동결건조한 키위분말을 첨가한 우육의 연육 효과 (A study on the Effect of Kiwi fruit powder after Freeze drying on Tenderization of Beef)

  • The purpose of this study was to investigate the effects of protease in kiwi fruit powder after freeze drying which has the ratio of 0%, 1%, 2%, and 3% on the tenderization of the bovine longissimus dorsi muscle. Beef loin chunks were marinated in distilled water (Control), 1% kiwi powder (K1), 2% kiwi powder (K2), and 3% kiwi powder (K3). As a result, the enzyme activities have shown to have higher activity (p<0.001) as the amount of freeze-dried kiwi powder increased. There are significant difference in pH (p<0.01), color of the beef were slightly different between the C (control) group and the sample groups. The cooking loss showed the highest value of K3 (p<0.001), and water holding capacity showed the highest value of K3. Furthermore, the sample groups exhibited lower shear force values compared with the control (p<0.001).

Evaluation of Feeding a Fibrolytic Enzyme to Lactating Dairy Cows on Their Lactational Performance during Early Lactation

  • Twenty eight multiparous lactating cows were utilized in an experiment to evaluate the response to an exogenous fibrolytic enzyme on their lactational performance during early lactation period (in terms of milk production, milk composition, feed intake, milking efficiency, body weight change) and the exact time of this response. Cows were randomized into two groups (14 each) with similar parities and were fed a concentrate ration of barley, ground corn, soybean meal, and wheat bran and roughage ration of alfalfa hay. One of the two groups was supplemented with the fibrolytic enzyme immediately after parturition up to 100 post partum. The experiment was of two phases with 50 days each. The enzyme, which has a cellulase/hemicellulase activity (derived from Trichoderma group), was added to the concentrate part of the ration in a dry powder form. Milk production, 3.5% fat corrected milk, energy corrected milk were higher (p<0.05) for cows fed treated diet. At the same time, No differences were observed in percentages of milk components, feed intake, body weight, body weight change, or rectal temperature for the whole experimental period or during any of the two phases. Efficiency of milk production was higher (p<0.05) for treatment group cows than for that of the control ones. However, efficiency was better during the second phase than during the first phase. Feeding enzyme treated diets to dairy cows improved lactational performance during early 100 day of the lactation period. However, the first 50 days of lactation looked to be the critical.

팽화홍삼분말이 벤조피렌을 투여한 마우스의 항산화 효소 활성에 미치는 영향 (Effects of Puffing Red Ginseng Powder on Antioxidant Enzyme Activities in Benzo(α)Pyrene-Treated Mice)

  • 팽화홍삼분말이 B($\alpha$)P의 투여에 의한 간 독성이 유발된 마우스에서의 글루타치온 및 과산화지질 함량, 항산화 효소활성에 미치는 영향을 살펴보았다. B($\alpha$)P 투여에 의해 증가된 혈청 내 ALT와 AST의 활성이 팽화홍삼분말을 투여하면 감소하였다. 간 조직중의 SOD, catalase 그리고 GSH-Px의 활성도 B($\alpha$)P 투여로 유의적으로 증가되었다가, 팽화홍삼분말의 투여로 이들 활성이 유의적으로 감소하였고, 반면, 글루타치온 함량과 GST 활성은 B $\alpha$)P 단독군에서는 감소되었다가 팽화홍삼분말 투여 시 유의적인 증가를 보였다. 그러나 cytochrome P-450 활성과 지질과산화물 함량은 B ($\alpha$)P 투여 시 증가되었다가 팽화홍삼분말의 투여 시 유의적으로 감소되었다. 특히 홍삼분말보다 팽화시킨 홍삼분말에서 더 큰 항산화능을 보였다. 이상의 결과로 팽화홍삼분말은 항산화 효소의 활성에 의한 B($\alpha$)P에 의한 간 손상에 대한 보호효과를 가지는 것으로 사료된다.

감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. 뇌조직의 리포푸신, 아세틸콜린 및 그 관련효소 활성에 미치는 실크 피브로인의 영향 (Effects of Silk Fibroin Powder on Lipofuscin, Acetylcholine and Its Related Enzyme Activities in Brain of SD Rats)

