BBP (Butyl benzyl phthalate), a widely used plasticizer. can enter the food and environment as consequence of its manufacture, use, and disposal. BBP was found to be developmental and teratogenic or endocrine disrupting chemical in rats. The effects of BBP were investigated in female rats (P) and second generation (F1) via lactations. Sprague-Dawley were given BBP by oral administration at 0, 5, 10, 100, 1000 mg/kg on day 0 to 21 of lactation period. The results were as follows : At maternal findings, there were some significant changes (p<0.05) in relative organ weight, especially liver and uterus weight by BBP administration. In estrous cycle, high treated group was inclined to be proestrus or estrus compared to control group. BBP indues estrous cycle earlier than the control group. At fetal findings, there were some significant changes in relative liver and spleen weight, especially 100, 1000 mg/kg administered groups. The relative weight of ventral prostate was decreased, so it was represent to dose-response tendency. Parent rats (P) were detected monobenzyl phthalate (MBeP) 3.21~5.81 $\mu\textrm{g}$/ml in 100, 1000 mg/kg dose groups. MBeP of male and female fetuses (F1) were detected at the level of 1.21~2.63 $\mu\textrm{g}$/ml of serum. Male serum concentration oi MBeP was higher than the females'. Estrogen receptor $\alpha$ expression by BBP and bisphenol A in uterus and testis of F1 were studied. The ER$\alpha$ expression were increased in F1 male testis and female uterus. F1 male showed distint ER$\alpha$ expression, especially in the combined exposrue. Synergistic ER$\alpha$ expression was found by combined treatment group of BBP and bisphenol A. From the above results, it could be concluded that the effects of dams and F1 by BBP administration during lactation period were estrogenic, and BBP can transfer to F1 via lactation, and make estrogenic at F1 reproductive organs.
Klotho (KL) is a single transmembrane protein composed of KL1 and KL2 repeats possessing ${\beta}$-glucuronidase activity and maintains calcium homeostasis in physiological state. It has been implicated in pigs that calcium is important for the establishment and maintenance of pregnancy, and our previous study has shown that transient receptor potential vanilloid type 6 (TRPV6), a calcium ion transporter, is predominantly expressed in the uterine endometrium during pregnancy in pigs. However, expression and function of KL in the uterine endometrium has not been determined in pigs. Thus, the present study determined expression and regulation of KL in the uterine endometrium during the estrous cycle and pregnancy in pigs. Real-time RT-PCR analysis showed that levels of KL mRNA decreased between Days 12 to 15 of the estrous cycle, and its expression showed a biphasic manner during pregnancy. KL mRNA was expressed in conceptuses and in chorioallantoic tissues during pregnancy. Explant culture study showed that expression levels of KL were not affected by treatment of steroid hormones or interleukin-1beta during the implantation period. Furthermore, levels of KL mRNA in the uterine endometrium from gilts carrying somatic cell nuclear transfer (SCNT)-derived embryos were significantly lower than those from gilts carrying natural mating-derived embryos on Day 12 of pregnancy. These results exhibited that KL was expressed at the maternal-conceptus interface in a pregnancy status- and stage-specific manner, and its expression was affected by SCNT procedure, suggesting that KL may play an important role in the establishment and maintenance of pregnancy in pigs.
Skim milk progesterone($P_4$) profiles in 74 dairy cows were determind to monitor postpartum ovarian activity by radioimmunoassay. Milk samples were collected from each cow every 5 days from 10 to 90 days postpartum. Signs of estrus were observed twice daily, and status of the ovaries and uterus were examined every 10 days by rectal palpation. Results are summarized as follows: 1. Cows were categorized into five types by the change of skim milk $P_4$ profiles; Type I(normal) : Cyclic changes of skim milk $P_4$ profiles appeared within 20 days postpartum(12 cows, 16.2%), Type II(cycle delayed) : Cyclic changes of skim milk $P_4$ profiles appeared from 21 to 60 days postpartum(39 cows, 52.7%), Type III(cycle ceased with low $P_4$) : Onset of the estrous cycle within 20 days postpartum but ceased later with low levels of $P_4$ (7cows, 9.5%), Type IV(cycle ceased with high $P_4$) : Onset of the estrous cycle within 20 days postpartum but ceased later with high levels(>3.0 ng/ml) of skim milk $P_4$ (4 cows, 5.4%), Type V(acyclicity) : Skim milk $P_4$ concentration remained low(<1.0 ng/ml) until 80 days postpartum(12 cows, 16.2%). 2. Out of the 17 cows classified as the Type III and Type V by skim milk $P_4$ profiles, 13 cows had inactive ovaries and remaining 6 cows had single or multiple follicular cysts in their ovaries by rectal palpation. All 4 cows of Type IV had a persistent corpus luteum in their ovaries. 3. Approximately eighty percent of the cows had begun ovarian activity by 60 days postpartum and 90.6% by 90 days by skim milk $P_4$ profiles, but only 39.2% by 60 days and 71.7% by 90 days had shown visible estrus signs. The mean days from parturition to the first, second and third ovulations determined by skim milk $P_4$ profiles was $28.0{\pm}11.0$, $46.4{\pm}13.3$ and $66.4{\pm}11.5$ days and the visible estrus signs were 9.3%, 38.1%, and 48.6%, respectively. The mean days from parturition to the first visible estrus was $57.2{\pm}15.9$ days. These results indicated that milk $P_4$ profile of each Types by radioimmunoassay can be utilized for monitoring postpartum ovarian and would be useful for the early detection of ovarian dysfunction in dairy cow.

