제주 용암해수를 용매로 한 제주 작물 추출물의 항산화 효능 (Antioxidant efficacy of Jeju crop extracts using Jeju lava seawater as a solvent)

  • 제주 용암해수에 함유된 풍부한 미네랄은 노화를 유발하는 활성산소를 제거하는 항산화 효과가 있는 것으로 알려져 있다. 이에 용암해수는 기능성 식품소재로서 상업적 가치가 높은 것으로 알려져 있다. 본 연구에서는 제주산 작물(당근, 블루베리, 감귤)추출물의 항산화 활성을 증류수 추출물과 용암해수 추출물을 이용하여 비교 분석하였다. 블루베리와 감귤의 총폴리페놀 및 플라보노이드 함량은 증류수 추출물보다 용암해수 추출물에서 더 높았다. 또한, 이들 작물의 항산화 효소 활성과 DPPH 라디칼 소거 활성은 증류수 추출물보다 용암해수 추출물에서 더 높았다. 특히, Vero 세포 이용한 블루베리와 감귤의 ROS 소거 효능은 용암해수 추출물에서 더 높게 나타났다. 한편, 당근의 항산화 활성은 총 폴리페놀과 플라보노이드 함량에 차이가 없었음에도 불구하고 용암해수 추출물에서 더 높았다. 이러한 결과는 용암해수가 기능성 식품 산업에서 용매로서의 가능성이 있음을 시사하였으며, 용암해수를 이용한 제주산 작물 추출물은 기능성 식품의 원료로 사용될 수 있는 것으로 판단된다.

열수 및 효소적 가수분해로 제조된 틸라피아 비늘 젤라틴 가수분해물의 ACE 저해 활성 (Angiotensin I Converting Enzyme Inhibitory Effects of Gelatin Hydrolysates Prepared from Tilapia mossambica Scales by Hot Water and Enzymatic Extraction)

  • Fish scales have potential in functional food preparation due to their antioxidant and antihypertensive properties. We investigated the angiotensin I converting enzyme (ACE) inhibitory activity of Tilapia mossambica scale extracts. Hydrolysates of tilapia scales were prepared by enzymatic extraction using five proteases (${\alpha}$-chymotrypsin, Alcalase, Kojizyme, Protamex and trypsin) after scales were treated with hot water for 3 hr. Scale enzymatic hydrolysates prepared using both hot water and enzyme treatments exhibited elevated hydrolysis (about 25%-55%) compared to only enzyme treatment (about 15%-45%). Enzymatic hydrolysates (1 mg/mL) prepared by both hot water and enzyme treatments also showed significantly increased ACE inhibitory activities from about 20%-75%. The pattern of ACE inhibitory activities was similar to the degree of hydrolysis. Alcalase and ${\alpha}$-chymotrypsin hydrolysates displayed the highest ACE inhibitory activities ($IC_{50}$ = 0.83 mg/mL and 0.68 mg/mL, respectively). In addition, the ACE inhibitory effects of $IC_{50}$-chymotrypsin hydrolysates increased with decreasing molecular weight (5 kDa>, 10 kDa> and 30 kDa>), with the 5 kDa> fraction displaying the highest ACE inhibitory activity (about 89.9% and $IC_{50}$ = 0.1 mg/mL). We suggest that the peptide compounds of enzymatic hydrolysates prepared from tilapia scale enhances ACE inhibitory activity and might be useful as an antihypertensive material.

수삼추출물 및 Glucose 또는 Arginine첨가 추출물의 특성과 항산화작용에 대하여 (The Characteristics and Antioxidant Activity of Non-enzymatic Browsing Products from Fresh Ginseng Bxtracts and Those with Arginine or Glucose)

