  • From both breast and leg muscle of 12 week-old broiler chicken held for aging in slushed ice and dry chilling at $33-35^{\circ}F$., myosin, actomyosin and other nitrogenous fractions were extracted with KCl-phosphate buffer for various periods from 1 hr. to 25 hr. post-mortem. The changes in extractable nitrogen occurred mainly as a result of decrease in extractability of myosin and to some extent, increase in extractability of actomyosin. Changes in stroma, sarcoplasmic and NPN fractions were small. Myosin extractability decreased rapidly during the first 3 hr. post-mortem and then reduced Continuously in both leg muscle and breast muscle during wet chilling. The decrease of myosin extractability in leg muscle was much more than that in breast muscle, and then the extractability increased after 17 hr. post-mortem in dry chilling. Actomyosin was extracted at low consistent level in wet chilling, while it increased considerably after 17 hr. post-mortem in dry chilling. The tendency was similar in both breast and leg muscle.

Comparative studies on Tenderness and Characteristics of Protein Obtained from Various Carcass grade in Korean native Cow (도체등급별 한우육의 연도와 단백질특성에 대한 비교연구)

  • This study was conducted to investigate the effects of carcass grade on the hardness, myofibrillar fragmentations index, protein extractability and Mg-ATPase activity of myofibril and actomyosin obtained from 1, 2, 3 and D carcass grade)subgrade) in Korean native cow. Proximate component, hardness, chewiness, myofibril fragmentation index, protein extractability and Mg-ATPase activity if myofibril or actomyosin were not significantly different between 1st and 2nd carcass grade loin. The hardness and chewiness of 2nd carcass grade loin's were significantly lower than 3th grade loin's, but the myofibril fragmentation index, sarcoplasmic protein extractability and Mg-ATPase activity of myofibril were higher. The myofibrillar protein extractability and Mg-ATPase activity of actomyosin obtained from 3th carcase grade loin's were significantly higher than D grade loin's, but the hardness, chewiness and stroma protein extractability were lower. In conclusion, the degree of toughness in Korean native cow's loin was not significantly different between 1st and 2nd grade, but 3rd and D carcass grade were significantly higher, regardless of before and after aging.

  • The effect of sugar addition and heat treatment on the myofibrillar protein extractability was studied. Maillard reaction was dependent on heating time significantly and glucose revealed the highest reactivity for Maillard reaction. The extractability of myofibrillar proteins was lowest in case of glucose addition and decreased according to increasing of heating time, Higher extractability was resulted in by digestion of myofibrillar proteins with enzymes after sugar addition and heat treatment than the undigested samples, as the sample was digested with trypsin that is the highest. And by digestion with trypsin, chymotrypsin and peptidase at a time the extractability of meat proteins increased remarkably.

Extractable Heavy Metals in Phosphogypsum

  • In addition to supplying the essential elements, Ca and S, phosphogypsum can have profound effect on both the physical and chemical properties of certain soils. However, no widespread use of by-product phosphogypsum will be made unless such uses pose no threat to the public health and soil contamination. In this study, the extractability of As, Cd, Cu, and Pb with water and DTPA solution from phosphogypsum samples of pH 3, 5, and 7 were investigated to estimate the availability of those metals. Contents of water extractable metals in ohosphogypsum were less than 5 mg/kg for all the heavy metals investigated. The extractability of metals in DTPA solution was not quite different but a little higher in comparison to the water extraction. And the extractability was decreased as the pH of phosphogypsum increased. In the phosphogypsum of pH 7, amounts of water extractable metals were nearly zero. There was no significant difference in the amount of extractable metals during the extraction period of 5 weeks. The length of extraction time did not affect heavy metal extractability. Therefore there may be small fractions of easily soluble or extractable forms of metals in the phosphogypsum and most of the metals would be present in very insoluble forms. These results suggest that the application of phosphogypsum at appropriate rates on agricultural lands appears of no concern in terms of hazardous element contamination in soil.

