마늘의 가공 조리방법에 따른 Lipoxygenase활성도 저해효과 -마늘 추출액이 Lipoxygenase 활성도 저해에 미치는 영향- (Inhibition of Lipoxygenase Activity by the Extract of Various Processed Garlic - Inhibitory Effect of Garlic Extracts on Soybean Lipoxygenase Activity -)

  • 동물체내에서 천식, 염증, 혈소판 응고 등에 관련된 매개체를 생성시키는 효소인 lipoxygenase작용기 전과 유사한 대두 lipoxygenase (Type IV)를 사용하여 생마늘의 수용액, 에탄올, 클로로포름 추출분획에 의한 lipoxygenase 저해 정도를 측정하였다. 효소를 클로로포름 추출 분획과 10분 preincuba-tion시킨 후의 효소 저해 양상은 비가역적 저해(1$_{50}$값, 55mg/$m\ell$)이었으며, 수용액 추출 분획의 경우는 주로 가역적 저해 양상(1$_{50}$값, 65mg/$m\ell$)을 나타내었다. 한편, diallyldisulfide와 dimethyldisulfide의 1$_{50}$값은 각각 1.3mM, 18mM 이었으며 이들은 가역적, 비가역적 저해 현상을 모두 나타내었다. 합성품 alliin은 비교적 높은 농도(10mM농도에서 22%저해)에서 저해하였으며, alliin의 분해산물은 비가역적 저해 양상을 나타낸 반면에, S-ethylcysteine sulfoxide는 효소를 거의 저해하지 않았다. 따라서 다진 마늘 속에는 가역적 저해제가 주로 함유되어 있고, 소량(25~30%)의 비가역적 저해제가 함유되어 있는 것으로 사료되었다.사료되었다.

p-Coumaric Acid Potently Down-regulates Zebrafish Embryo Pigmentation: Comparison of in vivo Assay and Computational Molecular Modeling with Phenylthiourea

  • p-Coumaric acid is an organic compound that is a hydroxyl derivative of cinnamic acid. Due to its multiple biological activities p-coumaric acid has been widely studied in biochemical and cellular systems and is also considered as a useful therapeutic candidate for various neuronal diseases. However, the efficacy of p-coumaric acid on zebrafish developmental regulation has not been fully explored. In this study, therefore, we first investigated the action mechanism of the p-coumaric acid on the zebrafish development in a whole-organism model. p-Coumaric acid treated group significantly inhibited the pigmentation of the developing zebrafish embryos compared with control embryos without any severe side effects. In addition, p-coumaric acid down-regulated more effectively in a lower concentration than the well-known zebrafish's melanogenic inhibitor, phenylthiourea. We also compared the molecular docking property of p-coumaric acid with phenylthiourea on the tyrosinase's kojic acid binding site, which is the key enzyme of zebrafish embryo pigmentation. Interestingly, p-coumaric acid interacted with higher numbers of the amino acid residues and exhibited a tight binding affinity to the enzyme than phenylthiourea. Taken all together, these results strongly suggest that p-coumaric acid inhibits the activity of tyrosinase, consequently down-regulating zebrafish embryo pigmentation, and might play an important role in the reduction of dermal pigmentation. Thus, p-coumaric acid can be an effective and non-toxic ingredient for anti-melanogenesis functional materials.

오공(蜈蚣)의 항산화효과에 관한 연구 (Antioxidative Effects of Scolopendra subspinipes)

  • Objective: The purpose of this study was to evaluate the antioxidative effects of the extract of Scolopendra subspinipes which has been used mainly for detoxication in the oriental medicine and reported to have sedative action, antiinflammatory effect, antihypertensive property and immunity enhancing activity. Method: Inhibitory activities on oxygen radical generating enzymes (aldehyde oxidase and xanthine oxidase) and increasing activities on oxygen radical scavenging enzymes (superoxide dismutase, glutathione peroxidase, glutathione-S-transferase) were investigated. Furthermore, the content of glutathione in the mouse brain, DPPH radical scavenging activity and also anti-lipid peroxidative effects in vivo and in vitro were estimated. Results: The extract showed weak inhibitory effects on the activities of aldehyde oxidase and xanthine oxidase which are oxygen radical generating enzymes. The extract inhibited lipid peroxidation with 26.1% against control group at 500 mg/kg in vivo and with 11.2% against control group at 10 mg/kg in vitro in a dose-dependent manner, which means this drug may protect radical-induced cell damages. The extract showed dose-dependently the scavenging effect on DPPH radical with 24.8% activity at 10 mg/ml in vitro. The extract enhanced the activities of superoxide dismutase, glutathione peroxidase and glutathione-S-transferase, which are oxygen radical scavenging enzymes, with 28.9%, 22.3% and 23.1%, respectively at 500mg/kg in vivo. Finally, this extract strongly increased the glutathione content in the mouse barin. Conclusion: Above results indicated that Scolopendra subspinipes can be useful for the protection or treatment of some diseases caused by reactive oxygen species.

