Influences of mineral contents in decoction water on the extraction efficiency of bioactive compounds from Eucommiae Cortex (전탕용수의 미네랄 함량이 두충의 유효성분 추출에 미치는 영향 분석)

  • Objective : Eucommiae Cortex is bark of Eucommia ulmoides Oliver, a deciduous tree native to China. Today, teas or herbal drugs made from E. ulmoides leaves are widely consumed as a healthy food especially in Eastern Asia including Japan, China and Korea. This study was aimed to verify the extraction efficiency of bioactive compounds from Eucommiae Cortex depending on characteristics of decoction water. Method : Six types of decoction water with various origin were prepared and the contents of minerals in water were analyzed by ICP-AES and ICP-MS. The contents of extracted bioactive compounds were analyzed by HPLC and the relationship between mineral contents and extracted bioactive compounds in the decoction water were analysed by multiple factor analysis. The decoction waters could be classified in three groups by hierarchical cluster analysis. Result : Extracted amounts of compounds in Eucommiae Cortex showed a positive relationship with Na, Si, K concentration in decoction water, while a negative relationship with Ca, Mg concentration. Conclusion : The study proved that the minerals in water influenced the extraction efficiency of bioactive compounds from Eucommiae Cortex.

Effects of Eucommiae Cortex on Osteoblast-like Cell Proliferation and Osteoclast Inhibition

  • Methanol extract (MeOH), n-hexane (Hx), chloroform ($CHCl_3$), ethyl acetate (EA), butanol (BuOH) and aqueous ($H_2O$) fractions of Eucommiae Cortex including geniposidic acid (GA), geniposide (GP) and aucubin (AU) were tested for their therapeutic efficacy on osteoporosis. The contents of GA, GP and AU in the cortex and leaf of Eucommia ulmoides Oliver were quantified by HPLC. The effect of Eucommiae Cortex on the induction of growth hormone (GH) release was studied by using rat pituitary cells. The proliferation of osteoblast-like cells increased by herbal extracts was assayed using a tetrazolium (MTT), alkaline phosphatase (ALP) activity, and [$^3H$]-proline incorporation assays. The inhibition of osteoclast was studied by using the coculture of mouse bone marrow cells and ST-2 cells. As a result, the GA, GP and AU were present in the cortex more than in the leaf of E. ulmoides Oliver. The MeOH (1mg/mL), Hx, $CHCl_3$ and EA fractions (each 20 $\mu$ g/mL) had potent induction of GH release. The $CHCl_3$ exhibited the potent proliferation of osteoblasts. The AU, GP and GA were increased proliferation of osteoblasts. In addition, GA ($IC_{50}: 4.43{\times}10^{-7}$M), AU and GP were significantly inhibited proliferation of osteoclast. In summary, it is thought that the components in a part of the fractions of Eucommiae Cortex participate in each step of mechanism for activating osteoblast to facilitate osteogenesis, and suppress osteoclast activity to inhibit osteolysis.

Quantitative Analysis of Glycine Semen Nigra and Eucommiae Cortex for Standardization of GCSB-5 Preparation (생약복합제 GCSB-5의 품질 표준화를 위한 흑두 및 두충의 함량 분석)

  • GCSB-5 preparation is a purified extract from a mixture six herbal medicines (Acanthopanacis Cortex, Achyranthis Radix, Saposhnikoviae Radix, Cibotii Rhizoma, Glycine Semen Nigra, Eucommiae Cortex) that have been widely used in traditional medicine to treat various bone disorders. This study was carried out to obtain the HPLC analysis method that can be used to establish quantitative analysis of Glycine Semen Nigra and Eucommiae Cortex for standardization of GCSB-5 preparation. HPLC analysis methods for the simultaneous determination of genistin (Glycine Semen Nigra) and geniposide (Eucommiae Cortex) were established for the quality control of herbal medicinal raw material and preparation. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline. As the result of quantitative analysis, the contents of genistin and geniposide in the raw material of GCSB-5 preparation were 0.0426-0.0427 mg/g and 0.431-0.432 mg/g. And GCSB-5 preparation contained genistin of 0.0202-0.0203 mg/capsule and geniposide of 0.211-0.212 mg/capsule, respectively.

