분화도 좋은 구강 편평상피세포암종에서 Dominant Negative p63 isoform의 발현 (EXPRESSION OF DOMINANT NEGATIVE p63 ISOFORM IN WELL-DIFFERENCIATED ORAL SQUAMOUS CELL CARCINOMA)

  • The p53 which is well known as tumor suppressor gene is located at 17p13. p53 is a sequence-specific DNA binding transcription factor that responds to certain cytotoxic stresses, such as DNA damage, by enhancing the transcription of genes that regulate cell-cycle progression as well as programmed cell death. The p63 gene that is located at 3q27-29, is recognized members of the p53 family, and responsible for the transcription of 6 isoforms. Three isoforms ($TAp63{\alpha}$, $TAp63{\beta}$, $TAp63{\gamma}$) contain an N-terminal transactivation (TA) domain and can induce apoptosis. The other 3 isoforms (${\Delta}Np63{\alpha}$, ${\Delta}Np63{\beta}$, ${\Delta}Np63{\gamma}$) lack the TA domain and may function in a dominant-negative fashion by inhibiting the transactivation functions of p53 and TAp63 proteins, and thus act as oncoproteins. A number of studies have investigated the role of p63 in human squamous cell carcinomas from different organs. Only a few studies have examined ${\Delta}Np63$ isoform in oral squamous cell carcinoma including normal epithelium. This study aimed to evaluate expression of ${\Delta}Np63$ isoform in human oral squamous cell carcinoma tissue and normal mucosa. The 3 cases of well differenciated oral squamous cell carcinoma specimen including adjacent normal mucosa were examined, and immunohistochemical study with monoclonal antibody(4A4) and tumor cell apoptosis analysis with Transmission Electon Microscopy were studied. And, RT-PCR analysis was done for expression of ${\Delta}Np63$ isoform. The results were as followed. 1. Normal gingiva showed the restricted p63 expression in basal cell layer. 2. Well differentiated squamous cell carcinoma showed mainly p63 expression in overall area of malignancy, especially in basal cell layer to adjacent stromal tissue. 3. Tumor cells around keratinized area with no p63 expression disclosed less micro-organelle in decreased size cytoplasm and severe chromatin margination with nuclear destruction that means apoptosis. 4. Comparison of mRNA expression of ${\Delta}Np63$ isoform by RT-PCR showed variable expression of ${\Delta}Np63$ isoform, but ${\Delta}Np63{\alpha}$ was most highly expressed in all 3 tumor specimen. From theses results, it should be suggested that ${\Delta}Np63$ isoform expression in well differentiated squamous cell carcinoma was closely related to tumor oncogenesis, expecially overexpression of ${\Delta}Np63{\alpha}$ is a most important factor in tumor genesis of oral squamous cell carcinoma.

ZNF424, a novel human KRAB/C2H2 zinc finger protein, suppresses NFAT and p21 pathway

  • Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK-293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions.

공예라는 전통과 캐논의 성립: 고미술과 미술공예 (The Invention of Tradition and Canon in Korean Crafts: Antiques and Art Craft)

  • 이 논문은 조선총독부의 고적조사와 일련의 사업을 통해 한국에 미술공예 개념이 수용된 과정과 그 과정에서 성립된 공예의 전통과 캐논에 대해 고찰한 글이다. 「고적 및 유물 보존규칙」과 「조선보물·고적·명승·천연기념물 보존령」의 제정을 통해 법률상으로 제시된 공예는, 조형물을 만들어낸 정교하고 뛰어난 기술, 더 나아가 회화, 조각과 같은 미술의 한 장르를 의미했다. 이왕가박물관과 총독부박물관은 미술공예라는 용어를 대중에 확산시키는 역할을 했고, 여기에 전시된 공예는 과거의 전통을 시각적으로 구체화하였다. 이와 같이 공예라는 용어는 고미술을 가리킬 때 빈번히 사용되었고, 구체적으로는 법률상의 보물로 지정되거나 박물관에 전시되어 지키고 보존해야 할 전통이 되었다. 공예의 전통과 캐논이 성립되자 당대에 제작되는 공예에 큰 영향력을 행사했다. 이 일련의 과정에는 일본의 문화 정책과 제국적 취향이 절대적으로 작용했다. 일반적으로 근대기에 성립된 공예 개념은 산업으로서의 공예와 미술로서의 공예, 두 가지로 구분되어 왔으나, 미술로서의 공예라는 개념 안에도 여러 층위가 존재했다. 캐논이 된 고미술은 동시대 제작의 준거가 되었으며, 실제로는 동시대에 제작되던 공예의 양상도 모두 같지 않았다. 이제까지 일괄적으로 논의되던 미술공예 개념을 세분화하는 작업이 공예에 있어서 '만들어진 전통'과 근대기 공예계의 양상을 정확히 파악하기 위한 기초가 될 것으로 기대한다.

