Growth Inhibition Effects of Ethanol and Sodium Chloride on Bacillus cereus (Ethanol과 NaCl에 의한 Bacillus cereus 생육저해 영향)

  • Agricultural foodstuffs, usually consumed without sterilization, are frequently contaminated with foodborne pathogen B. cereus. Ethanol and sodium chloride were used to inhibit this pathogen for its effective control. Though five minutes of exposure to 20% ethanol did not inhibit the growth of B. cereus, inhibition was detected to 30% ethanol solution. As exposed longer, B. cereus was more effectively inhibited than E. coli and S. typhimurium. B. cereus, E. coli and S. typhimurium were not inhibited when exposed in a 10% sodium chloride solution for five minutes. However, these bacteria were inhibited with a combination of 10% sodium chloride and 20% ethanol solution for five minutes. Much synergistic growth inhibition on B. cereus was found at the treatment. Its viable count was reduced from $10^8cfu/ml$ to $10^2cfu/ml$ after five minutes and showed no count after ten minutes. This trend was also confirmed for the wild types of B. cereus. This method may be applied for the effective pre-treatment of many agricultural foodstuffs, especially uncooked foodstuffs, without the hazards that accompany special sanitizers and the nutritional loss from harsh sterilization.

Functional Ingredients of Perilla Frutescens L. Britt Extracts and Preparation of PVA Nanoweb Containing Extracts (자소 추출물의 기능성 성분과 자소 추출물을 함유하는 PVA 나노 섬유의 제조)

  • The purpose of this study was to analyze the functional ingredients of Perilla Frutescens L. Britt extracts and to confirm the possibility of producing PVA nanofibers using extracts. Distilled water, 3% aqueous sodium hydroxide solution and ethanol were used as extraction solvents. The electrospinning was carried out at a PVA concentration of 12%, an applied voltage of 10 kV and a tip to collector distance of 15cm. The contents of volatile substances, essential oils, total polyphenols and flavonoids of the extracts were measured to examine the constituents of functional materials. Flavor components and esters were identified in 3% sodium hydroxide and ethanol extracts. The content of polyphenols and flavonoids in ethanol extracts was higher than that of medicinal plants. 1wt.% of Tween 20 was added to disperse the essential oil components of the ethanol extract. Addition of a dispersant made it possible to produce a homogeneous mixture by having some compatibility with the ethanol extracts and the PVA molecule. When the concentration of the ethanol extract was 0.25 and 0.5wt%, relatively uniform PVA nanofiber having an average diameter of 350 to 365nm could be produced. The results of FT-IR, XRD and DSC analysis confirmed that Perilla Frutescens L. Britt ethanol extract was well mixed with PVA molecules and was electrospun.

The Analysis of Ethanol Distribution Level in Biological Specimens (생체 시료중에서 Ethanol 농도 분포에 관한 연구)

  • Five kinds of packed column and two kinds of capillary column were used to get optimum condition for ethanol analysis by using fifteen different volatile, low molecular weight organic substances. Only two columns, Gaskuropack 54 and DB-1, showed good separation efficiency. In the adding salt-effect experiment 0.6N - perchloric acid, 1M - meta-phosphoric acid and saturated NaCl solution were used for alcohol concentration measurement of biological fluids and tissue specimens. Among adding salt experiment, adding saturated NaCl solution showed the most stable value of alcohol concentration. This fact might be due to the increased vaporization of alcohol in the saturated NaCl solution. In the time-course of blood alcohol concentration, the alcohol level was lineary decreased to the diameter of vessel containing specimens. This result was interpreted in view of ethanol level, weight, water content, and hematocrit value. The ethanol distribution levels were measured from samples of blood and tissue obtained from 25 postmortems cases investigated by NISI. This study showed that the distribution level was decreased in order of brain, blood, kidney, spleen, liver, and lung.

  • Poly (vinyl alcohol) (PVA) membranes crosslinked with poly (styrene-co-maleic anhydride) (PSMAn) have been prepared by the solution method. To investigate the separation behavior of the crosslinked PVA/PSMAn membranes in the pervaporation process, the selective sorption experiment and swelling measurements of the membranes in ethanol-water mixtures of 30~90 wt % ethanol contents were conducted with equipment that was able to measure the concentration and amount of the liquid absorbed in the membranes. The membranes prepared in this study exhibited good selectivity toward water component in sorption and permeation. Also, in the feed containing ethanol more than 50 wt %, the permeation selectivity of the membrane showed better correlation with the sorption selectivity than that with the swelling ratio changing toward the crosslinking content. This behavior was consistence with a solution-diffusion model correlating permeation and sorption selectivity, and led to the conclusion that the permeation selectivity was determined by sorption step rather than by diffusion step in the membrane.

