Mucin in cancer: a stealth cloak for cancer cells

  • Mucins are high molecular-weight epithelial glycoproteins and are implicated in many physiological processes, including epithelial cell protection, signaling transduction, and tissue homeostasis. Abnormality of mucus expression and structure contributes to biological properties related to human cancer progression. Tumor growth sites induce inhospitable conditions. Many kinds of research suggest that mucins provide a microenvironment to avoid hypoxia, acidic, and other biological conditions that promote cancer progression. Given that the mucus layer captures growth factors or cytokines, we propose that mucin helps to ameliorate inhospitable conditions in tumor-growing sites. Additionally, the composition and structure of mucins enable them to mimic the surface of normal epithelial cells, allowing tumor cells to escape from immune surveillance. Indeed, human cancers such as mucinous carcinoma, show a higher incidence of invasion to adjacent organs and lymph node metastasis than do non-mucinous carcinoma. In this mini-review, we discuss how mucin provides a tumor-friendly environment and contributes to increased cancer malignancy in mucinous carcinoma.

  • Bacterial infection is a very complex process in which both pathogens and host cells play crucial roles, and the host cells undergo drastic changes in their physiology, releasing various proteins in response to the pathogenic infection. Human airway epithelial surface serves as a first line of defense against microorganisms and the external environment. It is well known that bronchial epithelial cells secrete various chemokines and cytokines such as IL-6 and IL-8 to cope with various respiratory pathogens. (omitted)

Endoplasmic Reticulum Stress Induces CAP2 Expression Promoting Epithelial-Mesenchymal Transition in Liver Cancer Cells

  • Cyclase-associated protein 2 (CAP2) has been addressed as a candidate biomarker in various cancer types. Previously, we have shown that CAP2 is expressed during multi-step hepatocarcinogenesis; however, its underlying mechanisms in liver cancer cells are not fully elucidated yet. Here, we demonstrated that endoplasmic reticulum (ER) stress induced CAP2 expression, and which promoted migration and invasion of liver cancer cells. We also found that the ER stress-induced CAP2 expression is mediated through activation of protein kinase C epsilon (PKCε) and the promotor binding of activating transcription factor 2 (ATF2). In addition, we further demonstrated that CAP2 expression promoted epithelial-mesenchymal transition (EMT) through activation of Rac1 and ERK. In conclusion, we suggest that ER stress induces CAP2 expression promoting EMT in liver cancer cells. Our results shed light on the novel functions of CAP2 in the metastatic process of liver cancer cells.

Gene expression profiling after ochratoxin A treatment in small intestinal epithelial cells from pigs

  • Ochratoxin A (OTA) is a well-known mycotoxin that causes disease through the ingestion of contaminated food or feed, for example, in the porcine industry. The intestinal epithelium acts as the first barrier against food contamination. We conducted a study on the exposure of the porcine intestinal epithelium to OTA. We used the intestinal porcine epithelial cell line IPEC-J2 as an in vitro model to evaluate the altered molecular mechanisms following OTA exposure. Gene expression profiling revealed that OTA upregulated 782 genes and downregulated 896, totalling 1678 differentially expressed genes. Furthermore, immunofluorescence, quantitative real-time polymerase chain reaction, and western blotting confirmed that OTA damages the tight junction protein ZO-1. Moreover, OTA activated the expression of inflammatory genes (IL-6, IL-8, IL-10, NF-kB, TLR4, and TNF-α). In summary, this study confirmed that OTA alters various molecular mechanisms and has several adverse effects on IPEC-J2 cells.

Involvement of IKK/IkBα/NF-kB p65 Signaling into the Regulative Effect of Engeletin on MUC5AC Mucin Gene Expression in Human Airway Epithelial Cells

  • In this study, we examined whether engeletin exerts an effect on the gene expression of MUC5AC mucin, in human pulmonary epithelial NCI-H292 cells. The cells were pretreated with engeletin for 30 min and stimulated with phorbol 12-myristate 13-acetate (PMA), for the following 24 h. The effect of engeletin on PMA-induced nuclear factor kappa B (NF-kB) signaling pathway was also investigated. Engeletin suppressed the mRNA expression and production of MUC5AC mucin, induced by PMA through the inhibition of degradation of inhibitory kappa Bα (IkBα) and NF-kB p65 nuclear translocation. These results suggest engeletin inhibits the gene expression of mucin through regulation of NF-kB signaling pathway, in human airway epithelial cells.

