Esterification of High Concentration Free Fatty Acid in Rice Bran Oil (미강유 중 고농도 자유지방산의 에스테르화)

  • Characteristics of the esterification reaction between free fatty acid in rice bran oil and methanol was investigated in the presence of catalysts, such as PTS(p-toluene sulfonic acid), Amberlyst 15 dry and SCX(silica gel based strong cation exchange resin). While reaction temperature was kept constant at $65^{\circ}C$, initial feed content of free fatty acid was varied from 100% to 1% by addition of pure free fatty acid which was previously made from rice bran oil. Also, the effect of mole ratio of methanol to fatty acid on the final conversion was examined. When esterification of pure free fatty acid was catalyzed by several acids, final conversions were increased in order of Amberlyst 15 dry, SCX and PTS. Using PTS catalyst, initially the reaction proceeded in homogeneous 2nd oder reaction mechanism. However, phase of reaction mixture changed from homogeneous to heterogeneous along the reaction time and then reaction rate was retarded by mass transfer resistance of methanol. Final conversion of free fatty acid in reaction mixture was depended on initial feed content of free fatty acid, and had maximum value at 30% of initial feed free fatty acid content for all kinds of catalysts used. And the final conversion was increased with mole ratio of methanol by the improvement of reaction rate. When initial feed free fatty acid content below 10% and the reaction was catalyzed by PTS, concentration of free fatty acid in reaction mixture was increased in the middle of reaction time by hydrolysis of triglyceride in reaction mixture. Also, if silica gel was added into the reaction mixture which had initial feed free fatty acid content below 50%, final conversion was increased by the adsorption of moisture produced. The SCX catalyst made the esterification reaction of free fatty acid to progress like in case of PTS catalyst. However, when initial feed free fatty acid content below 10%, concentration of free fatty acid in. reaction mixture was decreased monotonically and not increased in the middle of reaction time on the contrary to the case of PTS. Thus, SCX catalyst accomplished more high value of final conversion than PTS catalyst for the initial feed fatty acid content range from 50% to 5% In case of initial feed free fatty acid content of 1% and mole ratio of methanol was 2, concentration of free fatty acid in reaction mixture increased over the initial feed free fatty acid content for all kind of catalysts used. Although SCX catalyst was added into reaction mixture which had 1% of initial feed fatty acid content, final conversion was hardly raised by mole ratio of methanol.

Effects of Different Concentrate and Roughage Ratios on Ruminal Balance of Long Chain Fatty Acids in Sheep

  • The effects of different feeding ratios of concentrate (C) and roughage (R) on balance of long chain fatty acids and microbial fatty acids in the rumen of sheep were investigated. The diets were divided into 8:2 (concentrate feeding), 4:6 (middle mixture) and 0:10 (roughage feeding) ratios (C:R). Duodenal digesta was collected through 24 hours after feeding. Biohydrogenation rate, total duodenal flow of fatty acids and microbial fatty acids were measured. Total duodenal flow of fatty acids was significantly (p<0.05) increased with increasing concentrate. Total duodenal flow of fatty acid was greater than intake of fatty acid in all diets. In comparison with intake of each fatty acid, duodenal flow of stearic acid ($C_{18:0}$) remarkably increased in all diets. Biohydrogenation rate for total C18 unsaturated fatty acids in the rumen tended to increase (p<0.10) when sheep were fed the middle mixture. In particular, biohydrogenation rate of linoleic acid ($C_{18:2}$) and linolenic acid ($C_{18:3}$) with the middle mixture were highest (p<0.05) compared with other diets. Duodenal flow of protozoal fatty acids was significantly (p<0.05) increased with the increased supply of concentrate. That of bacterial fatty acids was significantly (p<0.05) increased with both concentrate diets compared with the roughage feeding diet. $C_{18:0}$ occupied the greater part of both protozoal and bacterial fatty acids in all treatments. Results indicated that biohydrogenation of free unsaturated fatty acids was actively carried out when the middle mixture diet was supplied, and that microbial uptake and synthesis of fatty acids were accelerated by adding the supply of concentrate.

