감자의 지방질(脂肪質) 성분(成分)에 관한 연구(硏究) - 제(第) 3 보(報) : 유리(遊離) 및 결합(結合) 지질(脂質)중의 극성(極性) 지질(脂質)의 조성(組成)에 관하여 - (Studies on the Lipid Components of Potato Tubers - III. Composition of Polarlipids in Free and Bound Lipids -)

  • 시험 재배한 4가지 감자 품종에서 추출한 유리(遊離) 및 결합(結合) 지질(脂質)중의 당지질(糖脂質) 및 인지질(燐脂質)을 구성하는 지방질을 박층 크로마토그래피, 가스-액체 크로마토그래피에 의하여 분리 정량하여 비교한 결과 다음과 같은 결론을 얻었다. 1. 유리(遊離) 및 결합(結合) 지질(脂質) 중의 당지질(糖脂質)로는 esterified steryl glgcoside, monogalactosyl diglyceridc, steryl glycoside, digalactosyl diglyceride, trigalactosyl diglyceride 등의 성분을 분리 동정 하였고, 유리(遊離) 지질(脂質) 중의 당지질(糖脂質)에는 esterified steryl glycoside의 함량이 총 지방질에 대하여 5.8 %로 가장 많았으나 결합(結合) 지질(脂質) 중에는 trigalactosyl diglyceride가 6.1 %로 가장 많았다. 그리고 유리(遊離) 및 결합(結合) 지질(脂質) 중의 monogalactosyl diglyceride의 함량은 서로 비슷 하였으나 esterified steryl glycoside의 함량은 결합(結合) 지질(脂質)보다 유리(遊離) 지질(脂質) 중에 많은 편이었고, steryl glycoside, digalactosyl diglyceride 및 trigalactosyl diglyceride의 함량은 반대로 유리(遊離) 지질(脂質)보다 결합(結合) 지질(脂質)중에 많은 편이었다. 2. 유리(遊離) 및 결합(結合) 지질(脂質) 중의 인지질(燐脂質)로 phosphatidyl ethanolamine, phosphatidyl glycerol, phosphatidyl choline, phosphatidyl inositol의 성분을 분리 동정하였으나 phosphatidyl glycerol은 유리(遊離) 지질(脂質) 중에서는 존재하지 않았다. 유리(遊離) 지질(脂質) 중에는 phosphatidyl choline의 함량이 총 지방질에 대하여 3.3 %로 가장 많았으나 결합(結合) 지질(脂質) 중에는 phosphatidyl ethanolamine이 14.9%로 가장 많았다. 그리고 그 외의 각종 인지질(燐脂質)의 성분은 모두 유리(遊離) 지질(脂質)보다 결합(結合) 지질(脂質)중에 그 함량이 많았다. 3. 유리(遊離) 및 결합(結合) 지질(脂質) 중의 당지질(糖脂質)을 구성하는 주변 지방산 조성도 총(總) 유리(遊離) 및 결합(結合) 지질(脂質)을 구성하는 지방산 조성과 같았고 다만 유리(遊離) 지질(脂質) 중의 당지질(糖脂質) 중에는 총(總) 유리(遊離) 지질(脂質)보다 palmitic acid의 함량은 많았고 linoleic acid의 함량은 적었다. 또 당지질(糖脂質)을 구성하는 각 지방질의 주된 지방산 조성은 총(總) 당지질(糖脂質)을 구성하는 지방산 조성과 그 패턴이 거의 같았으나 다만 esterified steryl glycoside와 steryl glycoside 중에는 palmitic acid의 함량이 매우 많은것이 특이(特異)하였다. 4. 유리(遊離) 및 결합(結合) 지질(脂質) 중의 인지질(燐脂質)을 구성하는 주된 지방산 조성도 총(總) 유리(遊離) 및 결합(結合) 지질(脂質)을 구성하는 지방산 조성과 같았고 다만 유리(遊離) 및 결합(結合) 지질(脂質) 중의 인지질(燐脂質)에는 총(總) 유리(遊離) 및 결합(結合) 지질(脂質)보다 palmitic acid의 함량이 많았고 linoleic acid의 함량은 적었다. 또 인지질(燐脂質)을 구성하는 각 지방질의 주된 지방산 조성은 총(總) 인지질(燐脂質)을 구성하는 지방산 조성과 그 패턴이 거의 같았으나 phosphatidyl inositol 중에는 palmitic acid의 함량이 많았다.