Effects of Several Natural Medicines on Alkaline Phosphatase Activity in hFOB1 (수종의 생약제제가 hFOB1의 염기성 인산분해 효소 활성에 미치는 영향)

  • Recently, many natural medicines, which have advantage of less side effects and possibility of long-term use, have been studied for their capacity and effects of anti-bacterial, anti-inflammatory and regenerative potential for periodontal tissues. Cortex Eucommiae, Eupoly phaga, Semen Cuscutae, Halloysitum Rubrum have been traditionally used as medicines for treatment of bone disease in Korea. The objective of the present study is to examine the ability of alkaline phosphatase (ALP) activity in human fetal osteoblast cell line (hFOB1) with several natural medicines. hFOB1 added DMEM/F-12 were cultured with dexamethasone as a positive control, and with each natural medicine. ALP activity was measured by spectrophotometer for enzyme activity and naphthol AS-Bl staining was performed for morphometry. All of the natural medicines induced a higher ALP activity compared to negative control, especially, Cortex Eucommiae increased an ALP activity in all experimental groups (p<0.05). In naphthol AS-Bl staining, all of the natural medicines of this study increased the stained area compared to negative control. Especially, Cortex Eucommiae and Eupoly phaga showed statistical significance compared to negative control (p<0.05). These results indicate that Cortex Eucommiae, Eupoly phaga, Semen Cuscutae, Halloysitum Rubrum have an inducing ability of ALP synthesis on osteoblasts.

Dyeability of Colorant in Eucommiae Cortex - Analysis of Color Difference Value Depending on Dyeing Condition - (두충색소의 염색성 - 염색 조건에 따른 색차분석을 중심으로 -)

  • The color difference values of dyed fabrics with colorant in Eucommiae Cortex were as follows; One hour of dyeing depending on pH at $95^{\circ}C$ exhibited colors of dark brown, light brown, light beige and grey. The colors of wool, nylon and silk was darkest brown at pH 3 but light brown or yellow as pH increased. In all dyed fabrics the color was changed to darker brown as time prolonged, however; there was no further color change and value and chroma of wool declined. At pH 3, the increase in temperature turned colors of all dyed fabrics into dark brown from light yellow. As the result of repetitive dyeing, colors of all dyed fabrics gradually turned into darker brown. The methods of mordant resulted in color changes between light yellow and dark brown without various color changes.

Antioxidative Properties and Whitening Effects of the Eucommiae cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma (두충, 단삼, 진피 및 천궁의 항산화 활성 및 미백 효과)

  • In an attempt to find natural sources of antioxidants and whitening agents, comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Eucommiae Cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma were carried out. Comparison of the four ethanol extracts revealed that, Aurantii nobilis pericarpium had the highest electron-donating ability(79.0%),; however, Salviae miltiorrhizae radix had the highest SOD-like ability(21.9%). The xanthine oxidase experiment exhibited a hindrance effect of 79.3% in Salviae miltiorrhizae radix, 57.5% in Eucommiae cortex and 71.9% in Aurantii nobilis pericarpium. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 12.4% in the Eucommiae cortex, 22.8% in the Salviae miltiorrhizae radix, 27.5% in the Aurantii nobilis pericarpium and 59.5% in the Cnidii rhizoma. Based on these results, we suggest that the ethanol extracts of Eucommiae cortex, Salviae miltiorrhizae radix, Aurantii nobilis pericarpium and Cnidii rhizoma. can be used as food and cosmetic ingredients.

Comparison of Index Compound Content and Anti-inflammatory Activity of Eucommiae Cortex by Various Drying Methods (두충의 건조법별 지표성분 및 항염증 활성 비교)

  • Objectives Eucommiae cortex is one of the frequently used herbs for musculoskeletal disorders, with well-known anti-inflammatory effect. Meanwhile, powdered form of herbal drugs is more advantageous than decoction form, in that storing and taking is more convenient. Thus, the authors aimed to investigate whether the drying method affects the index compound level and anti-inflammatory effect of eucommiae cortex water extract. Methods Eucommiae cortex was extracted in distilled water at $100^{\circ}C$ for 3 hours, filtered, and then evaporated under vacuum. One half of the sample was freeze-dried at $-80^{\circ}C$. Another half of the sample was added with dextrin and then spray-dried at $180^{\circ}C$. To assess the possible change in index compound content, pinoresinol diglucoside was selected as the index marker. The content level of the index compound in various extract sample was quantified through high performance liquid chromatography. In addition, iNOS assay in LPS-induced RAW 264.7 cells was adopted to determine the anti-inflammatory effect of various extract samples. Furthermore, MTT assay was performed to confirm that the result of iNOS assay was not due to cytotoxicity. Results There was no significance difference in index compound content between extract samples obtained through two different drying methods. Anti-inflammatory activity of extract samples were similar at the matching concentration, regardless of the drying methods. Extract samples did not show any significant cytotoxicity. Conclusions Extract samples of eucommiae cortex were obtained through freeze-drying and spray-drying. Neither change in index compound content nor difference in anti-inflammatory activity was observed between drying methods.