Generation and analysis of whole-genome sequencing data in human mammary epithelial cells

  • Breast cancer is the most common cancer worldwide, and advanced breast cancer with metastases is incurable mainly with currently available therapies. Therefore, it is essential to understand molecular characteristics during the progression of breast carcinogenesis. Here, we report a dataset of whole genomes from the human mammary epithelial cell system derived from a reduction mammoplasty specimen. This system comprises pre-stasis 184D cells, considered normal, and seven cell lines along cancer progression series that are immortalized or additionally acquired anchorage-independent growth. Our analysis of the whole-genome sequencing (WGS) data indicates that those seven cancer progression series cells have somatic mutations whose number ranges from 8,393 to 39,564 (with an average of 30,591) compared to 184D cells. These WGS data and our mutation analysis will provide helpful information to identify driver mutations and elucidate molecular mechanisms for breast carcinogenesis.

  • Endoscopic submucosal dissection (ESD) is the standard treatment method for esophageal, gastric, and colorectal cancers. However, it has not been standardized for duodenal lesions because of its high complication rates. Recently, minimally invasive and simple methods such as cold snare polypectomy and underwater endoscopic mucosal resection have been utilized more for superficial nonampullary duodenal epithelial tumors (SNADETs). Although the rate of complications associated with duodenal ESD has been gradually decreasing because of technical advancements, performing ESD for all SNADETs is unnecessary. As such, the appropriate treatment plan for SNADETs should be chosen according to the lesion type, patient condition, and endoscopist's skill.

흉선종의 WHO 분류에 따른 예후인자의 임상적 고찰 (Analysis of Prognostic Factors according to WHO Classification for Thymic Epithelial Tumor)

  • 배경: 원발성 흉선상피세포종(primary thymic epithelial neoplasm)은 다양한 생물학적 및 형태학적인 특성을 가진 종격동 종양이다. 이 연구에서는 59예의 흉선종 환자를 새로운 WHO분류에 따라 재분류하고, Masaoka 임상적 병기를 비롯한 여러 가지 임상적인 특성을 분석함으로써 예후에 영향을 미치는 인자를 알아보고 WHO 분류가 독립적인 예후인자가 될 수 있는지를 알아보고자 했다. 대상 및 방법: 1986년 12월부터 2003년 8월까지 계명대학교 동산의료원에서 흉선종으로 진단 받은 후 수술적 치료를 받은 59예를 대상으로 생존율에 영향을 미치는 예후인자를 흉선종의 조직학적 분류(WHO classification)와 임상적 병기(Masaoka's clinical stage) 및 환자의 특성(성별, 나이, 중증근무력증 유무, 종괴의 크기, 종괴의 침습 유무, 재발, 전이)에 따라 분석하였다. 그리고 조직학적 분류와 임상적 병기사이의 연관성을 분석하였다. 결과: 남자는 32예, 여자는 27예였으며 평균연령은 50.1$\pm$14.2세였다. 모든 흉선종은 WHO 분류에 따라 A부터 C까지 재분류하였다. WHO 분류에 따라 Type A는 6명(10.2%)이었고 AB는 7명(11.9%), B$_1$은 7명(11.9%), B$_2$는 10명(16.9%), B$_3$는 7명(11.9%), C는 22명(37.3%)으로 나타났다. 여러 가지 임상적 특성 가운데 다변수 분석을 통해 완전절제(p=0.033)와 주변조직으로의 저침습성(p=0.001)이 좋은 예후와 연관이 있는 것으로 나타났다. 전체 흉선종의 5년 생존율은 53%였으며 WHO 분류에 따른 5년 생존율은 Type A+AB가 92.3%, B$_1$+B$_2$가 70.2%, B$_3$+C가 26.1%로 나타났다. WHO 조직학적 분류와 생존율 사이의 상관관계분석에서 WHO 조직학적 분류는 예후와 통제적으로 유의하게 연관성이 있는 것으로 나타났다(p<0.001). WHO 분류에 따른 Masaoka 임상병기 분석에서도 두 변수가 통계적으로 유의하게 연관성을 가지는 것으로 나타났다(p<0.001). 결론: WHO 분류는 단순한 조직학적인 분류일 뿐만 아니라 흉선종의 생존율에 중요한 영향을 미치며 종양의 침습성을 잘 반영하는 예후인자임을 알 수 있었다.

