Chemical Composition of Pine Sprouts and Pine Needles for the Production of Pine Sprout Tea (송순차 제조를 위한 송순 및 솔잎의 화학적 조성)

  • As a basic study for preparing pine sprout tea, chemical components in pine sprouts and pine needles were analyzed as follows: In proximate composition the contents of most components except for crude fat were different between in pine sprouts and pine needles. Moisture content in pine sprouts was higher than that in pine needles. Calcium and potassium were major minerals contained in pine sprouts and needles, and their contents in pine needles picked in December were higher than those picked in June. Soluble tannin and vitamin C contents in leaf part of pine sprouts were much higher than those in stem part and their contents in pine needles were increased according to their growth. Free sugars like fructose, glucose and sucrose were contained in both pine sprouts and needles, and their contents in pine sprouts were higher in stem part as compared that in leaf part. Although fourteen kinds of amino acids were detected in pine sprouts and pine needles, their contents were extremely low. Amino acid composition between pine sprouts and pine needles was different each other, but major amino acids contained in them were same, those are acidic amino acids such as aspartic acid and glutamic acid. Amino acid contents in pine needles were increased according to the growth. In fatty acid composition in leaf part of pine sprouts, saturated fatty acid contents were higher than unsaturated fatty acid contents, but in stem part unsaturated fatty acid contents were higher. In pine needles the amount of saturated fatty acid was increased with the growth, but the amount of unsaturated fatty acid was rather decreased.

  • The saturated fatty acids(arachidic acid, stearic acid, and palmitic acid) were repeatedly applied on water surface, and their ${\pi}-A$ isotherms and Maxwell displacement current(MDC) were measured to identify the effect of the number of carbons of fatty acids on the properties of Langmuir films. The saturated fatty which contain higher carbon number showed higher MDC peak, and the less carbon it contains, the higher surface pressure they show. It was found the maximum of MDC peaks was as the molecular area just before the surface pressure increases in compression cycles of saturated fatty acids.

Studies on Fatty Acid and Lipid Components of Foods (식품중 지방산 및 기타 지질성분에 관한 연구)

  • Nutritive value of lipids was associated with composition of fatty acid. Unvalance diet may increase the incidence of diseases. Abundant of EPA and DHA, polyunsaturated fatty acid, were contained in fishes. Studies on 10 kinds of fishes, lipid components were determined. The results were as follows; 1. The content of crude fat 18.1% were high in eels, on the contrary the content of EPA 0.1% and DHA 0.1% were low. The content of crude fat were low in mackeral and sadrine, but the content of poly unsaturated fatty acid were high. 2. Iodine value 189 was high in sadrine, consequently, this results showed the interrelation between Iodine value and the content of polyunsaturated number. 3. The content of crude fat 1.3% in flounder(sole) were low and the content of cholesterol 20 mg% were lowest in present studies.

A Study on the Cellular Fatty Acid Profiles of Listeria spp. Isolated from Foods (일반식품에서 분리된 Listeria spp.의 지방산 조성에 관한 연구)

  • The distribution of Listeria spp. in various foods and its fatty acid composition were examined. A total 60 samples of dairy products(15), seafoods(20), meat products(18), factory waster(2), and salades(5) were tested. Listeria spp. was found 10 samples, showing about 16.7% detection ratio; dairy products 0(0%),,seafoods 1(5%), meat product 7(38.9%), and factory wastes 2(100%). Whereas L. welshimeri was isolated from meat products 1(5.6%) and factory wastes 1(50%). The cellular fatty acid composition determined by gas chromatography was found not to differ among L. innocua isolated from food has similar fatty acid profiles when grown at 3$0^{\circ}C$,24 hrs on the tryptic soy plate with C15 and C17 anteiso branched acids accounting for about 80% of total.

  • This study was performed to investigate lipid content, melting point, refractive index, fatty acid composition and energy intake from lipid of commercial fried instant noodles. Sixty five samples were collected in Seoul area, and analyzed. The average of lipid content was 16.7%, melting point and refractive index were 29$^{\circ}C$ and 1.46, respectively. The fatty acid composition mainly consisted of C16:0, C18:1 and C18:2. The ratio of P/M/S in fried instant noodles was 0.19 : 0.77 : 1.00. The average energy intake from lipid of 100g fried instant noodles was 150.3kcal.

