Water quality and epiphytic diatom on the submerged stems of reed (Phragmites communis), which occupy 90% of the Donghwa wetland macrophytes were monthly monitored at three points such as inflow stream, high- and low-level wetlands, and outflow stream between March and October, 2005. 1) A diverse and high density of diatom species observed in the cold-season, especially Nitzschia palea and Nitzschia amphibia dominated the diatom community without wetlands. 2) High DAIpo and TDI indices were measured over the sampling periods and stations, regardless of nitrogen increase and phosphorus increase through the wetlands. 3) Higher density of diatom species in high wetland than low wetlands was attributed in the enough nutrients and light penetration by low growth of reed. Therefore, epiphytic diatom of reed stem in Donghwa wetland, where high nutrients released from the sediment and reed debris after the death of macrophytes, flourished with low canopy of low reed vegetation.
The effects of sediment and water movement on the survival and growth of Campylaephora hypnaeoides J. Agardh were examined in culture to elucidate why this species grows as an epiphytic alga. The survival and growth rates of Campylaephora tetraspores declined as the sediment concentration increased under both water-movement and no-water-movement treatments. After 5-day cultures under various sediment treatments, the respective survival and growth rates ranged from 27.32 to 78.63% and 0.22 to 0.29 $day^{-1}$ under the no-water-movement and from 15.66 to 82.69% and 0.19 to 0.31 $day^{-1}$ under the water-movement treatments. The maximum survival and growth of C. hypnaeoides tetraspores occurred at 20 and 40 ${\mu}mol$ photons $m^{-2}s^{-1}$, respectively. The survival of Campylaephora tetraspores and germlings declined with increasing dark period, but the tolerance of total darkness based on the survival rates was two-times greater for 5-day old sporelings than for tetraspores after 12 days. In conclusion, Campylaephora hypnaeoides grows as an epiphytic alga because its survivorship and growth decline rapidly with greater water movement and sedimentation and with lower light intensities.

Salmonellosis is a form of gastroenteritis caused by Salmonella infection. The main transmission route of salmonellosis has been identified as poorly cooked meat and poultry products contaminated with Salmonella. However, in recent years, the number of outbreaks attributed to contaminated raw produce has increased dramatically. To understand how Salmonella adapts to produce, transcriptomic analysis was conducted on Salmonella enterica serovar Virchow exposed to fresh-cut radish greens. Considering the different Salmonella lifestyles in contact with fresh produce, such as motile and sessile lifestyles, total RNA was extracted from planktonic and epiphytic cells separately. Transcriptomic analysis of S. Virchow cells revealed different transcription profiles between lifestyles. During bacterial adaptation to fresh-cut radish greens, planktonic cells were likely to shift toward anaerobic metabolism, exploiting nitrate as an electron acceptor of anaerobic respiration, and utilizing cobalamin as a cofactor for coupled metabolic pathways. Meanwhile, Salmonella cells adhering to plant surfaces showed coordinated upregulation in genes associated with translation and ribosomal biogenesis, indicating dramatic cellular reprogramming in response to environmental changes. In accordance with the extensive translational response, epiphytic cells showed an increase in the transcription of genes that are important for bacterial motility, nucleotide transporter/metabolism, cell envelope biogenesis, and defense mechanisms. Intriguingly, Salmonella pathogenicity island (SPI)-1 and SPI-2 displayed up- and downregulation, respectively, regardless of lifestyles in contact with the radish greens, suggesting altered Salmonella virulence during adaptation to plant environments. This study provides molecular insights into Salmonella adaptation to plants as an alternative environmental reservoir.

Occurrence of epiphytic dinoflagellates (EPDs) in coastal waters off Jeju was first reported in 2011 based on 45 substrate samples from 24 macroalgal species. When re-analyzing, the extreme heterogeneous distribution of whole and genus-specific EPDs was reconfirmed across the sampling stations and substrate macroalgal species, as well as even across substrate samples of the same species. Abundance maximum of an EPD genus (cells g-wwt-1) at a fixed surface-to-volume ratio (SA/V ratio) of the macroalgal species increased as the SA/V ratio increased up to 500 (cm2 cm-3). However, the abundance maximum of Ostreopsis further increased even in the MG2 (morphological group 2) macroalgae with the SA/V ratios over 500. The number of substrate macroalgal species on the plane of the MG and sampling station was more or less evenly scattered than the average EPD abundance, which was primarily driven by Gambierdiscus and Ostreopsis. Of the total EPD abundance of the five stations, 90.6% were represented by the two most common and abundant genera, Gambierdiscus and Ostreopsis, each accounting for 41.6% and 49.0%. Spatially, 95.9% of the total EPD abundance was found in St. 4 and St. 5, of which St. 4 with higher water temperature had more Ostreopsis spp. (31.8%), and St. 5 with higher salinity had more Gambierdiscus spp. (27.3%). Thus, the environmental transition to favorable T-S condition to MG2, the thin filamentous macroalgal group with very high SA/V ratios, is thus likely to support further success in EPD genera led by Ostreopsis in the coastal waters of Jeju.