  • 참깨박에 함유되어 있는 불용성 형태의 단백질을 Bacillus sp. CW-1121이 생성하는 효소를 이용하여 가용성 형태의 단백질로 용출시키기 위하여 효소를 작용시켰다. 효소 처리 참깨박 단백질의 기포 형성력은 염용성 단백질이 수용성 단백질에 비해 현저히 낮았다. 기포 안정성은 염용성 단백질이 10분 이내 크게 감소된 반면 수용성 단백질의 기포 안정성은 30분 까지 완만하게 감소하였다. 유화력은 각 단백질의 등전점 부근에서 가장 낮았으며, 염용성 단백질이 수용성 단백질에 비해 낮았다 유화 안정성은 유화력과 비슷한 크기로 나타나 $80^{\circ}C$에서의 30분 가열에서도 이들 단백질이 열에 안정함을 보였다. 유지 및 수분 흡착력은 염용성 단백질이 수용성 단백질 보다 높은 값을 나타내었다.

Concrete crack rehabilitation using biological enzyme

  • Concrete is a material popularly used in construction. Due to the load-bearing and external environmental factors during utilization or manufacturing, its surface is prone to flaws, such as crack and leak. To repair these superficial defects and ultimately and avoid the deterioration of the concrete's durability, numerous concrete surface protective coatings and crack repair products have been developed. Currently, studies are endeavoring to exploit the mineralization property of microbial strains for repairing concrete cracks be the repairing material for crack rehabilitation. This research aims to use bacteria, specifically B. pasteurii, in crack rehabilitation to enhance the flexural and compression strength of the repaired concrete. Serial tests at various bacterial concentrations and the same $Urea-CaCl_2$ medium concentration of 70% for crack rehabilitation were executed. The results prove that the higher the concentration of the bacterial broth, the greater the amount of calcium carbonate precipitate was induced, while using B. pasteurii broth was for crack rehabilitation. The flexural and compression strengths of the repaired concrete test samples were the greatest at 100% bacterial concentration. Compared to the control group (bacterial concentration of 0%), the flexural strength had increased by 32.58% for 1-mm crack samples and 51.01% for 2-mm crack samples, and the compression strength had increased by 28.58% and 23.85%, respectively. From the SEM and XRD test results, a greater quantity of rectangular and polygonal crystals was also found in samples with high bacterial concentrations. These tests all confirm that using bacteria in crack rehabilitation can increase the flexural and compression strength of the repaired concrete.

Pseudomonas sp.가 생산하는 Inulinase에 관한 연구 -효소의 정제와 성질 - (Purification and Properties of Extracellular Inulinase of Pseudomouas sp.)

  • Pseudomonas sp.로 동정된 토양분리균의 inulinase를 ammonium sulfate 분획, DEAE Sephadex A-50 Chromatography, Sephadex G 100 및 Sephadex G200 gel여과 등의 과정을 거쳐 정제 한 결과 inulinase PI과 PII의 두 isoenzyme으로 분리되었으며 각각 약 24배와 17배 정제되어 단일단백질로 분리되었다. PI, PII 두 isoenzyme은 다같이 sucrose, raffinose 및 levan은 분해하지 못하고 inulin만을 endo-type로 분해 절단하는 $\beta$-2,1-fructanfructano hydrolase(EC 3.2.1.7)임이 밝혀졌다. 효소활성 최적온도는 두 효소가 같은데 비해 (55$^{\circ}C$), 최적 pH는 PI이 pH5.5, PII가 pH6.0으로 약간의 차이를 보였다. Inulin에 대한 Km값은 PI ; 2$\times$$10^{-3}$M, PII : 5$\times$$10^{-3}$M로써 PI효소가 PII보다 약간 높은 친화력을 보였다.

Characterization of Lipases from Staphylococcus aureus and Staphylococcus epidermidis Isolated from Human Facial Sebaceous Skin

  • Two staphylococcal lipases were obtained from Staphylococcus epidermidis S2 and Staphylococcus aureus S11 isolated from sebaceous areas on the skin of the human face. The molecular mass of both enzymes was estimated to be 45 kDa by SDS-PAGE. S2 lipase displayed its highest activity in the hydrolysis of olive oil at $32^{\circ}C$ and pH 8, whereas S11 lipase showed optimal activity at $31^{\circ}C$ and pH 8.5. The S2 lipase showed the property of cold-adaptation, with activation energy of 6.52 kcal/mol. In contrast, S11 lipase's activation energy, at 21 kcal/mol, was more characteristic of mesophilic lipases. S2 lipase was stable up to $45^{\circ}C$ and within the pH range from 5 to 9, whereas S11 lipase was stable up to $50^{\circ}C$ and from pH 6 to 10. Both enzymes had high activity against tributyrin, waste soybean oil, and fish oil. Sequence analysis of the S2 lipase gene showed an open reading frame of 2,067 bp encoding a signal peptide (35 aa), a pro-peptide (267 aa), and a mature enzyme (386 aa); the S11 lipase gene, at 2,076 bp, also encoded a signal peptide (37 aa), pro-peptide (255 aa), and mature enzyme (399 aa). The two enzymes maintained amino acid sequence identity of 98-99% with other similar staphylococcal lipases. Their microbial origins and biochemical properties may make these staphylococcal lipases isolated from facial sebaceous skin suitable for use as catalysts in the cosmetic, medicinal, food, or detergent industries.

