온주밀감 및 당유자 진피 효소 추출물의 항산화 활성 (Antioxidative Activities of Enzymatic Digests from Dried Citrus unshiu and Citrus grandis Peels)

  • 온주밀감 및 당유자 진피 추출물의 추출수율은 탄수화물분해효소인 viscozyme 추출물과 단백질분해효소인 kojizyme 추출물에서 높았으며, 당유자의 효소 추출물들이 온주밀감에 비하여 높게 나타내었다. 총 폴리페놀 함량은 탄수화물분해효소인 ultaflo와 단백질분해효소인 alcalase 및 flavourzyme에서 높았으며, 온주밀감 추출물들이 당 유자에 비하여 높게 나타내었다. 총 플라보노이드 함량은 탄수화물분해효소인 ultaflo와 단백질분해효소인 alcalase 및 물 추출물에서 높았다. DPPH radical 소거활성은 온주밀감에 비하여 당유자의 효소 추출물에서 매우 높게 나타났으며, 탄수화물분해효소인 viscozyme과 단백질분해효소인 kojizyme에서 가장 높았다. 특히 viscozyme 추출물은 86% 이상의 매우 높은 활성을 나타내었다. Hydrogen peroxide 소거활성은 추출물의 농도가 증가됨에 따라 약간씩 증가하였으나, 2.0 mg/mL에서는 온주밀감 진피의 kojizyme 추출물을 제외하고는 91% 이상의 매우 높은 활성을 나타내었다. Alkyl radical 소거활성은 추출물의 농도가 증가됨에 따라 급격히 증가하였으며, 모든 효소 추출물들은 0.5 mg/mL 이상에서 매우 높은 활성을 나타내었다. Hydroxyl radical 소거활성은 추출물의 농도가 증가됨에 따라 급격히 증가하였으며, 모든 효소 추출물들은 다른 활성산소종들의 활성에 비하여 상대적으로 낮은 활성을 나타내었다.

Enzymatic Activities of Allergen Extracts from Three Species of Dust Mites and Cockroaches Commonly Found in Korean Home

  • Allergen extracts from dust mites and cockroaches commonly found in Korean homes were used to evaluate their enzymatic activity as they are believed to influence allergenicity. Allergen extracts were prepared from 3 dust mite species (Dermatophagoides farinae, D. pteronyssinus, and Tyrophagus putrescentiae) and 3 cockroach species (Blattella germanica, Periplaneta americana, and P. fuliginosa) maintained in the Korea National Arthropods of Medical Importance Resource Bank. Proteins were extracted in PBS after homogenization using liquid nitrogen. The activities of various enzymes were investigated using the API Zym system. No significant difference in phosphatase, lipase, or glycosidase activity was observed among the 6 allergen extracts, but much difference was observed in protease activity. Protease activity was assessed in more detail by gelatin zymography and the EnzChek assay. Extract from T. putrescentiae showed the highest protease activity, followed by those of the cockroach extracts. Extracts from D. farinae and D. pteronyssinus showed only weak protease activity. Gelatinolytic activity was detected mainly in a 30-kDa protein in D. farinae, a 28-kDa protein in D. pteronyssinus, a > 26-kDa protein in T. putrescentiae, a > 20-kDa protein in B. germanica, and a > 23-kDa protein in P. americana and P. fuliginosa. The information on various enzymatic activities obtained in this study may be useful for future studies. In particular, the strong protease activity found in cockroach extracts could contribute to sensitization to cockroach allergens, which is known to be associated with the development of asthma.

Polysaccharide Degrading Enzyme을 이용한 참모자반 효소분해 추출물의 생리활성 연구 (Biological Analysis of Enzymatic Extracts from Sargassum fulvellum Using Polysaccharide Degrading Enzyme)

  • SC092 strain, producing a polysaccharide degrading enzyme, was isolated from the seawater. This strain was identified as Microbulbifer sp. using the comparative sequence analysis against known 16S rRNA sequence. A polysaccharide degrading enzyme from this strain was used to acquire the enzymatic extracts of Sargassum fulvellum. DPPH radical scavenging and SOD activity of the enzyme extracts of S. fulvellum were about 61.9% and 82.9% at 2 mg/mL, respectively. Nitrite scavenging activities was 52.5% at 2 mg/mL on pH 1.2. In addition, ${\alpha}$-glucosidase inhibitory activity was also increased in a dose-dependent manner and was about 52.7% at 2 mg/mL. To determine the influence of enzyme extracts of S. fulvellum on alcohol metabolism, the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. ADH and ALDH activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}$-glucosidase inhibitory activity of enzyme extracts of S. fulvellum was remarkably increased in a dose-dependent manner and was about 52.7% at 2 mg/mL. These results indicate alcoholizing and ${\alpha}$-glucosidase inhibitory activities can be enhanced by the enzymatic extracts of S. fulvellum.