The C-terminal Phosphorylation Sites of eel Follicle-Stimulating Hormone Receptor are Important Role in the Signal Transduction

  • The large extracellular domain of glycoprotein hormone receptors is a unique feature within the G protein-coupled receptors (GPCRs) family. After interaction with the hormone, the receptor becomes coupled to Gs, which, in turn stimulates adenylyl cyclase and the production of cAMP. Potential phosphorylation sites exist in the C-terminal region of GPCRs. The experiments described herein represent attempts to determine the functions of the eel follicle-stimulating hormone receptor (eelFSHR). We constructed a mutant of eelFSHR, in which the C-terminal cytoplasmic tail was truncated at residue 614 (eelFSHR-t614). The eelFSHR-t614 lacked all potential phosphorylation sites present in the C-terminal region of eelFSHR. In order to obtain the eelFSHR ligand, we produced recombinant follicle-stimulating hormone ($rec-eelFSH{\beta}/{\alpha}$) in the CHO-suspension cells. The expression level was 2-3 times higher than that of the transient expression of eelFSH in attached CHO-K1 cells. The molecular weight of the $rec-eelFSH{\beta}/{\alpha}$ protein was identified to be approximately 34 kDa. The cells expressing eelFSHR-t614 showed an increase in agonist-induced cAMP responsiveness. The maximal cAMP responses of cells expressing eelFSHR-t614 were lower than those of cells expressing eelFSHR-wild type (eelFSHR-WT). The $EC_{50}$ following C-terminal deletion in CHO-K1 cells was approximately 60.4% of that of eelFSHR-WT. The maximal response in eelFSHR-t614 cells was also drastically lower than that of eelFSHR-WT. We also found similar results in PathHunter Parental cells expressing ${\beta}$-arrestin. Thus, these data provide evidence that the truncation of the C-terminal cytoplasmic tail phosphorylation sites in the eelFSHR greatly decreased cAMP responsiveness and maximal response in both CHO-K1 cells and Path-Hunter Parental cells expressing ${\beta}$-arrestin.

학교폭력의 실태분석과 개선 방안에 관한 연구 (A Study on Present Situation of Violence in School and Improvement Program)

  • 학교란 청소년들이 가장 많은 시간을 보내고 친구들과 교우관계를 형성하고 지도를 받으면서 사회로 나가기 위하여 많은 것을 배우는 터전이다. 이런 배움의 터전에서 폭행, 금품갈취, 집단 따돌림 등의 일들이 벌어지고 있으며, 이러한 일들의 결과로 자살, 살인, 성매매등의 극단적인 범죄로 까지 이어지고 있어 심각한 사회문제로 대두되고 있다. 따라서 본 논문에서는 학교폭력의 개념과 현황에 대하여 살펴보고 학교폭력에 대한 문제점을 파악하여 보다 실제적인 학교폭력에 대한 개선방안을 제시하고자 하는데 연구의 목적이 있다. 이에 본 연구를 통하여 얻어진 개선방안은 첫째, 인성교육의 강화로 현재의 현 실정에 맞는 인성교육의 재조명 및 필요성 부분에 대한 방안을 제시하였고 둘째, 가정의 기능과 가장교육역할 확대로 학교교육뿐만이 아니라 가정에서의 부모들의 올바른 가정교육에 대해 제시하여 가정교육의 중요성을 강조하였다. 셋째, 전문상담 인력의 확충과 사회적 환경개선으로 학교폭력 예방 및 해결에 있어 중요한 전문 상담인력 확충과 전문상담실 설치 운영에 대해 제시하였고, 넷째, 보안요원의 활용으로 전국적으로 학교 내에 학교보안요원 배치 후 적극 활용에 대한 부분 등을 학교폭력에 대한 대응방안으로 제시하였다.

