독성물질 검출을 위한 Plasmid Vector 개발

  • After DNA damage, umuDC is the only SOS operon that must be induced to promote SOS mutagenesis in Escherichia coli. The recombinant plasmid pBC401 and pBC402 were constructed to fuse the lac structural genes with promoter region of umuDC operon to induce the expression of lacZ gene by DNA damage. We transformed the plasmid pBC401 and pBC402 into E. coli MC1061, lacZ deleted strain and determined the activity of $\beta$-galactosidase for various mutagen; UV, mitomycin C (MMC), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), 4-nitroqunoline-1-oxide (NQO), ethyl methanesulfonate (EMS). The $\beta$-galactosidase activities of PBC401 and pBC402 for UV, MMC, and NQO were increased in proportion to expression time until 3 hours thereafter, the activities were constant or slightly decreased. The activities for MNNG and EMS were not so high as for UV, MMC, and NQO. When MNNG and EMS were treated, $\beta$-galactosidase activity of pBC402 was slightly lower than pBC401 but when UV, MMC, and NQO were treated in pBC402, $\beta$-galactosidase activity was slightly higher than in pBC401. Therefore, the pBC402 was better than the pBC401 in terms of sensitivity for frameshift mutagen. We suggest that the plasmid pBC401 and pBC402 are easy to detect mutagens which cause frameshift mutation rather than point mutation.

EMS 처리에 관한 콩 근류착생 및 기타 형질의 돌연변이 출현 양상 (Mutagenesis of Nodulation and Other Growth Characters with Ethyl Methanesulfonate in Soybean)

  • 황금콩과 백운콩에 30mM 및 50mM EMS를 각각 종자 1립 당 1ml 수준으로 실온에서 6시 간 처리하여 얻은 $M_1종자를 파종한 다음, 포장출현율 및 $M_1개체들의 형태적 특성의 분포 양상을 검토하고,$M_2세대에서 근류착생 및 기타 돌연변이 출현 양상을 조사한 결과를 요약하면 다음과 같다. 1.$M_1 종자의 포장출현율은 백운콩이 황금콩보다 비교적 높았으나 EMS 농도 처리간 차이는 없었으며, 엽록소 결핍 개체 발생율은 평균 0.7%였다. 2. 수확시 $M_1개체들의 경장, 개체당 협수 및 립수는 무처리개체에 비하여 감소되는 경향이었으며, EMS 처리 농도간 분포차이는 크지 않았다. 3. $M_2 세대에서 엽록소 결핍 개체 출현율에 의하여 판별된 돌연변이 유기율은 평균 2.2%이었으며, 돌연변이가 유기된 $M_1종자는 chimera 현상을 나타내었으며 돌연변이 세포의 비율은 5.3~84.2%로 나타났다. 4. EMS 처리에 의하여 근류착생이 많은 MA48계통과 근류착생이 거의 안되는 MD69계통을 선발하였는데 MA48계통은 엽연소함량이 많았고 MD69 계통은 엽록소 결핍 현상을 나타내었다.

EMS 처리에 의한 좀비비추[Hosta minor (Baker) Nakai]의 엽색변이 품종 '황나래' 육성 (New Cultivar 'Hwangnarae' of Leaf Color Variegated Hosta minor Developed by EMS Treatment)

  • '황나래(Hwangnarae)'는 국립수목원에서 육종한 품종으로 돌연변이 유기 화학물질인 EMS를 활용하여 돌연변이 개체를 유도하였다. 변이체 선발을 통해 잎 가장자리에 황록색의 테두리가 들어간 무늬종을 선발하였다. 이렇게 선발된 개체는 영양번식을 통하여 개체 수를 늘렸다. 2013년부터 2018년까지 6년간 특섬검정을 실시하여 잎 가장자리에 황록색 무늬가 균일하게 발현되는 것을 확인한 후 2019년 최종 선발되었다. '황나래 (Hwangnarae)'는 지피용이나 분화용으로 활용적 가치가 매우 높다.

Transcriptome Profiling and Characterization of Drought-Tolerant Potato Plant (Solanum tuberosum L.)

  • Potato (Solanum tuberosum L.) is the third most important food crop, and breeding drought-tolerant varieties is vital research goal. However, detailed molecular mechanisms in response to drought stress in potatoes are not well known. In this study, we developed EMS-mutagenized potatoes that showed significant tolerance to drought stress compared to the wild-type (WT) 'Desiree' cultivar. In addition, changes to transcripts as a result of drought stress in WT and drought-tolerant (DR) plants were investigated by de novo assembly using the Illumina platform. One-week-old WT and DR plants were treated with -1.8 Mpa polyethylene glycol-8000, and total RNA was prepared from plants harvested at 0, 6, 12, 24, and 48 h for subsequent RNA sequencing. In total, 61,100 transcripts and 5,118 differentially expressed genes (DEGs) displaying up- or down-regulation were identified in pairwise comparisons of WT and DR plants following drought conditions. Transcriptome profiling showed the number of DEGs with up-regulation and down-regulation at 909, 977, 1181, 1225 and 826 between WT and DR plants at 0, 6, 12, 24, and 48 h, respectively. Results of KEGG enrichment showed that the drought tolerance mechanism of the DR plant can mainly be explained by two aspects, the 'photosynthetic-antenna protein' and 'protein processing of the endoplasmic reticulum'. We also divided eight expression patterns in four pairwise comparisons of DR plants (DR0 vs DR6, DR12, DR24, DR48) under PEG treatment. Our comprehensive transcriptome data will further enhance our understanding of the mechanisms regulating drought tolerance in tetraploid potato cultivars.
