자발성 고혈압 흰쥐에서 찐콩과 청국장 분말의 혈압 및 지질대사 개선 효과 (Improving Effect of Powders of Cooked Soybean and Chongkukjang on Blood Pressure and Lipid Metabolism in Spontaneously Hypertensive Rats)

  • 자발성 고혈압 흰쥐(SHR)에서 찐콩과 청국장 분말이 혈압 및 지질대사에 미치는 효과를 조사하여 가열처리 및 발효과정을 거친 콩제품의 항동맥경화 효과를 평가하고자 하였다. 식 이의 단백질 급원으로써 카제인, 찐콩 분말, 그리고 청국장분말을 단백질 함량으로 각각 18% 되게 첨가한 semi-pu-rified diet를 SHR(n=10, 숫컷)에 6주간 공급한 후 혈압, 혈장과 간의 지질농도, 간의 HMG-CoA reductase 활성 및 분변의 담즙산 배설능을 분석하였다. 그 결과, 카제인 대조군에 비교하여 찐콩과 청국장 섭취군의 체중증가량은 차이를 보이지 않았으며, 식이효율은 오히려 높게 나타났다(p<0.05). 찐콩 및 청국장의 장기간 섭취는 자발성 SHR의 가령에 따른 혈압상승을 효과적으로 억제하였으며(p<0.05) 청국장은 찐콩보다 우수한 혈압 강하효과를 보였다. 혈중 콜레스테롤 함량은 실험군간에 유의적인 차이가 없었으나 중성지방의 농도는 대조군에 비해 찐콩 섭취군에서 다소 낮았으며 청국장 섭취군에서 유의적으로 낮았다(p<0.05). LDL-콜레스테롤 농도는 대조군에 비해 찐콩 및 청국장 섭취군에서 유의적으로 낮았으며 항동맥경화인자로 알려진 HDL-콜레스테롤 농도는 유의적으로 높게 나타났다(p<0.05). 따라서 동맥경화 지수는 대조군에 비해 찐콩과 청국장군에서 유의적으로 낮았으며, 청국장군에서 가장 낮은 동맥경화지수를 보였다(p<0.05). 간의 총지질과 중성지방 함량은 대조군에 비해 찐콩 및 청국장 섭취군에서 유의적으로 낮게 나타나(p<0.0.5) 혈장에서의 결과와 일치하였다. 간의 HMG-CoA reductase 활성은 찐콩과 청국장군에서 유의적으로 낮았다 (p<0.05). 분변 중 총지질량은 세 군간에 유의적인 차이가 없었으며 콜레스테롤 배설량은 찐콩군에서 가장 높았으며, 분변으로의 담즙산 배설능은 청국장군에서 유의적으로 높았다(p<0.05). 이상의 결과는 단백질 급원으로 카제인을 대체한 찐콩과 청국장의 섭취는 동맥경화 위험인자인 혈압과 고지혈증을 개선함으로써 순환기계질환을 예방하는데 도움을 줄 수 있음을 제시하였다.

PHYSIOLOGICAL RESPONSES, FEED INTAKE, URINE VOLUME AND SERUM OSMOLALITY OF AARDI GOATS DEPRIVED OF WATER DURING SPRING AND SUMMER

  • In order to evaluate the adaptability of Aardi goats to arid environment, 5 Aardi bucks were deprived of water for four days during spring and summer seasons. The rise in average maximum ambient temperature from $24.8^{\circ}C$ in spring to $35.8^{\circ}C$ in summer caused a significant rise in rectal temperature ($0.3^{\circ}C$), respiratory rate (62%), serum osmolaity (8%) and serum sodium concentration (17%). While, it resulted in a significant decline in dry matter intake (50%), urine volume (74%) and fecal water excretion (60%) compared with their values in spring, but had no significant effect on the volume of drinking water. At the end of the 4-days deprivation period during spring, respiratory rate, dry matter intake and urine volume were reduced by 18, 77 and 91% relative to their average in control goats. The corresponding reduction in summer were 58, 100 and 100%. Serum osmolaity was risen by 15% in spring deprived goats and 29% in summer deprived goats. Rectal temperature rose by a mean value of $1.3^{\circ}C$ only in goats deprived of water in summer. Percent of moisture in the feces declined from 64 in control goats, to 24% in water deprived goats during spring season. The corresponding values in summer were 25 and 6%. These responses of Aardi goats deprived of water in summer indicate that they possess a water economy mechanism enable them to tolerate infrequent drinking in hot-arid environment.

