Isolation of Keratinolytic Protease Producing Microorganism and Its Cultivation Condition (Keratinolytic protease 생산균, Pseudomonas sp. KP-364의 분리 및 배양)

  • A bacterial strain KP-364 producing extracellular keratinolytic protease was isolated from the soil of the poultry fac-tory. It was identified as Pseudomonas sp. based on its morphological and physiological characteristics, The optimal culture conditions for the production of keratinolytic protease by Pseudomonas sp. KP-364 were investigated. The composition of optimal medium for the keratinolytic protease was 2.0% glucose, 0.5% soybean meal. 0.5% $NaNO_3$ and 0.2% KCI Optimal initial pH for production of Keratinolytic protease production were 6.5 and $37^{\circ}C$ respec- tively. The keratinolytic protease production reached a maximum of 1,270 U/ml/hr after 48 hours cultivation under the optimal culture conditions.

  • Bacillus sp. SMMJ-2 producing extracellular keratinolytic protease was isolated from the Swedish soils. The optimal culture conditions for production of keratinolytic protease by Bacillus sp. SMMJ-2 were investigated. The optimal medium compositions for the keratinolytic protease production were 0.7% $K_2HPO_4$, 0.2% $KH_2PO_4$, 1.0% fructose,1.2% soybean meal (roasted), and 0.01% $Na_2CO_3$. Optimal initial pH and temperature for the production of keratinolytic protease were 7.0 and 30$^{\circ}C$, respectively. The keratinolytic protease production showed a maximum of 105 units/ml/min after 72 hours cultivation under the optimal culture conditions.

  • solvent-tolerant bacterium strain, MH6, was isolated by hydrophilic organic solvent DMSO enrichment in the medium and identified as Serratia marcescens. The extracellular protease with novel organic-solvent-stable properties from strain MH6 was purified and characterized. The molecular mass of the purified protease was estimated to be 52 kDa on SDS-PAGE. The open reading frame (ORF) of the MH6 protease encoded 504 amino acids with 471 amino acid residues in the mature protease. Based on the inhibitory effects of EDTA and 1,10-phenathroline, the MH6 protease was characterized as a metalloproteinase. The enzyme activity was increased in the presence of $Ni^{2+}$, $Mg^{2+}$, and $Ca^{2+}$. The protease could also be activated by the nonionic surfactants Tween 80 (1.0%) and Triton X-100 (1.0%). The protease showed remarkable solvent stability in the presence of 50% (v/v) solutions of long-chain alkanes and long-chain alcohols. It was also fairly stable in the presence of 25% solutions of hydrophilic organic solvents. Owing to its high stability in solvents and surfactants, the MH6 protease is an ideal candidate for applications in organic catalysis and other related fields.

Relationship between Sporulation and Synthesis of Alkaline Protease in Streptomyces sp. (방선균 일주에서 포자형성과 호알칼리성 단백질 분해효소의 생합성과의 관계성)

  • The aims of the present studies were to understand the physiolosical and genetic characters of Streptomyces sp. isolated from soil. It revealed that Streptomyces sp. SMF301 had very fast growth rate and produced extracellular protease and heavily sporulated on rich media. It also showed $\beta$-lactamase activity and pigment production. Nonsporulating mutants were isolated after NTG or acriflavin treatment and their characters were compared with the parent strain. It was found that the mutants obtained by acriflavin treatment and ghier characters were compared with the parent strain. It was found that the mutants obtained by acriflavin treatment lost the pigment formation and $\beta$-lactamase production. Protease actibity of the mutant was lowered and the pH optimum was changed toward neutral. It was found that the changes were resulted from the reduction of alkaline protease biosynthesis in the bald mutant. Therefore it is considered that sporulation, pigment formation, $\beta$-lactamase production, and alkaline protease production in Streptomyces sp. might be controlled with a closely related relationship.