  • Tyrosinase는 식물의 갈변현상에 관계가 있으며, 이의 방지를 위한 방법의 개발은 과학적인 관심사가 되고 있다. 솔잎으로부터 3개의 tyrosinase isozyme을 정제하여 그들의 활성에 영향을 주는 인자들을 분석하였다. L-ascorbic acid와 ${\beta}$-mercaptoethanol은 효소의 활성을 크게 저해하였다. 또한 sodium diethyldithio-carbamate도 효소활성에 대한 경쟁적 저해제였으며, $K_i$ 값은 $P_1$(0.030 mM), $P_2$(0.015 mM), $P_3$(0.019 mM)이였다. 반면 대부분의 금속이온들은 효소활성을 증가시켰다. L-DOPA를 기질로 사용했을 때 효소의 최적 pH는 9.0~9.5이며, 최적 온도는 $55{\sim}60^{\circ}C$를 나타내었다.

Anti-oxidative Activities of 3'-Hydroxygenkwanin from the Flower Buds of Daphne genkwa in Caenorhabditis elegans

  • In the course of screening for antioxidant compounds from natural plants in Korea by measuring the radical scavenging effect, a methanol extract of the flower buds of Daphne genkwa S. et Z. (Thymelaeaceae) was found to show a potent antioxidant activity. Subsequent activity-guided fractionation of methanol extract of D. genkwa led to the isolation of four compounds from the ethyl acetate soluble fraction. The chemical structures were elucidated as genkwanin (1), 3'-hydroxygenkwanin (2), apigenin (3), and tiliroside (4) by spectroscopic techniques. Among them, compound 2 showed the significant anti-oxidative effect on DPPH. And compound 2 showed the significant riboflavin-and xanthine-originated superoxide quenching activities. To verify the antioxidant enzymatic activities of compound 2, the SOD enzymatic activity was measured spectrophtometrically using prepared Caenorhabditis elegans homogenates. The results showed that compound 2 was able to elevate SOD activity of C. elegans in a dose dependent manner. Moreover, compound 2 decreased the intracellular ROS accumulation of worms.

Effects of Chitosan and Lactic Acid on Enzymatic Activities and Bioactive Compounds during Germination of Black Rice

  • The effect of chitosan on enzymatic activities and on bioactive compounds was characterized during germination at $25^{\circ}C$ for 7 days to search for a method to produce a germinated black rice. The germination rate was reduced by the addition of lactate and chitosan. The rotting rate was greatly decreased by chitosan, suggesting that the addition of chitosan into a germination solution might be an effective method for controlling fungal contamination during the germination of cereals. The addition of 100 and 200 ppm chitosan increased $\alpha$-amylase activity after 7 days by up to 152 % and 197 %, respectively. The activities of $\beta$-amylase and $\beta$-glucosidase were lower with 200 ppm chitosan than in distilled water and 100 ppm lactate. The amount of total soluble phenolics and total flavonoids decreased rapidly for four days and thereafter remained constant until the seventh day. The antioxidant activity of germinated black rice, in terms of hydrogen-donating activity, increased slowly and did not correspond to the changes of total soluble phenolics and total flavonoids. The amount of phytic acid was reduced by the addition of 200 ppm chitosan compared to distilled water, indicating that chitosan could be used as an elicitor for the increase of phytase activity during the germination of black rice.

Localization of Germin Genes and Their Products in Developing Wheat Coleoptiles

  • Germination is a process which characterized with nescient synthesis of genes. Among the genes synthesized during the germination of wheat embryos, germin genes, proteins and their enzymatic activity were defined. Germin is a water soluble homopentameric glycoprotein which is remarkable resistant to degradation by a broad range of proteases including pepsin. Germin proteins found to have strong oxalate oxidase activity which produces hydrogen peroxide by degrading oxalic acid. The current study, aimed to localize the germin genes, proteins and enzymatic activities in developing coleoptiles which is a rapidly growing protective tissue of leaf primordium and shoot apex. Non-radioactively abeled germin riboprobes were employed to localize germin mRNAs in situ. FITC (Fluorescein isothiocyanate) and alkaline phosphatase linked anti-germin antibodies were used to localize germin proteins under the fluorescence and light microscopy and finally germin enzymatic activity was localized by using appropriate enzyme assay. The results revealed that in coleoptiles germin genes, proteins and their enzymatic activity were predominantly associated with the cells of epidermis and vascular bundle sheath cells.

