Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향 (Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

Ethanol 급여 흰쥐의 심장 지질과산화에 미치는 Methionine의 영향 (Effect of Methionine on Heart Lipid Peroxidation in Rat with Alcohol Administration)

  • This study was designed to investigate the effects of methionine(Met) on the activities of heart lipid peroxidation related enzymes in ethanol administrated rats. Male Sprague-Dawley rats were fed on diets containing one of the three levels of Met(0%, 0.3%, 0.9% of kg diet) and ethanol(2.5g/kg of body weight) was administrated as 25v/v% ethanol to ethanol treated groups orally. The rats were sacrificed after 5 and 10 weeks of feeding. Xanthine oxidase(XO) and catalase activities increased with ethanol administration and those activities were higher n Met excessive and deficiency group than those of Met normal group at 5 and 10 weeks dieting. Superoxide dismutase(SOD) activity in heart decreased significantly in Met deficiency and Met excessive group as compared to that of control. Glutathione peroxidase(GSH-Px) activity in heart significantly decreased in Met deficiency group as compared to that of Met excessive and normal group. Glutathione S-transferase(GST) activity of heart tissue significantly increased by ethanol administration. Glutathione(GSH) content in heart decreased with ethanol administration and shwoed no significant differences with Met levels. Ethanol administration increased the content of lipid peroxide(LPO).

참취 분말이 에탄올을 투여한 흰쥐의 항산화계에 미치는 효과 (The Effect of Powdered Herb of Aster scaber Thunb. on Antioxidant System in Ethanol-Treated Rats)

  • In vitro 항산화 활성이 확인된 참취가 생체 내에서도 활성을 나타내는 지를 확인하기 위해 대조군과 5%및 10% 참취 분말을 첨가한 식이를 4주간 횐쥐에 투여하면서 마지막 1주간 알콜로 산화적 스트레스를 유발한 후 희생시킨 흰쥐의 체중 증가량과 함께 체중에 대한 장기의 상대 중량, 간, 신장에서의 지질과산화, 항산화효소 활성, glutathione 및 혈청 알부민의 함량을 분석하였다. 4주간의 사육 후 체중 증가량은 대조군, 5% 참취첨가군 그리고 10% 참취첨가군의 순으로 높았으나 유의적인 차이는 없었으며 체중에 대한 상대적인 장기 중량 중에서 간, 비장은 대조군에 비해 참취 첨가군의 경우 더 낮은 비율을 나타내었다. 간에서 지질과산화로 생성되는 TBARS 함량은 대조군(66 $\mu\textrm{g}$)에 비해 5%(45 $\mu\textrm{g}$) 및 10% 참취 분말 식이군(35 $\mu\textrm{g}$)에서 참취 첨가량이 증가할수록 낮았다. Mn-SOD와 catalase활성은 참취 첨가군들에서 대조군보다 낮았으나 Cu, Zn-SOD와 glutathione peroxidase 활성 및 glutathione 함량은 실험군들간에 유의적인 차이를 나타내지 않았다. 신장에서는 Mn-SOD와 glutathione peroxidase 활성이 대조군보다 참취 첨가군에서 유사하거나 낮은 경향을 나타내었으나 Cu, Zn-SOD와 catalase의 활성 및 TBARS, glutathione의 함량은 실험군들간에 유의적인 차이를 보이지 않았다. 한편, 혈청 알부민의 함량은 실험군들간에 큰 차이가 없었다. 이상과 같은 실험 결과를 종합할 때 참취는 에탄올에 의해 유발된 산화적인 스트레스를 적절하게 방어하여 특히 간장에서의 지질과산화 및 Mn-SOD활성의 감소를 가져왔으며 그 효과는 참취의 첨가량에 비례하는 경향을 나타내었으므로 생체 안에서 알코올과 같은 산화적 스트레스에 대한 항산화제로서의 역할을 효과적으로 수행할 수 있을 것으로 기대된다.

고 Isoflavone 콩나물이 만성 에탄올 투여 흰쥐의 지질대사에 미치는 영향 (Effects of Isoflavone-Rich Bean Sprout on the Lipid Metabolism of the Ethanol-Treated Rats)