Effects of Procedures for Preparing Fatty Acid Methyl Esters on Fatty Acid Composition of Korean Vegetable Oils (Sesame Seed Oils) (지방산 메틸에스테르 조제방법이 한국산 식물성 기름(참깨 기름)의 지방산 조성에 미치는 영향)

  • The effects of procedures for preparation of fatty acid methyl esters for gas chromatography were investigated. A quantitative comparison of four procedures for the preparation of the fatty acid methyl esters from Korean sesame seed lipids which can be representative of fatty acid ranges of Korean vegetable oils has been made. The procedures employed were $BF_3$-methanol, HCI-methanol, sodium methoxide-methanol, and tetramethylammonium hydroxide-methanol. Twelve fatty acids ranged from 14:0 to 24:0 were identified in the lipids from Korean white and black sesame seeds. All four procedures gave similar results for the fatty acids, 16:0, 18:0, 18:1, 18:2, and 18:3 present in the range of $1{\sim}44%$ but only in the HCI-methanol procedure, the fatty acids, 16:1, 20:0, 22:0, 24:0 present in the range of $0.02{\sim}1%$ showed the lowest values. When using tetramethylammonium hydroxide-methanol procedure for determination of total fatty acid composition from white and black sesame seed lipids, unsaponifiable matters including sesamol, sesamolin and sesamin present in the seed lipids are not removed from the resulting reaction mixture. Thus the transesterification mixture is used without further treatment for injection into the gas chromatography. However, the gas chromatographic analysis of the transesterification mixture showed that the unsaponifiable matters had no effect on the fatty acid composition of the seed lipids. From the results, it appears that the $BF_3$-methanol, sodium methoxide-methanol and tetramethylammonium hydroxide-methanol procedures can be used to prepare fatty acid methyl esters from Korean vegetable oils. Among the methods, the tetramethylammonium hydroxide-methanol procedure, which give total fatty acid composition, glyceride fatty acid composition and composition of free fatty acids present, appears to be a simple, convenient and quantitative procedure and applicable to samples containing broad ranges of fatty acids.

Effects of Feeding the Mixture of Lineseed and Sunflower Seed Oil on the Fatty Acid Composition in Lipid of Brain and Heart in Dietary Hyperlipidemic Rats (아마인유와 해바라기 종자유의 혼합급이가 식이성 고지혈증 흰쥐의 뇌 및 심장지질의 지방산 조성에 미치는 영향)

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks. then they were fed with the diet containing lard 3.0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil, and antihyperlipidemic drugs for 2 weeks. Analysis of the fatty acid composition of the brain and heart lipids showed following results. In the fatty acid composition of brain lipids, C20:4 and C22:6 were the major fatty acids but showed little difference among the groups. In the fatty acid of heart lipids,C18:2 was the major fatty acid. The proportion of C20:4 decreased gradually as n-3P/n-6P ratio of the test lipids increased in groups 5 (linseed oil 12.0%) to 9 (sunflower seed oil 12.0%) while the proportion of C22:6 was not affected by the fatty acid composition of the test lipids.

  • Enzymatic ethanolysis of fish oil with immobilized lipase was investigated for reducing the free fatty acid contents and enhancing the function of fish oil. Ethanolysis reactions were carried out in erlenmeyer flask (25ml) containing a mixture of squid viscera oil and 99.9% ethanol using 1% (based on w/w squid viscera oil) immobilized lipase. The reaction mixtures were incubated at $50^{\circ}C$ and shaken at 100rpm. Ethanol was added into the mixture by stepwise addition method of Shinmada[9]. Measurement of free fatty acid molar amounts was studied by Acid Value. Tendency of oil variation during transesterification was studied by TLC method. Enzymatic ethanolysis composed diglyceride, monoglyceride and fatty acid ethyl ester with reducing free fatty acid contents. Also, selective ethanolysis by Lipozyme TL-IM and Lipozyme RM-IM mostly did not react at the sn-2 position of squid viscera oil. Lipozyme RM-IM was more suitable enzyme to reduce the free fatty acid contents by ethanolysis than Lipozyme TL-IM. Squid viscera oil was transformed into suitable properties (5 in Acid Value) for functional fish oil production.

  • Maxwell displacement current (MDC) measuring technique has been applied on the study of monolayers of fatty acid and polyamic acid mixture. The displacement current was generated from monolayers on the water surface by monolayer compression and expansion. Displacement current was generated when the area per molecule was about 132 $^2$and 115 $^2$just before the initial rise of the surface pressure during the 1st and 2nd mixed monolayer compressions cycle, respectively. Maxwell displacement currents were investigated in connection with mixed monolayer compression cycles. It was found that the maximum of MDC appeared at the molecular area just before the initial rise of surface pressure in compression cycles. Ultra thin film of fatty acid and polyamic acid mixture was prepared on the hydrophilic quartz plate by Langmuir-Blodgett (LB) method. The precursor LB film was heated in a vacuum dry oven at 12$0^{\circ}C$ in order to convert it into the LB film of polyimide. The absorption spectra of LB films were also induced photoisomerization by UV and visible light irradiation.