A Novel Strain of Cucumber mosaic virus Isolated from Lilium longiflorum

  • A new strain of Cucumber mosaic virus (CMV) from easter lily (Lilium longiflorum), Ly2-CMV, was identified and compared to the well-characterized Mf-CMV (subgroupⅠ) and LS-CMV (subgroupⅡ) by host reaction in several indicator plants, dsRNA analysis, serological property, RT-PCR analysis, restriction enzyme profile of the PCR products and nucleotide sequence of coat protein (CP) gene. Remarkable differences in symptoms of Ly2-CMV were found between Mf-CMV or LS-CMV in tobacco plants and Datura stramoinium. Ly2-CMV induced small necrotic ringspots on the inoculated leaves of Nicotiana tabacum cvs. Xanthi nc and Burley 21 and D. stramonium, and failed to infect these species systemically. Of the indicator plants tested, N. benthamiana only reacted with systemic infection by inoculation of Lr2-CMV. In experiments of dsRNA analysis, serology and RT-PCR of CP gene, Ly2-CMV was come within subgroupⅠ CMV. However, restriction enzyme analysis of the PCR products using MspⅠ showed that Ly2-CMV was distinct to Mf-CMV. The CP gene of Ly2-CMV contains 657 nucleotides, and the nucleotide sequence is similar to that of Mf-CMV. There is also a high degree of conservation between their putative gene products in Ly2-CMV and Mf-CMV, with five amino acid changes in the 218 amino acids of the CPs.

데침과 열수의 침적과 Trehalose 처리가 오이 김치의 저장중 효소 활성의 변화와 관능 검사에 미치는 병용효과 (The Combined Effect of Enzyme Activity and Sensory Test of Blanching and Brining in Hot Solution and Trehalose Treatment on the Cucumber Kimchi for the Storage Period)

  • 1. 비가열 시료인 a와 b가 PE의 활성은 저장 둘째 주에 급격히 증가하다가 감소하였고, 가열 시료인 c 는 저장 넷째 주에 증가하다가 감소하였고, 병행 열처리와 trehalose 처리 시료는 저장 끝까지도 증가하였다. 2. PG의 활성은 비가열 시료인 a와 b는 저장 둘째 주부터 감소하다 저장 끝에는 증가하였고, 가열 시료인 c와 d는 저장 기간 동안 변화가 거의 없었다. 3. POD의 활성은 비가열 시료인 a와 b는 저장 둘째 주에 증가하다 감소하였고, 가열 시료인 c는 둘째 주에 약간씩 감소하였고, 병용 열처리하고 trehalose 처리군은 감소하였다. 4. 관능검사 결과 시료 a. b, d는 아작함과 기호도가 전 저장 기간동안 높았고, 시료 c는 갈색과 신맛의 증가로 기호도가 낮았다 저장기간에 따라서는 2주차에는 아작함과 형태가 좋았으나 4주 6주로 가면서 아작함과 기호도의 값이 감소하였다. 그리고 각 시료별 기호도에 가장 영향을 주는 항목은 아작함이었고 시료 c는 갈색, 맛, 조직감 등에서 음의 관계를 나타냈다.

Synthesis and Characterization of Magnetic Nanoparticles and Its Application in Lipase Immobilization

  • We demonstrate herein the synthesis and modification of magnetic nanoparticles and its use in the immobilization of the lipase. Magnetic $Fe_3O_4$ nanoparticles (MNPs) were prepared by simple co-precipitation method in aqueous medium and then subsequently modified with tetraethyl orthosilicate (TEOS) and 3-aminopropyl triethylenesilane (APTES). Silanization magnetic nanoparticles (SMNP) and amino magnetic nanomicrosphere (AMNP) were synthesized successfully. The morphology, structure, magnetic property and chemical composition of the synthetic MNP and its derivatives were characterized using transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR) analysis, X-ray diffraction, superconducting quantum interference device (SQUID) and thermogravimetric analyses (TGA). All of these three nanoparticles exhibited good crystallization performance, apparent superparamagnetism, and the saturation magnetization of MNP, SMNP, AMNP were 47.9 emu/g, 33.0 emu/g and 19.5 emu/g, respectively. The amino content was 5.66%. The AMNP was used to immobilize lipase, and the maximum adsorption capacity of the protein was 26.3 mg/g. The maximum maintained activity (88 percent) was achieved while the amount of immobilized lipase was 23.7 mg $g^{-1}$. Immobilization of enzyme on the magnetic nanoparticles can facilitate the isolation of reaction products from reaction mixture and thus lowers the cost of enzyme application.