Growth, Nutrient Utilization and Amino Acid Digestibility of Dairy Calves Fed Milk Replacers Containing Different Amounts of Protein in the Preruminant Period

  • This study was designed to examine the effects of different dietary protein levels on growth performance, nutrient utilization, amino acid (AA) digestibility and serum biochemical parameters of preruminant dairy calves. Fifteen healthy new-born calves were randomly allotted to three experimental groups and fed with different milk replacer that contained 18% (LP), 22% (MP) or 26% (HP) of protein. The results showed that final BW, net gain and ADG were significantly higher in the MP group than in LP and HP groups (p<0.05). In addition, the apparent digestibility of CP in the MP group was significantly higher than in the other two groups (p<0.05). The values of N intake and fecal N excretion were significantly increased following the increase of dietary protein content (p<0.05). However, in all three groups of animals, dietary protein content had no significant effect on urinary N concentration (p>0.05). BUN concentration, on the other hand, decreased as calves grew but increased following the increase of dietary CP content. Furthermore, no significant differences in digestibility of amino acids were observed among these three groups of animals (p>0.05). We concluded that calves fed with milk replacer containing 22% of protein had better growth performance and nutrient utilization as compared to animals treated with milk replacer containing either 18% or 26% of protein.

Digestibility of nitrogen and dry matter of oilseed meals and distillers dried grains supplemented in swine diets

  • This study was conducted to investigate the digestibility of dry matter (DM) and nitrogen (N) in oilseed meals and distillers dried grains (DDG) fed to growing-finishing pigs. As experimental animals, eleven barrows (initial body weight, $71.7{\pm}17.0kg$) were housed in individual metabolism cages. The experimental design consisted of an $11{\times}8$ incomplete Latin square with 11 dietary treatments and 8 replication periods. The diets were individually formulated with dehulled soybean meal produced in Korea (SBM-KD), soybean meal produced in India (SBM-I), soybean meal produced in Korea (SBM-K), corn high-protein distiller dried grains (HPDDG), tapioca distillers dried grains (TDDG), canola meal (CAM), corn germ meal (CGM), copra meal (COM), palm kernel meal (PKM), sesame meal (SM), and perilla meal (PM). Pigs with SBM-KD and SBM-K showed greater (p < 0.05) intake of N than SBM-I, HPDDG, and PKM. Total feces output was decreased (p < 0.05) in SBMs (SBM-KD, -I, and -K), HPDDG, and CGM compared with TDDG, SM, and PM. The DM in excreted feces was decreased (p < 0.05) in SBMs and CGM compared to TDDG, SM, and PM. Similarly, the SM and PM fed to pigs resulted in greater (p < 0.05) fecal excretion of N than the others. Apparent total tract digestibility (ATTD) of DM in SBMs and CGM was greater (p < 0.05) than TDDG, SM, and PM. The SBMs fed to pigs showed higher (p < 0.05) ATTD of N than TDDG, COM, SM, and PM. In conclusion, our results provided nutritional information about various ingredients and would be useful to contain more precise amounts of nutrients included in feed ingredients of pig diet.

한국인 성인남자의 리보플라빈 대사에 관한 연구 (A Study on the Metabolism of Riboflavin in Korean Men)

  • This study investigate the balance and biochemical status of riboflavin in Korean men. During the experimental period, four riboflavin diets with different levels of riboflavin(0.4, 0.6,0.8 and 1.0mg/1000kcal) were followed by eight healthy college men. the riboflavin status was assayed by erythrocyte glutathine reudcctase activity coefficient (EGRAC0 and urinary excretion of riboflavin. Riboflavin intake of the subjects who consumed a det was 0.46mg/1000kcal. the riboflavin intakes of the subjects who consumed the experimental diet with 0.4, 0.6, 0.8 and 1,0mg/1000kcal of riboflavin were 0.41, 0.60, 0.81 , 0.97mg, respectively. Fecal riboflavin loss, absorbed riboflavin , urinary riboflavin loss and retained riboflavin increased in the subjects consumed 0.4, 0.6, 0.8 and 1.0mg/1000kcal of riboflavin. The average EGRAC values for the subjects consumed 0.4, 0.6, 0.8 and 1.0mg/1000kcal of riboflavin were 1.303$\pm$0.029, 1.271$\pm$0.022, 1.239$\pm$0.013, 1.202$\pm$0.030, respectively and urinary riboflavin values ($\mu\textrm{g}$/g creatinine) were 86.89$\pm$ 20.07, 123.88$\pm$ 15.88, 240.70$\pm$57.14 and 393.36$\pm$76.94, respectively. Results indicate that 0.6mg/1000kcal is the level of riboflavin intake needed to maintain urinary riboflavin within the normal range. And above1.0mg/1000kcal of riboflavin is need to maintain urinary riboflavin with in the normal range. And above 1.0mg/1000 of riboflavin is needed to maintain the EGRAC within the normal range. The riboflavin intake correlated positively with urinary riboflavin value, but correlated negatively with the EGRAC value. The EGRAC value correlated negatively to protein intake as well as animal protein intake. The linear equation of between riboflavin intake and EGRAc was EGRA=-0.1667 $\times$riboflavin intake +1.3710. The riboflavin intake to maintain EGRAc below 1.20 was calculated 1.02mg/1000kcal by the above equation.

