목단피로부터 식품부패 미생물에 대한 항균성 물질의 분리 및 동정 (Isolation and Identification of Antimicrobial Compound from Mokdan Bark (Paeonia suffruticosa ANDR))

  • 우리나라 야산에서 손쉽게 구할 수 있는 구황식물인 목단피를 건조시켜 분쇄한 후 methanol과 여러 용매로 추출하여 식품 부패 미생물의 증식 억제 효과를 검색하고 그 항균물질을 분석하여 다음과 같은 결과를 얻었다. 목단피의 methanol 추출물은 1500$\mu\textrm{g}$/mL 농도에서 B. subtilis, L. monocytogenes, V parahaemolyticus의 증식을 100% 억제하였으며 S. aureus, E. coli도 같은 농도에서 각각 36.31%와 67.81%의 억제 효과를 보였다. 목단피의 ethylacetate 분획 추출물은 낮은 농도인 500$\mu\textrm{g}$/disc의 농도에서 식품 부패 미생물 5종 모두에 대하여 clear zone을 형성하여 항균력을 나타내었다. 특히 L. monocytogenes, E. coli균에 대해서는 낮은 농도인 500$\mu\textrm{g}$/disc에서 각각 12mm, 11mm의 clear zone을 형성하였다. 목단피의 ethylacetate추출물을 silica gel colum chromatography(7cm${\times}$1.2 cm)한 후 thin layer chromatography(TLC)를 실시하여 항균 실험을 한 결과 B. subtilis의 경우 500 $\mu\textrm{g}$/disc 농도에서 2nd fra. 1이 21mm, V. parahaemolyticus는 250$\mu\textrm{g}$/disc 농도에서 14mm를 형성하여 목단피의 methanol 추출물로부터 각 용매별로 계통 분획하여 분리한 ethylacetate 분획으로부터 silica gel column chromatograpy와 TLC를 하여 항균성을 보인 2nd fra. 1의 성분을 알아보기 위하여 HPLC로 단일분리하여 얻은 peak Ⅸ에 대한 compound를 GC-MS로 분석한 결과 peak Ⅸ에서는 isobutyl isopentanoate가 있는 것으로 추정되었다.

  • 우리나라 야산에서 쉽게 구할 수 있는 구황 식물인 질경이를 메탄올과 여러 용매로 추출하여 식품 부패 미생물에 대한 항균력을 실험하고 그 항균활성 물질을 분석하여 다음과 같은 결과를 얻었다. 질경이의 메탄올 추출물은 2000$\mu\textrm{g}$/ml 농도에서 B. subtilis 및 V. parahaemolyticus의 증식을 100% 억제하였다. 그리고 질경이의 ethylacetate 분획물은 1000$\mu\textrm{g}$/disc 농도에서 5종의 모든 실험 균주에 대하여 8.5~11mm 크기의 clear zone을 형성하여 가장 높은 항균력을 보였다. 질경이의 ethylacetate 분획물을 silica gel column chromatography와 TLC로 분리하여 얻은 fraction을 5가지 실험 균주에 대하여 항균 실험을 한 결과 3rd 분획물 중 4번째 분획물이 250$\mu\textrm{g}$/disc 농도에서 모든 균주에 대하여 10 mm 이상의 clear zone을 나타내었다. 항균력을 보인 질경이의 ethylacetate 3rd fraction No. 4를 HPLC로 단일 분리하여 얻은 peak IV로 부터 $^1$H-NMR 및 GC-MSD로 동정한 결과 hexadecanoic acid로 동정되었다.

참나물의 항균, 항산화 활성 및 대장암세포 성장억제 활성 평가 (Antibacterial, Antioxidative and Anti-proliferative Activity against Human Colorectal Cell of Pimpinella brachycarpa)

  • 참나물은 광범위하게 이용되는 식용산채임에도 불구하고 그 연구는 전 세계적으로 미미한 상태이다. 본 연구에서는 최근 고급 식용 산채로서 각광받고 있는 참나물의 메탄올 추출물을 조제하고, 이로부터 n-hexane, methylene chloride, ethylacetate 및 butanol을 이용하여 순차적 유기 용매 분획물과 물 잔류물을 조제하여 각각의 항산화, 항균 및 대장암세포 생육억제 활성을 평가하였다. 참나물 메탄올 추출물의 71.51%는 물 잔류물로 이행되어 추출물 대부분이 수용성 물질이었으며, n-hexane, methylene chloride, ethylacetate 및 butanol 분획 효율은 각각 18.71, 0.7, 0.56, 및 4.57%로 나타났다. 총 플라보노이드 및 총 폴리페놀 함량은, 분획수율이 가장 낮은 ethylacetate 분획에서 가장 높았으며 물 잔류물에서 가장 낮았다. 항균 활성 평가결과, ethylacetate 분획에서 그람양성 및 그람음성세균에 대해 광범위한 항균 활성을 나타내었으며, 특히 그람양성세균에는 n-hexane 분획이, 그람음성세균에는 ethylacetate 분획이 효과적이었다. 항산화 활성의 경우 ethylacetate 및 butanol 분획에서 양호한 DPPH 및 ABTS 소거능, 환원력을 나타내었으며, n-hexane및 methylene chloride 분획에서 우수한 nitrite 소거능을 나타내었다. 한편 butanol 분획에서는 다른 분획과는 달리 우수한 인간 대장암세포 생육억제 활성을 나타내었다. 본 연구 결과는 참나물을 이용한 기능성 식품개발 및 ethylacetate 분획을 대상으로 한 유용 소재개발의 기본 자료로 활용될 것이다.