  • This study was designed to investigate the effects of silk fibroin (Mw 500) powder (SFP) on lipofuscin, acetylcholine (ACh) and its related enzyme activities in brain of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (SFP-2.5 and SFp-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. In case of liver membranes, lipofuscin (LF) levels resulted in a considerable decreases (11.5% and 13.8%, respectively) in SFP-2.5 and SFP-5.0 groups compared with control group. But in case of brain as the most sensitive organ, LF levels were remarkably inhibited about 18.3% and 21.7% in SFP-2.5 and SFP-5.0 groups compared with control group. Acetylcholine (ACh) levels were considerable decrease (3.0% and 9.2%, respectively) in brain membranes of SFP-2.5 and SFP-5.0 groups compared with control group. choine acetyltranferase (ChAT) activities as a synthesis enzyme of ACh, and acetylcholinesterase (AChE) activities as a hydrolysis enzyme resulted in a slight increases (2.4% and 3.0%, 4.6% and 6.3%, respectively), but significance difference between ChAT and AChE activities by SFP administration could be not obtained. Monoamine oxidase-B (MAO-B) activities were significantly inhibited (9.5% and 12.6%, respectively) in brain of SEP-2.5 and SFP-5.0 groups compared with control group. These results suggest that inhibiting effects of LF accumulation and MAO-B activity of silk fibroin(SFP) may play a pivotal role in protecting learning memory impairments by attenuating a various age-related changes for improvement of brain function.

효소변형 전분기반 하이드로젤 모델 프리믹스 내 탑재된 커큐민의 소화과정 중 안정성 (Stability of the enzyme-modified starch-based hydrogel model premix with curcumin during in vitro digestion)

  • 본 연구에서는 커큐민 함유 에멀션의 안정성을 위해 필드하이드로젤로 쌀전분(RS), 1시간 효소처리한 전분(1GS), 24시간 효소처리한 전분(24GS), 96시간 효소처리한 전분(96GS)을 사용하였으며 이를 이용한 모델 프리믹스로 커큐민 탑재 필드하이드로젤 분말(FHP)을 제조하여 탑재된 커큐민의 안정성 및 in vitro 소화 후 커큐민 보유율의 변화를 관찰하였다. FHP는 커큐민 함유 에멀션분말(EMP)과 비교하여 재분산시 크리밍 현상없이 분산안전성을 보여주었다. 재분산 하이드로젤은 고온에서 녹으면서 오히려 액적(droplet)의 유착(coalescence)과 응집(flocculation)이 가속화되어 에멀션에 비해 열안정성이 떨어졌으나 UV 안정성은 RS-FHP와 1GS-FHP에서 유의적인 커큐민 보호 효과를 나타냈다. GS-FHP는 in vitro 소화 중 기름방울의 응집 및 유착이 발견되지 않았으며 소화 후 에멀션의 유상에 용해된 커큐민을 성공적으로 보호함을 확인하였다. 따라서, GS-FHP는 분말형태 프리믹스 개발에 새로운 소재로 사용 가능할 것이며 커큐민 외에 다른 다양한 소수성 기능성물질에 대해서도 적용을 확장할 수 있을것으로 생각된다.

뽕잎분말을 첨가한 당면의 생리활성 평가 (A Study on the Physicochemical Activities of Dangmyon (starch vermicelli) Added with Mulberry Leaves Powder)

  • This study was performed to investigate the physicochemical activities such as mineral contents, antioxidantant activities, ${\partial}$-glucosidase activity, and angiotensin I-converting enzyme(ACE) inhibitory effect of Dangmyon (starch vermicelli) prepared with mulberry leaves powder. Mulberry leaves powder was added in ratio (w/w) of 0% (CON), 0.5% (MD1), 1.0% (MD2), 1.5% (MD3), and 2.0% (MD4), and then mineral contents, total polyphenol contents, antioxidant, antidiabetic, and antihypertensive activities of the Dangmyon were measured. The mineral contents were significantly increased by the addition of mulberry leaves powder (p<0.001). The total polyphenol contents and antioxidant activities of Dangmyon were significantly increased by the addition of mulberry leaves powder (p<0.05). In terms of antidiabetic and antihypertensive activities showed MD1~MD4 samples higher contents of ACE and ${\alpha}$-glucosidase compared to control (p<0.05). Our findings suggest that addition of mulberry leaves powder to Dangmyon may improve the effects of anti-oxidation, anti-DM, and anti-hypertension and provide health benefits of consumers.