Nutrient Digestibility and Metabolizable Energy Content of Mucuna pruriens Whole Pods Fed to Growing Pelibuey Lambs

  • The nutrient digestibility, nitrogen balance and in vivo metabolizable energy supply of Mucuna pruriens whole pods fed to growing Pelibuey lambs was investigated. Eight Pelibuey sheep housed in metabolic crates were fed increasing levels of Mucuna pruriens pods: 0 (control), 100 (Mucuna100), 200 (Mucuna200) and 300 (Mucuna300) g/kg dry matter. A quadratic (p<0.002) effect was observed for dry matter (DM), neutral detergent fibre (aNDF), nitrogen (N) and gross energy (GE) intakes with higher intakes in the Mucuna100 and Mucuna200 treatments. Increasing M. pruriens in the diets had no effect (p>0.05) on DM and GE apparent digestibility (p<0.05). A linear reduction in N digestibility and N retention was observed with increasing mucuna pod level. This effect was accompanied by a quadratic effect (p<0.05) on fecal-N and N-balance which were higher in the Mucuna100 and Mucuna200 treatments. Urine-N excretion, GE retention and dietary estimated nutrient supply (metabolizable protein and metabolizable energy) were not affected (p>0.05). DM, N and GE apparent digestibility coefficient of M. pruriens whole pods obtained through multiple regression equations were 0.692, 0.457, 0.654 respectively. In vivo DE and ME content of mucuna whole pod were estimated in 11.0 and 9.7 MJ/kg DM. It was concluded that whole pods from M. pruriens did not affect nutrient utilization when included in an mixed diet up to 200 g/kg DM. This is the first in vivo estimation of mucuna whole pod ME value for ruminants.

Effect of dietary protamine on lipid metabolism in ruts

  • Protamine has been widely used as a pharmaceutical product and natural food preservative. However, few studies have been conducted to assess the beneficial function of dietary protamine. This study examined the effects of dietary salmon protamine on serum and liver lipid levels and the expression levels of genes encoding proteins involved in lipid homeostasis in the liver of rats. Groups of male Wistar rats were fed AIN93G diet containing 2% or 5% protamine. After 4 weeks of feeding these diets, markedly decreased serum and liver cholesterol (CHOL) and triacylglycerol levels were noted. Increased activity of liver carnitine palmitoyltransferase-2 and acyl-CoA oxidase, which are key enzymes of fatty acid ${\beta}$-oxidation in the mitochondria and peroxisomes, was found in rats fed on protamine. Furthermore, rats fed protamine showed enhanced fecal excretion of CHOL and bile acid and increased liver mRNA expression levels of ATP-binding cassette (ABC) G5 and ABCG8, which form heterodimers and play a major role in the secretion of CHOL into bile. The decrease in triacylglycerol levels in protamine-fed rats was due to the enhancement of liver ${\beta}$-oxidation. Furthermore, rats fed protamine exhibited decreased CHOL levels through the suppression of CHOL and bile acid absorption and the enhancement of CHOL secretion into bile. These results suggest that dietary protamine has beneficial effects that may aid in the prevention of lifestyle-related diseases such as hyperlipidemia and atherosclerosis.

Targeted and sustained delivery of hydrocortisone to normal and stratum corneum-removed skin without enhanced skin absorption using a liposome gel