재가장기요양기관 방문간호사의 간호 경험 (The Lives Experience of Visiting Nurses of Home-based Long-term Care Service Center)

  • 본 연구는 재가장기요양기관 방문간호사의 간호 경험의 의미와 본질을 탐구하기 위해 van Manen의 해석학적 현상학 연구방법론을 적용하였다. 연구 참여자는 방문간호 경력 1년 이상인 10명의 방문간호사들로 목적적 표집과 편의 표집, 눈덩이식 표집 방법을 통해 선정하였다. 자료 수집은 2017년 1월 7일부터 2018년 10월 12일까지 진행되었으며, 연구 참여자와의 면담은 총 23회에 걸쳐 이루어졌다. 자료 분석은 van Manen이 제시한 해석학적 현상학 방법론에 근거를 두고 전체론적(holistic) 방법과, 선택적(selective) 방법, 그리고 세분법(detailed)을 사용하였다. 연구 결과, 재가장기요양기관 방문간호사의 간호 경험은 6개 본질적 주제와 20개 하위주제로 도출되었다. 본질적 주제는 '대상자 및 보호자와 친밀한 관계를 형성함', '주도적 간호수행으로 방문간호의 기반을 다져감', '지원체계 미비로 원하는 만큼의 간호를 제공하기 어려운 현실에 직면함', '간호의 진정성이 전달되지 않을 때 속상함', '내 환자라는 책임감을 가지고 돌봄의 소명을 다함', '방문간호사로서 보람과 의미를 느끼며 오래도록 일하고자 함'이다. 본 연구는 재가장기요양기관 방문간호사들의 간호 경험을 탐색함으로써 지역사회 내에서 노인장기요양보험 방문간호사의 역할과 중요성을 이해하였다는데 의미가 있다.

한국 한의학교육의 발전방안 (Ways to Educational Improvement of Korean Oriental Medicine)

  • Problems and current situation of public health globally and domestically were analyzed in this study and based on these findings, ways to improve from western medicine and Oriental medicine can be deduced as follows: 1. Current problems of public health in Korea and the world 1) Increase of diseases resulted from daily habits and infectious diseases, many are at the brink of being ill. Quality of life from extended life span and unbalanced health care must be solved. 2) Natural and societal factors including host factors, public health service, and other external and internal factors play an important role in deciding healthy and being ill. 3) Some of the limits and problems of modem medicine include insufficient academic knowledge and incomplete theory, as well as misled approach to the treatment. Human itself isn't perfect organism and other realistic problems hinder one's well-being. 4) Regardless of western medicine or Oriental medicine, patients were approached as someone with diseases and disorders, and wholistic approach was disregarded. Lack of clinical training, absence of clear educational philosophy and goal are some of the reasons why the education isn't under concrete system 2. Important factors for the medical education and proper direction for the education of Oriental medicine 1) Important factors for medical education - Education should not be limited on the human health and illness, but also cover qualities such as well-being, social welfare, service, and happiness. Every aspects of human life must be considered and attended for more productive outcome. - Basic understanding of humanity must be included in the educational curriculum - Foundation of human diseases and pain are associated with inner life and surrounding causes including family, society, nature, race, culture, religion, politics, and etc., thus the education must be approached to recognize aforementioned criteria. 2) Proper direction for the education of Oriental medicine - Values of Oriental medicine for medical principles and importance of lifehood must be educated. - Educational goal, limits, and levels must be established for the school of Oriental medicine - Respect for life must be the top priorities of educational direction which should lead to solution based education for the human health. Latest medical theory and technology should be accommodated as well as prevention, treatment, and balancing of basic courses and clinical training for optimal education.

Astaxanthin 함유 기능성 식품소재의 유전독성 및 항산화능 검사 (Genotoxicity and Anti-Oxidative Effectiveness Study of Functional Food Additive Containing Astaxanthin)

