An endo-${\beta}$-1,4-glucanase gene, cel5L, was cloned using the shot-gun method from Bacillus sp.. The gene, which contained a predicted signal peptide, encoded a protein of 496 amino acid residues, and the molecular mass of the mature Cel5L was estimated to be 51.8 kDa. Cel5L contained a catalytic domain of glycoside hydrolase (GH) family 5 and a carbohydrate-binding module family 3 (CBM_3). Chromatography using HiTrap Q and CHT-II resulted in the isolation of two truncated forms corresponding to 50 (Cel5L-p50) and 35 kDa (Cel5L-p35, CBM_3-deleted form). Both enzymes were optimally active at pH 4.5 and $55^{\circ}C$, but had different half-lives of 4.0 and 22.8 min, respectively, at $70^{\circ}C$. The relative activities of Cel5L-p50 and Cel5L-p35 for barley ${\beta}$-glucan were 377.0 and 246.7%, respectively, compared to those for carboxymethyl-cellulose. The affinity and hydrolysis rate of pNPC by Cel5L-p35 were 1.7 and 3.3 times higher, respectively, than those by Cel5L-p50. Additions of each to a commercial enzyme set increased saccharification of pretreated rice straw powder by 17.5 and 21.0%, respectively. These results suggest CBM_3 is significantly contributing to thermostability, and to affinity and substrate specificity for small substrates, and that these two enzymes could be used as additives to enhance enzymatic saccharification.
To investigate a novel ${\beta}$-glucosidase from Bifidobacterium breve ATCC 15700 (BbBgl) to produce compound K (CK) via ginsenoside $F_2$ by highly selective and efficient hydrolysis of the C-3 glycoside from ginsenoside Rd, the BbBgl gene was cloned and expressed in E. coli BL21. The recombinant BbBgl was purified by Ni-NTA magnetic beads to obtain an enzyme with specific activity of 37 U/mg protein using pNP-Glc as substrate. The enzyme activity was optimized at pH 5.0, $35^{\circ}C$, 2 or 6 U/ml, and its activity was enhanced by $Mn^{2+}$ significantly. Under the optimal conditions, the half-life of the BbBgl is 180 h, much longer than the characterized ${\beta}$-glycosidases, and the $K_m$ and $V_{max}$ values are 2.7 mM and $39.8{\mu}mol/mg/min$ for ginsenoside Rd. Moreover, the enzyme exhibits strong tolerance against high substrate concentration (up to 40 g/l ginsenoside Rd) with a molar biotransformation rate of 96% within 12 h. The good enzymatic properties and gram-scale conversion capacity of BbBgl provide an attractive method for large-scale production of rare ginsenoside CK using a single enzyme or a combination of enzymes.

Five experimental diets were formulated to evaluate the effects of dietary enzymatically hydrolyzed tuna by-product on growth, non-specific immune responses, and hematology of juvenile rainbow trout (Oncorhynchus mykiss). A basal diet with 50% of fishmeal was used as control (CON) and four other diets replaced 12.5% ($TBB_{12.5}$), 25% ($TBB_{25}$), 37.5% ($TBB_{37.5}$), and 50% ($TBB_{50}$) of fish meal in the CON diet. Juvenile rainbow trout ($4.87{\pm}0.05g$) were randomly distributed into 15 tanks (50 L) and fed 3-4% of wet body weight two times a day. At the end of 7 weeks of feeding trial, weight gain, specific growth rate, feed efficiency, and protein efficiency ratio of fish fed CON diet were significantly higher than those of fish fed $TB_{50}$ diet (P < 0.05). But there were no significant differences among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets (P > 0.05). There were no significant differences in GPT levels among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets. Also, there were no significant differences in lysozyme, superoxide dismutase, glucose, and total protein levels in all experimental diet (P > 0.05). The broken-line analysis indicated that the minimum dietary level of enzymatically hydrolyzed tuna by-product to replace fishmeal could be 29.7% in rainbow trout. These results indicated that the optimum level of dietary enzymatically hydrolyzed tuna by-product could replace greater than 29.7% but less than 37.5% of fishmeal in juvenile rainbow trout diet.

Sweetpotato is rich in starch, which is converted to sugar during storage due to enzymatic hydrolysis. The sugar content of sweetpotato is a component related to taste and storability. In this study, the sugar content (fructose, glucose, maltose, sucrose and total sugar content) of 94 genotypes was evaluated and the GWAS (Genome-Wide Association Study) was conducted to search for candidate genes for sugar content. The fructose and glucose content were 0.2 ~ 8.8 and 0.2 ~ 9.4 g/100g, respectively. The maltose, sucrose and total sugar content were 0.2 ~ 9.1,3.2 - 30.0 and 7.9 ~ 40.2 g/100g, respectively. The fructose and glucose showed a positive correlation (0.98). The 94 genotypes were genotyped with genotyping-by-sequencing (GBS) and aligned against the reference genome sequences of sweetpotato. The GBS libraries from 94 genotypes were sequenced on an Illumina HiSeqXten system, and 1,339,892 SNPs (Single Nucleotide Polymorphism) were generated. Filtering for < 60% missing rate and > 0.05 minor allele frequency resulted in a total of 44,255 SNPs used in GWAS. The GAPIT (Genome Association and Prediction Integrated Tool) was used to conduct based on the mean of sugar content with a Bonferroni-corrected chromosome-wide significance threshold with a -logio(P) of 5.95. The significant SNPs were obtained with fructose (seven), glucose (six), maltose (four) and sucrose (nine). There were several genes related to sugar content around the significant SNPs such as sugar transport protein 8-like, probable galactose-1 -phosphate uridyltransferase-like and beta-amylase. These results will contribute to understanding of sugar content and conversion in sweetpotato.
