Studies on Enhanced Oxidation of Estrone and Its Voltammetric Determination at Carbon Paste Electrode in the Presence of Cetyltrimethylammonium Bromide

  • The electrochemical behaviors of estrone in the presence of various surfactants were examined with great details. It is found that a cationic surfactant, cetyltrimethylammonium bromide (CTAB), obviously facilitates the electro-oxidation of estrone at carbon paste electrode (CPE) from the significant peak current enhancement and the negative shift of peak potential. Additionally, chronocoulometry and electrochemical impedance spectroscopy (EIS) were also used for further investigation of the electrode process of estrone, indicating that low concentration of CTAB exhibits excellent enhancement effects on the electrochemical oxidation of estrone, greatly enhances the diffusion coefficient and the electron transfer rate. Based on this, an electrochemical method was proposed for the determination of estrone. The oxidation peak current is proportional to the concentration of estrone in the ranges over 9.0 × 10?8 - 8.0 × 10?6 mol/L, and a low detection limit of 4.0 × 10?8 mol/L was obtained for 180s accumulation at open circuit (S/N = 3). Finally, this proposed method was demonstrated using estrone tablets with good satisfaction.

Effect of Estrone Treatment on Proteinuria in Maturing Lean and Obese Male SHR/Mcc-cp Rats (Estrone 투여가 SHR/Mcc-cp 계통의 정상체중과 비만한 성장기 숫쥐들에서 단백뇨 증상에 미치는 영향)

  • Lean and obese male spontaneously hypertensive(SHR/Mcc-cp) rats were fed a ground rat chow diet with or without 0.001% estrone added from 6 to 18 weeks of age. Urine samples were collected weekly with 24 hour fasting. Both control lean and obese rats showed significantly higher urinary protein than their estrone treated counterparts. Treatment with 0.001% estrone diet was found to reduced the proteinuria in the maturing lean and obese SHR/Mcc-cp rats. Peaks in urinary protein level were noted at 16 weeks of age in both lean and obese control rats. Both control and estrone treated lean rats showed higher proteinuria than the obese rats. Therefore, obesity does not appear to be a contributing factor to the proteinuria in young male SHR/Mcc-cp rats.

The Effect of Exogenous Estrogen on the Pregnant Rats (Estrogen투여가 임신 rat에 미치는 영향)

  • The purposes of the present study were to evaluate the effect of estrone on the prevention of implantation and pregnancy and the concentrations of estradiol and progesterone or sodium and potassium in the plasma or uterine fluid of pregnant rats injected a single dose of 0, 0.1, 0.4 or 1.0 mg on days 5, 9, or 13 of pregnancy. The body weight gains of pregnant rats were significantly decreased in administering estrone on day 5 of pregnancyi no effect obtained on days 9 or 13. Numbers of viable fetuses were significantly decreased and pre- and post-implantation loss increased by administering estrone 0.1, 0.4 or 1.0 mg and numbers of implantation sites significantly increased by administering estrone 0.1 mg but significantly decreased or not occurred by 0.4 or 1.0 mg on day 5 of pregnancy, respectively, but no effect obtained on days 9 or 13. Plasma estradiol concentrations of pregnant rats were significantly increased on 1, 2 or 18 days after treatment with estrone (1.4 or 1.0 mg and 3 days after 0.4 mg on day 5 of pregnancy, respectively. But plasma progesterone concentrations had no effect on the treatment with estrone. Plasma sodium contents were significantly increased on 1 day after treatment with estrone 0.1 or 1.0 mg and 3 days after 0.1 mg on day 5 of pregnancy, respectively, except potassium content were no detectable change. Sodium contents in uterine fluid were significantly increased on 1 day after treatment with estrone 1.0 mg and 3 days after 0.1, 0.4 or 1.0 mg on day 5 of pregnancy, respectively. And potassium contents were significantly increased on 1 or 2 days after treatments with estrone 0.1 mg but significantly decreased on 3 days after 0.4 or 1.0 mg on day 5 of pregnancy, respectively. It is suggested that the prevention of implantation and failure of pregnancy could be induced by treatment with estrone 0.4 mg on day 5 of pregnancy and might be interrelated with changes of plasma estradiol concentration and sodium and potassium contents in uterine fluid.