The objective of this study was to characterize the litter sizes in Huanghuai goats with high prolificacy (HP, five or more kids born per litter on at least one occasion), and to compare their peripheral blood concentrations of progesterone and estradiol with those of goats with poor prolificacy (PP, up to three kids born per litter on any occasion). The circulating concentrations of progesterone and estradiol were measured by radioimmunoassay from daily blood samples taken during natural estrus cycles and at 1-5 days after ovariectomy. Estrus was synchronized using two doses of a prostaglandin analog. Litter size for the HP goats increased up to a parity of five and decreased thereafter. The percentage of goats with litter sizes of $\geq$4 from parities 3 to 6 ranged from 44.5% to 58.3%. Although small differences in litter size were obtained for goats of parities three, four and six relative to five, parity five does had the highest mean litter size. Progesterone concentrations began to rise earlier and were higher in the HP than in the PP goats on most days of the luteal phase, but not during the follicular phase of the cycle or after ovariectomy. There was a significant difference between the two groups (p<0.05) in the magnitude of the progesterone plateau. Mean estradiol concentrations in the HP group remained significantly higher than in the PP group (p<0.05) during the estrus cycle. There were two estradiol peaks in the HP goats during the early luteal phase, but only one in the PP group. Measurements of individual corpora lutea (CL) in vitro showed that there was a greater prevalence of small CL (<6 mm in diameter) in the HP group than in the PP group (p<0.05). After ovariectomy, the estradiol level on day 1 was significantly higher than at the nadir during the estrus cycle in both the HP (p<0.01) and PP (p<0.05) goats, while levels decreased by 12.3% and 26.2% respectively compared with the mid-luteal period in HP and PP goats (p<0.05). The overall mean estradiol concentrations in HP goats were lower than in the PP group, but no significant differences were found between groups at 1-5 days after ovariectomy.
The present study was carried out to determine the changes of the sex hormone levels in serum throughout the estrous cycle and the gestation period on the Landrace gilts. The blood samples were taken from the vein of six gilts. LH, FSH, prolactin, progesterone, $estradiol-17{\beta}$ and cortisol in serum were analyzed by the radioimmunoassay methods. The results obtained on this study were summarized as follows; 1. The age at puberal estrus was 179.5 days, the weight at puberal estrus was 88.2kg, the length of estrous cycle was 21.3days, the gestation length was 114days and the litter size was 9.5 head in the Landrace gilts. 2. During the estrous cycle, the serum LH and prolactin concentrations were below 1.56mIU/ml and 2.4ng/ml, respectively, under the limit of detection of the assay. The FSH concentrations ranged from 1.50 to 2.20mIU/ml for day 6~15 after the estrus and they were below 1.25mIU/ml from day 3 to day + 3, with day 0 being the first day of the estrus. 3. Progesterone concentrations were 1.90ng/ml at day 0 of the estrus and increased about 13.1ng/ml at day 3 of the estrus, and reached peak levels at day 9. $Estradiol-17{\beta}$ concentrations were below 27.2pg/ml throughout the luteal phase, and reached about 27.2pg/ml at day 0 and day 18. Cortisol concentrations reached peak levels at dey 0 and ranged from 24.65 to 28.57ng/ml throughout the luteal phase. 4. During the gestation period, the concentrations of LH, FSH and prolactin ranged of 3.10~4.37mIU/ml, 1.30~1.80mIU/ml and 2.60~6.70ng/ml, respectively. 5. Progesterone concentrations declined from 38.90~16.85ng/ml throughout the pregnancy to 1.90ng/ml at the time of parturition. $Estradiol-17{\beta}$ concentrations increased from 27.20pg/ml at 15 days after the pregnancy to 620.17pg/ml at the time of parturition. Cortisol concentrations reached peak levels at the time of parturition and ranged from 13.58 to 22.31ng/ml throughout the pregnancy.

