Characterization of a Bioflocculant Produced by an Isolate, Bacillus megaterium G31

  • A bacterial strain capable of producing a novel bioflocculant was isolated from a biofilm sample and identified as Bacillus megaterium G31. The highest biopolymer yield was achieved when the organism was cultivated in a medium containing acetate as the sole carbon source and ($NH_4)_2HPO_4$as the nitrogen source. In kaolin suspension, the flocculating activity was highest at 170 mg I$^{-1}$ and decreased at the higher bioflocculant concentrations. The crude bioflocculant produced by the organism was purified by ethanol precipitation and gel permeation chromatography. The FTIR spectrum of the purified bioflocculant revealed that the bioflocculant might be a heterogeneous polysaccharide composed of hexosamines and neutral sugars. The analysis of sugar components of the bioflocculant using high performance anion-exchange chromatography showed that the sugar constituents of the bioflocculant were glucosamine, fucose, galactosamine, galactose, glucose, mannose in approximate molar ratio of 4 : 1 : 6 : 3 : 8 : 19. Its flocculating activity was stimulated by various cations. The bioflocculant was thermo-stable and retained 64% of its original activity after heating at $100^{\circ}C$ for 50 min.

Novel Cationic Microbial Polyglucosamine Biopolymer from New Enterobacter sp. BL-2 and Its Bioflocculation Efficacy

  • A new bacterium BL-2 excreting a novel cationic polyglucosamine biopolymer was isolated from the spoiled leaves of Chinese cabbage and identified as Enterobacter sp. BL-2. The isolated Enterobacter sp. BL-2 was cultivated in pH-stat fed-batch culture using acetic acid as the feeding stock at pH 8.0, resulting in 17.11 g/l of cells and 1.53 g/l of an extracellular biopolymer after 72 h. The excreted biopolymer was purified by a three-step procedure, involving ethanol precipitation and deproteinizations, to a nearly homogeneous state, and its molecular weight was found to be 106 kDa. It was composed of glucosamine, rhamnose, and galactose at a molar ratio of 86.4:1.6:1.0, respectively, indicating a rarely found novel high-glucosamine-containing biopolymer. The FT-IR and $^{13}C-NMR$ spectra of the novel cationic polyglucosamine biopolymer PGB-l revealed a close identity with chitosan from crab shell. It can effectively flocculate various suspended solids, including kaolin clay, $Ca(OH)_2,\;Al_{2}O_3$, active carbon, microbial cells, and acidic dyes.

  • Paclitaxel is an effective antineoplastic agent against ovarian, colon and breast tumors. But there have been many difficulties to formulate this drug due to the extremely low aqueous solubility. Paclitaxel is currently formulated in a vehicle composed of Cremophor EL and absolute ethanol mixture which is $5\~20$ fold diluted in normal saline or $5\%$ dextrose solution before I.V. injection. However, this formulation has many problems such as allergic reactions and drug precipitation on aqueous dilution. To overcome these problems, we prepared the micelle and microemulsion systems for parenteral administration of paclitaxel by using glycofurol, $Soluto^{(R)}lHS$ 15 and oleic acid. Phase diagram, pH-rate stability, particle size distributions and pharmacokinetics of the systems were studied. Micelles and microemulsions formulated as nano-particle delivery system were physically and chemically stable. Therefore, these formulations might be the promising alternative candidate for the parenteral delivery of paclitaxel.

  • 한국에 야생하는 독버섯의 하나인 노란다발버섯을 물로 가열 추출한 후 추출액에 3배의 에탄올을 첨가하여 고분자 물질을 침전시켰으며 이를 다시 투석하여 정제 하였다. 이것을 냉동 건조하여 분말을 얻었으며 60mg/kg 용량을 백서의 복강내에 주사 하였던 바, 6일 후 부터 사망하기 시작하여 7일째에는 7마리중 4마리가 절명하였다. 이 유독 성분을 "네마톡신"이라 명명하고자 한다.

  • For thestudying and utilizing the thorn sea cucumber synthetically, the method preparing polysaccharide and polypeptide from sea cucumber was studied. The comparatively rational craft for preparing crude polysaccharide is fresh raw materials deal with oar form, $60\%$ of the ethanol precipitate after hydrolyzed by pretense and vibrated with ultrasonic wave auxiliary. The purification of polysaccharide and removal of protein fromcrude polysaccharide were made through precipitation method using the acetate. The polypeptide is obtained by concentrating in vacuum, freeze-drying after mixing precipitations of two times. Polysaccharide acute poisoning experiment indicate there is no bad reaction when LD50>5000g/kg, there strain rate of liver tumor H11 approach present generally acknowledged tumor treat medicine cyclophosphamide, and there is no side effect. The fatigue resistance function of polypeptide experiment also indicates that the fatigue resistance ability of mouse which fed on added sea cucumber polypeptide has a great improvement.