Citrus 펙틴을 효소로 가수분해시켜 얻은 펙틴 분해물이 35%가 되면서부터 항균력이 서서히 나타나기 시작하여 75% 이상에서는 급격히 증가하였으며 완전분해(100)%)된 펙틴 분해물을 배지에 2.0% 첨가하면 $35^{\circ}C$에서 24시간까지 Escherichia coli ATCC 11229의 생육을 완전히 억제하였다. 펙틴분해물의 항균력은 pH 4.9~5.5 범위에서 가장 좋았으며 균종별로는 젖산균을 제외한 대부분의 세균에 대하여 강한 항균력을 나타내었고 젖산균과 곰팡이류에 대해서는 항균효과가 약하였으며 효모에 대하여는 전혀 영향을 미치지 않았다. 즉, 대부분의 세균은 펙틴 분해물을 2.0~3.0% 첨가하므로써 $35^{\circ}C$에서 48시간까지 생육이 완전히 억제되었으나 Lactococcus Lactis ATCC 19435, Lactobacillus Bulgaricus, Pencillium funiculosum ATCC 11797은 대조구에 비하여 30~40% 만이 억제되었으며 Saccharomyces formosensis와 Saccharomyces cerevisiae는 전혀 영향을 받지 않았다. 한편, glycine, ethanol, sodium ascorbate, sodium chloride 또는 sodium acetate 등의 화학보존료를 병용하면 펙틴 분해물을 단독으로 사용하였을 때 보다 항균력이 상승하였다. 김치에 펙틴 분해물을 1.0% 첨가하여 $4^{\circ}C$에 저장하면 섭취하기에 알맞은 김치를 pH를 유지하는 기간이 대조군에 비하여 적어도 15일 이상 연장되었다. 또한 빵속에 펙틴분해물을 1.5% 첨가하면 $25^{\circ}C$에서 저장 4일째 대조구에 비하여 4 log cycles 그리고 저장 10일까지도 2 log cylces 정도 균의 성장이 억제되었다. 따라서, 펙틴 분해물은 천연식품보존제로 이용가능하며 특히, 젖산균이나 효모에 대해서는 항균작용이 미약하거나 없으므로 젖산균이나 효모를 이용하는 발효식품의 품질보존제로서 아주 유용할 것으로 생각된다. 또한 기존의 화학 보존료와 병용하므로써 화학 보존료의 첨가량을 감소시킬 수 있다는 점에서도 유용하다고 생각된다.
전통적인 발효와 유사한 가수분해 형태를 가지는 단백질 분해효소에 의한 최적 발효 조건을 검토하기 위하여 trypsin, chymotrypsin, Alcalase, Neutrase, Protamex와, 오징어 내장 및 간에서 추출한 조효소를 첨가하여 제조한 멸치 액젓 및 actomyosin의 가수분해 특성을 조사하였다. 멸치 젓갈의 휘발성 염기질소 값은 오징어 내장을 첨가한 젓갈이 저장 70일째 173.6mg/100g으로 가장 높았고, 저장 기간에 따른 과산화물가의 증가는 시료간에 큰 차이를 보이지 않았다. Alcalase와 Protamex를 첨가한 젓갈의 비단백태 질소 함량과 총 유리아미노산 증가가 가장 높게 나타났다. 숙성 60일째에 Alcalase와 Protamex는 육 단백질에 대해 약 $57\%$의 가수분해율을 보인 반면, 대조구는 약 $43\%$이었다. pH 5.0$\~$7.5 범위에서 멸치 actomyosin에 대하여 가장 높은 활성을 보이는 pH는 trypsin, chymotryosin, Alcalase, Neutrase 및 Protamex가 각각 7.5, 6.5, 6.5, 7.0 및 5.0이었고, 최적 활성을 보이는 온도는 trypsin, chymotryosin, Alcalase, Neutrase에서 각각 55, 45, 60 및 $55^{\circ}C$였으며, Protamex는 온도의 상승 ($20\~70^{\circ}C$)과 더불어 활성도 상승하는 것으로 나타났다. 아울러 Protamex는 사용한 효소 중 가장 높은 $K_m$(3.545)과 $V_{max}$값(2.688)을 나타내었고, NaCl에 대한 영향을 적게 받아 일반적인 젓갈 숙성 조건인 $20^{\circ}C$, $25\%$ NaCl에서도 $52.5\%$의 높은 잔여 활성을 나타내었다. 따라서 Protamex가 멸치 actomyosin의 가수분해에 효과적이라 판단된다.