  • This study was carried out to investigate lipid content, melting point, refractive index and fatty acid composition of commercial chocolate and imitation chocolate. Fifty-eight samples for test were 45 of domestics and 13 of foreign in Seoul. The results were as follows ; Mean value of lipid content was 35.1% in chocolate and 31.9% in imitation chocolate, and foreigns were lower than domestics. In melting point of lipid, chocolates and imitation chocolate were 26.4$^{\circ}C$ and 27.4$^{\circ}C$ as mean value, respectively. Mean value of refractive index of lipid was 1.4560, imitation chocolate of domestic were lower than another. The fatty acid compositions were chiefly consisted of {{{{C_18:1 , C_18 , C_16, C_16:1 and C_18:2. }} The ratio of SFA : MUFA : PUFA in chocolate and limitation chocolate were 8.2 :6.9 :1.0 and 3.1 : 4.3: 1.0 , respectively.

Characterization of Ion Fragmentations of Fatty Acids

  • Ion fragmentations of fatty acids such as stearic acid, palmitic acid, myristic acid, and lauric acid were studied using mass spectrometry and semiempirical calculations. The mass spectra of fatty acids showed the distributions of CH₃(CH₂)$_n^+$ and $[(CH_2)_nCO_2H]^+$ fragment ions. The relative ion abundance distributions of $[(CH_2)_nCO_2H]^+$ showed the local maxima at n=6, 10, and 14. The local maximum phenomena were also found in the mass spectra of methyl stearate but not in those of normal alcohols. These local maxima could be explained not by heats of reaction for fragmentation but by the cyclic structures of the molecular ions. The AM1 semiempirical calculations for fatty acids clearly show that the linear structures are more favorable than the cyclic ones for neutral molecules while the cyclic structures are more favorable than the linear ones for ionic molecules. The distances between carboxyl group and methylene of the cylic structures of ionized fatty acids were calculated. The methylene carbons with n=6, 10, and 14 were closer to the carboxyl group than adjacent ones.

Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat (어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

A Sensitive Determination of Plasma Free Fatty Acids Following Tert-butyldimethylsilyl Derivatization using Gas Chromatography-Mass Spectrometry for Screening of Fatty Acid Oxidation Disorders (지방산대사이상질환 스크리닝을 위한 TBDMS 유도체화 후 GC-MS를 이용한 혈장 중 유리지방산의 분석)

  • Purpose: A sensitive gas chromatography mass spectrometry (GC-MS) method was developed for screening of fatty acid oxidation disorders. Methods: The assay utilized a simple protein precipitation with sulfosalicylic acid followed by tert-butyl dimethylsilyl (TBDMS) derivatization of hydroxyl functional group by N-tert-butyldimethylsilyl-N-methyltrifluoroacetamide (MTBSTFA). Results: Calibration curves of spiked pooled plasma showed a linear relationship in the range of 0.01 ng -2 mg with correlation coefficient value greater than 0.98. Limits of detection (LOD) and limits of quantification (LOQ) were found in the range of 0.9-8.8 ng and 9-88 ng, respectively. Conclusion: The new developed method might be useful for a rapid, sensitive screening of inherited fatty acid oxidation disorders. In addition, the method expected to be one of the alternative method for screening newborns of metabolic disorders in the laboratories where expensive MS/MS is unavailable.

The Relationships between Muscle Fiber Characteristics, Intramuscular Fat Content, and Fatty Acid Compositions in M. longissimus lumborum of Hanwoo Steers

  • The objective of this study was to investigate the relationship between muscle fiber characteristics, intramuscular fat (IMF) content, and fatty acids composition in longissimus lumborum (LL) muscle from Hanwoo steers. The LL muscles were obtained from four quality grades (QG) carcasses and subjected to histochemical analysis. There were significant (p<0.05) differences in fiber number percentage (FNP) and fiber area percentage (FAP) of muscle fiber types among muscles from four QGs. Both FNP and FAP of type I increased while those of type IIB decreased with increasing QG from QG 2 to QG $1^{{+}{+}}$ (p<0.05). Also, with increasing QG, the saturated fatty acid (SFA) proportion decreased while monounsaturated fatty acid (MUFA) increased significantly (p<0.05). IMF content was positively correlated with both FNP and FAP of type I, but negatively correlated with those of type IIB. The proportions of SFA and MUFA were significantly (p<0.001) correlated with both type I and IIB composition. These results implied that muscle fiber type composition is an important factor influencing fatty acid composition in LL muscle of Hanwoo steer.