  • Estrone such as estriol and estradiol can not be determined by votammetric methods, because these are electrochemically inactive in the potential windows for mercury drop electrode. Nitro-derivatives of estrone are electro active and nitration of estrone is accomplished by heating the solution involving estrone and sodium nitrite in a water-bath at $100^{\circ}C$ for 30 min. Such nitro-derivatives are determined directly by voltammetry. The electrochemical behavior for nitrated estrone was investigated by cyclic voltammetry. The trace estrone was determinated by differential pulse adsorptive cathodic stripping voltammetry. Nitrated estrone gives a well defined voltammetric wave at ca. - 0.61 V (vs. Ag/AgCl electrode). The electrochemical reaction was irreversible process in sodium borate buffer at pH 11 and nitrated estrone was strongly adsorbed on the surface of mercury electrode. The optimal experimental conditions for the determination of nitrated esterone were found to be 0.05 M sodium nitrate, 0.01 M sodium borate, pH 11.0, and an accumlation potential of 0.10 V (vs. Ag/AgCl). The detection limit was as low as $1{\times}10^{-9}M$ for estrone with 2 min accumulation time.

Changes in Serum Concentration of Progesterone and Estrone Sulphate during Gestation in Korean Native Goats (한국재래산양의 임신기간중 혈중 Progesterone 및 Estrone Sulphate 농도의 변화)

  • This study was conducted to find out the changes of progesterone and estrone sulphate concentration in serum of Korean native goats throughout gestation, and to apply the serum levels of the hormones to pregnancy diagnosis. Serum concentration of progesterone and estrone sulphate were assayed by radioimmunoassay. Serum progesterone concentration was similar to its luteal phase values during early pregnancy and remained at the high level continually at 20∼140days and decreased rapidly at the day of parturition. Serum estrone sulphate concentration showed to increase markedly at 40∼50dyas gestation and steadily increased to the maximum of 7.13ng/ml at 140days, but declined sharply at the day of parturition. The accuracy of pregnancy diagnosis by the measurement of serum progesterone at 20∼24days after mating was 85.7∼92.3% and that of non-pregnancy diagnosis was 100%, when the serum progesterone levels higher and lower than 3.0ng/ml were supposed to indicate pregnancy and non-pregnancy, respectively. The accuracy of pregnancy diagnosis by the mearsurement of serum estrone sulphate was found to be nearly 100% since 50days after mating, when the serum levels of estrone sulphate higher then 0.5ng/ml were diagnosed to be pregnant. The optimal sampling time for pregnancy diagnosis was considered to be at 50 days after mating or to be later. It appears that estrone sulphate values above 7.0ng/ml at any time in gestation are highly indicative of twin. But there was found no significant difference(P<0.05) in serum estrone sulphate concentration and number of kids between does with single and twin kids.

Chemiluminescence Immunoassay for Measurement of Estrone-3-Sulfate Using Monoclonal Antibody to Estrone-3-Glucuronide (Estrone-3-Glucuronide에 대한 단일클론항체를 이용한 Estrone-3-Sulfate 측정을 위한 화학발광면역분석법)

  • This study was carried out to develop an immunoassay for the diagnosis of the pregnancy and ovarian function of domestic animals. Using 2E92C10 monoclonal antibody(McAb) generated against estrone-3-glucuronide(E1-3-G) and appeared a high cross-reactivity with estrone-3-sulfate(E1-3-S), chemiluminesence immunoassay (CIA) to detect E1-3-S was developed. 2E92C10 McAb cross-reacted with E1-3-S (30%) was purified from ascites fluid using protein G sepharose gel column. The purity of purified antibody fraction was monitored by SDS-PAGE and was better compared to that of crude ascite fluid. The soild and liquid phase CIA for E1-3-S were established utilizing 2E92C10 antibody and E1-3-G-ABEI conjugate used as a tracer. As the results, the titer of 2E92C10 antibody was 5g/ml in soild phase and 1:2000 in liquid phase. The sensitivity on soild and solid phase CIA were about 200 pg/ml. These results indicate that CIA for measurement of E1-3-S was successfully developed by using ant-E1-3-G McAb cross-reacted with E1-3-S and could be usefully used to research this area.