청조구폐탕(淸燥救肺湯)과 이음전(理陰煎)이 호흡기 접액분비에 미치는 영향 (Effects of Cheongjogupye-tang(淸燥救肺湯) and Yieum-jeon(理陰煎) on Secretion of Mucin from Respiratory Epithelial Cells)

  • Objectives : In this study, the author tried to examine whether Cheogjogupye-tang (淸燥救肺湯, CGPT) and Yieum-jeon (理陰煎, YEJ) significantly affect in vitro and in vivo mucin secretion, MUC5AC gene expression in airway epithelial cells and contractility of isolated tracheal smooth muscle of rabbit. Materials and Methods : For in vitro experiment, confluent hamster tracheal surface epithelial (HTSE) cells were chased for 30 minutes in the presence of CGPT and YEJ to assess the effects of the agents on mucin secretion by enzyme-linked immunosorbent assay (ELISA), with removal of oriental herbal medicine extract from each agent-treated sample by centrifuge microfilter. Also, the effects of the agents on TNF-alpha or EGF-induced MUC5AC gene expression in human airway epithelial cells (NCI-H292) were investigated. Possible cytotoxicities of the agent were assessed by examining both LDH release from HTSE cells and the rate of survival and proliferation of NCI-H292 cells. For in vivo experiment, hypersecretion of airway mucin and goblet cell hyperplasia was induced by exposure of rats to $SO_2$ over 3 weeks. Effects of CGPT and YEJ orally administered for 1 week on in vivo mucin secretion from tracheal goblet cells of rats and hyperplasia of goblet cells were assessed using ELISA and histological analysis after staining the epithelial tissue with alcian blue, respectively. Also, the effects of CGPT and YEJ on contractility of isolated tracheal smooth muscle were investigated. Results : (1) CGPT significantly inhibited in vitro mucin secretion from cultured HTSE cells. However, YEJ did not affect in vitro mucin secretion; (2) CGPT and YEJ did not affect hypersecretion of in vivo mucin and hyperplasia of tracheal goblet cells; (3) CGPT and YEJ slightly increased the expression levels of TNF-alpha or EGF-induced MUC5AC gene in NCI-H292 cells; (4) CGPT and YEJ inhibited acetylcholine-induced contraction of isolated tracheal smooth muscle of rabbit; (5) CGPT and YEJ did not affect LDH release from HTSE cells and the survival and proliferation of NCI-H292 cells. Conclusion : The results from the present study suggest that CGPT and YEJ mainly affect the expression of mucin gene rather than secretion of mucin and do not show remarkable cytotoxicity to respiratory epithelial cells.

꼬막 Tegillarca granosa 심신낭복합체의 미세해부학적 특징 (Microanatomy of the Heart-kidney Complex of Tegillarca granosa (Bivalvia: Arcidae))

  • 꼬막 심신낭복합체의 미세해부학적 구조를 해부 및 광학현미경을 이용하여 기재하였다. 심신낭복합체는 발 기저부 위쪽, 내장낭 등쪽 부근과 후폐각근 사이의 위심강 안에 위치하였다. 심장은 하나의 심실과 두 개의 심방으로 구성되었으며, 신장은 좌 우 한 쌍으로 구성되어 있었다. 심방과 심실은 심외막, 심근, 심내막으로 구성된다. 심외막은 단층상피층으로서 입방형 상피세포로 구성되며, 이들 상피세포의 핵은 중앙에 위치하고 강한 호염기성을 나타냈다. 심근은 다수의 근섬유 다발로 구성되며, 근섬유 다발은 심방보다 심실에서 더 치밀하게 나타났다. 심내막은 단층상피층으로서 편평상피세포로 구성되며, 핵은 중앙에 위치하고 강한 호염기성을 나타냈다. 심방에서 심내막의 두께는 $6.04({\pm}2.26){\mu}m$였고, 심실에서 심내막의 상피층 두께는 $7.36({\pm}3.21){\mu}m$로 심방에서보다 심실에서 더 두껍게 나타났다. 신장은 다수의 세뇨관으로 모인 집합적 구조였으며, 신장의 세뇨관은 단층상피층으로서 원주형 상피세포로 구성되었다. 핵은 기저부에 존재하였으며, 세포질에는 다수의 과립들이 관찰되었고 자유면에는 선조연이 발달되어 있었다.