Equilume light masks had no impact on hastening the resumption of estrous cyclicity in mares maintained in outdoor pastures on the mainland of Korea due to the cold weather conditions. Jeju Island is a major horse-breeding site in Korea and is warmer than the mainland during the winter season. Therefore, the primary objective of this study was to explore the efficiency of the Equilume light mask on the resumption of seasonal estrous cycles in Thoroughbred mares on Jeju Island. A total of 20 nonpregnant mares were randomly divided into the Equilume light mask (n = 9) and stable lighting (n =11) groups. The experiment was performed at seven different horse-breeding farms located on Jeju Island from November 15, 2020, to February 15, 2021. The mares were exposed to the respective lights from 16:00 to 23:00. Follicle size and uterine edema were measured by ultrasound scanning. Body condition scores (BCS) were also monitored during the experiment. Statistical analysis was conducted using the SAS and SPSS software, and p-values of < 0.05 were considered statistically significant. Two of the nine (22.2%) mares in the Equilume light mask group and three of the 11 (27.28%) mares in the stable lighting group were still cycling in December and January, which were considered as all-year-round cycling mares. On February 15, there was no difference between groups in the resumption of early seasonal estrus cycle, which was determined by follicles > 25 mm in addition to uterine edema. All mares in the Equilume light mask group and five of the eight mares (62.5%) in the stable lighting group had resumed cycling. Interestingly, six of the seven mares (87.5%) in the Equilume light mask and four of eight mares (50%) in the stable lighting group had already ovulated on February 15 (p > 0.05), as determined by the presence of a recent corpus luteum. No difference was observed in BCS and uterine edema between groups (p > 0.05). In conclusion, the Equilume light mask can be an effective approach to induce early seasonal estrus cycles of mares in Jeju Island, and it also enhances the efficiency of farm management by reducing labor.
The corpus luteum is a transient endocrine gland essential for regulation of the ovarian cycle as well as for establishing and maintaining pregnancy. Prostaglandin $F_{2{\alpha}}$ (PGF) initiates functional and structural regression of the corpus luteum and therefore is an important regulator of the estrous cycle. It is a matter of debate whether the endothelial cells of the bovine corpus luteum express PGFR, the cognate receptor for PGF. Therefore, the aim of this study was to assess the expression of PGFR in bovine endothelial cells. Endothelial cells were isolated from the bovine corpus luteum of the mid-luteal stage using magnetic beads and cultured in vitro. We demonstrate that this isolation procedure generates a pure culture of endothelial cells as confirmed by synthesis of Factor VIII and lack of expression of $3{\beta}$-hydroxysteroid dehydrogenase. By RT-PCR, Western blot and immunofluorescence analyses, we further show that the cultured endothelial cells produced PGFR. This model system can be utilized to provide an experimental system to investigate the role of PGF on endothelial cells during the reproductive cycle.
This study was performed to determine the estrous cycles by macroscopic observation of the ovarian changes using the laparoscopy and to make use of these results for embryo transfer in Korean black goat (Copra hircus aegagrus). Laparoscopic examinations of the ovaries were performed from 2 days after $CIDR^(R)$ removal to 22 days after ovulation. The serial morphological changes of follicles and corpus luteum (CL) were observed. CL was classified corpus hemorrhagicum(CH), corpus luteum (CL) and corpus albicans (CA) by its maturation and regression. On the day before ovulation (Day 0), Graafian follicles (GF) were found on one or both ovaries. On the day (Day 1) and $2^{nd}$day (Day 2) of ovulation, and ovulation depression (OD) and an early stage corpus hemorrhagicum $(CH_1)$ were observed at the site of GF, respectively. On Days 3 to 4, more developed and enlarged corpus hemorrhagicum $(CH_2\;and\;CH_3)$ arised from the ovulation of the GF with well vascularization. On Days 5 to 6, it was identified that mature corpus luteum $(CL_3)$ was grown on the ovary, and fully developed CL with adjacent follicles were occupied most part of the ovary on Days 17 and 18. Then the size of CL was diminished, and completely luteal regression $(CL_1\;or\;CA)$ with new large follicle was identified on Days 20 and 22. From these results, the 4 stages of the estrous cycle in Korean black goats were 1) estrus (Day 0) for 1 day, 2) metestrus $(Day\;1{\sim}4)$ for 4 days (stage of CH development), 3) diestrus $(Day\;5{\sim}16/17)$ for 12 or 13 days (luteal stage), and 4) proestrus $(Day\;17/18{\sim}20/22)$ for 4 or 5 days (stage of luteal regression and follicular growing). Laparoscopy for observation of ovarian changes was invasive than laparotomy. Additionally, it had advantages of reduced adhesion and quick operation time. It was considered that laparoscopic examination of ovarian changes will be useful for embryo transfer in the Korean black goats.