  • Several endocrine disrupting chemicals(EDCs) were monitored to evaluate the estrogenic activities and the concentrations by yeast two-hybrid assay and enzyme-linked immunosorbent assay(ELISA) in sewage treatment plant(STP) which consist of industrial and domestic line. In the influent of domestic line, estrone, 17$\beta$-estradiol, 17$\alpha$-ethinylestradiol and alkylphenolethoxylate(APE) were detected up to 167.1, 39.7, 7.3 and 145.4 ng/L, respectively. The average removal efficiency of 17$\beta$-estradiol after the activated sludge process was 77.5% and further removed to 80.8% after the sand filtration-ozonation step. These results suggests that the activated sludge process has limited potential to remove the estrogenic activity effectively. The contributions of the estrogenic chemicals to the estrogenic activities were 70.7, 23.3, 3.7 and 2.3% for estrone, 17$\beta$-estradiol 17$\alpha$-ethinylestradiol and APE, respectively, in the domestic line effluents. Therefore, 17$\beta$-estradiol and estrone contributed most of the estrogenic activity in the domestic line effluents.

  • Thin layer, paper and column chromatography were compared for the separation, detection and quantitation of three kinds of estrogen in urine of dairy cows. While thin layer chromatography utilizing silica gel was better for the detection of estrogens, column chromatography using celite 545 was preferable. Spectrophotometry was compared with fluorometry for determination of estrone, estradiol-17 ${\beta}$ and estriol eluted by paper chromatography and column chromatography. Optical density of three standard estrogens showed almost same curve at maximum absorption wave length of 230 and $282m{\mu}$. However, the former showed a higher peak. In fluorometry, the fluorescence intensity of estrone and estradiol-17 ${\beta}$ were rather strong, when the estrogens were dissolved in sulfuric acid, and showed higher sensitivity than that of the spectrophotometry. However, in the case of estriol was exceptional.

  • The release of steroids such as progesterone, testosterone, estrone and estradiol were investigated in two types of block copolymer matrices, which were AB type block copolymer composed of poly(γ-benzyl-L-glutamate)[PBLG] as the A segment and polyoxypropylene[POP] as the B segment and ABA type triblock copolymer composed of the A segment and poly(propylene glycol)[PPG] as the B segment. The release rates of progesterone, estradiol and testosterone through PBLG-POP1 and PBLG-POP2 matrices and those of progesterone, estrone and testosterone through GPG1 of ABA type triblock copolymer were higher in an acidic solution than in a basic one. However, the release rate of estrone through PBLG-POP1 and PBLG-POP2 matrices and that of estradiol through GPG1 of ABA type triblock copolymer were higher in a basic and neutral solutions than in an acidic one. The order of release rate in three different matrices was GPG1 > PBLG-POP1 > PBLG-POP2. The results also showed that the molecular weight of steroids is inversely proportional to the rate of release.

  • When 19-hydroxycholesterol acetate was added into CSD-10 in Nutrient Broth or in a mineral salts medium consisting of KH$_{2}$PO$_{4}$(0.1%), $K_{2}$HPO$_{4}$(0.1%), NH$_{4}$NO$_{3}$(0.1%), MgSO$_{4}$(0.02%), CaCl$_{2}$(0.002%), and FeCl$_{3}$(0.005%), a substantial amount of 19-hydroxyandrost-4-ene-3,17-dione was accumulated prior to accumulation of estrone. From this result and all of previous works, a tentative degradation pathway of 19-hydroxycholesterol acetate to estrone by CSD-10 was derived. 19-hydroxyandrost-4-ene-3,17-dione seems to be an attractive intermediate for the synthesis of 19-norsteroids.