  • 본 연구는 고 isoflavone 콩나물이 만성 에탄올 섭취 동물에서 지질대사와 항산화 효소계에 미치는 영향을 살펴보고 기능성 식품으로서의 가능성을 알아보기 위하여 수행되었다. 실험동물은 SD계 수컷쥐를 사용하였으며 40일간 Lieber-DeCarli 액체식이를 일부 변형한 실험식을 공급하여 고 isoflavone 콩나물과 isoflavone 추출물이 에탄올 투여 흰쥐의 지질대사와 항산화효소계에 미치는 영향을 조사하여 다음과 같은 결과를 얻었다. High SS군은 알코올 대조군에 비하여 체중증가량, 식이섭취량 및 식이효율이 모두 증가되었으나, low SS군과 IE군은 전반적으로 체중증가량, 식이섭취량 및 식이효율이 알코올 대조군보다 낮았다. 고 isoflavone 콩나물과 대두 isoflavone 추출물은 알코올 투여시 증가된 혈장 TG, TC, AI를 낮추었고, HDL-chol/TC의 비를 높였으며 간조직의 총 지질량, TG, TC 수준을 낮추었다. Isoflavone 추출물은 간조직의 TBARS 수준을 유의하게 낮추었으나 간조직 항산화효소 활성도를 증가시키지는 않았으므로 isoflavone의 항산화기전에 대한 연구가 필요할 것으로 보인다. 이상의 결과로부터 고 isoflavone 콩나물과 isoflavone 추출물은 혈장과 간조직의 지질 농도와 지질과산화를 낮추며 일부 항산화 효소계에 긍정적인 효과를 미치는 것으로 보인다.

사염화탄소와 에탄올에 의해 유발된 랫드의 간경변에서 GODEX (Hepadif-S capsule)의 치료 효과 (Therapeutic Effect of the GODEX on the Liver Chirrosis Induced by CCl4 and Ethanol in the Rat)

  • The hepato-protective activity of the GODEX (Hepadif-s capsule) has been studied in the rats against $CCl_4$-ethanol induced liver toxicity. The rats were oral1y treated with $CCl_4$ (corn oil/ $CCl_4$ 1:1, 1 mg/kg). And one week passes, $CCl_4$(0.4 mg/kg) administered two times a week for 7 weeks. The drugs have been administered every two days for 4 weeks after $CCl_4$ injection. The experimental groups have consisted of the GODEX (250 mg/kg), Hepadif (200 mg/kg), DDB complex (DDB 50 mg/kg and garlic oil powder 50 mg/kg), DDB (50 mg/kg), and vehicle control respectively. There was a significant decrement on the serum level of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and total bilirubin in all treated groups. Specially, ALT level of GODEX and Hepadif only treated groups was decreased c1early. Also, serum albumin level was significantly enhanced in GODEX treated group compared with control and DDB treated groups. In histological results, hepatocellular vacuolar degeneration, lobular restructure and necrosis of bile duct were severely showed in control. But other treated groups showed centerilobular degeneration and mild hyper-plasia. Hepadif or DDB has a effects of the recovery on serum parameters and structure ill liver injury. When it was compared GODEX to Hepadif alone or DDB complex or DDB, it suggested to have the best activity of the liver recovery.

Enhancement of Alcohol Metabolism by Sprouted Peanut Extract in SD Rats

  • Excessive ethanol intake is known to induce a number of physiological symptoms, including headache, dizziness and vertigo. In this study, we investigated the attenuation effect of sprouted peanut extract (SPE) on ethanol-induced hangover in male Sprague-Dawley rats. The animals were divided into five groups: the control group, which was administered ethanol only; the ethanol plus SPE experimental groups, which were administered ethanol and 100, 200, or 400 mg SPE/kg b.w.; and the positive control group, which was administered ethanol plus DAWN808$^{(R)}$, a commercial product. SPE-suspended water was delivered to rats via gavage 15 h and 30 min before the administration of ethanol. Blood was collected from the tail 0, 1, 3, and 5 h after ethanol administration. The results showed that serum ethanol concentrations were significantly lower in SPE treated groups than in the control group. Furthermore, hepatic alcohol and acetaldehyde dehydrogenase activities were enhanced by SPE in a dose dependent manner. These results suggest that SPE could be useful in attenuating hangover after alcohol consumption.

시령탕(柴苓湯)이 에탄올 투여로 유발된 흰쥐의 간손상에 미치는 방어효과 (Effects of Shiryung-tang Extract on the Liver Injury induced by Ethanol in Rats)

  • Alcoholic liver disease (ALD) is a major cause of morbidity and mortality around the world. Although much progress has been made in understanding the pathogenesis of ALD, there remains no effective therapy for it. Accumulated evidence indicates that oxidative stress is the main pathological factors in the development of ALD. Ethanol administration causes accumulation of reactive oxygen species (ROS), including superoxide, hydroxyl radical, and hydrogen peroxide. ROS, in turn, cause lipid peroxidation of cellular membranes, and protein and DNA oxidation, which results in hepatocyte injury. In addition to pro-oxidants formation, antioxidants depletion caused by ethanol administration also results in oxidative stress. The objective of this study is to investigate the effects of Shiryung-tang extract on the chronic alcoholic liver injury induced by EtOH. Male Sprague Dawley rats were used in this study. All rats were maintained under standard laboratory conditions ($23{\pm}1^{\circ}C$, 12h light/12h dark cycles). All animals (n=30) were randomly divided into following groups: (1) Normal group, treated with distilled water (n=10); (2) Control group, treated with ethanol (n=10); (3) Sample group, treated with ethanol + pharmacopuncture (n=10). For oral administration of ethanol in Control and Sample group, the ethanol was dissolved in distilled water in concentrations of 25%(v/v). Throughout the experiment of 8 week, the rats were allowed free access to water and standard chow. Sample group were administrated by Shiryung-tang extract daily for 8 weeks. Control group were given normal saline for same weeks. As a results, the oral administration of ethanol for 8 weeks leads to hepatotoxicity. The levels of hepatic marker such as HDL-cholesterol, triglyceride, aspartate aminotransferase and alanine aminotransferase were altered. The ethanol also increased lipid peroxidation and depletion of antioxidant enzyme activities as well as hepatic tissue injury. However, the treatment of Shiryung-tang extract prevented all the alterations induced by ethanol and returned their levels to near normal. These data suggest that Shiryung-tang extract could have a beneficial effect in inhibiting the oxidative damage induced by chronic ethanol administration. Therefore, Shiryung-tang extract can be a candidate to protect against EtOH-induced liver injury.