Enzymatic Hydrolysis for Effective Extraction of Antioxidative Compounds from Hizikia fusiformis

  • Hizikia fusiformis hydroysates by five carbohydrases (Viscozyme, Celluclast, Termamyl and Ultraflo) and five proteases (Protamex, Kojizyme, Neutrase, Flavourzyme and Alcalase) were investigated for their extraction efficacy (yield and total total polyphenolic content) and antioxidative activity (DPPH radical and hydrogen peroxide scavenging activity). Termamyl and Ultraflo of the carbohydrases and Flavourzyme and Alcalase of proteases were selected by their high eficacy of extraction and antioxidative activity. Selected enzymes were used to investigate the optimum enzymatic reaction time and dosage (enzyme/substrate ratio) suitable for hydorolysis. Optimum reaction time for the enzymatic hydrolysis was 3 days and optimum dosage of hydrolysis was observed as 5%. Simultaneously, Ultraflo of the two carbohydrases and Alcalse of the two proteases were selected as the most effective enzymes. Combination of Ultraflo and Alcalase under optimum hydrolysis conditions could intensify the extraction efficacy of antioxidative materials form H. fusiformis. The hydrolysate obtained by combining the enzymes was separated into four different molecular weight fractions (<1kD, 1-10 kD, 10-30 kD and >30 kD) and recorded the polyphenolic content distribution and respective antioxidative ability. The fraction <1kD was identified as less effective and those fractions > 1kD indicated comparatively higher antioxidative activities related to their polyphenolic content.

  • This study was carried out to investigate the characteristics of extracellular cellulase and xylanase from 4 selected different species, such as enzyme activity and stability by pH, temperature and metal ions, for application into enzymatic deinking system. The optimal temperature and pH for enzyme activity of Bacillus pumilus I, B. subtilis I, B. pumilus II and B. subtilis II were mainly $40{\sim}60^{\circ}C$ and pH $6.0{\sim}7.0$, respectively. Certain metal ions, calcium and cobalt, elevated enzyme activity, even though there were different results of enzyme activities based on various metal ions in 4 different species. With these results we suggest that enzymatic deinking system should be proceed at $50^{\circ}C$ with neutral pH condition.

블루베리 (Vaccinium corymbosum L.) 유래 효소 추출물의 항산화성 (Antioxidant Potential of Enzymatic Extracts from Blueberry (Vaccinium corymbosum L.))

  • 제주지역에 서식하고 있는 블루베리 (Vaccinium corymbosum L.)로부터 수용성 추출물을 효과적으로 제조하기 위하여 5가지 종류의 탄수화물 분해효소 (AMG, Celluclast, Termamyl, Ultraflo, Viscozyme)와 또 다른 5종의 단백질 분해효소 (Alcalase, Flavourzyme, Kojizyme, Neutrase, Protamex)를 이용하여 효소적 추출을 시도하였다. 이들 효소를 이용하여 제조된 여러 가지 블루베리 효소 추출물에 대하여 1,1-diphenyl 2-picrylhydrazyl (DPPH), 활성산소종 (ROS), 일산화질소 $(NO{\cdot})$ 등의 소거 활성, 금속 킬레이트결합능, 그리고 지질 과산화 저해능과 같은 항산화 효과를 검토하였다. 모든 효소 추출물의 페놀함량은 $517.85\~597.96\;mg/100 g$ 건조시료이었으며, 항산화 효과에 대한 평가에서 특히 DPPH 및 $NO{\cdot}$ 소거활성, 그리고 금속 킬레이트 결합능 등이 우수한 것으로 나타났다. Viscozyme 추출물은 DPPH 소거활성 $(0.046{\pm}0.002\;mg/mL)$이, 그리고 AMG 추출물은 $NO{\cdot}$ 소거활성$(0.339{\pm}0.011\;mg/mL)$ 및 지질 과산화 억제활성 $(0.28{\pm}0.01\;mg/mL)$이 각각 우수하였다. 과산화 수소 소거활성에 있어서는 블루베리 효소 추출물들이 천연 항산화제인 a-토코페롤보다 상대적으로 높았다. 이러한 결과로 볼 때 블루베리는 항산화 효과가 우수한 화합물들을 함유하고 있을 것으로 판단되며, 향후 천연 항산화 자원으로서 이용이 가능할 것으로 사료된다.