애기달맞이꽃(Oenothera laciniata Hill) 추출물의 항균활성 (Antibacterial Activities of Oenothera laciniata Extracts)

  • The solvent extracts of Oenothera laciniata, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservative. The O. laciniata extract by 80% ethanol was sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and butanol. The antibacterial activities and cell growth inhibition were investigated on each strain with the different concentrations of O. laciniata extracts. Antibacterial activities were shown in ethanol, ethylacetate, and butanol fraction of O. laciniata. However n-hexane, dichloromethane and water fraction showed weak antibacterial activity against the tested microorganisms. Among the five fractions, ethylacetate fraction showed the highest antibacterial activities against microorganisms tested, such as Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella Enteritidis and Salmonella Typhimurium. The polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess high antibacterial activity. The polyphenolic compounds from ethanol, n-hexane, dichloromethane, ethylacetate, butanol, and water fraction were 63.96 mg/g, 8.49 mg/g, 28.11 mg/g, 172.64 mg/g, 114.56 mg/g, and 34.91 mg/g, respectively. There are some relationships between antibacterial activity and polyphenol content in natural plant. The ethylacetate fraction could be suitable for the development of a food preservative.

천연약용식물의 활성산조종에 의한 산화적 손상의 보호 효과 (Protective Effect of Natural Medicinal Plants against Oxidative Damage Induced by Reactive Oxygen Species)

  • In order to evaluate anti -oxidant activities and protective effect against oxidatve damage, DPPH radical scavenging activity and lipid peroxidation inhibitory activity were measured among methanol extracts prepared from natural medicinal plants. Fourteen natural medicinal plants which were reported to have anti -oxidative or anti-inflammatory effects were selected based on our previous report. In addition to the total methanol extracts, n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions were prepared from each total extract. DPPH radical scavenging assay was performed against 14 total extracts and all samples showed dose-dependent activities in various extent. Among those, 6 samples, methanol extracts of Euryale ferox, paeonia suffruticosa, Areca catechu var. dulcissima, Cinnamomun cassia, Alpinia katsumadai and Betula platyphlla var. japonica showed IC$\sub$50/ value lower than 6.0 $\mu\textrm{g}$/ml. The highest DPPH radical scavenging activity was found in ethylacetate fraction of paeonia suffruticosa with IC$\sub$50/ value of 1.1 $\mu\textrm{g}$/ml. Analysis of lipid peroxidation inhibitory activity on hydrogen peroxide-induced oxidative damage in Chinese hamster lung fibroblast (V79-4) cells revealed that the highest inhibitory effect was observed in methanol extract of Betula platyhpylla var. japonica. Lipid peroxidation inhibitory activity was observed as a dose-dependent manner in all samples used in this study. Among fraction samples, ethylacetate fraction of Alpinia katsumadai had the strongest inhibitory activity with IC$\sub$50/ value of 0.9 $\mu\textrm{g}$/ml.

싸리버섯 추출물의 항돌연변이성 및 암세포 성장 억제 효과 (Antimutagenic and Anticancer Effects of Ramaria botrytis(Fr.) Rick Extracts)

  • The inhibitory effect of Ramaria botrytis on the mutagenicity in Salmonella assay and cytotoxicity on cancer cell were studied. Ramaria botrytis methanol extracts showed antimutagenic effects of 60~90% on B(a)P and AFB1 in S. typhimurium TA98 and TA100. These extracts showed 73~85% antimutagenicity on TA100 against MNNG. The methanol extracts with strong antimutagenic activities were further fractionated by ethylacetate and water, the ethylacetate fraction were found to be stronger antimutagen icity against MNNG than water fraction. Ramaria botrytis methanol extracts and ethylacetate fraction revealed the highest cytoxicity against HT 29 human colon adenocarcinoma cells in which cell growth was inhibited by 57~88% and 69~94% at 0.25~1.0mg/ml, respectively. These methanol extract and ethyl acetate fraction exhibited 53~79% and 66~87% inhibition against HepG2 human hepatocarcinoma cells, respectively. But water fraction showed only 10~24% inhibition. However, these extract and fractions did not show cytotoxic effect against human chang liver cells. From these results, it is considered that Ramaria botrytis has stronger antimutagenic and anticancer effects in vitro.