천련자 에탄올 추출물이 Streptozotocin으로 유발된 당뇨 흰쥐에 대한 혈당, 지질대사, 당대사 효소 활성과 항산화 작용에 미치는 영향 (The Effect of Meliae toosendan fructus Ethanol Extract on Blood Glucose, Lipid metabolism, Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats)

  • The ethanol extraction yield of Meliae toosendan fructus(MT) was about 24.5% by extract apparatus. This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of MT in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) were significantly decreaed in MT treated group compared to the those of STZ-control group, also content of Total cholesterol was decreased. High density lipoprotein (HDL)-cholesterol was increased in MT treated group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in MT treated group. Also the activities of glucose-6-phosphate dehydrogenase(G-6-PDH) and glucokinase(Gk) were increaed in MT treated group. The content of hepatic glycogen was significantly increaed in MT treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in MT treated group. Also, content of glutathione(GSH)was dereased in MT treated froup. whereas, activity of catalase(CAT) was significantly increaed in MT treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of MT would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

흰쥐에게 Ethanol과 Xylene 병행 투여시 간손상에 미치는 영향 (Effect of Liver Damage by Combined Administration of Ethanol and Xylene in Rats)

  • 흰쥐에게 ethanol과 xylene을 투여시 간손상 정도를 알아보기 위해 정상적인 S-D계 수컷 흰쥐를 4군으로 나누어 실험을 수행하였다. 생화학적 검사 결과에서 혈청 AST, ALT, LDH활성은 대조군보다 ethanol 투여군, xylene투여군, ethanol+xylene투여군으로 갈수록 활성이 유의하게 증가되었으며, 그 증가율은 ethanol+xylene투여군에서 가장 높게 나타났다. 산화적 손상정도를 알아보기 위한 혈청 MDA함량은 대조군보다 ethanol투여군과 xylene투여군 그리고 ethanol+xylene투여군으로 갈수록 활성이 증가되었으며, 그 증가율은 ethanol+xylene투여군에서 가장 높게 나타났다. 그러나 혈청 ALP는 대조군에 비해 모두 감소하였다. Xylene의 투여시 최종대사산물인 요 중 m-methylhippuric acid는 대조군에 비해 xylene투여군과 ethanol+xylene투여군에서 증가되었으며, 그 증가율은 xylene투여군이 ethanol+xylene투여군보다 더 높게 나타났다. 간 조직검사에서는 xylene투여군은 지방변화를, ethanol+xylene 투여군은 경도~중등도의 괴사와 염증을 보였다. 이상의 혈청학적 검사와 조직학적 검사결과 xylene 단독 투여에 비하여 ethanol과 xylene을 병행 투여했을 때 간손상이 더 크게 초래된 것으로 나타났다.

Effects of Taraxacum mongolicum Extract on Blood Glucose Levels and Lipid Profiles in Streptozotocin-Induced Diabetic Rats

  • This study was designed to evaluate antihyperglycemic and antihyperlipidemic effects of ethanol extracts of Taraxacum mongolicum(T.m.) on streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley rats were randomly assigned to five groups: normal (NC), STZ-control (DC), and three experimental groups. Diabetes was induced in Sprague-Dawley rats with a single intravenous injection [45 mg/kg body weight (b.w.)] of STZ. An ethanol extract of T.m. was orally given to diabetic rats for 14 days. Three experimental groups were additionally treated with T.m. extract at doses of 1 g/kg b.w./day for T.m.-1, 2 g/kg b.w./day for T.m.-2, and 3 g/kg b.w./day for T.m.-3. Oral administration of T.m.-2 significantly increased their body weights. T.m.-1 and T.m.-2 significantly decreased aspartate aminotransferase (AST) levels than DC. T.m.-1 and T.m.-2 group significantly decreased blood glucose levels. Total cholesterol, triglycerides, and free fatty acids were significantly decreased whereas high-density lipoprotein cholesterol was significantly increased in groups treated with T.m. extract than those in the DC group. These results support the fact that administration of T.m. extract can reduce hyperglycemia and hyperlipidemia risk in diabetic rats.
