  • In the type II system. there are two elongation enzymes in E. coli, FabB is well-known to its ability to elongate cis-3-decenoly-ACP (C10:1) in unsaturated fatty acid synthesis, whereas FabF is important for the thermal regulation of fatty acid composition by its ability to elongate palmitoleic acid to vaccenic acid. based on their genetic mutation anaylsis. Radiochemical enzyme assay was performed using myristoyl-ACP as a substrate, which is known for general substrate of FabB and FabF. (omitted)

한국산 및 중국산 참깨로부터 착유한 참기름의 주요성분 비교 (Comparison of Major Components of Sesame Oil Extracted from Korean and Chinese Sesames)

  • Korean and Chinese sesames were subjected to microscopic observation and instrumental determination of fatty acid composition and sesamolin/sesamin ratio to obtain basic data for discriminating each other. The overall appearance of both samples was differently observed by stereo microscope (X8). Fatty acid composition of sesame oils, extracted from both samples with different roasting degrees, showed a similar pattern although Chinese samples cointained about 6% higher content of stearic acid and 47% lower content of linolenic acid then Korean ones. The sesamolin/sesamin ratio was remarkably lower(.039) in Chinese samples than Korean (0.67∼0.72). showing a variation depending on producing districts. Roasting degrees of raw sesames little influenced their composition of fatty acid and sesamolin/sesamin ratio. Based on the above results, it is considered that the comparison between domestic and Chinese sesames in view of their stearic and linolenic acid contents and sesamolin/sesamin ratio might be one of the potential criteria in discriminating their production origins.

Fatty Acid Content in Perilla Cultivars and Commercial Oils Determined by GC Analysis

  • The content analysis of fatty acids in Perilla cultivars and commercial oils is conducted through gas chromatography with a flame ionization detector. Results show that Perilla cultivars, such as Deulsaem and Daesil, contain high amounts of ${\alpha}-linolenic$ acid (262.22 and 261.97 mg/g, respectively). Among commercial oils, Perilla oil contains a higher amount of ${\alpha}-linolenic$ acid (515.20 mg/g). Accordingly, ${\alpha}-linolenic$ acid is a major fatty acid of Perilla cultivars and oil. Therefore, Perilla cultivars could be used as a food supplement for nutritional and pharmaceutical purposes.

  • Albinism is a phenomenon that color of the body surface is changed to white or faint brown from the specific color to the species by difficiency of pigments due to mutation or disease. This study was undertaken to investigate the experimental basis on the appearance of albinism in cultured flounder Paralichthys olivaceus. The skin and muscle, from the normal and albinic flounder were used by measuring contents of lipid and fatty acid, Contents of lipid and fatty acid in the skin and muscle are different from normal and albinic flounder. Contents of lipid in the skin were higher than those of muscle from flounder. The major fatty acids in both skin and muscle were palmitic acid, oleic acid docosahexaenoic acid (DHA) and eicosapentaenoic acid(EPA).

  • Feasibility of surface plasmon resonance (SPR) method to evaluate soybean frying oil quality was evaluated. Free fatty acid value, p-anisidine value, conjugated dienoic acid content, conjugated trienoic acid content, peroxide value, iodine value, total polar compound (TPC), ratio of unsaturated fatty acid to saturated fatty acid, refractive index, dielectric constant, smoking point, and L, a, and b of Hunter color system were selected as parameters related to conventional evaluation of soybean frying oil quality. SPR scale mapped with conventional quality parameters well described free fatty acid value, conjugated dienoic acid content, dielectric constant, TPC, and b of Hunter color system, as shown by correlation and linear regression analyses.

해동피의 화학성분 및 생리활성 (Chemical Constituents and Biological Activity of Kalopanacis Cortex)

  • The study on phytochemical analysis and the biological activity of Kalopanacis Cortex was carried out in this research. As a phytochemical result, liriodendrin as a lignan glycoside was isolated and characterized. Two subfraction separated from the acidic substance of $CHCl_3$ fraction were saturated and unsaturated fatty acid, respectively. Saturated fatty acid mixture identified from GC-MS tool was as follows: palmitic acid, stearic acid, arachidic acid, heneicosanoic acid, docosanoic acid, tricosanoic acid, tetracosanoic acid, pentacosanoic acid, hexacosanoic acid and octacosanoic acid. Unsaturated fatty acid was found to be linoleic acid on the basis of spectroscopic method. An active principle of liriodendrin exhibited significant antihepatotoxic activity but failed to show a considerable antiedemic activity. In this paper, the result of writhing test on liriodendrin was also described.