Extraction and Bioassay of Allelochemicals in Jerusalem Artichoke

  • Helianthus tuberosus has been known to inhibit the growth of weeds and other plants sharing its habitat. This study was conducted to identify the allelochemicals of Helianthus tuberosus which were extracted with water and solvents. Aqueous extracts of leaf, stem, root, tuber and tuber peel of Helianthus tuberosus except tuber did not show significant differences in phytotoxicity to alfalfa seedlings. It was considered that Helianthus tuberosus contained fewer or less potential water-soluble substances that were toxic to alfalfa. Methanol extract of leaves of Helianthus tuberosus was sequentially partitioned in increasing polarity with n-hexane, ethylacetate and n-butanol. Each extract had a yield of 148, 12, 15.7 and 9.5g, respectively. Inhibitory effects on germination of alfalfa seeds treated with four fractions were not significantly different. But the significant reductions on hypocotyl length were observed for all the solvent extracts. Among the four fractions, the ethylacetate fraction showed the most significant inhibition effect on bioassay with alfalfa. Further separation of the active ethylacetate fraction by open column chromatography led to the 25 subfractions. In bioassay of each sub-fraction with alfalfa seeds, sub-fraction No. 13 showed the most inhibitory effect on seedling growth. $^1$H NMR and gas chromatography-mass spectrometry analysis revealed that sub-fraction No. 13 was the mixture of straight-chain saturated fatty acids.

황벽나무 추출물의 유기질 문화재 오염균에 대한 항균성 및 항산화 활성 (Antifungal Activities on Organic Heritage Fungi and Antioxidative effect of Phellodendron amurense Extractives)

  • Antifungal and antioxidative activities of Phellodendron amurense extracts were investigated for use as a natural preservative. After separation of P. amurense into phloem, xylem, leaf and fruit each part was subjected to methanol extraction. Each MeOH extract was further fractionated with several solvents(n-hexane, methylene chloride and ethylacetate). Among the methanol extracts, extracts of phloem and leaf inhibited effectively the growth of mold fungi and rot fungi, respectively. Especially, ethylacetate fraction from phloem showed the highest growth inhibitory effects against fungi tested, such as P. citreonigrum H3, P. toxicarium H4, P. corylophilu H5, A. clavatus, P. osteatus, S. commune, and G. lucidum. The fractions of fruit, which had lower antifungal activities mostly than those of phloem, strongly inhibited rot fungi such as G. lucidum, T. versicolor, and T. palustris. Compared to ferulic acid which is well known antioxidant, ethylacetate fraction of fruit showed high antioxidative activities on concentration of 1 to $100{\mu}g/m{\ell}$ in DPPH radical scavenging activity. As a result, antifungal and antioxidative activities of P. amurense suggest that its extract and fraction have a possibility as conservative of cultural heritage because it might get conservation effects against deteriorating microorganisms of cultural heritage.

호장근의 사염화탄소로 유도된 지질과산화 저해활성 (Antilipoperoxidant Activity of the Root Polygonum cuspidatum on Carbon Tetrachloride Induced Hepatotoxicity)

  • The root of Polygonum cuspidatum (Polygonaceae) has been used as treatments of hyperlipidemia, dermatitis, gonorrhea, favus athlete's foot, inflammation in traditional medicine. In order to examine anti-lipidperoxidation activity, hexane, EtOAc, BuOH and water fractions of its methanol extract were administered to carbon tetrachloride intoxicated rats. Ethylacetate fraction exhibited antilipidperoxidative effect on liver lipid homogenate and the radical scavenging effect on DPPH. Serum transaminase, AlP, triglyceride and total cholesterol contents significantly decreased by administrations of ethylacetate fraction.

민들레로부터 항균성 화합물의 분리 및 동정 (Isolation and Identification of Antimicrobial Compound from Dandelion(Taraxacum platycarpum D.))

  • 최근 식품 산업의 발달로 장기간 식품 보존을 위하여 합성 보존제의 사용이 크게 증가하고 있는 실정이다. 그러나, 인체에 대한 유해성이 문제로 대두되면서 이를 대체할 천연 보존제의 개발이 절실히 요고되고 있는 실정이다. 따라서 본 연구는 우리나라 야산에서 쉽게 구할 수 있는 구황 식물인 민들레를 메탄올과 여러 용매로 추출하여 식품부패 미생물에 대한 항균력을 실험하고 그 항균 활성 물질을 분석하여 다음과 같은 결론을 얻었다. 민들레의 메탄올 추출물은 $2000\mu\textrm{g}/ml$ 농도에서 s. aureus를 완전히 억제하였고, L. monocytogenes는 98.43% 억제하였다. 그리고 민들레의 ethylacetate 분획물은 $1000\mu\textrm{g}/disc$ 농도에서 5종의 모든 시험균주에 대하여 clear zone을 형성하여 가장 높은 항균력을 보였다. 민들레의 ethyl­-acetate 분획물을 silica gel column chromatography와 TLC를 통해 분리하여 얻은 fraction을 5가지 시험균주에 대한 항균 실험 결과 3번째 3rd fraction의 항균력이 가장 높아 $250\mu\textrm{g}/disc$ 농도에서 E. coli를 제외한 모든 균주에 대하여 14mm 이상의 clear zone을 나타내었다. 항균력을 보인 민들레의 ethylacetate 분획의 3번째 3rd fraction을 먼저 HPLC로 분리하여 얻은 peak II로부터 $^{l}H-NMR$ 및 GC-MS로 분석한 결과 benzoic acid로 동정 되었다.