Isolation, Purification and Characterization of Polysaccharides that induce in vitro Immuno-Stimulation of Macrophases derived from Liquid Culture of Cordyceps militaris

  • The crude polysaccharides(C-CPF, C-CPM, C-CPB) derived from fruiting body, mycelia and mycelia free broth of cordyceps militaris were obtained by ethanol precipitation of hot water extracts. After a batch fermentation of C. militaris was carried out in a 5 L jar vessel, endo-polysaccharide and exo-polysaccharide were obtained. They were demonstrated as the hetero polysaccharides which were composed of glucsose, galactose and mannose by performed with HPAEC(high pH anion exchange chromatography) and conformation of random coil by its complex forming ability with congo red reagent. They were purified by ion exchange (DEAE-cellulose) and gel filtration chromatography. They were monitered by phenol-sulfuric acid method and Bradford method. The NO induction activities of crude polysaccharides and purified polysaccharides derived from mycelia free broth were enhanced rather than LPS(lipo polysaccharide) which was used as a general NO inducer. These effects presumably contibute to the antitumor activities. The homogenieties and molecular weights of polysaccharides were determined by using Sepharose CL-6B. The yield, molecular weights and NO induction activities of C-CPFN Fr.III, C-CPMN Fr.III, C-CPBN Fr.II were 0.387, 0.408 and 0.153, 127 K 210 K and 36 K, 40.79%, 88.72%, and 104.17%, respectively.

Variation in Fucoidan Contents and Monosaccharide Compositions of Korean Undaria pinnatifida (Harvey) Suringar (Phaeophyta)

  • Three different forms of Undaria pinnatifida, the southern form (U. pinnatifida f. typica), the northern form (U. pinnatifida f. distans), and Samcheok form (recently cultivated strain), were examined for the contents and compositions of fucoidans. Fucoidans were extracted from the dried edible portions of three forms of U. pinnatifida in low pH condition, mainly by ethanol precipitation and CaCl2 treatment. It was shown that Samcheok form contains 1.8 and 3.5 times more fucoidans than the northern and the southern forms, respectively. The monosaccharide compositions of individual fucoidans were also varied. The fucoidans from the southern and the northern forms were shown to be composed of mainly fucose and galactose with the molar percentage ratios of 83.5%:16.5% and 87.4%:12.6%, respectively, indicating that these are F-type fucoidans. The fucoidan from Samcheok form, however, consisted of fucose (62.7%), galactose (32.9%), and small amount of glucose (4.4%). The results of this study showed that both amount and monosaccharide compositions of fucoidans are variable depending on U. pinnatifida forms.

  • 멥쌀식혜와 찹쌀식혜의 한계덱스트린을 알코올침전, Biogel P-2의 겔 크로마토그래피, Superose 12 겔 크로마토그래피 칼럼을 사용한 FPLC로 정제하여 1H-NMR 분석을 행하였다. 멥쌀식혜의 한계덱스트린은 $\alpha$-1,4-글루코시드 결합과 $\alpha$-1,6-글루코시드 결합의 비율이 1:4.5, 찹쌀식혜의 한계덱스트린은 1:5.9를 나타냈다. Pullulanase 소화로 멥쌀식혜 및 찹쌀식혜의 한계덱스트린은 말토오스, 말토트리오스, 말토테트라오스, 말토펜타오스, 말토헥사오스까지 나타내 이들이 서로 조합하여 한계덱스트린을 만들고 있는 것으로 분석되었다.

Effects of the Antitumor Component, F-D-P, Isolated from Elfvingia applanata on the Immune Response

  • An antitumor component, F-D-P, was purified from the hot water extract of the carpophores of Elfvingia applanata by precipitation with ethanol, dialysis, and passage through a column of DEAE-cellulose ion exchange. F-D-P inhibited the growth of Sarcoma 180 in mice showing the tumor inhibition ratio of 88.3% in doses of 20 mg/kg for ten days. Chemical analysis of F-D-P showed that it was composed of polysaccharide(65.3%) and protein(6.5%0, and that the monosaccharides consisting of the polysaccharide was glucose(89.1%) and mannose(10.9%). The immunomodulatory activities of F-D-P were explored by determining its effect on the proliferation of the whole and subpopulations of lymphocytes, and on the generation of natural killer(NK) cell activity in vitro. F-D-P was mitogenic to total lymphocytes and B cells, but not to purified T cells, even in the presence of accessory cells. F-D-P did not increase NK cell activity when added to cultures of resting lymphocytes. From these results, it is clear that F-D-P modulates primarily the humoral immune responeses.

  • Proteolytic activities were compared using three species involving in squid jeotkal fermentation and showing positive reaction upon casein test : Pseudomonas D2, Flavovacterium odoratum and Acinetobacter calcoaceticus. Pseudomonas D2 produced highest activity of protease at 72h when incubated in our own modified medium(polypeptone, 0.5% ; tryptone, 0.5% ; NaCl, 3% ; pH, 7.5). Thus, this specie was selected for the further study. The growth pattern was coincided with the production of protease. Thus purification of protease was proceeded by ethanol precipitation, sephadex G-100 gel filtration, and DEAE sepharose ion exchange chromatography. The purified protease showed highest activity at pH 7.0 and 5$0^{\circ}C$. The enzyme was very stable over the wide ragnes of the temperature ; even with one hour heat treatment at 7$0^{\circ}C$, the enzyme showed substantial amount of the activity toward casein. In addition, the enzyme was stable over the wide range of pH. Molecular weight of the protease was determined to be 17.4 kD by SDS-PAGE.
