lard on lipid metabolism of insulin dependent diabetic mice. Streptozotocin-induced diabetic mice were fed three kinds of experimental diets with 20% lipid from lard(L), soy bean oil(SBO) and rice gem oil(RGO) for 7 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in diet intake, body and organs weights among experimental groups. But the concentrations of serum triglyceride of SBO and RGO groups, and of serum total cholesterol were lower in RGO group than in the other groups. The hepatic total lipid and total cholesterol levels of RGO group were significantly lower than those of the other groups. The contents of total lipid and total cholesterol excreted in feces of SBO and RGO groups were higher than those of L group, and the significance was shown only in RGO group. These results suggested that soy bean oil and rice germ oil can reduce serum triglyceride and total cholesterol levels and hepatic total lipid concentration of insulin dependent diabetic mice compared with lard as a animal fat source by increasing fecal lipid excretion of these groups. But the significant reducing effects on serum and liver lipid levels were shown only in RGO group, and we need to investigate the hypolipidemic effect of this oil by supplementary level and period.

Three barrows weighing 45.0 kg, fitted with simple T-cannulas in both the duodenum and terminal ileum, were assigned to diets in a $3{\times}3$ Latin Square design experiment to determine the effect of two calcium levels (0.8% vs 0.4%) on phytase activity and nutrient balance in growing pigs. The control diet contained 0.8% calcium, with no added inorganic phosphorus (0.45% total phosphorus) and no added phytase. The two additional experimental diets contained microbial phytase (750 phytase units/kg) and supplied either 0.8% or 0.4% calcium. With added microbial phytase, ileal and total tract digestibility of rotal phosphorus were improved by 20.9 and 13.8 percentage units, respectively (p=0.01). The apparent duodenal and ileal digestibility of phytate phosphorus were increased by 51.8 and 49.7 percentage units (p=0.01). Lowering dietary calcium in the presence of microbial phytase increased the digestibility of phytate phosphorus by an additional 10.9 (p=0.001) and 5.7 percentage units for duodenal and ileal digestibility, respectively. Supplementation with microbial phytase significantly reduced fecal excretion of nitrogen and phosphorus and increased the percentage of these nutrients retained by the pig. Lowering dietary calcium further increased the percentage of dietary phosphorus retained. Overall, reducing dietary calcium appeared to increase the effectiveness of added microbial phytase in degrading phytate phosphorus. As a result, care should be taken to avoid high levels of dietary calcium when supplementing swine diets with microbial phytase.