본 연구에서는 미더덕 껍질(SCT)의 전처리과정에 사용되는 수산화나트륨을 농도별로 처리하여 알칼리처리에 따른 SCT 셀룰로오스의 물리화학적 특성 및 효소학적 특성에 대한 연구를 수행하였다. 알칼리처리 한 SCT를 주사전자현미경으로 분석한 결과 알칼리처리에 의해 표면에 농도 의존적으로 상당히 많은 손상을 초래하는 것을 확인할 수 있었으며, X-선 회절 분석을 통한 결정화도의 분석으로 알칼리처리에 따라서 SCT 셀룰로오스의 결정화도가 증가함을 알 수 있었다. Celluclast를 이용한 효소의 초기반응에서는 알칼리처리에 따른 SCT 표면 손상과 효소반응이 밀접한 관계가 있었으나 장시간 동안 완전분해를 하는 경우에는 알칼리처리로 인한 결정화도의 증가로 인해 분해되지 않고 남는 SCT가 많음을 확인할 수 있었다. 따라서 SCT의 전처리 시에 알칼리 사용을 최대한 적게 사용하거나 결정화도를 감소시킬 수 있는 대체 기술에 대한 연구가 필요할 것으로 판단된다.
펩타이드는 Asp, Glu, Leu을 주요 구성 아미노산으로 하는 한우 혈청을 단백질 분해효소로 분해 후 한외여과를 거쳐 제조하였다. 단백질 분해효소는 alcalase, esperase, flavourzyme, protamex를 이용하였고 alcalase가 가장 좋은 분해력을 나타내어 폡타이드 생산을 위한 혈청 분해에 활용하였다. 한우 혈청으로부터 분리한 펩타이드와 킬레이트 결합한 칼슘 (chelate-Ca)과 게르마늄 (chelate-Ge)을 방울토마토에 적용하여 흡수 변화를 확인하고자 시험을 수행하였다. Chelate Ca을 500, 1,000, 2,000배로 희석하여 방울토마토 재배에 적용한 결과, 처리 농도 및 횟수 증가에 따라 엽과 과실의 칼슘함량이 다소 증가하여 대조처리 Calciolid Ca-300 (3409.1, $67.5mg\;kg^{-1}$)이 Chelate Ca-200 (3781.1, $78.0mg\;kg^{-1}$) 보다 처리농도가 높음에도 불구하고 그 함량이 낮아 킬레이트 칼슘의 흡수율이 더 많았다. Chelate Ge을 50, 100, 200배로 희석하여 적용한 결과, 방울토마토 엽의 게르마늄 함량이 처리 농도가 높을수록 많아져 대조구와 Ge $132^{(R)}$-20 보다 모든 처리구에서 2배 이상의 높은 함량을 나타내었다. 본 연구를 통해 펩타이드와 킬레이트 결합한 칼슘과 게르마늄을 방울토마토에 시용할 경우 그 흡수율이 증가되는 것을 확인하였다.

Renewable energy resources and technologies have the potential to provide long-lasting solutions of the global energy-requirements faced by the economic and environmental sectors of a nation. Therefore, waste money bills were used as renewable energy source for the production of bio-ethanol. In this study, different concentrated NaOH 0.5%. 1.0%, 2.0%, 3.0% and 0.0% (as a control) were used for 10, 20 and 30 mins at $121^{\circ}C$/15 psi in an autoclave. Saccharification and fermentation (aerobic and anaerobic) were carried out through commercial enzyme Celluclast 1.5 L, Novozymes 188 and Saccharomyces cerevisiae KCCM 11304 respectively. The results of pretreatment showed that the NaOH pre-treated substrate enhanced enzyme action and released more amount of glucose. The amount of glucose was found with the increasing concentration of NaOH and time $44996.95{\pm}6.30$, $46763.10{\pm}3.56$, $53421.32{\pm}4.72$, $63431.25{\pm}6.95$ and $56850.98{\pm}6.75\;ng/{\mu}l$ for 30 min respectively. As for bioethanol, the conversion rate of NaOH resulted $1010.08{\pm}4.71$, $1050.25{\pm}4.37$, $1109.49{\pm}4.39$, $1139.25{\pm}3.26$ and $1020.77{\pm}3.89$ ppm for aerobic; $16730.54{\pm}6.67$, $17076.45{\pm}6.25$, $17516.17{\pm}4.49$, $19782.68{\pm}6.19$ and $17973.39{\pm}7.50$ ppm for anaerobic and $18935.02{\pm}4.59$, $19895.45{\pm}5.39$, $21912.95{\pm}4.83$, $24895.21{\pm}6.72$ and $18961.21{\pm}4.90$ ppm for anaerobic condition with benzoic acid for respective condition. Thus, the results of the present work clearly revealed that with the increasing of alkali concentration might be more effective for bio-ethanol production from waste money bill, which is economic and environmental friendly.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