  • Astaxanthine is a pigment that belongs to the family of the xanthophylls, the oxygenated derivatives of carotenoids whose synthesis in plants derives from lycopene. Astaxanthine is also a carotenoid widely used in salmonid and crustacean aquaculture to provide the pink color characteristic of that. Recent study reported that astaxanthine has the role as a detoxicant against the free radicals. On our study, we estimated the genotoxicity in ICR mice and possibility as antioxidant reagents of mutant Phaffia rhodozyma strain over expressing the astaxanthine by gamma-lay and carophyll pink including astaxanthine in apoE knock out mice, respectively. In our study, we administered Phaffia rhodozyma (2 mg and 3 mg) and carophyll pink for 4 and 8 week. The clinical sign and mortality were not detected compared with control groups. In the mutant frequency of hprt gene and chromosome aberration in splenic cells, there was not detected abnormality. There was not critical change in hematological and serum biochemical test compared to control. In expression level of repair enzyme, increase of catalase were detected and increase of expression level of Nrf-2 was detected in Phaffia rhodozyma (3 mg) and carophyll pink in 8 week treated group. In GSH level, the group of treated with Phaffia rhodozyma (3 mg) showed the increase of the GSH. In conclusion, mutant Phaffia rhodozyma and caphyll pink may be applied to the effective food additives to reduce the free radical.

Impact on Inflammation and Recovery of Skin Barrier by Nordihydroguaiaretic Acid as a Protease-Activated Receptor 2 Antagonist

  • Atopic dermatitis is a chronic, inflammatory disease of the skin with increased transepidermal water loss. Both an abnormal inflammatory response and a defective skin barrier are known to be involved in the pathogenesis of atopic dermatitis. Protease activated receptor 2 (PAR2) belongs to a family of G-protein coupled receptors and is activated by both trypsin and a specific agonist peptide, SLIGKV-$NH_2$. PAR2 is expressed in suprabasal layers of the epidermis and regulates inflammatory responses and barrier homeostasis. In this study, we show that nordihydroguaiaretic acid (NDGA) inhibits the PAR2-mediated signal pathway and plays a role in skin barrier recovery in atopic dermatitis. Specifically, NDGA reduces the mobilization of intracellular $Ca^{2+}$ in HaCaT keratinocytes by down-regulating inflammatory mediators, such as interleukin-8, thymus and activation-regulated chemokine and intercellular cell adhesion molecule-1 in HaCaT keratinocytes. Also, NDGA decreases the protein expression of involucrin, a differentiation maker of keratinocyte, in both HaCaT keratinocytes and normal human epidermal keratinocytes. We examined NDGA-recovered skin barrier in atopic dermatitis by using an oxazolone-induced atopic dermatitis model in hairless mice. Topical application of NDGA produced an increase in transepidermal water loss recovery and a decrease in serum IgE level, without weight loss. Accordingly, we suggest that NDGA acts as a PAR2 antagonist and may be a possible therapeutic agent for atopic dermatitis.

저농도 혈중연과 혈장레닌활성도 및 혈압과의 관련성 (The Relationship of Lew-Level Blood Lead to Plasma Renin Activity and Blood Pressure)

  • 일상생활을 통한 연섭취로 인한 고혈압 발생 가능성 및 이때 연이 혈압에 영향을 미칠 수 있는 기전을 밝히기 위한 연구의 일환으로 환자 대조군 연구를 실시하였다. 대상자로서 울산 및 그 인근지역의 직업적 연폭로 과거력이 없는 남자 근로자 중 고혈압 환자 72명, 대조군 69명을 선택하였다. 혈중 연과 혈장레닌활성도(Plasma renin activity, PRA)를 측정하였으며 동시에 흡연유무, 음주유무, 고혈압 가족력 유무를 확인하고 body mass index(BMI), 혈청 소디움, 포타시움, 이온화 칼슘, 크레아티닌 및 총 콜레스테롤을 측정하였다. 고혈압군의 혈중 연농도는 $19.8{\pm}5.5{\mu}g/dl$로서 대조군의 $12.5{\pm}4.7{\mu}g/dl$에 비해 유의하게 높았고(p<0.01) 또한 혈중 연의 고혈압발생에 대한 교차비(odds ratio)가 1.38로서 역시 유의하였다(p<0.01). PRA나 In(PRA)가 고혈압군과 대조군 사이에 유의한 차이가 없었으나 고혈압군에서 혈중 연농도가 증가함에 따라 PRA도 어느정도 증가하는 양상을 보였고(p<0.1), In(PHA)를 종속변수로 하는 중회귀분석을 실시하였을 때 혈중 연농도(회귀계수 ; 0.037)가 10% 수준에서 유의하였다. 본 연구를 통해 볼 때, 지금까지 알려진 정상치 범위 이내의 혈중 연과 고혈압 발생간의 인과 관계에 대해 계속 연구해 볼 필요성을 느끼며 이때 PRA의 변화가 작용 기전의 일부에 관여할 가능성이 있을 것으로 추측된다.