The study was conducted to find out the changes of progesterone and 20$\alpha$-dihydroprogesterone (20$\alpha$-OHP) levels during the oestrous cycle in Korean native goats. 4 cyclic goats were offered for this experiment. Blood samples were taken from jugular vein on day 0, 1, 3, 5, 7, 9, 11, 13, 15, 17 and 19 during the oestrous cycle, then the next oestrous day. The serum levels of progesterone and 20$\alpha$-OHP were measured by radioimmunoassay. The progesterone concentration in seurm of the cyclic goats was 0.29$\pm$0.06ng/ml on the first day of oestrous (day 0), increased to 5.29$\pm$0.73ng/ml on day 9, reached to a peak level of 5.73$\pm$0.61ng/ml on day 13, and thereafter decreased to 0.35$\pm$0.30ng/ml on day 1. The serum level of 20$\alpha$-OHP during the oestrous cycle was 0.42$\pm$0.33ng/ml on day 0 and then decreased to 0.28$\pm$0.01ng/ml on day 5. This basal level was maintained until day 13, increased gradually, and reached a peak level of 0.62$\pm$0.05ng/ml on day 19. From the above results, it was suggested that the enzyme 20$\alpha$-hydroxysteroid dehydrogenase(20$\alpha$-HSD) catalyzing the conversion of progesterone to a biologically inactive steroid, 20$\alpha$-OHP, should be active in the luteal cells during the oestrous cycle in Korean native goats.
The aim in this study was to obtain the basic data for estimation of optimal mating time and ovulation time by finding out the relationship between vaginal cytology and reproductive hormone profiles during the estrous cycle in 36 Korea Jin-do bitches. A characteristic features of vaginal cytology during the estorous cycle were the high proportion of superficial cell, anuclear cell and erythrocyte in proestrus, superficial cell, anuclear cell and erythroucyte in estrus, parabasal cell, small and large intermediate cell and leukocytes in diestrus, and parabasal cell and small intermediate cell in anestrus, respectively. Cornification index(CI) was the high proportion in proestrus and estrus, then it was decreaed in diestrus and anestrus. It indicated that the CI was significantly high proportion in proestrus and estrus in comparison with the other phases. Plasma progesterone concentration was below 1.0ng/ml at the first day of vulval bleeding in pregnant and non-pregnat bitches, and then it was increaed slowly. When Day 0 was timed from the day that plasma progesterone concentration was first increased above 4.0ng/ml, plasma progesterone concentrations at Day 0 in pregnant and non-pregnant above 4.0 ng/ml, plasma progesterone concentrations at Day 0 in pregnant and non-pregnant bitches were $5.2{\pm}0.3ng/ml and 5.7{\pm}0.5ng/ml$$46.5{\pm}3.3 ng/ml$ in pregnant bitches and at Day 20 with $39.8{\pm}0.1ng/ml$ in nonpregnant bitches. It indicated that plasma progesterone concentration was higher in pregnant bitches than in non-pregnant bitches. Hereafter plasma progesterone concentration was decreased steadily. At Day 63 which is parturition day, plasma progesterone concentration was decreased below 1.0ng/ml with $0.8{\pm}0.2ng/ml$ in pregnant bitches whereas in the non-pregnant bitches at Day 75 were decreased below 1.0ng/ml with 40.5{\pm}0.4ng/ml$. Plasma progesterone concentrations was maintained below 1.0ng/ml during anestrus in all of them. The plasma estradiol-17 $\beta$ concentration was increased above 1.0 pg/ml at the first day of vulval bleeding and it showed a peak Day-1 with 38.2 pg/ml. Thereafter it was sharply decreased after Day 0, which was the day that plasma progesterone concentration was first increase above 4.0ng/ml, and was maintained below basal levels. In relationship between CI and reproductive hormones, plasma estradiol-17 $\beta$concentration showed a peak at Day-3 and CI showed a peak at Day-1 which was the second day after plasma estradiol-17 $\beta$ peak, and plasma progesterone concentration was first increased above 4.0ng/ml at Day 0 which was the first day after CI peak. CI was first increased above 80% at Day-6 which was the third day before plasma estradio-17 $\beta$ peak and it was maintained above 80% between Days-6 and Day 3 during 10 days, and showed above 90% at Day-3 which was the day that plasma estradiol-17 $\beta$peak and was maintained above 90% between Day-3 and Day 3 during 7 days. In conclusion, ovulation in Koran Jin-do bitches occurred at the first day after CI peak, at the third day after plasma estradiol-17 $\beta$peak and the day when plasma progesterone concentration was first increased above 4.0 ng/ml. And it was estimated that the optimal mating time was the day when the CI was maintained above 90% and plasma progesterone concentration was between 3.0~8.0ng/ml. Therefore plasma progesterone concentration measurement was used for determination of an accurate ovulation time and the optimal mating time but also vaginal cytology, which is low-priced with equipment and is the simple examination method, was reliable method for estimating estrous cycle, optimal breeding time and ovulation time in Korea Jin-do bitches.
