Antimicrobial Effect of Cutellaria baicalensis George Extracts on Food-Borne Pathogens (황금(Cutellaria baicalensis George) 추출물의 식중독성 미생물에 대한 항균효과)

  • This study was performed to investigate the antimicrobial effect of the Cutellaria baicalensis George extracts against food-borne pathogens. First, the Cutellaria baicalensis George was extracted with methanol at room temperatures, and fractionation of the methanol extracts from Cutellaria baicalensis George was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Cutellaria baicalensis George extracts was determined using a paper disc method against food-borne pathogens and food spoilage bacteria. The ethyl acetate extracts of Cutellaria baicalensis George showed the highest antimicrobial activity against Staphylococcus aureus and Shigella dysenteriae. The synergistic effect has been found in combined extracts of Cutellaria baicalensis George and Portulaca oleracea as compared to each extracts alone. Finally, the growth inhibition curve was determined using ethyl acetate extracts of Cutellaria baicalensis George against Staphylococcus aureus and Shigella dysenteriae. The ethyl acetate extract of Cutellaria baicalensis George showed strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. The 4,000 ppm of ethyl acetate extract from Cutellaria baicalensis George retarded the growth of S. aureus more than 24 hours and Shigella dysenteriae up to 36 hours. The ethyl acetate extracts of Cutellaria baicalensis George has been shown the antimicrobial effect against Staphylococcus aureus and Shigella dysenteriae.

Study on Antioxidant Effects of Fractional Extracts from Ligularia stenocephala Leaves (곤달비(Ligularia stenocephala) 잎 분획물의 항산화활성 평가)

  • Ligularia stenocephala leaves were subjected to extraction by using several solvents with different polarities, which were then investigated for their antioxidant and antimicrobial activities. Extraction yield of ethanol extract of Ligularia stenocephala was 21.36%. The water fraction showed the highest extraction yield of 60.35%, followed by the n-butanol, n-hexane, and ethyl acetate fractions. Total polyphenolic content was the highest (254.00 mg/g GAE) in the ethyl acetate fraction of Ligularia stenocephala extract. The ethyl acetate fraction had $IC_{50}$ values of 0.28 mg/mL for DPPH radical scavenging, and 96.$67{\pm}0.09%$ for ABTS radical scavenging activity. Further, FRAP value was significantly higher in the ethyl acetate fraction. The ethyl acetate fraction showed antimicrobial activities against B. cereus, and the diameter of the zone of inhibition was 10.2 mm at 5 mg/disc. These results suggest that the ethyl acetate fraction of Ligularia stenocephala possesses the antimicrobial activities against B. cereus, and its high antioxidant activity could be applicable to food additives and as a natural cosmetic ingredient.

Anti-oxidative Effects of Ethyl acetate Fraction of Saururus chinensis in Caenorhabditis elegans (삼백초 Ethyl acetate 분획물의 예쁜꼬마선충 내의 항산화 효과)

  • Saururus chinensis (Lour.) Baill. (Saururaceae) has been used as a traditional medicine for the treatment of dysuria, leukorrhea, eczema, jaundice and ascites in Korea, China and Japan. Ethanol extract of S. chinensis was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions the ethyl acetate soluble fraction showed the most potent DPPH radical scavenging and superoxide quenching activities. To verify antioxidant activities of ethyl acetate fraction, we checked the activities of superoxide dismutase (SOD) and catalase, and intracellular ROS level and oxidative stress tolerance in Caenorhabditis elegans. Furthermore, to see if increased stress tolerance of worms by treating of ethyl acetate fraction was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain. Consequently, ethyl acetate fraction elevated SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, ethyl acetate fraction-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.

Nitric Oxide Production Inhibitory Effect and Antibacterial Activity of the Extract and Fractions from Paeoniae Radix (작약 메탄올 추출물 및 분획물의 Nitric Oxide 생성 억제 효과와 피부질환 원인균에 대한 항균활성)

  • In this study, we investigated antibacterial activity and nitric oxide production inhibitory effect of the methanol extract and its fractions from Paeoniae Radix. In antibacterial activity by the disc diffusion assay against S. aureus, S. epidermidis and P. aeruginosa, the ethyl acetate fraction showed stronger antibacterial activity than other fractions and the extract. Moreover, the ethyl acetate fraction showed strong nitric oxide (NO) production inhibitory effect in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell. However, in NO scavenging ability, the chloroform fraction was higher than the other fractions and the extract. In the lactate dehydrogenase (LDH) assay against RAW 264.7 cell, the extract and fractions were exhibited normal LDH release level as nontoxic result without the ehtyl acetate fraction of 100 ${\mu}g/ml$. As a result, the ethyl acetate fraction and chloroform fraction of the methanol extract from Paeoniae Radix could be applicable to functional materials for antibacterial and anti-inflammatory related fields, respectively.

Antifungal and Antioxidant Activities of Volatile Organic Compounds Generated During the Drying Process of Chamaecyparis obtuse (편백나무 건조 중 발생하는 휘발성 유기화합물의 항진균 및 항산화 활성)

  • In this study, we evaluated antifungal and antioxidant activities of condensed liquid of volatile organic compounds generated during the drying process of Chamaecyparis obtuse. Condensed liquid of volatile organic compounds were extracted with hexane and ethyl acetate, respectively. The extracts were used for antifungal activity at range from 100 to 500 mg/L Antifungal activity of extracts increased as the extracts concentration increased, the activity against tested dermatophytes was high at 500 mg/L. Among extract solvents, antifungal activity was higher at hexane extract than the ethyl acetate. Especially, the antifungal activity against Trichophyton rubrum was higher than other tested dermatophytes. Antioxidant activity was 90% above 25 mg/L regardless of the extractive solvents, while the hexane extract showed high antioxidant activity below 25 mg/L. Major compounds of hexane extract were torreyol, alpha-cadinol, and tau-cadinol. However, major compounds of ethyl acetate extract were alpha-amorphene, alpha-cadinol, and gamma-cadinene.