Epifluorescence microscopy was used to observe epiphytic bacteria directly on plant leaf surfaces as well as indirectly in the leaf liberating solution by staining with fluorochromes of 4',6-diamidino-2-phenylindole (DAPI) and acridine orange(AO). Epiphytic bacteria could not be well observed on the leaf surface by staining with AO due to an intrusive orange or red background fluorescence. However, DAPI gave us clear epifluorescent images of the bacteria on the leaf. On the contrary, epiphytic bacteria in the liberating leaf solution were well observed on filters stained by both types of fluorochrome, although DAPI showed better fluorescent images than AO and not necessarily required a washing step of the filters stained. The optimum conditions of the DAPI stains were 5 $\mu$g/ml for 5 min both for leaves and for filters of the liberating solution. It was confirmed that a critical step in the epifluorescence microscopy of leaf surfaces was to minimize release of water from the leaf. For this, the stained leaf samples were put on a filter paper, kept in a dry oven at $70^{\circ}C$ for 2 min instead of air-drying, and then immediately observed by epifluorescence microscopy. The established technique was applied to enumerate epiphytic bacteria on oak tree leaf surfaces.
Elachista tenuis Yamada, epiphytic on Sargassum confusum, is reported for the first time in Korea. It forms hemispherical tufts by pseudoparenchymatous medulla and assimilatory filaments. The cells ofassimilatory filament are not constricted at septa, and nearly equal in breadth from base to apex. Plurilocular sporangia are rather abundant, while unilocular sporangia rare.
The total methylotrophic population associated with rice plants from different cultivars was enumerated at three different stages: vegetative, flowering, and harvesting. The bacterial population in the leaf, rhizosphere soil, endophytic in the stem and roots, and epiphytic in the florets and grains were determined from four rice cultivars, Il-mi, Nam-pyeoung, O-dae, and Dong-jin, sampled from three different field sites. The methylotrophic bacteria isolated on AMS media containing 0.5% methanol as the sole carbon source uniformly showed three distinct morphologies, which were recorded as separate groups and their distribution among the various samples was determined using the ecophysiological index. The growth stage at the time of sampling had a more significant effect on the methylotrophic population and their distribution than the field site or cultivar. A similar effect was also observed for the PPFMs, where their population in different plant parts increased from V10 to R4 and then decreased towards stage R9. A canonical discriminant analysis of the PPFM population from different parts of rice showed clear variations among the cultivars, sampled sites, and growth stages, although the variations were more prominent among the growth stages.
Plant leaf surface is an important niche for diverse epiphytic microbes, including bacteria and fungi. Plant leaf surface plays a critical frontline defense against pathogen infections. The objective of our study was to evaluate the effectiveness of a starch-based super-absorbent polymer(SAP) combination, which enhances water potential and nutrient availability to plant leaves. We evaluated the effect of SAP on the maintenance of bacterial populations. In order to monitor bacterial populations in situ, a SAP mixture containing Pseudomonas syringae pv. tabaci that expressed recombinant green fluorescent protein(GFPuv) was spray-challenged onto whole leaves of Nicotiana benthamiana. The SAP combination treatment enhanced bacterial robustness, as indicated by disease severity and incidence. Unexpectedly, bacterial numbers were not significantly different between leaves treated with the SAP combination and those treated with water alone. Furthermore, young leaves treated with the SAP combination had more severe symptoms and a greater number of bacterial spots caused by primary and secondary infections compared to young leaves treated with the water control. In contrast, bacterial cell numbers did not statistically differ between the two groups, which indicated that measurement of viable GFP-based bacterial spots may provide a more sensitive methodology for assessing virulence of bacterial pathogens than methods that require dilution plating following maceration of bacterial-inoculated leaf tissue. Our study suggests that the SAP combination successfully increased bacterial aggressiveness, which could either be used to promote the ability of biological agents to control weedy plants or increase the robustness of saprophytic epiphytes against competition from potentially harmful microbes.