  • Color is one of the most important quality factors of red ginseng (Hong-sam) which is processed from fresh ginseng (Panax ginseng C. A. Meyer). Therefore, a study of characteristics of browning mixtures of aqueous fresh ginseng extracts, factors which accelerate the browning of the aqueous extracts, and the antioxidant activity of the browning mixtures may contribute to the improvement of the color and other quality of red ginseng and other ginseng products such as ginseng extracts. In the present study, factors which affect the Maillard-type browning reaction of aqueous extracts of fresh ginseng roots were investigated firstly by adding various concentrations (0.001-0.5M) of arginine or glucose solutions, by varying the browning reaction temperatures and durations. Secondly, some characteristics such as brown color intensity, amounts of water-soluble and ether-soluble extracts, amounts of non-dialyzable materials, pH, viscosity, and reactivity with 2,2'- diphenyl -1 - picrylhydrazyl and antioxidant activity of the browning mixtures of the aqueous fresh ginseng extracts with small amounts of 0.1 M arginine, 0.1 M glucose, and distilled water at various browning temperatures and reaction time were studied. The results of the present study are as follows. 1. Color intensity (absorbance at 470 nm) of the browning mixtures was increased by adding various concentrations of arginine solution to the fresh ginseng extract, but the addition of the same amount of glucose solution did not increase the color intensity. 2 The amounts of water- or ether-soluble extracts, amounts of non-dialyzable materials were slightly greater in case of the browning mixtures of the fresh ginseng extract with 0.1M arginine solution than in case of the browning mixtures of the fresh ginseng extract with the same amount of 0.1 M glucose solution. In the process of the browning reaction, the pH of the browning mixtures of the fresh ginseng extract with 0.1 M arginine solution decreased slightly, while that of the browning mixtures with 0. 1 M glucose solution was almost constant. 3. The color intensity (absorbance at 470 nm) of the browning mixtures of the fresh ginseng extract with 0.1 M arginine or 0.1 M glucose solutions did not correlate well with the reducing power or the antioxidant power of the browning mixtures. The antioxidant activity of 90% ethanol extracts from the earlier stages of the browning mixtures of the fresh ginseng extract with the arginine solution was almost comparable to that of the 90% ethanol extracts from the later stages of the corresponding browning mixtures. The browning mixtures of only the fresh ginseng extract or of the fresh ginseng extract with the glucose solution showed considerable antioxidant activity, although both showed less brown color intensity than the fresh ginseng extract with he arginine solution.

Efficient Recovery of Lignocellulolytic Enzymes of Spent Mushroom Compost from Oyster Mushrooms, Pleurotus spp., and Potential Use in Dye Decolorization

  • This study was conducted in order to perform efficient extraction of lignocellulolytic enzymes amylase (EC 3.2.1.1), cellulase (EC 3.2.1.4), laccase (EC 1.10.3.2), and xylanase (EC 3.2.1.8) from spent mushroom compost (SMC) of Pleurotus ostreatus, P. eryngii, and P. cornucopiae. Optimal enzyme recovery was achieved when SMCs were extracted with 50 mM sodium citrate (pH 4.5) buffer at $4^{\circ}C$ for 2 hr. Amylase, cellulase, and xylanase activities showed high values in extracts from P. ostreatus SMC, with 2.97 U/g, 1.67 U/g, and 91.56 U/g, respectively, whereas laccase activity and filter paper degradation ability were highest in extracts from P. eryngii SMC, with values of 9.01 U/g and 0.21 U/g, respectively. Enzymatic activities varied according to the SMCs released from different mushroom farms. The synthetic dyes remazol brilliant blue R and Congo red were decolorized completely by the SMC extract of P. eryngii within 120 min, and the decolorization ability of the extract was comparable to that of 0.3 U of commercial laccase. In addition, laccase activity of the SMC extract from P. eryngii was compared to that of commercial enzymes or its industrial application in decolorization.

Comparison of Superoxide Dismutase and Peroxidase Activities in Rice Varieties

  • Fifty-four Korean native and 28 foreign varieties harvested in 1998 and 1999 were examined for superoxide dismutase (SOD) and peroxidase (POD) activities. The SOD and POD activities of leaves extracts in Korean native and foreign rice varieties showed variation at the heading stage. The activities of SOD and POD changed with growth stage. In comparison of storage duration, the SOD and POD activities of the extract from three months stored seeds in Korean native (CV=53.3%) and foreign rice (CV=57.9%) varieties were higher than that of stored rices for a year in seed extracts although the activities among varieties did not show significant variation. Also, the averaged activity of foreign rice varieties (SOD=12.9%) was relatively higher than that of korean native rice varieties (SOD=10.7%). The test of activity at the enzymatic level related to antioxidative activity suggests that the rice varieties with higher antioxidative potentials can be developed and also may provide information with rice breeder to breed rice variety with a high antioxidative activity.