  • Antimicrobial activity of bamboo leaves extract on microorganisms related to kimchi fermentation was investigated. Bamboo leaves were extracted with several ogranic solvents such as methanol, acetone, ethyl ether, and ethyl acetate. The bamboo extract with ethyl acetate showed the strongest antimicrobial activity among them. Strong antimicrobial activities of the extract against microorganisms related to kimchi fermentation and food spoilage indicated that the extract had a wide range of antimicrobial spectrum. The antimicrobial activity was especially strong against Brettanomyces custersii, Klebsiella oxytoca, Pichia membranaefaciens which cause kimchi softening. In addition, the antimicrobial activity of bamboo leaves extract was higher than that of 0.5% and 1.0% sorbic acid, and moreover it was stronger in pH 5 compared to pH 7.

A Study on the Effect of the Dendropanax Mobifera Extract on Anti-Hypertensive (황칠나무 추출물이 고혈압에 미치는 영향에 관한 연구)

  • This study examined the effects of an extract of Dendropanax morbifera on blood pressure, Angiotensin II, Angiotensin Converting Enzyme, Aldosterone, and lipid levels of spontaneously hypertensive rats. The groups were as follows: Control group, Hypertension control group, Water extract treated group, Ethanol extract treated group, n- hexane fraction treated group, Ethyl acetate fraction treated group, n- butanol fraction treated group, and Water fraction treated group. The blood pressure, and Angiotensin II, and Angiotensin Converting Enzyme, and Aldosterone levels were lower in the Ethyl acetate fraction treated group than in the hypertension control group. The change in blood pressure was lower in the Water extract treated group, Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group. The concentration of Angiotensin II was lower in the Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group(p<0.05). The level of Angiotensin Converting Enzyme was lower in the Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group. The concentration of Aldosterone was lower in the Ethyl acetate fraction treated group, and n- butanol fraction treated group than the hypertension control group(p<0.05). In addition, the concentration lower in the Water extract treated group, Ethanol extract treated group, n- hexane fraction treated group than the hypertension control group. Overall, the effect of the Dendropanax morbifera extracts might be useful as anti-hypertensive, and functional food agents.

Antioxidative Activity of Rooibos Tea(Aspalathus linearis) Extracts (용매에 따른 Rooibos Tea(Aspalathus lineais) 추출물의 항산화 효과)

  • Total phenolic compounds and antioxidative activities of rooibos tea(Aspalathus linearis) fractions were studied. Three extracts, using hexane, ethyl acetate, and ethanol, were prepared. Total phenolic compounds were 3069.3 mg/100 g extract in the hexane fraction, 18604.4 mg/100 g extract in the ethyl acetate fraction, and 13458.8 mg/100 g extract in the ethanol fraction. Levels of vanillic acid, caffeic acid, syringic acid, 4-coumaric acid, and ferulic acid were analyzed by RP-HPLC, and totals of 3452.6 and 3156.1 mg/100 g of extract were found in the ethanol and ethyl acetate fraction, respectively. In the DPPH assay, the ethanol fraction(82.2% of contol) and the ethyl acetate fraction(78.9%) showed the highest free radical scavenging capacities. The induction period of each tea fraction in the fish oil rancimat assay was measured. When 500 ppm of the ethanol fraction was applied, a 1.19 h induction period was observed. This was 2-fold greater than the induction period of the control.

Involvement of Heme Oxygenase-1 Induction in the Neuroprotective Activitiy of Extract of Siegesbeckia Herba in Murine Hippocampal HT22 Cells (희렴 추출물의 Heme Oxygenase-1 발현을 통한 생쥐 해마 유래 HT22 세포 보호효과)

  • Siegesbeckia Herba is known to have anti-oxidant, anti-inflammatory, anti-allergic and anti-tumor. The objective of this study is to explore the neuroprotective effect of Siegesbeckia Herba against glutamate-induced oxidative stress in mouse hippocampal HT22 cells. Siegesbeckia Herba 70% ethanol extract and solvent fractions have the potent neroprotective effects on glutamate-induced nerotoxicity by induced the expression of heme oxygenase (HO)-1 in the mouse hippocampal HT22 cells. Especially, ethyl acetate fraction showed higher protective effect. In HT22 cell, Siegesbeckia Herba ethyl acetate fraction makes the nuclear accumulation of Nrf2. Further, we found that treatment with c-JUN N-terminal kinase (JNK) inhibitor (SP600125) reduced Siegesbeckia Herba ethyl acetate fraction induced HO-1 expression and Siegesbeckia Herba ethyl acetate fraction also increased JNK phosphorylation. In conclusion, the ethyl acetate fraction of 70% ethanol extract of Siegesbeckia Herba significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2 and JNK pathway in mouse hippocampal HT22. Taken together these finding suggest that Siegesbeckia Herba ethyl acetate fraction good source for taking active compounds and may be a potential therapeutic for brain disorder by targeting the oxidative stress of neuronal cell.