함침농도와 CO2 가스 유입농도에 따른 활성탄의 흡착특성 (Adsorption Characteristics of Activated Carbons According to Impregnation Concentrations and Inlet CO2 Gas Concentrations)

  • The adsorption characteristics of $CO_2$ gas on impregnated activated carbons with MEA (Mono-ethanolamine) and AMP (2-Amino 2-methyl 1-propanol) were studied to improve the adsorption ability of $CO_2$ gas on activated carbon. The equilibrium adsorption capacity of $CO_2$ gas was increased by increment of impregnation concentration up to 40 %, but decreased above 50 %. The adsorption capacity of activated carbon impregnated with AMP was higher than activated carbon impregnated with MEA. The breakthrough was fast according to increment of inlet concentration of $CO_2$ gas.

Binding Subsites In the Active Site of $Zn^{2+}$-Glycerophosphocholine Cholinephosphodiesterase

  • The properties of binding sites in the active site of $Zn^{2+}$-glycerophosphocholine cholinephosphodiesterase were examined using substrates and inhibitors of the enzyme. Phosphodiesterase hydrolyzed p-nitrophenylphosphocholine, p-aminophenylphosphocholine, and glycerophosphocholine, but did not hydrolyze either acylated glycerophosphocholine or bis (p-nitrophenyl)phosphate, suggesting a size limitation for interaction with a glyceryl moiety-binding subsite. The hydrolysis of p-nitrophenylphosphocholine was competitively inhibited by glycerophosphocholine and p-aminophenylphosphocholine, while glycerophosphoethanolamine was a weak inhibitor. The enzyme was also inhibited by choline, but not by ethanolamine. Thiocholine, a much more potent inhibitor than choline, was more inhibitory than cysteamine, suggesting a strict specificity of an anionic subsite adjacent to a $Zn^{2+}$ subsite. Of all oxyanions tested, the tellurite ion was found to strongly inhibit the enzyme by binding to a $Zn^{2+}$ subsite. The inhibitory role of tellurite was synergistically enhanced by tetraalkylammonium salts, but not by glycerol. Deactivation of the enzyme by diethylpyrocarbonate was partially protected by choline, but not by glycerophosphate. It is suggested that the active site of phosphodiesterase contains three binding subsites.

ZnO Wire의 형상 제어를 통한 전계 방출 특성 연구

  • ZnO는 수열합성법을 사용하여 저비용으로 일차원 형태의 잘 정렬된 nanowire의 제작이 가능하며 높은 화학적, 열적 안정성을 가져 전계 방출 소자로써 많은 연구가 진행되고 있다. 본 연구는 수열합성법에 사용되는 ZnO 전구체(Zinc Nitrate, HMTA, PEI & Ammonium Chloride), 안정제(Ethanolamine)의 농도에 따른 ZnO의 형상 변화와 이에 따른 전계방출 특성을 비교 분석하였다. 전구체의 농도 변화에 따라 ZnO Film, Rod, Tip과 같은 형상 변화를 확인 할 수 있었으며, 안정제를 사용함으로써 고종횡비를 가지는 ZnO Tip을 제작하였다. 제작된 ZnO 에미터의 전계방출특성을 고진공 챔버에서 측정하였고, 에미터 형상이 전계방출에 미치는 영향을 비교 분석하였다.

복합아민 적용에 따른 원전 2차 계통 부식생성물 거동평가 (Evaluation of Corrosion Product Behavior in NPP Secondary System with Complex Amine)

  • The aim of the study was to evaluate the water treatment of pressurized water reactor secondary side by the mixed amine of ammonia and ethanolamine, from the standpoint of corrosion control, as compared with all volatile treatment of ammonia. The pressurized water reactor systems have switched a secondary side pH control agent to minimize the corrosion in the moisture separator/reheater and feedwater heater systems and the transport of corrosion products into steam generator. As results of field test, pH was increased in the steam generator and the wet steam area of moisture separator/reheater and the concentration of Fe were decreased by more than 50% as compared with water treatment of ammonia.