  • Judging from hydrocortisone concentration in dosing area, the extent of absorption was reduced in the liposome-gel formulation. However, higher and sustained skin concentrations of hydrocortisone were achieved for the liposome-gel as compared to the ointment. Drug concentration in both viable and deep skin reached its maximum within 0.5 h after application of both formulations to both skin types. Drug concentrations in both skins from the ointment declined with time, while those from the liposome-gel were greatly sustained. The sustainment by the liposome-gel was more remarkable in the viable skin than in the deep skin. Drug concentration in the viable skin could be maintained at a nearly constant level for over 8 h by applying the liposome-gel. As a result, a 5-fold higher viable skin drug concentration was obtained from the liposome-gel than from the ointment at 8 h after the application to the SC-removed skin. However, the plasma concentration of hydrocortisone at 4 h from the liposome-gel was only one-fourth (p<0.01) the value from the ointment when the drug was applied to the SC-removed skin, consistent with. the lower urinary (one-third, p<0.05) and fecal (one-half, p<0.05) excretion. Conclusions : Retarded diffusion of the drug from the skin to the systemic blood stream appears to be a potential factor in the sustained skin concentration of hydrocortisone from the liposome-gel, Interaction of hydrocortisone in the skin with phosphatidylcholine, a component of the liposomes and skin, may well be a factor in retarding the diffusion of the drug in the skin.

동물성과 식물성단백질이 한국인 젊은여성의 체내 철분이용도에 미치는 영향 (Effects of Dietary Animal Protein and Plant Protein on Iron Bioavailability in Young Korean Women)

  • This study was performed to investigate the effect of sources of protein on iron bioavailability in 10 healthy young Korean women. The 18-day metabolic study consisted of a 6-day adaptation period, 6-day moderate protein(60g protein/day, 18mg Fe/day) and 6-day high protein period(90g protein/day, 18mg Fe/day). During the moderate and high protein period, 5 subjects were fed the high plant protein meals(80% plant protein). Fecal excretion of dietary iron was significantly higher(p<0.05) in high protein high plant diet group(HPP, 9.48$\pm$1.61mg/day) than in high protein high animal diet group (HPA, 14.40$\pm$0.89mg/day). Apparent absorption and bioavailability of iron was also significantly higher(p<0.10) in HPA(40.7$\pm$5.3%, 6.46$\pm$1.61mg/day) than in HPP(14.4$\pm$5.3%, 2.39$\pm$0.89mg/day). But there was no significant difference between the high animal protein group and high plant protein group in moderate protein period. Serum iron concentration and transferrin saturation increased as animal protein intake increased, from 106.0$\pm$5.1ug/이 and 30.6$\pm$1.5% for MPA to 129.1$\pm$6.7ug/이 and 37.1$\pm$1.3% for HPA. Statistically positive correlations were shown not only between the level of dietary heme iron and apparent absorption(r=0.95, p<0.05), but also between serum iron concentration and apparent absorption(r=0.64, p<0.05). Negative iron balance was shown in two subjects fed the moderate protein meals. These results suggest that recommanded dietary allowances of iron may be under the need to maintain the positive balance, and iron bioavaliability increase by only high level of animal protein intake.

Effects of Cellulase Supplementation on Nutrient Digestibility, Energy Utilization and Methane Emission by Boer Crossbred Goats

  • This study examined the effect of supplementing exogenous cellulase on nutrient and energy utilization. Twelve desexed Boer crossbred goats were used in a replicated $3{\times}3$ Latin square design with 23-d periods. Dietary treatments were basal diet (control, no cellulase), basal diet plus 2 g unitary cellulase/kg of total mixed ration dry matter (DM), and basal diet plus 2 g compound cellulase/kg of total mixed ration DM. Three stages of feeding trials were used corresponding to the three treatments, each comprised 23 d, with the first 14 d as the preliminary period and the following 9 d as formal trial period for metabolism trial. Total collection of feces and urine were conducted from the 4th d of the formal trial, and gas exchange measures were determined in indirect respiratory chambers in the last 3 d of the formal trial. Results showed that cellulase addition had no effect (p>0.05) on nutrient digestibility. Dietary supplementation of cellulase did not affect (p>0.05) N intake and retention in goats. Gross energy (GE) intake, fecal energy and urinary energy excretion, heat production were not affected (p>0.05) by the cellulase supplementation. Total methane emission (g/d), $CH_4$ emission as a proportion of live weight or feed intake (DM, organic matter [OM], digestible DM or digestible OM), or $CH_4$ energy output ($CH_4$-E) as a proportion of energy intake (GE, digestible energy, or metabolizable energy), were similar (p>0.05) among treatments. There was a significant (p<0.001) relationship between $CH_4$ and live weight (y = 0.645x+0.2, $R^2$ = 0.54), $CH_4$ and DM intake (y = 16.7x+1.4, $R^2$ = 0.51), $CH_4$ and OM intake (y = 18.8x+1.3, $R^2$ = 0.51) and $CH_4$-E and GE intake. Results from this study revealed that dietary supplementation of cellulase may have no effect on nutrient digestibility, nitrogen retention, energy metabolism, and methane emission in goat.