Effects of Eucommiae Cortex (杜冲) on the Bone Histomorphometry and Metabolism in Ovariectomized Rats (두충(杜冲)이 난소적출(卵巢摘出) 흰쥐의 골조직(骨組織) 형태(形態)와 골대사(骨代謝)에 미치는 영향(影響))

  • Objective : This study was undertaken to investigate the effects of Eucommiae Cortex on the osteoporosis in ovariectomized rats. Materials and Methods : We used Sprague-Dawley female rats in 8-week-old. They were divided into three groups. Sample group was ovariectomized and administered with 10 mg/100 g/day Eucommiae Cortex extract solution for 10 weeks. Control group was ovariectomized and sham group was conducted by sham's operation. And control group and sham group were administered with normal saline as the same way. We measured rats's body and uterus weight and also measured the serum levels of Ca, phosphorus and ALP. We stained the specimens of rat's tibial bones with Goldner's modified Masson's Trichrome and then examined bone histomorphometry with Bioquant computer program of image analysis system. We measured the thickness of osteoid and callus as static parameters and measured bone volume and mineral apposition rate as dynamic parameters. We observed the expressions of RANKL and OPG mRNA of the tibial bone by RT-PCR. Results : The body weight was significantly (p<0.05) increased in control and sample groups compared with sham group, respectively. The uterus weight was significantly (p<0.05) decreased in control and sample group compared with sham group. In the change of Ca, phosphorus and ALP there were no significant changes among three groups. There were no significant changes of trabecula cortical and osteoid bones' thickness and volume. But trabecula mineral apposition rate (MAR) was significantly (p<0.05) increased in sham and sample group compared with control group. In the expression of RANKL mRNA, sample group was decreased compared with control group, and in that of OPG mRNA, sample group was increased compared with control group. Conclusion : This study shows that Eucommiae Cortex has the beneficial effects on bone histomorphometry and metabolism in the ovariectomized rats. We suggest that Eucommiae Cortex be useful for the treatment of osteoporosis.

Effects of Traditional Drugs on $CCl_4-induced$ Cytotoxicity in Primary Cultured Rat Hepatocytes (수종의 전통약제가 일차 배양 간세포에서 $CCl_4$ 유발 세포독성에 미치는 영향)

  • 80% Methanol extracts of 44 traditional drugs used for the treatment of liver diseases or tonic effects were screened for anti-hepatotoxic activity by in vitro assay using $CCl_4-induced$ cytotoxicity in primary cultured rat hepatocytes. $CCl_4-induced$ cytotoxicity was evaluated by determination of LDH, GOT or GPT activity in the medium. Rehmaniae Radix Preparata and Gelantina nigra inhibited the release of LDH, GOT or GPT from $CCl_4-treated$ hepatocytes. Gibotii Rhizoma and Eucommiae Cortex showed inhibitory effect on release of LDH from normal hepatocytes as well as $CCl_4-treated$ hepatocytes. Eucommiae Cortex and Lili Bulbus decreased release of GOT and LDH from normal hepatocytes, respectively. Astragali Radix inhibited release of GPT in $CCl_4-treated$ hepatocytes. Phlomidis Radix, Imperatae Rhizoma, Cistanchis Herba, Broussonetiae Fructus, Asparagi Tuber, Trigonellae Semen and Polgonati Rhizoma inhibited release of LDH from $CCl_4-treated$ hepatocytes. Among 44 traditional drugs, most of them released LDH, GOT or GPT at the dose of 1 mg/ml in normal hepatocytes, and Drynariae Rhizoma, Acanthopanacis Cortex, Longanae Arillus, Atratylodis Rhizoma and Ecliptae Herba increased $CCl_4-induced$ cytotoxicity.