Development of Chicken Immunoglobulin Y for Rapid Detection of Cronobacter muytjensii in Infant Formula Powder

  • The present study was aimed to produce a chicken polyclonal antibody against Cronobacter muytjensii and to develop an immunoassay for its detection. Purification of anti-C. muytjensii IgY from egg yolk was accomplished using various methods such as water dilution and salt precipitation. As a result, sodium dodecyl sulfate-polyacrylamide gel electrophoresis produced two bands around 30 and 66 kDa, corresponding to a light and a heavy chain, respectively. Indirect competitive enzyme-linked immunosorbent assay (IC-ELISA) was performed to determine the effectiveness of the chicken IgY against C. muytjensii. The optimum conditions for detecting C. muytjensii by indirect ELISA and checkerboard titration of the antigen revealed an optimum average absorbance at the concentration of 18 ${\mu}g/mL$, having ca. $10^8$ coated cells per well. The anti-C. muytjensii IgY antibody had high specificity for C. muytjensii and low cross-reactivity with other tested pathogens. In this assay, no cross-reactivity was observed with the other genera of pathogenic bacteria including Escherichia coli O157:H7, Salmonella Typhimurium, Staphylococcus aureus, Bacillus cereus, Enterobacter aerogenes, Salmonella Enteritidis and Listeria monocytogenes. In addition, detection of C. muytjensii in infant formula powder showed a low matrix effect on the detection curve of IC-ELISA for C. muytjensii, with similar detection limit of $10^5$ CFU/mL as shown in standard curve. These findings demonstrate that the developed method is able to detect C. muytjensii in infant formula powder. Due to the stable antibody supply without sacrificing animals, this IgY can have wide applications for the rapid and accurate detection of C. muytjensii in dairy foods samples.

Rhizopus oryzae에 의한 생전분 분해에 관한 연구 -분리균주에 의한 효소 생산 조건 및 에탄올 발효- (Studies on Digestion of Raw Starch by Rhizopus oryzae - Optimum Condition of Enzyme Production and Ethanol Fermentation -)

  • 토양등으로부터 생전분 분해효소 생산능이 있는 409주의 곰팡이와 62주의 세균 및 2주의 방선균을 분리하고 이들 중에서 7-BU 균주를 선정하여 동정하고 밀기울을 기본배지로 한 효소 생산조건 및 생전분을 이용한 알코올 발효 실험 등을 실시하여 다음과 같은 결과를 얻었다. 1. 생전분 분해효소 생산능이 강하다고 인정된 7-BU 균주는 Rhizopus oryzae로 동정되었다 2. 7-BU 균주의 효소생산 최적온도는 3$0^{\circ}C$이고 pH는 4.0이었으며 배양 72시간 후에 최고의 역가를 보였고 밀기울 배지에 옥수수전분을 0.5% 첨가하였을 때 효소생산이 더욱 촉진되었다. 3. 조효소액에 의한 각종 생전분의 가수분해율은 반응 4일에 쌀전분 89.2%, 옥수수전분 85%, 고구마전분 82%, 감자전분 36%이었고 쌀가루는 86%, 옥수수가루 81%. 고구마가루 79%, 감자가루 30%이었다. 4. 옥수수가루, 쌀가루, 고구마가루와 감자가루를 원료로 하여 조효소액과 효모를 첨가한 후 알코올 발효 실험을 한 결과 담금 4일 후 쌀가루구 9.4%, 옥수수가루구 9.0%, 고구마가루구 8. 1%, 감자가루구 5.4%의 에탄을 생성을 보였다.

Effects of age and diet forms on growth-development patterns, serum metabolism indicators, and parameters of body fat deposition in Cherry Valley ducks

  • Objective: This study was conducted to investigate the effects of age and diet forms on growth-development patterns, serum metabolism indicators, and parameters of body fat deposition in Cherry Valley ducks. Methods: According to the hatching age and initial weight, a total of 150 1-day-old male SM3 Cherry Valley ducks were randomly assigned to two diet forms (pellet vs powder form). Each treatment had with 5 replicates per treatment and 15 meat ducks per replicate. The study lasted 42 d, which was divided into two periods (1 to 21 vs 22 to 42 d). Results: Our results showed that compared with powder group, ducks in pellet group had greater growth performance during different period (p<0.05). The inflection point was 24 d and was not numerically affected by diet forms. Increasing age (42 vs 21 d) significantly increased the weight of body fat and hepatic fat metabolism related enzyme activities in ducks (p<0.05), meanwhile, increasing age (42 vs 21 d) improved serum metabolism indicators and decreased mRNA expression levels of fat metabolism-related genes in liver (p<0.05). Ducks fed different diets (pellet vs powder form) increased growth performance as well as the weight of body fat and improved serum metabolism indicators (p<0.05). In addition, interactions were found between age and diet forms on the levels of serum metabolism indicators in ducks (p<0.05). Conclusion: In conclusion, powder feed reduced growth performance of ducks, and the day of inflection point was 24 days old. Ducks with higher age or fed with pellet diet showed higher fat deposition. The effect of age and feed forms on body fat deposition might result from changes in the contents of serum metabolism indicators, key enzyme activity of lipid production, and hepatic gene expressions.