Extraction of Cd and Pb from Soil by Anionic Surfactant and Ligand NaI (NaI 리간드화 계면활성제에 의한 토양내 Cd과 Pb 추출 연구)

  • Heavy metals, Cd and Pb, in soil were extracted by using anionic surfactants such as AOS (alpha olefin sulfonate), SDS (sodium dodecyl sulfate), and LAS (linear alkyl benzene sulfonic acid). Metal extractability from soil was affected by the carbon number and solution pH of surfactants. LAS showed higher metal extractability due to the acidic solution condition. Although SDS has a fewer carbon number than AOS, it would produce smaller micelles and resulted in more efficient extraction of metals by increased soil contact. Cd extractability of surfactant was twice enhanced by adding NaI as a ligand. However, Pb extractability of surfactant was sometimes reduced by adding NaI. Those ligand effects were dependent on solubility of metal-ligand. The column experiment also showed that SDS having smaller micelles resulted in higher metal extractability than AOS.

Contrast Effect of Citric Acid and Ethylenediaminetetraacetic Acid on Cadmium Extractability in Arable Soil

  • Chelating agents have been proposed to improve the efficiency of phytoextraction of heavy metal hyperaccumulator. However, little studies to elucidate mechanism of chelating agents to increase cadmium (Cd) extractability have been conducted. The objectives of this study were to evaluate effect of different chelating agents on Cd extractability and to determine mechanism of Cd mobilization affected by these agents. An arable soil was spiked with inorganic Cd ($CdCl_2$) to give a total Cd concentration of $20mgCdkg^{-1}$. Ethylenediaminetetraacetic acid (EDTA) and citric acid (CA) were selected and mixed with the arable soil at the rates of 0 and $5mmolkg^{-1}$. The mixture soils were incubated at $25^{\circ}C$ for 4 weeks in dark condition. Concentration of F1 Cd fractions (water soluble) significantly increased with addition of EDTA but did not changed with addition of CA. Especially; concentration of F5 Cd fractions (residual) significantly increased with addition of CA. Increase in water soluble with EDTA might be attributed to complexation of Cd and EDTA. Dissolved organic carbon concentration significantly increased with EDTA addition, but did not with CA implying that considerable amount of CA was decomposed to inorganic carbon by microorganism. Log activity of carbonate ($CO_3{^{2-}}$) which might be generated from CA increased with addition of CA. Increase in residual Cd fraction might be due to precipitation of Cd as $CdCO_3$. As a result, EDTA was effective in increasing Cd extractability, by contrast CA had significant effect in reducing Cd extractability.

ATPase Activity and Solubility of Actomyosin Extracted from Muscle of Silky Fowl (오골계에서 추출한 Actomyosin 의 ATPase 활성 및 용해도)

  • Investigation on the extractability, Mg2+-, Ca2+ , EDTA-ATPase activity and solubility of actomyosin prepared from leg and breast muscle of silky fowol were as follows. The extractability of actomyosin in leg and breast muscle was 779mg/100g and 1, 318mg/100g respectively, breast muscle was higher than leg muscle . Mg2+-ATPase activity of actomyosin was high inionic strength 0.02-0.10 and Mg2+ATPase activity of low ionic strength was higher than high ionic strength not related to the part. Ca2+ ATPase activity was high in ionic strength 0.05-0.13, the activity of leg muscle was higher that breast muscle. And EDTA-ATPase activity showed low in low ionic strength and showed high in high ionic strength, and increased greatly depend ionic strength up to 0.4. The solubility of actomyosin was not different in leg and breast muscle , the solution started in KCI concentration of 0.3M and ended in DCI concentration of 0.4M.