Antioxidant and anti-inflammatory functionality of ten Sri Lankan seaweed extracts obtained by carbohydrase assisted extraction

  • Enzyme-assisted extraction is a cost-effective, safe, and efficient method to obtain bioactives from plant materials. During this study, 10 different marine algae from Sri Lanka were individually extracted by using five commercial food-grade carbohydrases. The enzymatic and water extracts of the seaweeds were analyzed for their antioxidant and anti-inflammatory activities. The highest DPPH, hydrogen peroxide ($H_2O_2$) and intracellular $H_2O_2$ scavenging abilities were observed from the Celluclast extract of Sargassum polycystum (CSp). CSp exerted protective effects against oxidative stress-induced cell death in hydrogen peroxide-induced Chang cells and in model zebrafish. The Celluclast extract of Chnoospora minima (CCm) showed the strongest anti-inflammatory activity against lipopolysaccharide (LPS)-induced NO production in RAW 264.7 macrophages ($IC_{50}=44.47{\mu}g/mL$) and in model zebrafish. CCm inhibited the levels of iNOS, COX-2, $PGE_2$, and TNF-${\alpha}$ in LPS stimulated RAW 264.7 macrophages. Hence, CSp and CCm could be utilized in developing functional ingredients for foods, and cosmeceuticals.

식물유래 물질이 뽕나무버섯(Armillaria mellea) 균사체 생장 및 혈전분해 활성에 미치는 영향 (Effect of Various Plant Extracts on the Mycelial Growth and Fibrinolytic Activity of Armillaria mellea)

  • 뽕나무버섯 균사체의 생산 및 혈전분해활성을 증대시키고자 배양기질에 7종의 식물체추출물을 첨가하여 증식시킨 결과 가시오가피 뿌리, 유근피, 홍삼박추출물의 첨가가 균사체 중식에 양호하였으나 인삼추출물 첨가에 의해서 유의적으로 증식이 억제되었다. 또한 가시오가피 가지, 유근피, 홍삼박추출물 첨가에 의하여 단백질분해 활성이 무첨가구보다 $36.8{\sim}46.1%$ 정도 증가되어졌으며 옻피, 가시오가피 가지, 홍삼박 첨가구의 혈전분해 활성은 각각 0.70, 0.71, 0 73 plasmin $unit/200\;{\mu}g$ protein으로 무첨가구의 0.44 plasmin $unit/200\;{\mu}g$ protein에 비하여 $59{\sim}65%$ 증가시킬 수 있었다. 뽕나무버섯 균사체의 조효소는 fibrinogen의 $A{\alpha}-chain$에 대해 높은 분해특성을 가지고 있었을 뿐만 아니라 무첨가구가 fibrinogen의 모든 단위체를 완전 분해시 까지 3시간이 소요되었던 반면 홍삼박 첨가구는 2시간 후 모든 단위체가 분해되어 대조구보다 높은 활성을 나타내었다. 현미경을 통한 관찰에서 역시 홍삼박 추출물첨가구가 무첨가구보다 높은 분해 활성을 보였다.

보리의 효소처리가 보리차의 품질 및 수율에 미치는 영향 (Effects of Enzyme Treatments on Quality and Yields of Barley Tea)