Encapsulation of Plasmid DNA in Pegylated Liposome

  • The purpose of the study was to prepare the pegylated liposome carrying plasmid DNA with optimal encapsulation efficiency. Plasmid DNA (pCEP4 clone 790, 10.6 kb) was entrapped in the pegylated liposome composed of neutral lipid, POPC (l-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine), cationic lipid, DDAB (dimethyl dioctadecyl ammonium bromide) and anionic lipids, DSPE-PEG 2000 (distearoyl phosphatidyl ethanolamine polyethylene glycol 2000) and DSPE-PEG 2000-maleimide by freezing/thawing method. Free plasmid DNA was separated from the encapsulated one by Sepharose CL-4B column chromatography. The DNA amount encapsulated into the pegylated liposome was increased as cationic lipid concentration, initial amount of plasmid DNA and total lipid amount were increased.

Pseudomonas sp. CH-414에 의한 Eicosapentaenoic Acid 생산 (Production of Eicosapentaenoic Acid by Pseudomonas sp. CH-414)

  • The identification of eicosapentaenoic acid (EPA) by Pseudomonas sp. CH-414 were investigated under the optimal culture conditions. The maximum dry cell weight and phospholipid production showed about 10 mg/ml and 0.6 mg/ml, respectively, at the 48 hr cultivation. The phospholipid form produced by Pseudomonas sp. CH-414 was elucidated as a phosphatidyt ethanolamine by thin layer chromatography. EPA was contained about 15% in the. extractable lipid, and the amount of EPA was about 83 $\mu $g/ml from the culture broth. Also myristic, palmitic, palmitoleic, stearic and oleic acids were identified with gas chromatography.

Prolonged Systemic Delivery of Streptokinase Using Liposome

  • To prolong the biological half-life of streptokinase, a thrombolytic agent, streptokinase-bearing liposome with and distearolyphosphatidyl ethanolamine-N-poly (ethylene glycol) 2000 (DSPEPEG 2000) was prepared and evaluated. Streptokinase-bearing liposomes composed of distearolyphosphatidylcholine (DSPC), cholesterol and cholesterol-3-sulfate with DSPE-PEG 2000 was prepared by the freeze-thawing method and administered via femoral vein to rats (15000 IU/kg). The activity of streptokinase in plasma was determined by the method based on the amidolytic activity of streptokinase-plasminogen complex. Pharmacokinetic parameters of streptokinase incorporated in liposomes were compared with those of streptokinase alone. The $T_{1/2}$ and $AUC_\infty$ streptokinase incorporated in DSPC-PEG liposome increased 16.3- and 6.1-fold, respectively, compared with those of streptokinase alone. Streptokinase-bearing long-circulating liposome could increase the circulation time of streptokinase in blood and expect longer thrombolytic activity compared with streptokinase alone.

  • The effect of ginseng glycosides and their aglycones on the thermodynamic characteristics of membranes from dipalmitoylphosphatidylcholine (DPPC) was investigated. Total saponins (TS) from Korean red ginseng, Panax ginseng C.A. Meyer, interacted with the Eel Phase of lipid in the Polar region and did not penetrate the deeper glycerol backbone of lipid molecule. From the all investigated components of TS (aglycons and ginsenosides), only 20-(S)-panaxadiol (PD) had an effect similar to TS. High concentration of TS penetrated in hydrophobic Cl-C8 region. The presence of cholesterol did not influence the interaction of TS with DPPC. An elimination of transition, however, took place at 10~100 $\mu\textrm{g}$/ml of TS. DPPC had a low ability to interact with cholesterol (CHL) as compared with other lecithins except ethanolamine. From our results, only TS and PD, at high concentrations (100 mol%), influenced the phase transition of mixture of DPPC:CHL.

  • The 6,7-dichloroquinolone-5,8-dione(I) was reacted with ethyl acetoacetate in the presence of sodium ethoxide to yield 6-(${\alpha}$-acetyl-${\alpha$-ethoxycarbonyl-methyl)-7-chloro-quin oline-5,8-dione(II). When this compound II was reacted with some alkylamine (methylamine, ethylamine, propylamine, isopropylamine, cyclopropylamine, methoxyethylamine, ethanolamine, benzylamine, furfurylamine), 1N-alkyl-2-methyl-3-ethoxycarbonyl-pyridino(2,3f)-indole-4,9-dione(IIIa-i) were obtained via intramolecular cyclization.
