Antioxidant Effects of Cheonggukjang Containing Phellinus linteus Extract

  • This study was carried out to examine the antioxidant effects of cheonggukjang combined with Phellinus linteus extract. The electron-donating activity (EDA) of cheonggukjang containing 0.3% P. linteus extract (0.3% CPLE) was higher than that of cheonggukjang only. EDA of the ethanol extract from cheonggukjang was higher than that of the water extract. The water and the ethanol extracts showed strong antioxidant activity with regard to peroxide value. However, the ethanol extract showed a higher peroxide value than the water extract. The nitrite scavenging activity of the ethanol extract was greater than that of the water extract, crresponding to the EDA and peroxide values for each extract. Therefore, the antioxidant effects were enhanced by adding 0.3% of extract from P. linteus in manufacturing cheonggukjang. It is suggested that P. linteus extract could be put into practice as an effective antioxidant agent.

Comparison and Analysis of Several Main Components between Hot Water Extract and Alcohol Extract of Gamichungsangbohatang (가미청상보하탕(加味淸上補下湯) 열수추출물(熱水抽出物)과 알코올추출물(抽出物)의 지표물질(指標物質) 및 패턴의 비교분석(比較分析))

  • Objectives : To find out more pharmacologically efficient way of extraction of herbal dicoction, Gamichungsangbohatang (GMCSBHT). Methods : Several main components of GMCSBHT was compared and analysed between hot water extract of GMCSBHT and Alcohol(70% ethanol) extract of GMCSBHT via HPLC method. Results : Hot water extract of GMCSBHT showed relatively more component content than ethanol extract of GMCSBHT. Also weighted mean of main components of hot water extraction of GMCSBHT was higher than that of alcohol extract of GMCSBHT. But from chromatographic pattern analysis of matching ratio and similarity ratio showed that these two forms of extracts might have different chemical composition, and 3D PDA plot of alcohol extract of GMCSBHT showed high peaks near UV $190{\sim}220nm$ which was invisible in hot water extract of GMCSBHT. Conclusion : Alcohol extract of GMCSBHT may have some special components which do not exist in hot water extract of GMCSBHT.

Effect of Gastrodiae Rhizoma Extract on Blood Pressure and Plasma Catecholamine Level in Unanaesthetized Spontaneously Hypertensive Rats (천마(天麻)엑기스가 Spontaneously Hypertensive Rat 에서 혈압(血壓) 및 혈장(血漿) Catecholamine 함량의 변화(變化)에 미치는 영향(影響))

  • The present study was carried out to investigate the effect of water extract and 70% ethanol extract from Gastrodiae Rhizoma on cardiovascular activities and plasma levels of catecholamines in unanaesthetized spontaneously hypertensive rats. The depressor response in SHR was observed during three to six hour period after an oral administration of water extract from Gastrodiae Rhizoma(GR). There was a statistically significant correlation between the magnitude of the depressor response induced by an oral administration of water extract from GR and the initial control blood pressure level. The increase in blood pressure induced by norepinephrine was less in Wistar rat treated with GR water extract than those without GR extract. No significant change in heart rate was observed in SHR receiving either water extract or ethanol extract from GR. Associated with the depressor response, there was a concomitant reduction in plasma levels of norepinephrine in SHR at 4 hour after an oral administration of water extract from GR. Plasma levels of norepinephrine and epinephrine were decreased slightly at 2 hour after an oral administration of ethanol extract from GR. These results suggest that the depressor effect of water extract from GR may be due, in part, to a decreased sympathoadrenal activity.

  • The effects of Arctii fructus extract on allegenic inflammation were investigated using in vivo and in vitro test models. Firstly, the cytotoxicity of Arctii fructus extract was validated using MTT assay. As a result, Arctii fructus extract showed no cytotoxic potential, while SDS, a positive control, revealed strong cytotoxic effect. In LLNA assay, Arctii fructus extract showed no skin allergenicity. Next, the anti-allergic actions of Arctii fructus extract were evaluated using rodent experimental models. The oral, intraperitoneal and intradermal administration of Arctii fructus extract significantly inhibited the compound 48/80-induced vascular permeability documented by Evans blue extravasation. In addition, Arctii fructus extract showed potent inhibitory effect on passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE when orally administered. In an in vitro study, Arctii fructus extract revealed to possess inhibitory potential on the compound 48/80-induced histamine release from rat peritoneal mast cells. Moreover, Arctii fructus extract inhibited the IL-4 and TNF-${\alpha}$ mRNA induction by PMA and A23187 in human leukemia mast cells, HMC-1. Finally, it revealed that Arctii fructus extract significantly suppressed histamin-provoked antigenic inflammation reactions in human prick test. Taken together, these results suggest that anti-allergic action of Arctii fructus extract may be due to the inhibition of histamine release and cytokine gene expression in the mast cells.

Screening of Antimicrobial Activity and Proteolytic Enzyme Stability of Extract of the Blue Mussel Mytilus edulis (진주담치(Mytilus edulis) 추출물의 항균활성 및 단백질 분해효소에 대한 안정성 탐색)

  • This study was performed to screen the antimicrobial activities and proteolytic enzyme stability of the acidified extract of the Blue mussel Mytilus edulis. The acidified extract showed potent antimicrobial activities against Gram-positive bacteria, Bacillus subtilis, and Gram-negative bacteria, Escherichia coli D31, but had no activity against Candida albicans. Treatment of extract with trypsin completely abolished all or significant antibacterial activity against the tested bacteria, but slightly decreased antimicrobial activity against B. subtilis, and treatment of extract with chymotrypsin retained almost antibacterial activity against the tested bacteria except for E. coli D31. To confirm the additional enzyme stability of the extract, antimicrobial activity of the extract was tested after treated with several enzymes. Enzymes treated extract showed potent antimicrobial activity against B. subtilis and its activity was also retained for 5 h after trypsin treatments. Non-proteinaceous materials in the acidified extract also showed strong DNA-binding ability but did not show bacterial membrane permeabilizing ability. All our results indicate that mussel extract might contain the proteinaceous or non-proteinaceous antibacterial materials target not bacterial membrane but intracellular components. These results could be used to develop mussel extract as an additive for the improvement of stability or antimicrobial activity of antibiotics against specific bacteria.