  • 시판(市販) 고가(高價)의 기름에 저렴(低廉)한 기름을 혼입(混入)시키는 일이 흔히 일어나므로 이것을 쉽게 검색(檢索)하기 위한 방편(方便)으로 각 식용유(食用油)의 지방산조성(脂肪酸組成)을 gas chromatography 법(法)에 의해서 조성비(組成比)를 조사(調査)하였으며 이들 지방산(脂肪酸)의 조성(組成)을 보면 참기름은 myristic, palmitic, stearic, linoleic acid와 미량(微量)의 linolenic acid로, 미강유(米糠油)와 대두유(大豆油)는 myristic, palmitic, stearic, oleic, linoleic 및 linoleic acid, 들기름은 palmitic, stearic, oleic, linoleic, linolenic acid, 해바라기 기름은 palmitic, stearic, oleic acid 및 linoleic acid로 이루어져 있다. 이상의 결과로 볼 때 참기름에도 linolenic acid가 극미량(極微量) 밖에 존재하지 않으므로 이것이 풍부한 타(他)의 기름이 혼입(混入)되었을 경우 쉽게 검출(檢出)할 수 있을 것이다.

  • 1. 재배시기이동에 따른 유채종자유에 따른 지방산조성은 지방산의 종류에 따라 함량이 높은 Erucic acid가 가장 큰 차이를 나타냈으며 함량이 적은 지방산의 변동은 적었다. 2. 추파재배한 유채종자가 춘파재배한 것보다 Erucic acid함량이 높은데 대해서 Oleic acid와 Linoleic acid함량은 반대로 낮었다. 이들 차이는 1%수준에서 유의차가 있었다. 3. 추파재배된 유채종자가 Erucic acid함량이 높았으며 이는 등숙기간의 적산온도가 낮고 강수량이 많은데 있다. 이들 기상조건과 Erucic acid간에는 높은 상관관계가 있었다. 4. 단쇄지방산인 Oleic acid와 Linoleic aci의 증감경향은 항상 정상관관계가 있으며 장쇄지방산인 Erucic acid와는 부의 상관관계에 있었다.

Arabidopsis thaliana로부터 지방산 불포화효소 유전자의 분석 (Characterization of a fad3 cDNA Encoding Microsomal Fatty Acid Desaturase from Arabidopsis thaliana)

  • Linoleic acid를 linolenic acid로 전환시키는 지방산불포화효소의 유전자(fad3)를 유채의 fad3 DNA probe를 이용한 plaque hybridization방법으로 $\lambda$ZAPII Arabidopsis thaliana cDNA expression library로부터 분리하였으며 1.8 kb-EcoRI DNA조각을 지니는 lambda clone을 pGEM7으로 subcloning하여 염기서열을 분석하였다. 그 결과로부터의 아미노산서열분석에 의하면 fad3 유전자는 open reading frame이 386개의 아미노산으로 이루어졌으며 44,075 Da의 분자량이 예측되고 있다. 엽록체의 $\omega$-3 지방산불포화효소(fad7)와 endoplasmic reticulum의 지방산불포화효소(fad2)와의 비교시 각각 70%와 58%의 유사성이 나타났다. 특히 82-151의 아미노산서열과 276-333의 아미노산지역은 보존성이 높았으며 이는 불포화지방산효소의 기능에 필수적인 지역으로 보여진다. 한편 대장균을 이용한 IPTG에 의한 Fad3단백질의 유도생성은 대장균의 생육에 독성효과를 나타내는 것으로 나타났다.

발아의 경과에 따른 참깨 종실내 주요성분의 변화 (Changes of Major Componets During Germination of Sesame (Sesamum indicum L.) Seeds)

  • These studies were undertaken to investigate changes of major components occuring during germination of sesame (Sesamum indicum L.) seeds, Changes of total lipid and protein contents, and fatty acid composition were determined. Also, the correponding values of various components in cotyledons, hypocotyls and roots were measured according to germination stage. The results were summarized as follows; During germination, total lipid and protein contents decreased. In particular, protein contents rapidly decreased to the 3 days after gemination(DAG), and then total lipid contents rapidly decreased. In changes of total lipid and protein of cotyledons, hypocotyles and roots detected at the 10, 15 and 20 DAG, some variations were determined. The contents of lipid and protein in hypocotyls rapidly decreased, but since than no changes were observed. In contract, in roots similar changes patterns were observed, while since 15 DAG a rapidly increase was wxamined. In fatty acid composition of total lipid ,saturatedmfatty acids such as palmitic acid increased during the germination. On the other hand, unsaturated fatty acid such as olic acid and linoleic acid decreased during the same periods. In changes of fatty acid composition of total lipid of cotyledons, hypocotlys and roots, saturated fatty acids such as palmitic acid and stearic acid increased during the germination. However, linoleic acid decreased during the same germination suggesting that this may be due to the rapid degradation. However, linoleic acid decreased during the same periods. According to SDS-PAGE analysis, there was no detectible polypeptide bands on the gel before seed germination suggesting that this may be due to the rapid degradation of the storage peotein in the mature seed by hydrolytic enzymes during the stag. As germination continued polypeptide bands, one with 40KD, two with 32∼34Kd and one with 24KD, were detected on the gel.