Factors Affecting the Extractability of Protein of Silkworm Larvae (번데기 단백질(蛋白質)의 추출성(抽出性)에 영향을 미치는 요인(要因))

  • This experiment was designed to investigate the optimum conditions for the extraction of protein from the silkworm larvae by using the oven-dried and freeze-dried samples. The extractability was examined on the basis of kinds of extractants, ratio of sample to extractants, extraction time, temperature, and the effect of pH. The following results were obtained: (1) About 56% of proteins were extracted when 0.05M NaOH was used and the highest extractability was obtained at the ratio of 1 : 40 (sample: extractant). (2) The amount of nitrogen extracted was gradually increased when the extraction temperature was increased from $10^{\circ}C$ to $50^{\circ}C$ in both oven-dried and freeze-dried silkworm larvae. The highest amount of nitrogen was extracted at $50^{\circ}C$ in freeze-dried sample and at $60^{\circ}C$ in oven-dried sample. In addition, sixty min. of extraction time showed the highest extractability of nitrogen, however, not much difference in extractability was found after 10 min of extraction time. (3) More than 60% of nitrogens were extracted at pH 13.0, but less than 20% were extracted from pH 1.0 to 11.0. There was only slight difference in extractability of nitrogen between freeze-dried and oven-dried samples.

Effect of Packing Method on Physico-chemical Properties of Frozen Chicken (포장방법이 동결계육의 이화학적 특성에 미치는 영향)

  • This experiment was carried out to find out the effect of packing methods on physico-chemical properties of breast and thigh meats in chicken, which was dried by air spray chilling method. The chicken carcass was cut into breast and thigh muscles, which were either vacuum packed or atmosphere packed, and stored at -2O˚C for 1, 4, 8, 12 and 16 wk after quick freezing at -45˚C for 35 min. The pH values of atmosphere-packed breast meat and vacuum-packed breast meat after one wk of storage were higher than those of atmosphere-packed thigh meat and vacuum-packed thigh meat(P< .05). The pH values increased as storage period extended, but no significant difference was detected between two packing method(vacuum vs. atmosphere). Total moisture contents of breast meats after one wk of storage were higher than those of thigh meats. The total moisture contents decreased slowly as storage period extended, but no significant difference was detected between two packing method(vacuum vs. atmosphere). The shear force value of thigh meat was higher than that of breast meat. The shear force values of both meats decreased as storage period extended, regardless of packing method. The water soluble protein extractability of thigh meats was higher than that of breast meat, and the water soluble protein extractability of all treatments decreased until 8 wk after storage, but increased gradually after 8 wk of storage period. The salt soluble protein extractability of breast meat was higher than that of thigh meat, and the salt soluble protein extractability of all treatments decreased as storage period extended. With regard to the packing method, the vacuum packing showed higher value than that of atmosphere packing method until 8 wk of storage. Total lipid contents of atmosphere- and vacuum-packed thigh meats at 1 wk of storage were higher than those of breast meats, and the total lipid contents of all of treatments decreased as storage period extended. However, no significant difference was detected between two packing methods. The fatty acid contents of breast and thigh meats were in order of o1eic(33,5~42.4), palmitic(19.7~30.8) and linoleic acid(10.8~17.4).

Changes in Actomyosin ATPase Activities Extracted from Beef Meet during Postmortem Storage (저장기간에 따라 추출된 쇠고기 Actomyosin의 생물활성 변화)

  • This study was carried out to compare the extractability and ATPase activity of actomyosin extracted shank, rib and loin muscle of beef meat stored at 8$^{\circ}C$. The extractability of actomyosin in shank, rib and loin muscle were 36.74, 72.55 and 56.77mg/g early in the storage, respectively. The extractability of the rib and loin muscle were similar, the shank muscle was processed differently with their. The Mg- and Ca-ATPase activity of the shank muscle rised to 3 days, but decreased the 6th day. And Mg- and Ca-ATPase activity of the rib muscle was similar during storage period, the loin muscle made a slow descent. The strength of Mg- and Ca-ATPase activity showed in the order shank, rib and loin muscle. The EDTA-ATPase activity of the shank and rib muscle was difference according to storage period and ionic strength, but the loin muscle was increase in succession with magnitude of ionic strength.