  • 보리차의 수율을 높이고 보리를 효율적으로 이용하며 품질을 향상시켜 한국인의 기호에 맞는 보리차를 개발하기 위하여 나맥인 영산보리를 조쇄하고 20분 침지하여 $250^{\circ}C$에서 6분 볶아 세 가지의 복합효소(1,000, 2,000 및 5,000)을 일정한 시간 반응시켜 물리화학적 성질 및 관능경사를 실시하였다. 고형물의 수율은 ${\alpha}-amylase$의 활성이 높은 복합효소 5,000이 가장 좋았고 복합효소 1,000, 2,000의 순이었다. 수율이 높은 효소일수록 반응시간이 짧아도 대조구에 비하여 수율이 높았으며 수율이 낮은 효소는 반응시간을 10분에서 40분까지 계속 증가시킬 때 수율은 직선적으로 증가하였다. 점도는 복합호소 5,000<1,000<2,000 순이었으며 D.E.는 이와 반대이었고 탁도는 점도와 같은 순이었으나 점도, 탁도 및 D.E.는 호소반응시간에 따라, 일정한 경향은 있었으나 큰 차이를 고이지는 않았다. 24시간 $4^{\circ}C$ 저장시 복합효소 5,000은 거의 침전물이 생성되지 않았다. 보리차의 색은 수율이 높을수록 L값은 낮고 a, b값은 높아져 전체적으로 색이 엷어졌다. 관능검사 결과 효소반응시간이 $30{\sim}40$분 일 때 전반적인 관능적 성질이 향상되었고 복합효소 5,000이 가장 효과적이었다.

담쟁이덩굴의 항산화 방어계의 탐색 (Screening of the Antioxidant Defense Systems from Parthenocisuss tricuspidata PLANCH)

  • 담쟁이 덩굴 추출물로부터 항암예방제로서의 물질을 탐색코자 항산화 비효소활성 및 효소활성을 조사해 본 결과, 부위별 항산화력 및 효소활성들은 차이를 나타내었다. 부위별 에틸아세테이트 분획물은 잎에서 DPPH 소거활성이 가장 높았으며, 실리카겔 컬럼크로마토그래피에서 잎, 줄기, 뿔리에서 각각 7.06, 6.99, 12.39($\mu\textrm{g}/m\ell$)이었다. 에틸아세테이트 분획에서 활성이 가장 높은 분획을 Sephadex LH-20 column chromatography로 분리 정제한 결과 잎, 줄기 그리고 뿔리에서 각각 5.5, 6.11, 5.29 ($\mu\textrm{g}/m\ell$)의 DPPH 활성이 높은 분획을 얻었다. Sephadex LH-20에 의해 활성이 가장 높은 분획을 GC/MS로 분석한 결과, Benzoic acid(Gallic acid), 1,2,3-Benzenetriol, 4-amino-Benzenemethanol과 같은 페놀화합물의 항산화물질들이 동정되었다. 줄기와 뿌리에서 POD활서은 잎에서 보다 활성이 높았고, SOD 활성은 잎, 줄기 그리고 뿌리 순으로 높은 활성을 나타내었으며 특히, 잎은 뿌리보다 2배 이상의 높은 활성을 보였다.

진주담치(Mytilus edulis) 추출물의 제조 및 품질특성 (Evaluating Cultured Sea Mussels Mytilus edulis Extractions Methods and Extract Quality Characteristics)

  • Extraction methods for cultured sea mussels Mytilus edulis and the quality characteristics of resulting extracts were investigated. The crude protein, carbohydrate and volatile basic nitrogen content of raw sea mussels was 15.2%, 1.9%, and 11.2 mg/100 g, respectively. Extracts were prepared using three different methods: hot-water extract (WE), scrap enzymatic hydrolysate extraction (SE), and complex extraction (CE). The respective extracts contained 5.5%, 8.6%, and 6.6% crude protein; 281.7, 366.0, and 343.0 mg/100 g amino nitrogen,: and 2.0%, 1.1% and 1.8% salinity. Their extraction yields were 689, 323, and 1,012 mL/kg. The CE method was superior to the traditional WE method in terms of extraction yield, amino-nitrogen content, and organoleptic qualities, but not odor. Active taste components were evaluated and the total free amino acid content of the WE and CE methods was 5,667.0 and 7,006.3 mg/100 g, respectively. The concentrations of major components (for WE and CE methods, respectively) were as follows: glutamic acid (1,244.0 and 955.4 mg/100 g), taurine (987.9 and 746.8 mg/100 g), glycine (721.2 and 847.0 mg/100 g), alanine (341.9 and 423.8 mg/100 g), arginine (265.5 and 376.5 mg/100 g), lysine (199.8 and 270.4 mg/100 g), and proline (253.9 and 220.3 mg/100 g). In conclusion, these results demonstrate that there is potential for using the CE method to expand the commercial utilization of sea mussels as a flavoring substance resource.