Effect of Ginseng Residue Extract on Yeast Growth (효모생육에 미치는 홍삼박의 영향)

  • To evaluate the possible utilization of ginseng by-products, chemical components of ginseng residue, reducing ability of DPPH, effect of residue extract on the yeast growth, amino acid contents of yeast cell, increase of residue extract yield by enzyme treatment were studied. Alcohol and water extract residue contained 43-46% total reducing sugar and 14-15% crude protein, while alcohol extract residue had 0.18% n-BuOH extract. Water extract of alcohol extract residue had about 45% reducing ability of DPPH in comparison with that of alcohol extract from ginseng roots. Essential nutrients for the yeast growth were found in extract when Saccharomyces cerevisiae was cultured in Czapeck medium, a compound medium, with the residue. The addition of residue extract to malt medium, a natural medium, enhanced 30-40% yeast growth. And content of each amino acid in yeast cell cultured on malt medium with ginseng residue extract was much more than that of the cell cultured without ginseng extract, but amino acid composition of yeast cell did not differ from one another. The treatment of alcohol extract residue with cellulase increased 250% yield of residue extract.

  • The effect of dietary white ginseng on growth of silkworm larva was studied with several young and grown silkworm larvae divided into four groups and fed ad basal artificial diet containing different levels of ginseng extract. The four levels of ginseng extract added to basal artificial diet were: 0 (control), 46.6, 139.9 and 279.8 mg per g of dry diet. The results obtained were as follows: 1. The body weight of silkworm larvae fed ad basal artificial diet containing different levels of ginseng extract was increased in 46.6mg ginseng extract level, 139.9mg ginseng extract level and 279.8mg ginseng extract level order. The body weight was greatly increased in 46.6mg ginseng extract level than in control. It is, therefore. considered that a little white ginseng was effective for larval growth in silkworm rearing with artificial diet. 2. The blood sugar content of silkworm larvae fed on an artificial diet containing several levels of ginseng extract was remarkably decreased as compared wilt that of the control silkworm larvae.

  • This study has been carried out to investigate the effects of Ukgansan(UGS) extract on anti-convulsive, antipyretic, analgesic, sedative and GABAergic system of experimental animals. The results of this study were as follows : 1. UGS extract prolonged significantly the beginning time to convulsion and death induced by strychnine. 2. UGS extract prolonged significantly the time to death induced by electrical shock of ECT unit.(3 sec, 200 F, 25 mA) 3. On the experiment of hypothermic effects of UGS extract on the rectal temperature of mouse, UGS extract decreased significantly the rectal temperature of mouse 4. On the experiment of antipyretic effects of UGS extract on the febrile induced by the subcutaneous injection of $150\;{\mu}g/kg$ endotoxin in mouse, UGS extract decreased significantly the rectal temperature of mouse. 5. On the experiment of analgesic effects of UGS extract on the writhing syndrome induced by intraperitoneal injection 0.7% acetic acid 1 ml/100g in mouse, the writhing syndrome induced by acetic acid was reduced significantly by administration of UGS extract. 6. On the experiment of effects of UGS extract on spontaneous motor activity measured by wheel cage method in mice, the spontaneous motor activity was reduced significantly by administration of UGS extract. 7. On the experiment of effects of UGS extract on the activity of GABA-transaminase(GABA-T) in mouse brains after 21 days of oral administration of UGS extract. the activity of GABA-T was reduced significantly by administration of UGS extract. 8. On the experiment of effects of UGS extract on the activity concentration of GABA in mouse brain after 21 days of oral administration of UGS extract, the activity concentration of GABA was reduced significantly by administration of UGS extract. 9 On the experiment of effect of UGS water extract on the activity of GAD in mouse brain after 21 days of oral administration of UGS extract, the activity of GAD was reduced significantly by administration of UGS extract. According to the these results, Ukgansan extracts reveal the effects on the anti-convulsive, antipyretic, analgesic, sedative and GABAergic system.

Effect of Chicory Root Extract on Cholesterol Metabolism in Rats

  • Effects of water-soluble extract from roasted-chicory root on the cholesterol metabolism in rats fed cholesterol diet were investigated. Sprague-Dawley rats received a hypercholesterolemic diets without (control group) or with 5.0% water-soluble extract from roasted chicory root for 2 weeks. Roasted chicory extract group showed significantly higher body weight gain and food intake compared with the control group. The concentrations of total cholesterol and LDL+VLDL cholesterol in serum were significantly lower in rats fed roasted chicory extract diet. However, HDL-cholesterol concentration, and atherogenic index were not significantly affected by the dietary roasted chicory extract. Fecal net weight, fecal cholesterol, and bile acid excretion were significantly higher in the chicory extract group. The results suggest that the hypocholesterolemic effect in rats fed roasted chicory extract may be caused by an alteration in the absorption of cholesterol by an increase in the fecal excretion of cholesterol and bile acid.