  • A protease isolated and purified 51 fold from the culture filtrate of a soil bacterium, Bacillus sp. KUN-17, which was appeared to be a monomeric protein with molecular weight of 38, 000 daltons, was suggested to be involved in the serine (-alkaline) protease (E.C 3.4.21.14) since its activity was selectively inhibited by phenylmethylsulfonyl fluoride (PMSF) and required 40$\circ$C and pH 10.5 for optimal condition. The half-life of the enzyme activity was 1 hr at 55$\circ$C, and the activity was maintained even under high concentrations of SDS or urea. The enzyme was indicated to perform random proteolysis from the fact that most of the chromogenic substrates employed were hydrolyzed by the enzyme. The affinity of the enzyme for natural proteins was approximately 10-times higher than ester compounds, and both substrates showed mutual inhibitory effect competitively for the enzyme activity.

  • Several protease-deficient mutants of Aspergillus niger have been isolated by halo-screening on skim milk plate after UV irradiation of conidiospores. The extracellular proteolytic activities of the mutant strains grown in an optimized medium varied from 3% to 85% of that of the parental strain. Especially, two mutant strains named as ANPD-129 and ANPD-153, which had 3% and 49% of acid protease activity of the parental strain, respectively, were further characterized both physiologically and genetically. The growth rates of the mutants, ANPD-129 and ANPD-153, were similar to that of the parental strain, unlike other protease-deficient mutants. The diploid formed between the two mutants restored protease activity to a similar level of that of the parental strain. This result revealed that ANPD-129 and ANPD-153 had mutations at different loci. Using master strains with marked chromosomes these loci were assigned to linkage groups. The mutation locus (prt129) in ANPD-129 was assigned to linkage group VI and the locus (prt153) in ANPD-153 to linkage group III.

  • A thermophilic bacteria showing proteolytic activity against defatted soybean was isolated from soil. It was identified as Bacillus amyloliquefaciens based on its morphological and physiological characteristics. The Bacillus amyloliquefaciens NS 15-4 was cultivated at 50$\circ$C by rotary shaking in a medium containing defatted soybean. An extracellular protease from this strain was purified to homogeneity by ammonium sulfate precipitation, ion exchange, and hydrophobic interaction chromatographies. The molecular weight of the enzyme was estimated to be approximately 30,000 by SDS-PAGE and the N-terminal amino acid sequence of the enzyme was turned out to be AQSVPYGISQIKAPA. The optimum temperature and pH for the enzyme reaction were 60$\circ$C and 11, respectively, and its thermostability was increased by the addition of calcium ion. The enzyme was inactivated by phenylmethylsulfonylfluoride, suggesting it be a serine protease. Comparing with other commercial proteases, the enzyme showed relatively high proteolytic activity against defatted soybean, a water-insoluble protein substrate.

  • Alkaline bacterium producing a high pro-tease activity at low temperature was isolated by using enrichment culture from soil samples and identified as Bacil-lus sp. WRD-1 Cell growth was maximal at 10 hours and the optimal initial pH and culture time of culture condition for enzyme production was pH 7 and 10 hours, respectively. Temperature range of high enzyme activity were $10~40^{\circ}C$. The optimal pH and temperature for the enzyme activity were pH9 and $30^{\circ}C$.

  • Effect of iron on the extracellular proteolytic activity of live Uronema marium was determined by fluorescence polarization (FP) method. Supplementation of 0.5 and 5.0 μM iron significantly increased caseinolytic activity of live U. marinum. In contrast, supplementation of 50 μM iron showed no significant differences in FP values compared to the control. The present result suggests that iron in cultured water or skin tissue of olive flounder may influence on the penetration and establishment of U. marinum, correlating with modulation of extracellular protease activity of the ciliates.

Characterization of Partially Purified Extracellular Protease of Local Bacteria BAC-4

  • To achieve the aim of this investigation, the extracellular protease was isolated from bacteria BAC-4, a strain was cultivated in the medium for the production of penicillin acilase in a period of 32 hours. The enzyme was first purified by aceton precipitation method, followed by ion exchange chromatography on DEAE-sephacel column. The highest specific activity of the aceton fraction was found to be 2.19 unit per mg, with degree of purification of 13 times. Further purification of the enzyme on DEAE -sephacel had a specific activity of 58.6 unit per mg and degree of purification of 344 times compared to its crude extract. The optimum pH of the enzyme was 8.4, and the potimum temparature was 37$^{\circ}C$. The K$\_$M/ and $V_{max}$ calculated at experiment conditions were found to be 0.66%(W/V) and 3.61 unit per mL respectively.