Effect of ascorbic acid, ethanol and acetone on adhesion between the treated fiber posts and composite resin cores

  • PURPOSE. The aim of the present study was to assess the effect of ascorbic acid, ethanol and acetone on microtensile bond strength between fiber posts pre-treated with hydrogen peroxide and composite resin cores. MATERIALS AND METHODS. Twenty four fiber posts were pre-treated with 24% hydrogen peroxide and divided into 4 groups as follows: G1: no treatment, as control group; G2: treatment with10% ascorbic acid solution for 5 minutes; G3: treatment with 70% ethanol solution for 5 minutes; and G4: treatment with 70% acetone solution for 5 minutes. Each fiber post was surrounded by a cylinder-shaped polyglass matrix which was subsequently filled with composite resin. Two sections from each sample were selected for microtensile test at a crosshead with speed of 0.5 mm/min. Statistical analyses were performed using one-way ANOVA and a post hoc Tukey HSD test. Fractured surfaces were observed under a stereomicroscope at ${\times}20$ magnification. The fractured surfaces of the specimens were observed and evaluated under a SEM. RESULTS. Means of microtensile bond strength values (MPa) and standard deviations in the groups were as follows: G1: $9.70{\pm}0.81$; G2: $12.62{\pm}1.80$; G3: $16.60{\pm}1.93$; and G4: $21.24{\pm}1.95$. G4 and G1 had the highest and the lowest bond strength values, respectively. A greater bond strength value was seen in G3 compared to G2. There were significant differences between all the groups (P<.001). All the failures were of the adhesive mode. CONCLUSION. Application of antioxidant agents may increase microtensile bond strength between fiber posts treated with hydrogen peroxide and composite cores. Acetone increased bond strength more than ascorbic acid and ethanol.

Characteristics of Greenish Pigments from Silkwarm Excrement by Ethanol Extraction (누에 배설물에서 Ethanol 추출한 녹색색소의 일반적 성질)

  • A study of characteristics of greenish pigments from silkwarm excrement by ethanol extraction. Through visible absorption scanning, it showed two absorption peaks at 415 and 657nm, and it was shown to be greenish color. In the presence of light, the stability of pigments rapidly degraded, but in the presence of Al-foil, green and blue cover were very stable. It was shown to be stabilized at the temperature of 7$0^{\circ}C$ until 20days and presence of metal ions, such as Na+, K+, Ca2+, Mg2+ and Zn2+. The pigments was shown to be stabilized in 5% acetic acid solution, but in the presence of lactic acid, citric acid and tartaric acid solution were unstable. On the result of TLC analysis, pigments were shown to be composed of eight color fractions, and main color fractions were F-1, F-2 and F-3. In F-1, F-2 fractions were revealed green color and F-3 fraction were revealed yellow color.

  • Ethanol fermentation from the chemically gelatinized starchy material was examined. The critical concentration of sodium hydroxide solution for gelatinization was dependent on the species of starch; 0.4M for potato and 0.6M for tapioca at room temperature. For alcohol fermention the starchy material was gelatinized by addition of sodium hydroxide solution, neutralized by sulfuric acid, and then yeast was added. The amount of $CO_2$ evolved during ethanol fermentation indicates that non-fermentable material was not produced from the starch by chemical gelatinization. In ethanol fermentation of potato and tapioca starch no significant difference was observed between the thermal and the chemical gelatinization.

  • In the present study adding hydroxypropyl celluose as dispersant to the ethanol solution of Zr-butoxide prior to starting the hydrolysis reaction, spherical and submicrometer sized hydrated zirconia powders were synthesized successfully. But syntesized hydrated zirconia powders were soluble considerably in ethanol used as washing medium. Washing once the powder with plenty amount of water after washing thrice it with acetone, reaction mother solution remained between the particles was effectively removed and the particles were converted to insoluble state to ethanol at the same time. As a result of such washing processes it was able to prevent the formation of polydispersed, agglomerated and multiplet particles almost always even when such concentrated solutions of Zr-n-butoxide as 0.5 M were hydrolyzed.

  • The effect of capsaicin on the toxicity of ethanol in mice were studied. Capsaicin was administered i.p. every other day for 4 weeks and 5% ethanol was provided ad libitum by tap water for 4 weeks. The administration of capsaicin 3.0 mg/kg showed the increase of body weight gain, ratio of liver wt./body wt., s-GPT. s-triglyceride and s-cholesterol, and showed the decrease of BUN as compared to control group. Capsaicin administered 3.0 mg/kg showed severe moth eaten appearance. eosinophilic necrosis and cholangitis in mouse liver The administration of 5% ethanol showed the decrease of body weight gain, ratios of liver, kidney and spleen wt./body wt., s-tryglyceride and s-cholestrol. Ethanol administered 5% solution showed little fatty change, moth eaten appearance, Kupffer cell proliferation, spotty necrosis and nuclear regeneration. The administration of capsaicin and ethanol together decreased the influence of ethanol on body weight gain, ratios of liver, kidney and spleen wt./body wt., s-triglyceride and s-cholesterol, and showed the less severe moth eaten appearance, eosinophilic necrosis and cholangitis. It might be concluded that the administration of capsaicin and ethanol together decreased the toxicity caused by capsaicin or ethanol respectively.

Use of Selective Ethanol Adsorption for Ethanol Concentration (선택적 에탄올 흡착을 활용한 에탄올 농축공정개발)

  • In this study, we developed simple process for ethanol concentration. We developed magnetically separable polyanilline nanofiber (PAMP) for selective ethanol adsorption. PAMP can adsorbed 80% of ethanol in the solution. After adsorption, the ethanol was recovered with simple magnetic separation and centrifugation process. After 10 times recycle of PAMP, the ethanol adsorption maintained 92% of its initial adsorption capacity. Using ethanol concentration process, the ethanol concentration increased up to 197.6 g/L from 46 g/L which was 4.3 folds increase.