  • We were investigated by cyclic voltammetry to the stability through the electrochemical characteristics of Langmuir-Blodgett films mixed with myristic acid and stearic acid. Fatty acid mixture monolayer LB films was deposited by the LB method on the indium tin oxide(ITO) glass. The electrochemical properties was measured by cyclic voltammetry with a three-electrode system in 0.01 N $NaClO_4$ solution. The measuring range is continuously oxidized to 1650 mV, with an initial potential of -1350 mV was reduced. Scanning rates of 50, 100, 150, 200, and 250 mV/s was set. As a result, LB monolayer films of fatty acid mixture was appeared on irreversible processes by the oxidation current from the cyclic voltammogram. Diffusion coefficient (D) of fatty acid mixture was calculated $7.9{\times}10^{-2}cm^2s^{-1}$ at 0.01 N $NaClO_4$ solution.

  • We carried out this subject to observe photoisomerization using 1,2-dioleoyl-sn- glycero-3-phosphocholine(DOPC) mixed with fatty acid containing azobenzene group which has reversible to cis-trans by light irradiation. Spreading solutions for the LB films were prepared in chloroform($5.0{\times}10^{-5}$mol/L).We investigated the photoisomerization and property of the organic ultra thin film of fatty acid containing azobenzene was prepared on the hydrophilic ITO(idium tin oxide) glass plate by LB method. As a result, the absorption spectra of 8A5H and DOPC of mixture LB films was induced to photoisomerization by alternating irradiation of ultraviolet and visible light, because the condensation of pure azobenzene monolayers was loosened by the introduction of phospholipid into the monolayers, and the molecular high aggregation in pure azobenzene monolayers is also weakened by the introduction of phospholipid. We found that it was reversibly induced to cis-trans photoisomerization in several solvents and mixture LB films.

  • This study was carried out to investigate the effect of the feeding mixture of linseed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hyperlipidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter and cholesterol for 3 weeks, then they were fed with the diet containing lard 3 .0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil and antihyperlipidemic drugs for 2 weeks. Analysis of the fatty acid composition of the serum lipoprotein fractions showed following results. In the fatty acid composition of serum lipoprotein , the proportion of C18:2 was dominant in all fractions, C20:5 in LDL and HDL fraction and C22:6 in chylomicron fraction. The ratio of n-3P/n-6P tended to increase gradually as it of the test lipid increased in groups 5 to 9 group and was affected by the fatty acid composition of the test lipids.

  • This study was carried out to investigate the effect of the feeding mixture of linesed oil, rich in n-3 PUFA and the sunflower seed oil, rich in n-6 PUFA on the lipid metabolism in the dietary hypprlidemic rats. After male Sprague-Dawley rats were induced hyperlipidemia by feeding the diet containing lard, butter, and cholesterol for 3 weeks, then they were fed with the diet containing lard 3.0% and butter 12.0% for control, the mixture in different proportion of both linseed oil and sunflower seed oil, and antihyperlipidemic durgs for 2 weeks. Analysis of the lipid component and the fatty acid composition of the liver showed following results. Concentration s of the total cholesterol and phospholipid in liver were significantly higher in group 2 (olive oil 12.0%) and lower in the other groups than in the control group, especially lower in groups 3 (cholestyramine 2.0%) and 9 (sunflower seed oil 12.0%) . Concentration of triglyceride was lower in the other groups except group 4 (liparoid), especially lowe rin group 9 than in the control group. In the fatty acid composition of liver lipids, C18:2 was the major fatty acid. Contents of n-6 PUFA increased , while those of n-3 PUFA decreased in groups composition of the test lipids. From the data on concentration s of total cholesterol. Phospholipid and triglyceride in liver, we concluded that the feeding mixed with 3.0% lard and 12.0 % sunflower seed oil were most effective for the improvement of the live lipids. The fatty acid composition in liver lipids were affected by the fatty acid composition of the test lipids.
